1.The role of TLR4 acetylization in its inflammatory pathway in the pathogenesis of gestational diabetes mellitus

Objective To probe into the pathogenesis of gestational diabetes mellitus (GDM) by studying expression of TLR4 acetylation in peripheral blood mononuclear cells of gravida with GDM as well as its role in TLR4 inflammatory pathway. Methods 30 normal gravidas and 30 gravidas with GDM were enrolled in the study, 15 mL peripheral blood from every participant was collected for extraction of mononuclear cells via density gradient centrifugation and culturing in vitro by LPS. The expression of TLR4 acetylation in the mononuclear cells was detected using immunoprecipitation and western blot and the levels of inflammatory cytokines, TNF-α, IL-1 and IL-10 in the supernatant were detected by ELISA. Results TLR4 acetylation appeared positive in GDM group, and negative in the control group. After the LPS intervention, the degree of TLR4 acetylation was enhanced significantly and the latter was significantly higher than those of the GDM group and the normal + Lps group (P < 0.05). The differences in the levels of inflammatory cytokines, TNF-α, IL-1, IL-10, were statistically significant (P < 0.05). Conclusion TLR4 acetylation exists in mononuclear cells of gravida with GDM, and mediates the release of inflammatory cytokines by influencing the activation of TLR4 inflammatory pathobenesis, which contributes to the promoted antiinflammatory - proinflammatory imbalance in gravidas. In this way, it is involved in the growth of GDM.
2.Error Analysis of X—knife Treatment Plan

Objective To investigate the probability of verifying the X-knife treatment plan by the radiotherapy simulator,and to analyze and report the errors of the X-knife treatment plan for reference.Methods The radiation fields of the X-knife treatment plan were observed in the whole simulating treatment process by the radiotherapy simulator.Results The error rate of X-knife treatment plan were in the simulating treatment process was 5.9%(22/372).Among the errors,3 cases of X-knife plans were found in the simulating treatment process,and the errors of isocenter in Z direction were 10cm.Conclusion This is a special error of X-knife TPS,which deserves more attention in stereotactic radiotherapy.
3.Influence of MBL on the expression of Bcl-2 and p53 gene in thyroid cancer cell lines

Objective To observe the effect of recombinant human mannose-binding lectin (RhMBL)on the expression of Bcl-2 and p53 gene in thyroid cancer cell lines.Methods Two kinds of thyroid cancer cell lines( K1 cells and FTC-133 cells) were dealed with different concentrations of rhMBL( control group:0 mg/L;experimental group:1 mg/L),then total protein was extracted to detect the expression of Bcl-2 and p53 by Western Blot.Results The expression level of p53 and Bcl-2 were observed to be obviously down-regulated after incubation with RhMBL.( 1 ) K1 cells:There was significant difference on the expression of p53 and Bcl-2 between control group and experimental group [ p53:( 1.51 ± 0.29) vs (0.74 ±0.07),t =7.63 ; P =0.040;Bcl-2:( 1.20 ± 0.07 ) vs ( 0.59 ± 0.04 ),t =6.28,P =0.001 ] ; ( 2 ) FTC-133 cells:there was also significant difference on the expression of p53 and Bcl-2 between control group and experimental group [ p53:( 0.88 ±0.14) vs(0.35 ±0.08),t =3.29,P=0.003; Bcl-2:(1.07±0.11) vs (0.33 ±0.06),t =5.73,P=0.000 ].Conclusion Bcl-2 and p53 may play an important role on the induction of thyroid cancer cell apoptosis and inhibition its proliferation by RhMBL.
4.The initial investigation of the change of myeloid dendritic cells level in ankylosing spondylitis after treatment with TNF-α blocker

Objective To investigate the role of myeloid dendritic cells(mDC)in the pathogenesis of ankylosing spondylitis (AS),and to study the mechanism of tumour necrosis factor (TNF)-α blocker on the treatment of AS by counting the number of mDC before and after the treatment.Methods Peripheral blood from 21 AS patients treated with 50 mg rhTNFR-Fc and 15 AS patients treated with placebo were investigated at week 0,week 2 and week 6 in a randomized,double-blind,placebo-controlled trial.Three-color flow cytometry analysis was used to investigate the change of the number of mDC before and after the treatment.And their correlation with the clinical parameters was analyzed.Results MHC Class Ⅰ positive mDC(Lin-/CD11c+/HLA-abc+)in AS patients was slightly less than in healthy controls (not statistically significant).No significant change in MHC Class Ⅰ mDC number was observed after drug treatment.The number of MHC Class Ⅱ mDC cells was not correlated with clinical parameters.Conclusion The treatment with rhTNFR-Fc in AS induces a significant upregnlation of MHC-Ⅱ DC.
5.Rapid Screening of Carbamates Pesticide Residues in Vegetables by Electrospray Ionization Mass Spectrometry Based on Principle of Enzyme Inhibition

