1.The effect of thiopental anasthsia on T lymphocyte snbsets in mice

ObjectiveTo observe the effect of thiopental on T lymphocyte subsets in mice. MethodsT lymphocyte subsets were measured by flow cytorneter and monoclonal antibodies and immunofluorescence technic. ResaltsThe CD4 T cell and the CD4/CD8 ratio decreased after thiopental anaesthesia in mice. At the same time the whole amount of white blood cells has no statistical significant difference between test group and control group. ConclusionThiopental anaesthesia inhibits immune function.
2.Medical personnel health status and related factors analysis

Objective To understand the health status of medical staffs in a tertiary hospital of Chongqing City to provide the basic information and scientific basis for formulating the interventional measures.Methods The physical examination data of 1 397 medical staffs in 2013 and 2 178 medical staffs in 2015 were selected as the research objects and performed the summary analysis according to the abnormal indexes distribution,sex and age.Results A total of 1 397 persons completed the physical examination in 2013,the abnormal indexes were detected in 1 250 people,the detection rate of sub-health status was 89.48%;2 178 people in 2015 completed the physical examination,the abnormal indicators were detected in 2 046 people,the detection rate of sub-health status was 93.94%.The comparison of two-year health examination data showed the different degrees of increasing trend in bilateral breast hyperplasia,gallbladder polyps,thyroid cysts,low density lipoprotein cholesterol.Conclusion The health status of medical staffs is not optimistic,so the self health care consciousness of medical staffs needs to be elevated.
3.Analyse on polymorphism of endothelial nitric oxide synthase gene(CA)_n in type 2 diabetes mellitus patients

Objective To explore the relationship of the (CA)n dinucleotide repeats polymorphism of the eNOS gene with type 2 diabete mellitus (DM) and the serum NO level in DM patients. Methods The eNOS genotype was assessed in 179 unrelated Chinese of Han nationality,including 93 patients with DM and 86 normal controls(NC).Genotype was determined by polymerase chain reaction(PCR) and denature polyacrylamide gel electrophoresis (PAGE) with silver stain.Fasting serum NO level was measured by nitrate reductase and fasting serum NOS level was measured by spectrophotometer. Results Twenty two different alleles were identified containing 19-40 CA repeats,in which the frequency of (CA) 30 (166bp)was the highest. The heterozygosity (H) and polymorphism information content (PIC) were 0.85 and 0.83 respectively. Frequency of allele (CA) 34 was higher in DM (7.5%) than in NC (2.9%) significantly. Compared with NC,the serum NO level was significantly lower in T2DM with genotype (CA)n 1 (CA)n 2 when n 1≥34 or n 2≥34. Conclusions The allele(CA) 34 is associated with the susceptibility to DM and may be involved in the development of DM. The eNOS gene polymorphism is connected with the serum NO level.
4.Clinical significance of CXCL9 and IL-22 expression in peripheral blood of patients with psoriasis vulgaris

Objective To detect the expression level of CXCL9 and IL‐22 in peripheral blood of patients with vitiligo vulgaris and explore its role in the pathogenesis of vitiligo vulgaris .Methods The level of CXCL9 and IL‐22 in peripheral blood from 35 vit‐iligo patients (20 active cases and 15 stable cases) and 20 normal controls were measured by enzyme linked immunosorbent assay . Then ,we compared the differences in different groups and analyzed their correlation with ages ,duration ,psoriasis area and severity index (PASI) .Results The contents of CXCL9 and IL‐22 in active cases and stable cases were obviously higher than those in con‐trols respectively by statistical analysis ,there was significant difference (P< 0 .05) .The contents of CXCL9 and IL‐22 in active ca‐ses were obvious higher than those in stable cases ,there was significant difference(P< 0 .05) .Conclusion CXCL9 and IL‐22 may be involved in the pathogenesis of vitiligo vulgaris .
5.Effect of BaoYuan Ⅱ on expression of Smad in Kidney of diabetic rat

