1.Expression of C5aR in renal tissue of rats with unilateral ureteral obstruction

Objective To observe the effect of unilateral ureter obstruction(UUO) on the expression of complement 5a receptor(C5aR) in rat renal tissue,and to investigate the roles played by C5aR in the development of renal intersititial fibrosis.Methods Thirty six male Sprague Dawley rats were randomly assigned into two groups(18 each): sham-operated group and UUO group.The left ureter of the rats in UUO group was obstructed,while in sham-operated group the left ureter was not.Six rats in each group were executed respectively on the 5th,10th and 15th day after surgery.Histological changes in renal tubular interstitium were observed with H.E.and Masson staining,and the expression of C5aR in renal tissues was detected by iminunohistochemical ELiVisionTM plus at each time point.Results Renal tubular dilatation and partial interstitial cells degeneration were found in UUO group on the 5th day after surgery.Necrosis of the interstitial cells was found in renal tubule,and the interstitial fibrosis was found on the 10th day.On the 15th day after surgery diffuse thickening and shrinkage of renal tubular basement membrane and obvious fibrosis were found(P0.05).Conclusion The C5aR expression in renal tissue increases significantly at the early stage of UUO,while the process is slowed down in the later stage.C5aR may take part in the process of renal intersititial fibrosis at the early stage.
2.Meta-analysis on Serological Effect of the Primary Vaccination for Japanese Encephalitis of the Literatures

Objectives To summary and compare the different seroconversion rates after the primary vaccination for the Japanese encephalitis (JE),and to evaluate the serological effect of 3 kinds of JE vaccines.Method Searching "CHKD","Wanfang" database and "EMCC" databases,the studies of the immunogenicity after the primary JEV vaccination,all randomized controlled trials or non-randomized controlled trials were included,and statistical analysis were made by RevMan 4.2.10 software.Results A total of 12 literatures were included,7 studies had control groups.The seroconversion rates after the primary vaccination,JEV-L,JEV-I (Vero) and JEV-I(PHK),were 86% (95% CI:80% ~ 91%),83% (95% CI:72% ~ 94%) and 64% (95% CI:58% ~ 69%) respectively.Comparing the seroconversion rates of the 3 kinds of vaccines after primary immunization,the rate of JEV-I (Vero) was significantly higher than the rate of JEVI(PHK),other comparisons were no significant difference.Conclusion The serological effects of JEV-L and JEV-I (Vero) after the primary vaccination were higher than that of JEV-I (PHK).
3.THE INHIBITION EFFECT OF ANTI-CD81 ON THE PROLIFERATION OF ASTROCYTES

Objective To investigate the effect of anti-CD81(antibodys against CD81) on the proliferation of astrocytes. Methods Purified astrocytes from newborn rats' cerebral cortex were divided into 6 groups and added with anti-CD81 different concentrations(0,0.1,0.5,1,5,10?mg/L).The activity of astrocytes was tested by methyl thiazolyl terazolium(MTT).Three significative groups were chosen based on MTT result and added with anti-CD81 of different concentrations(0,0.5,5mg/L).After administration for 24 hours,the cell cycle of the astrocytes was measured by flow cytometer.The corresponding data were analyzed with SPSS statistical software. Results 1.By MTT,the average optical density(AOD) values of astrocytes were reduced after administration with anti-CD81 of different concentrations for 24 hours,that is,the number of astrocytes was reduced,which indicated anti-CD81 inhibited the proliferation of astrocytes and the effect showed a dose-dependent pattern.2.By cell cycle analysis,a progressive dose-dependent decrease was found in the index of cells in G-0/G-1 phase and an increase in S phase.Such as,the index of cells in G-0/G-1 phase,was 82.73 in 0,is 82.16 in 0.5?mg/L,was 78.58 in 5?mg/L.Conclusion Anti-CD81 inhibits the proliferation of astrocytes and the number of astrocytes is reduced.Further more,the index of cells decreases in G-0/G-1 phase and increases in phase S after administration with anti-CD81.This study shows that anti-CD81 doesn't restrain the cells from G-1 phase to S phase but the cells are arrested in S phase.
4.Determination of Chlorogenic Acid in Qingyan Granules by RP-HPLC

OBJECTIVE:To establish a RP-HPLC method for the determination of chlorogenic acid in Qingyan granules.METHODS:Waters Breeze System was used with a Hypersil BDS C18 column(5?m,4.6mm?150mm).The mobile phase was composed of methanol and 0.4%(V/V)H3PO4 water solution(20∶80,V/V),flow rate:1.0ml/min;wavelength of UV detector:327nm,column temperature 20℃.RESULTS:The calibration curve of chlorogenic acid was linear within the range of (174~1 392)ng(r=0.9 999).The average recovery was 100.10%,RSD=1.60%(n=5).CONCLUSION:This method is simple,sensitive,as well as reliable,and is available to quality control.
5.Trans-gastric peritoneoscopy with technique of natural-orifice transluminal endoscopic surgery for diagnosis of tuberculosis peritonitis: a report of 20 cases

