1.Effects of injection needle scalpel combined with Tongbi tincture hot compress on the erythrocyte sedimentation rate, CRP and IL-1 in knee osteoarthritis

Objective To observe the effect of injection needle scalpel combined with Tongbi tincture heat deposited on the erythrocyte sedimentation rate, CRP and IL-1 in knee osteoarthritis.Methods 96 cases of knee osteoarthrosis according to the treatment divided into the control group(n=48) and experimental group(n=48), the control group was treated with injection needle scalpel, the experimental group on the basis of injection needle scalpel combination of tongbi tincture heat deposited, all lasted for 14 days.The clinical efficacy, CRP, IL-1, blood sink, knee joint functional impairment score, and safety was observed in the two groups.Results The total effective rate of the experimental group was 93.75%,higher than the control group 79.17%(P<0.05).After treatment, the CRP, IL-1, blood sedimentation of experimental group was lower than the control group[(7.09±0.97)mg/L,(6.54±0.82) ng/L,(16.47±2.03) mm/h vs.(10.35±1.28) mg/L,(7.21±0.93) ng/L,(18.25±2.21) mm/h](P<0.05).The scores of knee joint function impairment in the experimental group was higher than that in the control group(P<0.05).There was no significant difference in safety between the two groups.Conclusion The therapeutic effect of injection needle scalpel combined with Tongbi tincture heat deposited in treatment knee osteoarthrosis is better than injection needle scalpel.It can promote the absorption of inflammation, relieve knee function.
2.Effects of MAPK on a human inflammatory breast carcinoma cell line

Purpose:To invastigate the changes of the related signaling pathways and it's mechanism of reguation of mitogen activated protein kinase cascade(MAPK) by SUM149 cells induced plasmid of dominant-negative E-cadherin mutant H-2k~d-E-cadherin.Methods:Using western blot method,we examined the signal pathway of mitogen activated protein kinase(MAPK) between the dominant-negative mutant E-cadherin transfected SUM149 cells and controls.Results:Compared with the controls,the phosphorylated extracellular signal regulated kinase P44/42 expresion of the SUM149 cells which expressed higher dominant-negative mutant E-cadherin was significantly down-regulated.Conclusions:In this cell line model,the down-regulation of P44/42 may involve the regulation of MMPs.
3.Stump anorectal longitudinal incision and colorectal heart-shape anastomosis for the treatment of Hirschsprung′s disease

Objective[WT5”BZ] To evaluate the effect of stump anorectal longitudinal incision and colorectal heart shape anastomosis for the treatment of Hirschsprung′s disease (HD). Methods Ninety eight children with HD underwent colorectal resection and GI reconstruction by our method from April 1991 to April 2000. Results All patients were followed up for 5~96 months. There were no postoprative infection in pelvic and celiac cavities or wound infection,nor anastomotic strictures. One patient had an anastomotic fistula that was treated successfully by early repair. Recurrent constipation in 3 cases was successfully treated by dilatation. One patient experienced occasional fecal seepage. Enterocolitis occured in six patients. The overall complication rate was 11 2%.[WT5”HZ] Conclusion[WT5”BZ] Anorectal longitudinal incision and heart shape colorectal anastomosis has the advantage of satisfactory clinical result and less complication in the treatment of HD.
4.Antimicrobial use in 33 township central hospitals

Objective To investigate antimicrobial use in 33 township central hospitals,and improve the rational use of antimicrobial agents. Methods Thirty-three township central hospitals in 1 1 counties were randomly select-ed,7 920 outpatient prescriptions,medical records of 465 non-surgery patients and 213 surgery patients were inves-tigated and analyzed. Results Of 33 hospitals,antimicrobial usage rate in outpatients and inpatients was 56.60%and 89.68% respectively,combined antimicrobial usage rate was 24.16% and 43.58% respectively. Antimicrobial use density in inpatients was 147.25DDDs. Antimicrobial usage rate in surgery patients was 97.18% ,combined an-timicrobial usage rate was 59.90% ,the percentage of one drug,two-drug combination and three-drug combination was 40.10% ,47.82% ,and 12.08% respectively. The percentage of antimicrobial use in patients of type Ⅰ,Ⅱ, and Ⅲincision was 97.56% (40/41),96.93% (158/163)and 100.00% (9/9)respectively,combined antimicrobial us-age rate was 30.00% ,67.72% ,and 44.44% respectively.Conclusion The overuse of antimicrobial agents exists in 33 township central hospitals,antimicrobial usage rate,combined usage rate,antimicrobial use density and antimi-crobial prophylaxis in typeⅠincision operations are all high.
5.Effects of antioxidant and NF-κB on the induction of iNOS gene in rat pulmonary microvascular endothelial cells in vitro

