2.Changes of Painful P300 in Rabbits with Adjuvant Induced Arthritis
Chinese Journal of Rehabilitation Theory and Practice 2012;18(3):233-235
Objective To explore the changes of the painful P300 under the state of pain induced by adjuvant induced arthritis (AIA) inrabbits. Methods The painful P300 and pain threshold in 10 rabbits were recorded before Freud's adjuvant injection, before and after morphineinjection on the 7th day and the 21st day after Freud's adjuvant injection. Results With the pain threshold lowed (P<0.01), the latencyof painful P300 was differentially shortened on the 7th day and 21st days after adjuvant injection (P<0.01). After morphine injected, painthreshold increased (P<0.01) and the latency of painful P300 was prolonged (P<0.01). On the 7th day, the amplitude of painful P300 differentiallyincreased (P<0.05), and decreased (P<0.01) after morphine injected. The amplitude of painful P300 on the 21st day was significantlylower than that of the 7th day (P<0.01). The pain threshold correlated with the latency of painful P300 (r=0.516, P<0.01). ConclusionPainful P300 is related with the condition of pain in rabbits with AIA.

Background and purpose:Proteasome inhibitors constitute a novel class of antitumor agents that has a complex mechanism of action.Previous studies have confirmed that proteasome inhibitor MG-132 can significantly inhibit Hep-2 cell growth and induce cell apoptosis in a manner that is dependent on dosage and time. But it also induced p-STAT3 protein expression. The aim of this study was to explore whether the STAT3 gene can,by transfecting short hair pin RNA(shRNA),enhance the anti-tumor effect of MG-132 on human laryngeal carcinoma cells.Methods:Hep-2 cells were plated into 96-well and 6-well plates and incubated overnight. Then, they were treated with MG-132 alone and combined with pshSTAT3.Their cell growth was detected by MTT assay, and apoptosis was examined with flow cytometry. The protein expression of p-STAT3 was detected by Western blotting. Results:MTT assay showed that a combined group inhibited the proliferation of Hep-2 cells compared to the MG-132 group and pshSTAT3 group(P<0.01).Flow cytometry showed that apoptosis of the combined group was significantly higher than the MG-132 group and pshSTAT3 group (P<0.01).Western blotting showed that the p-STAT3 protein expression up-regulation was observed in the MG-132 group, whereas down-regulation was expressed in the combined group and pshSTAT3 group. Conclusion:The shRNA targeting STAT3 gene can prevent the up-regulation of p-STAT3 protein following a MG-132 treatment thereby significantly enhancing the anti-tunlor effect of the protease inhibitor,MG-132,on human laryngeal carcinoma cells.

Journal of Central South University(Medical Sciences) 2007;32(1):63-68
Objective To derive hematopoietic stem cells with functional properties of hematopoietic reconstitution from murine embryonic stem (ES) cells. Methods ES-D3 cells by formation of the day-4 embryoid bodies (4dEBs) were induced into hematopoietic stem cells by co-culture with murine bone marrow endothelial cell-conditional medium (mBMEC-CM) and the fetal liver stromal cell-conditional medium (FLSC-CM). This experiment was designed to 4 groups (mBMEC-CM + FLSC-CM group, mBMEC-CM group, FLSC-CM group, and the control group). Results The total cell numbers, CD34 + cell numbers, and colony numbers formed in the mBMEC-CM + FLSC-CM group were the highest among the 4 groups. The cells in the mBMEC-CM + FLSC-CM group resumed the hematopoietic system of the mice after being transplanted with the inducing cells. Conclusion The culture condition combing mBMEC-CM with FLSC-CM can promote murine ES cells differentiating into hematopoietic stem cells with functional properties of hematopoietic reconstitution.

Journal of Central South University(Medical Sciences) 2013;38(9):915-919
Objective:To explore the relationship between social support and negative emotion in parents of children with congenital heart disease before the operation.
Methods:Social support rating scale (SSRS), self-rating anxiety scale (SAS), and Center for epidemiological survey-depression scale (CES-D) were used to assess the association between social support and anxiety and depression in 226 parents.
Results:The mean SSRS score was signiifcantly lower than the normal. There was signiifcant difference in social support of parents in monthly family earning and knowledge about surgery. Parents with monthly family income≤1000 yuan had lower scores than the other groups. Parents who knew nothing about the surgery approach had lower scores. Social support was significantly correlated to negative emotion (anxiety and depression). Social support could predict anxiety and depression.
Conclusion:Social support is widely and significantly correlated to negative emotion in parents of children with congenital heart disease before the surgery.