Objective: To observe the expression of Smad in diabetic rat’s kidney and the effect of BaoYuan II to it.Methods: SD rats were randomly divided into four groups: control group,model group,Benazepril-treatment group and BaoYuan II-treatment group.The latter three groups were induced to be diabetic nephropathy by intraperitoneal injection with streptozotocin and then the drugs were treated respectively.After eight weeks,the plasma albumin,blood glucose,urinary protein,urinary?2-M were determined and expression of mRNA of TGF-?1,Smad2,Smad3,Smad7 was detected by RT-PCR.Results: Compared with diabetic rats group,the level of plasma albumin was increased and the level of blood glucose,urinary protein,urinary?2-M was depressed in BaoYuan II-treatment group.The expression of TGF-?1 and Smad2,Smad3 were down-regulated and Smad7 was up-regulated in model group.The effects of BaoYuan II were similar to Benazepril.Conclusion: BaoYuan II had protected effect to diabetic nephropathy rats.It acted by regulating TGF-?1 and Smad2,Smad3,Smad7 possibly.
6.Effects of jinlongshe granules on apoptosis of MKN-45 human gastric cancer cells orthotopically transplanted in nude mice

OBJECTIVE: To assess the effects of Jinlongshe Granules (JLSG) on tumor growth of gastric carcinoma. METHODS: Fifty nude mice orthotopically transplanted with MKN-45 human gastric cancer cells were divided into five groups: untreated group, 5-fluorouracil (5-FU)-treated group and high-, medium-, and low-dose JLSG-treated groups. Corresponding antitumor drugs were administered in each group except the untreated group. The antitumor effects in vivo were evaluated. Cell cycle distribution and apoptosis of MKN-45 human gastric cancer cells were determined by using flow cytometry (FCM) and Annexin V-fluorescein isothiocyanate/propidium iodide (Annexin V-FITC/PI) staining assay. The ultrastructure of MKN-45 gastric cancer cells was observed by transmission electron microscope. RESULTS: In the mice treated with high-, medium-, and low-dose JLSG, the growth inhibition rates of gastric cancer were 68.13%, 55.94% and 50.31% respectively, and this antitumor effect was dose-dependent. In the mice treated with intraperitoneal injection of 5-FU, the growth inhibition rate of gastric cancer was 53.43% and not much different from those treated with JLSG. The apoptotic rates in the high-, medium-, and low-dose JLSG-treated groups were 22.81%, 28.27% and 38.54% respectively, in a dose-dependent manner, with the cell cycle arrested at G(0)/G(1) phase. An Annexin V-FITC/PI staining assay revealed that the percentages of early apoptotic cells in the three dose JLSG-treated groups were all significantly higher than that in the 5-FU-treated group, whereas the late apoptotic and necrotic cells were much more in the 5-FU-treated group than those in the three dose JLSG-treated groups. CONCLUSION: Jinlongshe Granules exert an inhibiting effect on MKN-45 human gastric cancer cell.
7.Effects of Rhizoma kaempferiae volatile oil on tumor growth and cell cycle of MKN-45 human gastric cancer cells orthotopically transplanted in nude mice

OBJECTIVE: To evaluate the effects of Rhizoma kaempferiae volatile oil on tumor growth and cell cycle of MKN-45 human gastric cancer cells orthotopically transplanted in nude mice. METHODS: One hundred and five nude mice orthotopically transplanted with MKN-45 human gastric cancer cells were randomly divided into seven groups: untreated group, normal saline-treated group, dissolvant-treated group, cyclophosphamide (CTX)-treated group and high-, medium-, and low-dose Rhizoma kaempferiae volatile oil-treated groups. Corresponding interventions were implemented in each group except the untreated group. The antitumor effects in vivo were evaluated. Cell cycle distribution and apoptosis of MKN-45 human gastric cancer cells were determined by using flow cytometry (FCM). The ultrastructure of MKN-45 gastric cancer cells was observed by a transmission electron microscope. RESULTS: In the high-, medium-, and low-dose Rhizoma kaempferiae volatile oil-treated groups, the growth inhibition rates of gastric cancer were 57.2%, 28.0% and 5.0% respectively, and the gastric cancer cells were arrested at G(0)/G(1) phase. This antitumor effect was dose-dependent. The apoptotic cells occurred more frequently in the high-dose Rhizoma kaempferiae volatile oil-treated group and the CTX-treated group than those in the medium- and low-dose Rhizoma kaempferiae volatile oil-treated groups. CONCLUSION: The Rhizoma kaempferiae volatile oil is an effective composition for growth inhibition of gastric cancer, and its mechanism may be related to regulating the cell cycle and inducing apoptosis.
8.Role of JLP on the epithelial to mesenchymal transition in renal tubular epithelial cells

