1.Expression and function of tumor susceptibility gene 101 in multidrug-resistant gastric cancer cells

Objective To investigate the expression and possible function of tumor susceptibility gene 101 (TSG101) in multidrug-resistant cell lines of gastric cancer.Methods The expression of TSG101 was examined in gastric cancer cell line SGC7901 and its vincristine(VCR)-resistant subline SGC7901/VCR with semi -quantitative RT-PCR and Western blot. TSG101 eukaryotic expression vector was transfected into SGC7901 cells by lipofectamine TM 2000. The expression levels of TSG101 in SGC7901 and the transfectants were detected with Western blot. Fluorescence-activated cell sorting was applied to examine the cell cycle alteration and the intracellular mean fluorescence intensity of adriamycin (ADR). Growth curve and drug sensitivity of cells to VCR and ADR were analyzed by MTT assay.Results TSG101 was highly expressed in VCR resistant gastric cancer cells. The expression of TSG101 in TSG101 transfectants was up-regulated compared with that in SGC7901 transfected with empty vector or in SGC7901 cells. TSG101 transfectants were accumulated in S phase, with a concomitant decrease of cell population in G 1 phase. MTT assay showed that TSG101 transfectants proliferated rapidly and were more resistant to VCR and ADR than control cells. Conclusions The over-expression of TSG101 could promote the multidrug resistance phenotype of sGC7901 cells. TSG101 may play a certain role in multidrug resistance of gastric cancer.
2.The role of 18F-FDG PET/CT in evaluation of radiosensitivity enhancement by irisquinone

Objective To evaluate the value of 18F-FDG PET/CT in assessing radiosensitivity enhancement by irisquinone (IR) on rabbit xenografted VX2 lung tumor models.Methods Twenty-four tumor-beating rabbits were randomly divided into 3 groups (8 rabbits/group):group A with radiotherapy alone,group B with combined radiotherapy and IR,and group C without radiotherapy (the control group).18F-FDG PET/CT imaging was performed before radiotherapy and 24 h and one week after radiotherapy.The tumor SUVmax on delayed imaging was calculated in all rabbits.Two rabbits in each group were sacrificed after PET/CT imaging.HE staining was used to assess the differences in cancer cells among groups.Paired t test,one-way analysis of variance and Kaplan-Meier analysis were performed to analyze the data using SPSS 13.0.Results Before radiotherapy,the tumor SUVmax of all the 24 rabbits on standard and delayed imaging were 2.200 ± 0.761 and 3.162 ± 0.833 (t =-5.582,P < 0.01).At 24 h post-radiotherapy,the delayed SUVmax of groups A,B and C were 2.614 ± 0.654,2.349 ± 0.869 and 5.663 ± 1.144,respectively.The differences between pre-radiotherapy and 24 h post-radiotherapy were statistically significant in all three groups (t =2.527,3.620,11.011,all P <0.05).One week after radiotherapy,the delayed SUVmax of groups A,B and C were 3.625 ± 1.064,3.058 ±0.850 and 7.424 ± 1.751,respectively.The differences among groups A,B and C were statistically significant (tA∶ B =2.652,tA∶C =3.799,tB∶C =4.366,all P <0.05).The cancer cells of group B were fewer than those of groups A and C by pathological findings,which was consistent with 18F-FDG PET/CT results.The survival times of groups A,B and C were (62.375 ±4.534),(69.000 ±4.660) and (54.125 ±5.276) d,respectively.Kaplan-Meier survival curves revealed better survival of group B as compared to groups A and C,respectively (Log-rank,x2 =7.355,16.943,both P < 0.01).Conclusion 18 F-FDG PET/CT is able to evaluate the effect of irisquinone on tumor radiosensitivity enhancement.
3.Special prosthetic replacement in limb salvage treatment of bone tumors bone tumors

Thirty-three patients with bone tumor(malignant in 10 cases and benign in 23 cases)selected from Department of Bone Tumor,the Second Affiliated Hospital of Mongolian Medical College from October 1999 to August 2006 underwent special prostheric replacement at neoplastic segment.The prosthesis was made of stainless steel, alloy of cobalt-chrome-molybdenum,or titanium alloy,which are provided by Beijing Lidakang Technology Company Limited.All patients provided the conftrmed consent.In the follow-up of 6-70 menths,one patient with giant cell tumor of haunch bone,two patients with malignant synovioma,and one patient with malignant fibrous histiocytoma died due to pulITlonary metastasis.In all those patients who survived,none have local recidivation,transferring,or fracture.Among them,one patient had postoperative infection and rigor at knee joint.Three patients had ache at middle part of thigh(junction between prosthesis and femoral bone)after knee joint replacement.According to the Ennekhag scores after reconstruction of surgical oncology,fineness rate of upper limbs was 80%,and that of lower limbs was 86.4%.Odynolysis rate of shoulder ioint was 90%.Active movement was increased and activity of femoral articulation Was generally normal.Extension and flexion of knee joint ranged from 0 to 100 degree.Rejection occurred in stainless steel prosthesis of knee joint in one patient.Junction between prosthesis and femoral bone Was looge in two cases.All other prostheses had great biocompatibillty to tissues.
4.Study on the changes and clinical significance of the DNT cells and T lymphocyte subtype in children with infectious mononucleosis

