Objective To investigate of the expression of vascular endothelial growth factor receptor 3 (VEGFR-3) and its relationship with the prognosis of esophageal carcinoma. Methods VEGFR-3 in 156 cases of esophageal carcinoma tissues was detected by immunohistochemistry. The follow-up data for 3 years in 47 cases were analyzed. Results The VEGFR-3 expression in esophageal carcinoma tissues with lymph node metastasis was higher than that in those without lymph node metastasis. VEGFR-3 expression was associated with the clinicopathological stages. Analysis of the follow-up data also showed that there was a statistically positive correlation between VEGFR-3 expression and lymph node metastasis in primary tumors (P

3.A STUDY ABOUT THE GENETIC OF PRIMARY HEPATOCELLUAR CARCINOMA

74 primary hepatocellular carcinoma patients (PHC) and 75 controls are tested for HLA and investigated heritability. The results showed the frequency of Bw39 antigen was significantly higher in PHC patients than that in the controls. (Fisher P0.05); but the frequeney of Bw60 antigen, was lower (Fisher P0.05 ). The result of heritability proved the role ofgenetic factor in PHC development.
4.Metabolic syndrome, metabolic syndrome components and thyroid nodule

Objective To evaluate the relationship of metabolic syndrome (MS) and MS components with thyroid nodule. Methods A total of 10 357 subjects ( age > 18 years old) who received physical checkup at the Second Affiliated Hospital of Dalian Medical University between June 2009 and June 2010 were enrolled in this retrospective study. Anthropometric parameter, fasting plasma glucose (FPG),serum lipid profile, blood uric acid, alanine aminotransferase and thyroid ultrasonography were measured. Results The prevalence of thyroid nodule,MS,and thyroid nodule + MS was 46. 96% ,23. 6%,and 11.6%, respectively. The prevalence of thyroid nodule was significantly higher in MS patients than in non MS patients ( 75.9% vs 38. 0%, P < 0. 05 ). Multifactor logistic analysis showed that MS, body mass index (BMI) and FBG (β vales were 0. 78,1.22,and 0. 62,respectively; odds ratios were 4. 167,3. 876,and 2. 359, respectively; all P < 0. 05 ) were independently correlated with the development of thyroid nodule. Conclusions Significantly increased prevalence of thyroid nodule could be found in MS patients. BMI and FBG may be independent risk factors of thyroid nodule.
5.Determination of Metamizole Sodium and Chlorphenamine Maleate in Zhongganling Tablets by Solid-Phase Extracting HPLC

Objective: To establish a method for the determination of metamizole sodium and chlorphenamine malete in zhongganling Tablets. Methods: The sample was determined by ion pair HPLC after it was purified on Sep Pak C 18 microcolumn. The chromatographic conditions included: Hypersil DBS C 18 chromatographic column (250mm?4.6mm, i.d.5?m) as an anlaytical column, methanol mixed solution of sodium heptanesulfonate and glacial acetic acid (600∶400) as a mobile phase, the detection wavelength at 264nm and 1.0mL?min -1 of flow rate. Results: The average recoveries of metamizole sodium and chlorphenamine maleate were 99.6% (RSD was 2.1% and n was 6) and 98.0% (RSD was 1.5% and n was 6), respectively. Conclusion: Metamizole sodium and chlorphenamine maleate can be determined respectively by HPLC with the same mobile phase when Sep Pak C 18 microcolumn solid phase extraction method is used to substitute for the traditional sample pretreatment methods refluxing, extracting and concentrating, and sodium heptanesulfonate ion pair reagent in acid condition is selected.
6.Study on the Quality Standard for Xiaobai Capsules

To establish the quality standard for Xiaobai capsules. Methods: Paeoniae Radix Alba and Angelicae Sinensis Radix were identified by TLC. The contents of psoralen and isopsoralen were determined by HPLC. Results:The qualitative i-dentification was with high resolution and without interference from the negative substances. The linear range of psoralen was 1. 082-108. 200 μg·ml-1 . The average recovery was 99. 82% with RSD of 1. 16%. The linear range of isopsoralen was 0. 818-81. 800μg· ml-1 . The average recovery was 99. 85% with RSD of 1. 81%. Conclusion:The method can be used in the quality control of Xiaobai capsules.
7.Effect of Preoperative Aprotinin Infusion on Blood Loss and Brain Edema in Patients Undergoing Meningioma Resection

Objective To observe the effects of preoperative aprotinin infusion on blood loss and brain edema in patients undergoing meningioma resection.Methods 80 were randomized to receive intravenous normal saline(control group) or aprotinin 2.0×106 KIU(aprotinin group) before operation.The intraoperatively hemostatic appearance was assessed by the surgeon in the form of quantitative scoring.The postoperative degree of brain edema was assessed by radiologist according to the appearance of computer tomography.Results There were no statistic differences in general information,intraoperatively infused volume,urine volume,and the quantitative score of postoperatively brain edema(P>0.05).The operation duration was(209±63) min and(305±93) min,blood loss was 420(150~3270) ml and 610(110~2430) ml in the aprotinin group and the control group respectively(P<0.05).There was statistic difference in the score of intraoperatively hemostatic apperance(P<0.05).Conclusion In the resection of meningeoma,aprotinin was infused with a dose of 2.0×106 KIU preoperatively can shorten the duration of operation and reduce the intraoperative blood loss,but it cannot relieve the degree of postoperatively brain edema.
8.Detecting multiple lysosomal enzymes in dried blood spots by tandem mass spectrometry