Arapidscreeningmethodforthedeterminationofcarbamatepesticides(CBPs)residuesin vegetables by measuring acetylcholinesterase ( AChE ) inhibition rate using electrospray ionization mass spectrometry ( ESI-MS ) has been established. After pretreatment by QuEChERS method, sample solution reacts with AChE using acetylthiocholine as substrate. AChE inhibition rate was calculated by determination of the conversion of substrate to product ( thiocholine) using ESI-MS. The temperature, time and concentration conditions of enzymatic reactions have been optimized. The relationship between the concentration of 10 kinds of common CBPs and AChE inhibition rate was researched. Matrix effects of real vegetables were studied. The limit of detection ( LOD) , which was measured by 3 times of enzyme inhibition rate of pesticide-free vegetable samples, was 0. 01-0. 05 mg/kg. The results showed that the method was better than the current national standard method of china for rapid screening of pesticide residues and fully meet the requirements of maximum residue limits( MRL) for pesticides in food of national food safety standard. False positive results were avoided effectively due to its good ability of resistance matrix interference. The reliability was proved by analyzing vegetables with liquid chromatography-tandem mass spectrometry. The method is simple, rapid, sensitive, reliable, and can be used for the rapid, high-throughput screening of CBPs in vegetables.
6.High Throughput Screening of 86 Kinds of Glucocorticoids in Cosmetics Using QuEChERS and Isotope Dilution-Liquid Chromatography Copuled to High Resolution Time-of-Flight Mass Spectrometry

A high throughput screening method based on QuEChERS purification and stable isotope dilution-liquid chromatography coupled to high resolution time-of-flight mass spectrometry was developed for the simultaneous rapid determination of 86 kinds of glucocorticoids (GCs) in cosmetics.The analytes were extracted by acetonitrile, and then the extracts were purified using an improved QuEChERS method.The chromatographic separation was performed on a novel multiple chromatographic retention mechanisms column of Poroshell 120 PFP (100 mm × 2.1 mm, 2.7 μm) with gradient elution using 0.2% (V/V) acetic acid and acetonitrile as mobile phase.The accurate mass database of parent ions and mass spectra library of fragment ions of 86 GCs were established under positive ionization mode with electrospray ionization source.Based on the method described above, the qualitative identifications of the 86 GCs were accomplished without the contrast of standard substances.The results demonstrated that the linear range of this method was from 2 μg/L to 200 μg/L with good correlation coefficients of R2>0.99.The average recoveries of the 86 GCs ranged from 66.2% to 112.8%, and the relative standard deviation (RSDs) was 4.6%-13.9% at three different spiked levels.The limit of detection (LOD) and quantification (LOQ) were 0.006-0.015 mg/kg and 0.02-0.05 mg/kg, respectively.The method is simple, efficient, reliable and accurate, and is suitable for high throughput screening of 86 GCs added illegally in cosmetics.
7.Correlation among subjective well-being, social support, and coping style in empty-nest elderly in Hangzhou

Objective To explore the relationships among subjective sense of happiness and social support, coping style in empty-nest elderly in Hangzhou. Methods A total of 225 empty-nest elderly ( convenient sampling method) were assessed by scales including happiness scale made by Memorial University of Newfoudland, perceived social support scale and trait coping style questionnaire. All data utilized descriptive statistics, Pearson correlation analysis and stepwise multipe regression analysis. Results The positive affection for empty-nest elderly acquired (4. 955 ± 2. 593) scores, negative affection (2. 600 ± 2. 865) scores, positive experience (9. 080 ± 3. 106) scores, negative experience ( 4. 322 ± 3. 025 ) scores. The knowing of social support and each dimensions had positive correlation with the subjective sense of happiness, and the same with positive response, but negative response had negative correlation with the subjective sense of happiness. Positive response and family support were positive predictive factors for subjective sense of happiness. Conclusions Social support and coping style are the important influencing factors to subjective sense of happiness for empty-nest elderly in Hangzhou.
8.Effects of individualized psychological nursing on patients with Non-Hodgkin lymphoma undergoing chemotherapy