Objective To study on the changes of the DNT cells and T lymphocyte subtype in children with infectious mononucleosis (IM) and its clinical significance.Methods The flow cytometry (FCM) was used to detected the DNT cells and other T lymphocyte subtype in 48 cases of IM.Results The study showed that DNT cells( 9.39 ± 4.89 )% were greatly increased in comparison with normal controls (NC) (4.26 ± 1.68)% ( P <0.01 ).CD4 cells(21.45 ±9.87)% were decreased ( P <0.01 ) and CD8 cells increased in comparison with NC (32.43 ± 5.07) % ( P < 0.01 ).Conclusion DNT cells and T lymphocyte subtype can be used to evaluate the immune function of children with infectious mononucleosis (IM) and provide guidance for adoptive immunotherapy.
5.Clinical observation of valsartan on chronic pulmonary heart disease with heart failure

Objective To study and analyze the clinical effect of valsartan on chronic pulmonary heart disease. Methods 100 cases of heart failure patients with pulmonary heart disease treated in our hospital from January 2015 to October 2016 were selected and randomly divided into the control group and the experimental group,50 cases in each group.Two groups of patients were given the basic treatment of diuretics,low salt diet, digitalis and nitrates,so as to ensure that the patients had enough rest.Patients in the control group were treated with benazepril,and the patients in the experimental group were treated with valsartan.The clinical indexes of the experimental group and the control group were compared and analyzed. Results After corresponding treatment,the number of effective treatment was 48 cases.In the control group,the total effective number was 41 cases. The effective rate of the control group (82.0%) was significantly lower than the experimental group(96.0%) (P<0.05).The incidence of adverse reactions in the experimental group was 8.0%, and the fatality rate was 10.0%. The incidence of adverse reactions in the control group was 10.0%, and the fatality rate was 12.0%.There was no significant difference between two groups.After treatment,the indexes of LVEDD and LVESD in the experimental group were better than those in the control group,with statistical significance(P<0.05).Conclusion Valsartan was effective in the treatment of chronic pulmonary heart disease with heart failure.It can improve the treatment efficiency and security in a certain extent,and has the significance of further popularization and application.
6.Applied value of susceptibility-weighted imaging in acute ischemic stroke

Objective To evaluate the applied values of susceptibility weighted imaging (SWI) in acute ischemic stroke.Methods Totally 40 patients with acute ischemic stroke were included within 72 hours after symptom onset.They adopted multimodal magnetic resonance imaging examination,including regular magnetic resonance imaging sequence (T1WI,T2WI),time of flight MR angiography (TOF-MRA),diffusion weighted imaging (DWI) and susceptibility weighted imaging (SWI).Furthermore,12 of the 40 patients received dynamic susceptibility contrast enhanced perfusion weighted imaging (DSCPWI) examination.Results Of the 40 patients,12 ones had positive results for multiple lesions of cerebral microbleeds (CMBs) by SWI,and the detection rate was 30%.There were 24 patients showed susceptibility vessel sign (SVS) in SWI,and the susceptibility rate was 60%.In the 12 patients received DSC-PWI examination simultaneously,there were 7 cases had the anomaly range by SWI larger than that by DWI while similar to that by DSC-PWI.Conclusion SWI can show SVS and CMBs clearly,estimate ischemic penumbra,know intracranial conditions completely and supply reliable basis for individualized treatment.
7.Clinical significance of changes of T lymphocyte subtype and serum levels of leukotriene B4 in children with acute leukemia

Objective To study the changes of T lymphocyte subtype levels and serum levels of leukotriene B4 ( LTB4 )in children with acute leukemia (AL). Methods The activity of T lymphocyte subsets:CD3+ T cells,CD4+ T cells,CD8 + T cell levels and NK cell were detected in 40 cases of AL and 40 control by Flow Cytometry(FCM). The serum level of LTB4 was measured by ELISA in all subjects. ResultsCompared with normal control , the percentages of CD3 + T cells ( [ 49.51 ± 6.53 ] % vs [ 67.63 ± 5.23 ] % ), CD4 + T cells ([22.15±4.87]% vs [34.43 ±5.07]%),CD8+ T cell ([23.41 ±3.96]% vs [28. 17 ±4.61]%),NK cell ( [5.41 ± 4.25 ] % vs [ 13.10 ± 4.91 ] % ) were significantly lower in children AL patients, ( Ps <0.01 ). However,the serum levels of LTB4 increased significantly in AL patients compared with normal control ( [ 25.23 ± 6.24 ] ng/L vs [ 8.28 ± 2.53 ] ng/L, P < 0.01 ). ConclusionT lymphocyte subtype and serum level of LTB4 showed significant changes in children AL patients compared with normal control, which revealed an important theoretical basis to AL pathogenesis.
8.Impact of individualized diabetes education on disease awareness and blood glucose control in type 2 diabetic patients

