Objective To explore the adeno-associated virus (AAV)gene transduction and cellular endocytosis mediated by ultrasound combined with microbubbles in two types of cells. Methods HeLa and NIH/3T3 cells were infected by rAAV2-EGFP at a concentration gradient to get the optimal concentrations for enhancement. At these concentrations,HeLa and NIH/3T3 cells were infected by rAAV2-EGFP mediated by ultrasound combined with microbubbles. The gene transduction efficiency were observed and measured by fluorescence microscopy and flow cytometry at 48 h after treatment. The cell viability was tested by CCK-8.The number and distribution of cellular clathrin-coated endocytic pits were observed by confocal fluorescence microscopy and transmission electron microscopy on 45 min after treatment. Results The optimal concentrations for HeLa and NIH/3T3 cells were 2000 v.g./cell and 10000 v.g./cell.Ultrasound combined with microbubbles significantly enhanced the transduction efficiency of rAAV2-EGFP (P <0.01) without significant cell viability decrease (P > 0.05 ).Confocal fluorescence microscopy and transmission electron microscopy demonstrated that clathrin-coated endocytic pits were more obviously increased in ultrasound combined with microbubbles mediated AAV transduction group than AAV transduction group. Conclusions Ultrasound combined with microbubbles can efficiently enhance the gene transduction of AAV,whose cellular transportation depends on cellular endocytosis,in two types of cells.Stimulating cellular endocytosis might be one of the mechanisms of enhanced cellular transportation of AAV mediated by ultrasound combined with microbubbles.

Objective: To investigate the effects of low molecular weight heparin(LMWH) on vascular endothelial growth factor(VEGF) expression of early diabetic nephropathy. Methods: Ninety-five male SD rats were randomly divided into three groups: normal control rats, STZ-induced diabetic rats and diabetic rats treated with LMWH. The renal tissues were subjected to immunohistochemical staining after 1,2,4,6,and 8 weeks’ treatment respectively to quantify the VEGF expression. Results: Immunohistochemical staining demonstrated an increasing in VEGF positive cells in diabetic rats. It was found that there were significant differences in VEGF staining intensity between diabetic rats and normal control rats and between LMWH treated rats and untreated diabetic rats after two weeks treatment. Conclusion: The inhibition of VEGF expression may be one of the mechanisms of LMWH’s renal protective effects on early diabetic nephropathy.

Objective To evaluate the effects of remifentanil on the proliferation, the cell cycle and apoptosis of human liver carcinoma cell line HepG2 in vitro. Methods Human liver carcinoma cells HepG2 were cultured in vitro. The HepG2 cells of the test group were incubated in the RPMI-1640 medium with remifentanil at different concentration(0.001,0.01,0.1,1,10,100,200 μmol/L).The HepG2 cells of the control group were incubated in the RPMI-1640 medium for 48 hours. The level of the cell proliferation was evaluated with methylthiazolyldiphenyl-tetrazolium bromide (MTT) method. The cell cycle was detected with flow cytometry (FCM).The morphological change of apoptosis cell was observed by fluorescence microscopy after staining by Hoechst33258.Results Remifentanil inhibited the proliferation of the HepG2 cells with a dose-dependent effect.Compared with control group, the cell proliferation capability was apparently decreased in the test group (P < 0.05) when the concentration of remifentanil was over 1 μmol/L (P <0.05).However,no significant difference in cell proliferation was found when remifentanil was 100 and 200 μmol/ L.The ratio of G0/G1 phase of HepG2 cells was significantly enhanced and the ratio of S phase of HepG2 cells was significantly decreased when remifentanil was over 1 μmol/L.The fluorescent microscopy stained by the Hoechst33258 showed part of HcpG2 cells apoptosis in test group, and the apoptotic rate was significantly increased when remifentanil was over 1 μmol/L (P < 0.05).Conclusions The data suggest that remifentanil would inhibit the proliferation of HepG2 cells and induce apoptosis when remifentanil was over 1 μmol/L.

Objective To investigate the serum levels and dietary intakes of copper, iron, and zinc among adult residents in Dahongshan copper mine area of Yunnan Province in China. Methods Serum levels of copper, iron, and zinc were measured by atomic absorption spectrometry among 180 subjects collected randomly from Dahongshan mine area, among which 171 samples were valid. Dietary intakes of copper, iron, and zinc of 60adults collected randomly from these 180 subjects were detected by chemical analysis. Results Serum levels of copper, iron, and zinc concentrations of 171 valid samples were (0.90±0.18), (1.25±0.93) , and (0.75±0. 28) mg/L, respectively. Serum copper was negatively correlated with serum iron (r = - 0. 26, P < 0. 001). Serum copper was positively correlated with age (r = 0. 25, P < 0. 001). Females had significantly higher serum copper than males (P=0.011). The dietary intakes of copper, iron, and zinc among these 60 subjects were (1.74±1.09),(16.29±10.73), and (7.56±3.38) mg/d, respectively, in which females had significantly lower dietary intakes than nales (all P < 0. 05). Significant regional differences were observed both in serum levels and dietary intakes of copper, iron, and zinc. Conclusions The average serum levels of copper and iron are normal among adult residents in Dahongshan copper mine area, while the average serum zinc level is low. Dietary intakes of these three minerals do not exceed tolerable upper intake levels, and are low in some subjects.
