1.Effects of Haikangling Mixture,a Traditional Chinese Medicine on Immunological Function in Immunosuppressed Mice

OBJECTIVE:To explore the effects of Haikangling Mixture on immunological function in immunosuppressed mice induced by cyclophosphamide. METHODS: Mice were given Haikangling Mixture intragastrically for 14d, and the level of hemolysin was determined by sheep erythrocyte (SRBC) immunization to exploer the influence of Haikangling on humoral immunity in mice; splenic lymphocytes were measured by MTT to observe the influence on cell immunity; the clearance of macrophage were measured by carbon particle clearance method to explore the influence on non-specificity immunity. RESULTS: The level of hemolysin, the cell reproductive activity, splenic lymphocytes and the phagotrophy of mononuclear macrophage were improved obviously by Haikangling Mixture in immunosuppressed mice induced by cyclophosphamide. CONCLUSION: Haikangling Mixture can enhance the immunological function in immunosuppressed mice induced by cyclophosphamide.
2.Analysis on Status Quo and Tendency of Chinese Materia Medica Injection Used in Our Hospital during 2002~2004

OBJECTIVE:To evaluate the drug use status of Chinese materia medica injection(CMMI)in our hospital.METHODS:Chinese materia medica injection(CMMI)used in our hospital from2002to2004was analyzed statistically in terms of consumption sum,defined daily dose(DDDs)and the average daily consumption sum(DDDc).RESULTS:The consumption sum of CMMI in3years increased year by year with annual increase rates of29.7%and32.3%,respectively.The cardio-cerebral vascular CMMI steadily dominated the first place in terms of consumption sum and DDDs in3years with a steady increase year by year from a high starting point.The DDDc of antitumor CMMI ranked the toppest.As for the consumption sum for single variety,xuesaitiong steadily ranked the first place in3years.CONCLUSION:The fast increase of the consumption of CMMI shows that it has a great market demand.
3.Application of CAD/ CAM techniques in repair of defects and correction of deformity on the jaw bones

Objective To apply the techniques of CAD/CAM in the repair of defects and correction of deformity on the jaw bones so as to improve the accuracy and controllability of operations.Methods With CAD techniques,3D bone model was reconstructed based on CT images.Then,a repairing model was created with mirror operation on original model.The physical models of original model and repairing model were fabricated through rapid prototyping.Based on physical models,operation plan and model surgery with pre-forming plate could be designed.Results The designed operation plan and pre-forming plate techniques of CAD/CAM were successfully applied in eight patients,with satisfactory results.Conclusions With techniques of CAD/CAM,reconstructing and fabricating the original bone model and mirror repair model are very helpful for designing operation plan for maxillofacial surgery.It also can improve the surgery accuracy and save operation time obviously.
4.Study on the pharmacokinetics of magnesium isoglycyrrhizinate injections in Chinese healthy volunteers

Aim To investigate the pharmacokinetics of intravenous injection of magnesium isoglycyrrhizinate in single dose in Chinese healthy volunteers.Methods 9 healthy volunteers received magnesium isoglycyrrhizinate injections in single dose of 100,200,300 mg respectively.The concentrations of magnesium isoglycyrrhizinate in plasma at different time were assayed with HPLC-UV method.The pharmacokinetic parameters of magnesium isoglycyrrhizinate injections were calculated with program 3P87.Results It was found that the plasma concentration-time curves of the preparetion fitted two-compartment model.The main pharmacokinetic parameters were as follows: C_(max) were(28.79?3.54),(67.56?8.84) and(99.28?17.57) mg?L~(-1);T_(12?) were(1.72?0.27),(1.46?0.35) and(1.13?0.33) h;T_(12?)were(23.10?3.30),(23.95?4.72) and(24.25?4.12) h;V_d were(3.332?0.471),(2.921?0.382) and(2.921?0.622) L;CL were(0.209?0.041),(0.186?0.048) and(0.166?0.039) L?h~(-1);k_(10) were(0.063?0.012),(0.064?0.016) and(0.057?0.009) h~(-1);AUC_(0-72) were(448.68?75.06),(1015.29?225.14) and(1688.42?367.44) mg?h ?L~(-1).Conclusion The pharmacokinetics of the drug in the dosage range of 100~300 mg in human body approximately fit linear dynamic features.Compared with glycyrrhizic acid and other glycyrrhizinate salts,magnesium isoglycyrrhizinate is eliminated more slowly.This is beneficial to the treatment of chronic hepatitis.