AIM:To investigate the role of NF-κB in the activation of inducible nitric oxide synthase (iNOS) gene by tumor necrosis factor alpha (TNF α) and lipopolysaccharide (LPS) in endothelial cells and effect of antioxidant on the induction of iNOS. METHODS:Rat pulmonary microvascular endothelial cell (RPMEC) was cultured and the cells were identified with antiendothelial cell antibody CD31 using immunohistochemistry(ABC). The concentration of nitrite in the culture media was determined based on Griess reaction. iNOS mRNA was analyzed using RT-PCR and Northern blot. NF-κB in cell nuclei was detected with electrophoresis mobility shift assay (EMSA). RESULTS:A marked production of nitrite in RPMECs was found after 24 hours treatment with TNF α(105 U/L) and LPS (1 mg/L) (P<0.01). The level of iNOS mRNA increased significantly after adding TNF α(105 U/L) and LPS (1 mg/L) to the cell media for 2 hours (P<0.05). Pretreatment with cycloheximide (CHX, 10 mg/L) or antioxidant, PDTC (0.1 mmol/L) or NAC (20 mmol/L) significantly decreased nitrite production and iNOS mRNA expression induced by TNF α(105 U/L) and LPS (1 mg/L) (P<0.05). Furthermore, there was a dose-effect relationship between PDTC/NAC and inhibitory effect. TNF α (105 U/L) and LPS (1 mg/L) triggered the activation and translocation of NF-κB. This effect was blocked by adding PDTC (0.1 mmol/L) or NAC (20 mmol/L) to the cell media for 1.5 hours.CONCLUSION:1.TNFα and LPS may induce iNOS gene expression at transcriptional or posttranscriptional level. The upregulation of iNOS depends on new protein synthesis. 2. The induction of iNOS gene expression by TNFα and LPS is dependent on the activation of NF-κB. 3. Antioxidants may inhibit the induction of iNOS gene through the inhibition of NF-κB activation.
6.Nursing of patients with primary open angle glaucoma underging implantation of EX-PRESS device

Objective To determine the nursing measures of patients with primary open angle glaucoma undergoing implantation of EX-PRESS device.Methods 12 glaucomatous patients with EX-PRESS miniature glaucoma device received appropriate hygienic-education preoperatively,intensive nursing services during and after surgery,as well as health instructions in the follow-up period.Results 12 patients finally recovered from the surgery.The main post-operative complications were low tension and shallow anterior chamber(2 eyes),obstruction of the device by iris (1 eye).The intraocular pressures of all these patients remained normal levels before discharge and the visual acuity were the same as those pre-operation.Conclusions The effective nursing services for patients with EX-PRESS implantation is beneficial to the rehabilitation by increasing the adherence to the follow-up schedule,contributing to better understanding the common sense of the disease and the choice of the surgery.
7.Study of aspirin on proliferation and apoptosis of human lung Adenocarcinoma cell lines SPCA-1 in vivo

AIM To study the effects of ASA on proliferation and apoptosis of human lung Adenocarcinoma cell lines SPCA-1 in vivo. METHODS Cytotoxicity assay was tested by MTT method.Cell cycle was analyzed by flow cytometry(FCM).The morphology of the treated cells was observed by wright exclusion,Hoechest/PI exclusion, electron microscope. Apoptosis landder was evaluated by agarose gel electrophoresis of DNA. RESULTS ASA inhibited SPCA-1 cell proliferation in a time-and dose-dependent fashion(1.0~12.5 mmol?L -1,24 h~72 h). ASA increased the number of cells in G 0/G 1 and G 2/M phases,degrased the population of S phases at on 24 h and incresed apoptosis cells number. CONCLUSION ASA may inhibit the proliferation of SPCA-1 cell lines through effects on cell cycle and apoptosis.
8.Effect of antioxidant and NF_?B on the induction of iNOS gene in rat pulmonary microvascular endothelial cells in vitro