This study was to evaluate the effect of individualized health education on blood glucose control in type 2 diabetic(T2D)Patients.Two hundred and thirty-eight T2D patients were randomly received individualized diabetes education(intervention group)or general education(control group).At 48 week,change of blood glucose control from baseline was observed.The blood glucose level and HbA1c in the intervention group were lower than those in the control group[(7.1±0.8)vs (9.3±6.5)mmol/L,(7.0 ±1.3)%vs(8.0%±1.0)%;P<0.05].Thus,individualized diabetes education might be helpful for blood glucose control in T2D patients.
9.Analysis on factors influencing the number of clinical pathways implemented at public hospitals

Objective To analyze the key factors influencing the number of diseases or conditions in which clinical pathways ( CPs) were implemented at public hospitals of China. Methods Based on the questionnaire survey of 51 public hospitals that had implemented CPs in Shanghai, Hubei province, and Gansu province, a multivariate logistic model was used to analyze the factors that influenced the number of CPs implemented in hospitals. Results In terms of the 14 issues and difficulties found in CPs′implementation, the overall acceptance rate was 38. 8% on average among surveyed hospitals, yet with a great variation(from 7. 1% to 100. 0%). A multivariate logistic model showed that the recognition of public hospitals on the issues and difficulties in CPs′implementation did not affect the number of implemented CPs, and tertiary public hospitals had significantly higher number of CPs implemented than secondary hospitals. Conclusions The resources and management capabilities of public hospitals in China are the determinants of CPs′promotion.
10.The expression and significance of trefoil factor 3 and SDF-1/CXCR4 biological axis in papillary thyroid carcinoma.

OBJECTIVE:
To study the expression of trefoil factor 3 (TFF3)with stromal cell-derived factor (SDF-1) and its receptor (CXCR4) in papillary thyroid carcinoma(PTC), and investigate the function of TFF3, SDF-1/ CXCR4 and the relationship among them during the tumor genesis,development and outcome of PTC.
METHOD:
Detecting the expression of TFF3 and SDF-1/CXCR4 by immunohistochemical method (SP) in 92 cases of PTC and para-carcinoma tissue. Semiquantitative analysis of the results of immunohistochemistry was conducted by image analysis software.
RESULT:
(1) TFF3 protein was expressed in the cytoplasm of cancer cells,while TFF3 was negative or weakly positive in follicular cells of para-carcinoma tissue. The positive expression rate of TFF3 was 92.39%, of which the strong positive rate of clinical stage III-IV accounted for 71.79% (42/59) and that of clinical stage I-II was 33.33% (11/33) (P < 0.01). The positive rate of TFF3 was significantly higher in the cases with lymph node metastasis than those without lymph node metastasis (100.00% vs 86.27%, P < 0.05). The AOD value of TFF3 was higher in PTC than in para-carcinoma tissue, that in cases with lymph node metastasis was higher than those without lymph node metastasis, and that in stage III-IV was higher than those in I-II (P < 0.05 or P < 0.05). (2) There was high expression of SDF-1 in the cytoplasm of malignant tissues. The para-carcinoma tissue was weakly positive or negative to SDF 1 and metastatic lymph nodes was weakly positive to SDF 1. The positive rates and AOD values of SDF-1 protein were similar to those of TFF3 in PTC,that is to say the positive rate and AOD values were higher in PTC than in para-carcinoma tissue, those in cases with lymph node metastasis were higher than those without lymph node metastasis, those in stage III-IV were higher than those in I-II, and those in patients older than 45 years old was obviously higher than those in patients under 45 years old (P < 0.05 or P < 0.01); CXCR4 was also mainly expressed in cytoplasm with few expression in nuclei, while negative or weakly positive in para-carcinoma. The positive rate and AOD values of CXCR4 in PTC were similar to SDF-1, meaning that they were higher in PTC than in para-carcinoma tissue and associated with the clinical stage, lymph node metastasis and age (P < 0.05 or P < 0.01). (3) There was positive relationship between TFF3 and SDF-1 as well as between SDF-1 and CXCR4 in PTC (r = 0.971, P < 0.01).
CONCLUSION
The high expression of TFF3, SDF-1 and CXCR4 in PTC are correlated with carcinogenesis and progression, and may play a significant role in evaluating the malignancy degree and progression of PTC.