6.Investigation on Tolerance of Magnesium Isoglycyrrhizinate Injection in Healthy Volunteers

OBJECTIVE:To evaluate the safety and tolerance of magnesium isoglycyrrhizinate(MgIG)injection in Chinese healthy volunteers,and provide safe and utility dosage scheme.METHODS:36healthy volunteers were randomly divided into4groups,each group administered respectively with single-dose intravenous infusion of100,200,300mg of MgIG and mul?ti-dose intravenous infusion of200mg of MgIG,qd for7consecutive days.Clinical symptoms,vital signs,electrocardiograms,routine blood and urine tests,as well as hepatic and renal function were observed before and after IV infusion of MgIG.RESULTS:In single-dose IV infusion of MgIG groups,the vital signs and laboratory indices did not change in the subjects immediately after or at1h,8h,24h after administration,as compared with before,while in multi-dose constant intravenous infusion group,the vital signs and laboratory indices at4days and8days after administration were in the normal range,as compared with before.CONCLUSION:Tolerance of healthy subjects to MgIG is good,and daily dosage of100mg to200mg is recommended for clinical use.
7.Determination of the Content of Stilbene Glycoside in Zhipingkang Misture by HPLC

OBJECTIVE:To establish an HPLC method for the determination of Stillbene glycoside (THPG) in Zhipingk-ang misture.METHODS:The chromatographic column was Hypersil C18(250 mm?4.6 mm,5 ?m).The mobile phase consisted of acetonitribl-water(25∶75) at a flow rate of 1.0 mL?min-1 with detection wavelength set at 320 nm. RESULTS: There was a good linear relationship within the concentration range of 0.5~8.0 ?g for THPG. The average recovery of THPG was 100.03%(RSD=1.07, n=6).CONCLUSION: The adoptd method is accurate, reproducible and suitable for the quality con-trol of Zhipingkang misture.

Objective To explore the correlation of meteorological parameters with the epidemic of acute Moraxella ca-tarrhalis respiratory infection in hospitalized children in Suzhou. Methods A total of 8143 children with acute respiratory infec-tion were participated in the trial during 2006 to 2010, and the secretions of nasopharynx were collected for bacterium culture. Moraxella catarrhalis was identified according to the routine technique of culture. Meteorological parameters including mean temperature, relative humidity, rainfall amount, duration of sunshine and mean wind velocity were collected monthly during the same period. The relationship between the epidemic of Moraxella catarrhalis and metrorological parameters were analyzed by seasonal decomposition method, the Spearman rank correlation and stepwise regression analysis. Results Moraxella catarrhalis was identified in 4.04% of 8 143 specimens. The prevalence of acute Moraxella catarrhalis respiratory infection was higher during winter and spring. The monthly infection rate of Moraxella catarrhalis was negatively correlated with mean temperature as well as duration of sunshine and wind velocity. Wind velocity was independent risk factor for Moraxella catarrhalis infection. Conclusions Moraxella catarrhalis is a primary pathogen in respiratory tract infection in children in Suzhou. The epidemic of Moraxella catarrhalis is closely related to meteorological parameters.

Objective To investigate the phenotype and the immunoregulatory function of CD8αα+TCRαβ+regulatory T cells in peripheral blood samples from mice.Methods The distribution profile and the phenotype of CD8αα+TCRαβ+regulatory T cells in C57BL/6 mice were detected by flow cytometry.The cytokines released by CD8αα+TCRαβ+regulatory T cells upon the stimulation with anti-CD3 antibody were analyzed by cytometric bead array.The in vitro immunosuppressive activity of CD8αα+TCRαβ+regulatory T cells on activated CD4+T cells was analyzed by using flow cytometry and carboxyfluorescein succinimidyl ester ( CFSE ) .An adoptive cell transfer assay was set up to evaluate the immunoprotective effects of CD8αα+TCRαβ+ regulatory T cells in a mouse model of experimental autoimmune encephalomyelitis ( EAE) .Results CD8αα+TCRαβ+regulatory T cells were detected in liver, spleen and peripheral blood samples collected from na?ve C57BL/6 mice.Compared with CD8αβ+TCRαβ+regulatory T cells, CD8αα+TCRαβ+regulatory T cells showed a memory-activated phenotype of CD25+CD122high CD44high CD62Llow CD69high NK1.1+DX5+.CD8αα+TCRαβ+regulatory T cells could produce IL-2 after 24 hours stimulation with anti-CD3 antibody, followed by producing IFN-γ, TNF-α, IL-4, IL-17A and traces of IL-6 and IL-10. In vitro, CD8αα+TCRαβ+regulatory T cells specifically suppressed the proliferation of activated CD4+T cells ( P<0.01 ).Moreover, they could delay the onset of EAE in mice and reduce clinical score (P<0.01).Conclusion CD8αα+TCRαβ+regulatory T cells were a unique population with immunoregula-tory function, which could be used as a potential therapeutic target in the treatment of autoimmune disease.