Objective To observe the effect of JLP on transdifferentiation of human renal proximal tubular epithelial cells (HK-2),and to investigate the role of p38 MAPK signaling pathway in this process.Methods The knock-down plasmids of JLP were constructed.HK-2 cells were randomly divided into four groups:negative control cells (Ctrl-shRNA group),knock-down jlp cells (jlpshRNA group),negative control cells with FGF-2 treatment (FGF-2 group) and knock-down jlp cells with FGF-2 treatment(jlp-shRNA +FGF-2 group).The expressions of JLP,E-cadherin,TGF-β1,α-SMA,p-p38 MAPK protein were detected by Western blotting.After the induction of FGF-2 for 24 hours,the expressions of α-SMA,COL-Ⅰ,FN were detected by immunocytochemistry.Results Compared with Ctrl-shRNA group,the expression of JLP protein was significantly down-regulated in FGF-2 group.Compared with FGF-2 group,the expressions of TGF-β1,α-SMA,p-p38 MAPK protein were significantly up-regulated,while E-cadherin protein was significantly down-regulated (P < 0.05).Compared with FGF-2 group,the expressions of α-SMA,COL-Ⅰ,FN immunostaining increased markedly in jlp-shRNA+FGF-2 group.Conclusion Scaffolding protein JLP is critical in preventing EMT in the course of fibrosis through the inhibition of p-p38 activation in HK-2 cells.
9.PROTECTIVE EFFECT OF VITAMIN E ON LIVER AGAINST ACUTE DAMAGE INDUCED BY CCI_4 IN RATS: A Histochemical Study

The protective effect of vitamin E and the change in activity of enzymes at hepatic injury by carbon tetrachloride(CCl_4) were studied with histochcmical methods. 60 rats were devided into three groups(A,B,C). The group A was untreated control; the animals of group B and C were injected(i. m.) with 0.9% NaCl 1 ml/100g or vitamin E 1 ml/100g body weight qd for 7 days respectively and then treated with 30% CCl_4 0.05ml/l00g by adding to the solution of NaCl or vitamin E from 8th to 14th days. On the 15th day, rats were sacrificed for histochemical study of the liver parenchyma. The results showed that the activity of SDH, G-6-Pase, ATPase, 5-NT or AcP in liver lobules of groups B and C animals were changed conspicuously after treatment with CCl_4. The quantitative histochemistry of SDH and G-6-Pase was also consistent with these results. The activity of SDH, G-6-Pase, ATPase and 5-NT were inhibited, but of AcP was elevated. As compared with those in the rats of group A, the changes however in group B appeared more obviously than those in group C. The results indicated that CCl_4 of the given dose induced acute damage in liver cells, while vitamin E had important protective effect against acute liver damag induced by CCl_4.
10.Protective Effect of Heat Shock Protein 27 on Cardiomyocytes when Ischemic Preconditioning Performed in Rat

ObjectiveTo investigate the mechanism and the protective effect of heat shock protein 27 (HSP27) on rat cardiomyocytes when ischemic preconditioning performed.MethodsCultured rat cardiomyocytes were divided into four groups: control group, ischemic group,ischemic preconditioning group and cyclohexamide group. Cell viabilities were analyzed by MTT. The apoptosis was evaluated with DNA ladder and flow cytometry Annexin V Flous staining. Western Blot was used to determine the expression of HSP27 and caspase-3 in cardiomyocytes.ResultsIschemic preconditioning could improve cell viability. The apoptosis ratio in ischemic preconditioning group was significantly less than that in ischemic group. These were accompanied by an increase in the expression of HSP27 and a decrease in caspase-3. The expression of the increased HSP27 and the protective effect induced by ischemic preconditioning were completely abolished by the presence of cycloheximide, a translation inhibitor.ConclusionThe expression of HSP27 induced by ischemic preconditioning plays an important role in protecting cardiomyocytes, and the mechanism is possibly related to the inhibition of cell apoptosis.