Objective To investigate the diagnostic value of trans-gastric peritoneoscopy with technique of natural orifice transluminal endoscopic surgery(NOTES)for tuberculosis peritonitis.Methods Clinical data of 20 patients with tuberculosis peritonitis diagnosed by trans-gastric peritoneoscopy via NOTES were retrospectively analyzed.Results All diagnoses were confirmed by biopsy.The findings of peritoneoscopy were defined as miliary type with miliary nodes scattered in ascites and on peritoneum,adhesive type with thickening of peritoneum and adhesion between peritoneum and intestines,cheese-like type with parietal peritoneal ulcer and cheese-like substances,and mixed type with 2 or 3 of above mentioned types.Positive findings in other laboratory examinations were hemoglobin decrease in 10(50%)patients,blood sedimentation rate increase in 16(80%),C reactive protein increase in 13(65%),CA125 increase in 18(90%),and positive tuberculin test in 9(45%).Abnormal findings were detected by chest X-ray in 8(40%)patients,by abdominal ultrasonography examination in 2(10%),by abdominal CT in 7(35%),and by colonoscopy in 1(5%).No abnormal results were found in all patients in anti-tuberculosis antibody test,ascites bacteria culture and gastroscopy.Conclusion Trans-gastric peritoneoscopy via NOTES with biopsy is effective for diagnosis of tuberculosis peritonitis.
6.Trausgastric peritoneal endoscopy in diagnosis of ascites with unknown origin

Objective To investigate the diagnostic value of transgastric peritoneal endoscopy in diagnosis of ascites with unknown origin.Methods Endoscopy was introduced into peritoneal cavity through gastric wall in 23 patients with exudative ascites which was able to be diagnosed by routine methods and biopsy was made through endoscopy to get pathological diagnosis.Results Definite diagnosis was made in 22 patient (95.7%),of which 12 (54.6%) were malignant tumors,8 (36.4%) were tuberculosis peritonitis,1 (4.5%) was spontaneous peritonitis associated with liver cirrhosis and 1 (4.5%) was eosinophilic enteritis.Conclusion Natural orifice transluminal endoscopy combined with biopsy is an effective and accurate procedure for diagnosis of ascites of unknown canses.

8.Nucleotide sequencing of E/NS1 gene segment of dengue type 1 viruses isolated in Guangdong Province.

OBJECTIVETo analyse the genetic relationship of local strains of dengue type 1 viruses isolated in different years and regions from Guangdong Province, and to explore the genetic links with strains of adjacent countries by comparing with the sequences of relevant strains in Genbank.
METHODSThe viral RNAs were extracted and used for one-step reverse transcriptase polymerase chain reaction (RT-PCR) to amplify the partial nucleotide fragments in E/NS1 gene junction which were then cloned into the plasmid pBluescript II SK for sequencing, the results were analysed by DNASTAR software.
RESULTSThe phylogenetic tree of the sequenced 14 strains of dengue type 1 viruses branches into two genotypic groups. The nucleotide sequences showed a maximal homologies of 99.2% with Indonesia strains, 100% with Philippines strains and 98.8% with Thailand strains.
CONCLUSIONSThe dengue type 1 viruses of Guangdong Province are closely related to the Philippines, Indonesia and Thailand strains, which may indicate the possibility of importation from those countries.

9.Analysis on the epidemiologic features of Dengue fever in Guangdong province, 1990-2000.

OBJECTIVETo determine the epidemiological characteristics and risk factors of Dengue fever in Guangdong province in 1990-2000, and to develop the strategy for its prevention and control.
METHODSSequential of the partial nucleotide fragments in E/NS1 gene junction was analysed.
RESULTSA total of 9,747 cases were reported with three deaths in Guangdong province, 1990-2000. The average incidence was 1.27 per 10,000 (range: 0-9.75 per 10,000). Outbreaks were commonly seen, with 13 cities (61.9%) involved, but most of the cases occurred in Guangzhou, Chaozhou, Zhaoqing and Foshan cities. Numbers of cases reported monthly were gathered with sporadic imported cases reported from January to June while the period of epidemics was from July to December. Sex ratio of patients was 1.04, but all age group was susceptible. Four sero-types of Dengue virus were found during these epidemics. The partial nucleotide fragments in E/NS1 gene junction of 12 isolated viruses were sequenced and 2 sub-gene types were identified in Guangdong province. The clinical symptoms seemed typical. Social and natural factors were responsible for the epidemic of Dengue fever in the regions.
CONCLUSIONThe epidemic of Dengue fever in Guangdong was closely related to the situations of neighboring countries, indicating the possibility of importation of virus from these countries. The epidemics usually started with imported cases. However, the endemic nature of the disease had not been identified yet.

10.Investigation on acute nitrite poisoning in Yangjiang city, Guangdong province, China.

OBJECTIVETo determine the cause of acute poisoning occurred in a factory in Yangjiang city, Guangdong province.
METHODSIn a cross-sectional study, interviews were conducted with the administrators of the factory and the local physician. A review was conducted on the water system used for industrial purposes and a separate system used by workers for drinking water. Treatment and discharge of industrial waste water were examined. Face-to-face interview was conducted to identify risk of exposure for illness among workers.
RESULTSA total number of 36 cases were identified in the plant and the attack rate was 16.4% (36/220). The incubation period (time between drinking polluted water and the onset of symptoms) had a median of 90 minutes (range: 30 - 230 minutes). Consuming water at the factory increased the attack rate and a dose-response effect was identified (chi(2)(trend) = 79.115, P < 0.01). The nitrite content of residuals in drinking water exceeded the WHO standard (1 ppm).
CONCLUSIONSThe accident of acute poisoning was due to drinking water contaminated with sodium nitrite. The prevention of drinking water contaminated by toxic chemicals like sodium nitrite, and the design of industrial and potable water supply system need to be carefully reviewed. Regulations should be developed and enforced to minimize the impact of industrial waste water discharges to guarantee the access to clean drinking water.