AIM:To investigate the role of nuclear factor ?B (NF ?B) in the induction of iNOS gene by TNF? and LPS in endothelial cells and the effect of antioxidant on the induction of iNOS. METHODS:Nitrite was determined based on Griess reaction. iNOS mRNA was analyzed using Northern blot. NF ?B in the cell nucleole was detected with electrophoretic mobility shift assay.RESULTS:(1)NO production and iNOS mRNA expression induced by LPS and TNF? was blocked by pyrrolidine dithiocarbamate(PDTC) or N-acetylcysteine(NAC). (2)LPS and TNF? triggered the activation and translocation of NF ?B, and PDTC or NAC inhibited the activation of NF ?B induced by LPS and TNF?.CONCLUSIONS:(1)The induction of iNOS gene by TNF? and LPS is dependent on the activation of NF ?B.(2)Antioxidants may inhibit the induction of iNOS gene through the inhibition of NF ?B activation.

Objective: To study the mechanism and effect of Chinese medicine Yiqi Tongyang Huatan prescription(益气通阳化痰方,YQTYHT) on hyperlipidemia and atherosclerosis(AS).Methods: The rabbit′s model was established by feeding the food containing high cholesterol,and 32 rabbits were randomly divided into four groups(each n=8): normal group,model group,Huoxue Huayu prescription treatment group(活血化瘀方,HXHY group) and YQTYHT treatment group(YQTYHT group).Before the experiment,at the end of 6 th and 12 th weeks after the experiment,the following items were measured: blood lipids platelet aggregation rate,platelet adhesion rate,malondialdehyde(MDA),endothelin-1(ET-1),nitric oxide(NO),the ratio of the lipid plaque area and the thickness of the endothelial plaque.Results: After feeding the food containing high cholesterol,the blood lipids,platelet aggregation rate,platelet adhesion rate,MDA,ET1,the ratio of lipid plaque area and the thickness of plaque were elevated significantly,while serum NO3-/NO2-decreased significantly in model group.Compared with the model group,the blood lipids were lower in the two treatment groups,and the blood lipid regulating effect in YQTYHT group was significantly better than that in HXHY group.Platelet aggregation rate and platelet adhesion rate of YQTYHT group and HXHY group were lower than those in the model group,but the degree of lowering in YQTYHT group and HXHY group had no significant differences.The serum MDA of YQTYHT group and HXHY group was lower than that in model group,and the lowering effect in YQTYHT group was better than that in HXHY group.YQTYHT group and HXHY group had a significant increase on the level of serum NO3-/NO2-and a decrease on the content of plasma ET-1,furthermore,the effects in the former group were significantly better than those of the latter group.The area of atherosclerotic plaque coverage in aorta and the thickness of plaque of HXHY group were larger than those in YQTYHT group(both P
10.Evaluation of the teaching activity on the courses correlated with public health from the clinical medical students in Sun Yat-sen university

Objective To evaluate the teaching activity on public health courses from clinical medical students in our university in order to provide a scientific basis for improving the curriculum design and teaching reform. Methods The “Questionnaire on Teaching Evaluation in Public Health Courses”, including teaching attitude, teaching content, teaching methods and teaching effectiveness was designed, and a general investigation was conducted among the clinical medical students of five-year program (840 students) and eight-year program (278 students) in these three aspects to under-stand students' evaluation to the course, who had finished the public health courses, including Preven-tive Medicine, Medical Statistics and Epidemiology (hereinafter referred to as: statistics, epidemiology, prevention) in Sun Yat-sen University. Statistical analysis was made using SPSS 13.0 software. Data analysis methods contain descriptive analysis, T-test, ANOVA, LSD, SNK, hierarchical logistic regres-sion analysis, etc. Results The overall score of teaching evaluation is (4.04±0.60). Differences exist between the evaluation in the five-year medical students and the eight-year medical students.