Objective:To explore the effects of individualized psychological nursing on improving patients with Non-Hodgkin lymphoma (NHL) undergoing chemotherapy.Methods:Totally 96 NHL patients admitted in Huzhou Central Hospital from May 2016 to May 2018 were selected by convenient sampling and divided into the control group and the study group according to the random number table, with 48 cases in each group. Patients in the control group received routine care, while patients in the study group received individualized psychological nursing according to the patients' psychological characteristics Self-Anxiety Scale (SAS) and Self-Depression Scale (SDS) were used to compare the psychological status between the two groups and investigate their satisfaction with nursing care.Results:After intervention, the SAS score (41.6±5.8) and SDS score (40.9±4.9) of the study group were lower than those of the control group, and the difference between the two groups was statistically significant ( t=4.849, 6.347; P<0.01) . The study group's satisfaction with nursing care was 97.9%, and that of the control group was 81.3%, and the difference between the groups was statistically significant ( P<0.05) . Conclusions:The use of individualized psychological nursing intervention can effectively reduce the anxiety and depression of chemotherapy patients, improve their psychological status, and enhance their recognition of nursing work.
9.Comparative Analysis of Chemical Composition in Methanol and Petroleum Ether Fractions from Curcuma longa of Different Habitats

OBJECTIVE:To compare the chemical compo sition difference in methanol and petroleum ether fraction from Curcuma longa of different habitats. METHODS :The ultrasonic method was used to extract C. longa from 7 defferent producingareas(S1-S7),and methanol and petroleum ether fraction were obtained and calculated yield. The curcumin compounds in methanol fraction were determined by LC-MS ;The chemical components in petroleum ether fraction were analyzed by GC-MS , and the relative percentage content was determined by peak area normalization method after determining its structure by comparing NIST 2005 standard mass spectra and Wiley 275 standard mass spectra. SPSS 25.0 software was used for principle component analysis(PCA)and cluster analysis of relative percentage content of common components in petroleum ether fraction from C. longa of different habitats. At the same time ,the influence of latitude of the habitats on the content of total tumerone (by tumerone and ar-tumerone )was analyzed. RESULTS :The yield of methanol fraction were 1.35%-8.90% from C. longa of 7 habitats;the yield of petroleum ether fraction were 0.81%-4.90%,which were the highest in C. longa from Longyan of Fujian Province. There was no significant difference in the relative content of curcumin compounds(reference peak area )from S 1,S3-S7,which was in descending order as follows as curcumin >desmethoxycurcumin>bisdemethoxycurcumin. There was slightly different in curcumin compounds of C. longa from S 2,mainly manifesting as the content of bisdemethoxycurcumin was higher than that from other producing areas. Totally 48 chemical compositions were identified from petroleum ether fraction in C. longa from different habitats , mainly being sesquiterpenoids and monoterpenoids. 23,10,15,18,11,14,15 chemical compositions were identified from S1-S7,accounting for 94.49%,96.09%,95.66%,98.98%,99.24%,89.05% and 97.27%. There were 4 common compositions in C. longa from different habitats ,which were tumerone (17.90%-43.07%),ar-tumerone(6.97%-33.66%),(6R,7R)-bisabolone (1.60%-4.28%),curlone(6.80%-20.63%). PCA analysis showed that accumulative contribution rate of former 6 principle components was 100%. Cluster analysis showed that S1,S2, S6 was clustered into a category ,respecrively;and others intoa category. Total content of total tumerone decreased first and then increased as the increase of latitude ,which was the highest in Mianyang of Sichuan province (64.28%)and the lowest in Zhangzhou of Fujian province (26.92%). CONCLUSIONS : There are difference in composition and content of methanol and petroleum ether fractions in C. longa from different habitats.
10.The current situation and influencing factors of patient perception for humanistic care in 30 provincial hospitals

Objective To understand the current status and influencing factors of patient perception for humanistic care in China hospitals,and to provide a basis for developing nursing humanistic care measures and improving the quality of nursing humanistic care services.Methods A total of 30,099 outpatients and inpatients from 107 hospitals in 30 provinces(autonomous regions and municipalities)from July to August 2022 as survey subjects.A general information questionnaire and the Relational Caring Questionnaire-Patient Form were used for a cross-sectional survey,and a single-factor analysis was used to analyze the influencing factors of patient relationship care.Results Finally,29 108 valid questionnaires were collected,and the effective questionnaire recovery rate was 96.7%.The patient evaluation of relationship care was(65.72±8.61)points.Single-factor analysis showed that gender,age,marital status,children's situation,education level,occupation,place of residence,average family income,medical insurance type,visiting department,and location of the visiting hospital,and whether or not surgery were influencing factors of patient relationship care(P<0.05).Conclusion The evaluation score of caregiver-patient relationship care among Chinese hospital patients is above average,but there is still room for improvement in western and rural regions,seriously ill and outpatient patients,low-income and low-medical insurance reimbursement populations,and non-surgical patients.Medical institutions at all levels should optimize and improve nursing humanistic care services based on influencing factors,and further enhance patients'perception of nursing humanistic care.