Objective Establish a method for measuring the activities of Galactocerebrosidase (GALC), α-Glucosidase(GAA), α-Galactosidase (GLA) and α-L-Iduronidase (IDUA) in dried blood spots specimen by tandem mass spectrometry ( MS/MS ).Methods A total of 2175 dried blood spot samples forinborn errors of metabolism in neonatalscreening center of Shanghai Xinhua hospital were collected in July and November, 2013.And twenty dried blood spot samples from patients withlysosomalstorage disorders( LSDs) of Shanghai Xin Hua Hospital were collected from September 2012 to January 2014.The extraction of DBS was incubated with enzyme substrates and internal standards.After liquid-liquid and solid-phase extraction, the extraction solution was dried under nitrogen and reconstituted.Then enzyme reaction products and internal standards were analyzed by MS/MS.Linearity, precision, accuracy and the limit of detection were evaluated.2175 dried blood spot samples were detected to establish the normal reference range for the activities of four enzymes according to 0.5th to 99.5th percentiles.20 specimens from patients withLSDs were detected to verify the reference range inclinical judgment.Results The intraassay and interassay precisions ranged from 1.7%to 11.8%, and the intraassay and interassay accuracies ranged from 85%to 115%.The linear coefficients for measured concentration of enzyme products/internal standards and theoretical concentration were 0.997-0.999.The limits of detection forGALC, GAA, GLA and GLA were 0.03 μmol/(L· h), 0.09 μmol/(L· h), 0.12 μmol/(L· h) and 0.16 μmol/(L· h) .The normal reference values for GALC, GAA, GLA and GLAwere 0.51-8.51μmol( L· h) ,1.99-22.22μmol/( L· h),1.68-41.59 μmol/(L· h) and 2.36-19.21 μmol/(L· h).The enzymes of 20 patients with LSDs were remarkably decreased compared to the normal range.The Krabbe, Pompe, Fabry, MPSⅠpatients can be effectively detected by this MS/MS method.Conclusions A MS/MS method for measuring GALC, GAA, GLA and IDUA enzyme activities in DBShas been established.
9.Study of the relationship among the uric acid,lipid and insulin resistance in patients with different glucose levels

Objective To study the levels of senlm uric acid(UA)in normal glucose(NC),impaired glucose regulation(IGR)and type 2 diabetes mellitus(T2DM),and investigate its relationship with insulin resistance and dyslipidemia.Method The levels of blood glucose,lipids,fasting insulin(HNS)and serum UA were measured in patients of 45 T2DM(T2DM group),20 IGR(IGR group)and 29 NC(NC group).Status of insulin resistance and insulin secretion function was evaluated by HOMA-IR and ISI.Results The levels of triglyceride(TG)and UA in T2DM group and IGR group were significantly higher than those in NC group[(3.34±8.77),(1.85±0.67),(1.26±0.38)mmoi/L and(316.71±96.20),(403.62±76.80),(325.45±94.43)mmol/L](P<0.01).HDL-C levels in T2DM group were significantly lower than those in IGR and NC group[(1.05±0.30),(1.07±0.21),(1.12±0.20)mmol/L](P<0.01).NO significant difference of FINS levels was found in the three groups.HOMA-IR level in T2DM and IGR group was higher than that in NC group(3.84,3.77,2.34)(P<0.01).ISI in T2DM and IGR group was lower than that in NC group(-4.52±0.79,-4.44±0.19,-4.03±0.58)(P<0.01).Correlation analysis indicated that the level of UA was positively related with BMI.TG and negatively related with HDL-C.Conclusion Increased UA in IGR indicates that hyperurieacidemia developes before DM.
10.Effects of adiponectin on atherosclerotic plaque evaluated by intravascular ultrasound in rabbits

Objective To investigate the effects of adiponectin on the development of atherosclerosis.Methods The in vivo role of adiponectin on the development of atherosclerosis in rabbits was investigated mainly using adiponectin-producing adenovirus(Ad-APN)and intravascular ultrasound(IVUS).On day 14 after Ad-APN local transfer to the intima of abdominal aortas,abdominal IVUS images were recorded and then rabbits were sacrificed.Abdominal aortas were collected and performed histochemical analysis.Results Ultrasonography revealed a significantly reduced atherosclerotic area,in abdominal aortas of rabbits infected through intima with Ad-APN,by 36.39% compared with the area in adenovirus expressing β galactosidase gene(Ad-βgal)treated rabbits(P<0.01),and by 37.50% compared with that before treatment(P<0.01).The lumen area stenosis was also reduced by 23.37%(P<0.05)and 33.15%(P<0.01),respectively.In rabbits with Ad-APN infection,the atherosclerotic plaque area as seen on Oil Red O staining was reduced significantly,by 30.70%(P<0.05)and the greatest thickness of plaque was reduced,by 20.83%(P<0.05)as compared with those in Ad-βgal-treated rabbits,respectively.Conclusions Intravascular ultrasound analysis is a valuable method in the in vivo study about the effects of adiponectin on the development of atherosclerosis.
