Objective To investigate the role of nuclear factor-KB( NF-KB) in the expression of monocyte chemoattractant protein-i (MCP-1) in human mesangial cells induced by oxidized low density lipoprotein(Ox-LDL) . Methods Human mesangial cells were used as target cells. Levels of inhibitory KBK (IKB?) and MCP-1 protein were measured by ELISA. Activity of transcriptional factors NF-KB was determined by electrophoresis mobility shift assay ( EMSA) . Immunohistochemistry was used to detect the translocation of p65. Results Ox-LDL increased the DNA-binding activition of NF-KB and decreased the expression of IKB?. 2.The value of Duke treadmill score in coronary heart disease

Objective To discuss the value of Duke treadmill score in the diagnosis of coronary heart disease.Methods 94 inpatients underwent treadmill test and coronary angiography,the relationship between the Duke treadmill score and the results of coronary angiography was analyzed.Results The prognostic accuracy and positive predictive value of Duke treadmill score to male and female were 88.9%,85.0% ;those of traditional electrocardiogram ST standard were 89.6%,82.5 %,the differences were significant (x2 =4.27,P < 0.05).Conclusion The Duke treadmill score is much more significant than the traditional ST standard in the prognostic accuracy and positive predictive value of diagnosing coronary heart disease.

4.Study the effects of losartan on brain natriuretic peptide and C-reactive protein in diabetic cardiomyopathy patients

Objective To explore the effects oflosartan on brain natriuretic peptide(BNP)and C-reactive protein(CLRP)in diabetic cardiomyopathy patients. Methods Sixty diabetics were divided into diabetic control group(30 cases)and diabetic cardiomyopathy group(30 cases), and the levels of BNP and CRP in plasma were determined. Then, diabetic cardiomyopathy group were divided into conventional treatment group(15 cases)and losartan treatment group(15 cases)by random digits table to take corresponding treatment for 12 weeks. The changes of cardiac function, the levels of BNP and CRP in plasma were detected and compared. Results The levels of BNP and CRP in diabetic cardiomyopathy group [(331.27 ±65.64)ng/L,(26.10±10.13)mg/L]were significantly higher than those in diabetic control group[(76.09±39.14)ng/L,(7.03±2.71)mg/L](P<0.01). After treatment 12 weeks, the levels of BNP and CRP were significantly lower, left ventricular ejection fraction was significantly higher and left ventricular end-diastolic diameter was significantly lower in losartan treatment group than those in conventional treatment group(P<0.05).Conclusions The increasing levels of BNP and CRP can reflect clinical severity of diabetic cardiomyopathy.Losartan can decrease the levels of BNP and CRP, and improve heart function, and it has better effective on diabetic cardiomyopathy.
5.Influence of Baduanjin on mild cognitive impairment in elderly diabetic patients

Objective To explore the influence of Chinese Qigong--Baduanjin on mild cognitive impairment (MCI) in elderly diabetic patients.Methods A total of 78 elderly type 2 diabetic patients complicated with MCI were divided into intervention group and control group by random digits table method.The control group with 41 patients was received routine treatment and discharged guidance after discharging from hospital.The intervention group with 37 patients was received Baduanjin exercise for 12 months based on the routine treatment in control group.Montreal Cognitive Assessment (MoCA) and Activity of Daily Living (ADL) scores in two groups were compared before intervention and 1,3,6,9 and 12 months after intervention.Results In intervention group,MoCA scores were significantly increased at different times after intervention compared with those before intervention,F=72.782,the differences were statistically significant,P<0.05.At the same time,MoCA scores were all significantly higher in intervention group than those in control group,F=2.231,2.972,4.362,5.085,5.373,the differences were statistically significant,P< 0.05.In intervention group,ADL scores had significantly improved since 6 months after intervention compared with those before intervention,F=93.126,the differences were statistically significant,P<0.05,and compared with those in control group,the scores were significantly higher since 6 months after intervention,F=3.853,4.561,7.162,the differences were statistically significant,P<0.05.Conclusion Chinese Qigong--Baduanjin in treatment of elderly diabetic patients complicated with MCI can delay the MCI progress and improve ADL of the patients.
6.Effect of Cluster Needling of Scalp Acupuncture Combined with Rehabilitation on Depression after Stroke following Motor Aphasia

ObjectiveTo explore the effect of cluster needling of scalp acupuncture combined with rehabilitation(Tang's Approach) on depression of stroke patients with motor aphasia.Methods70 stroke patients were randomly divided into the acupuncture with rehabilitation group and the control group. The cluster needle of scalp point with speech rehabilitation training was used in the acupuncture with rehabilitation group, while speech rehabilitation training was used only in the control group. The degree of depression and aphasia were observed in the two groups 4 weeks after treament.Results 4 weeks after the treatment, the classification of aphasia was getting better in the acupuncture with rehabilitation group than in the control group(P<0.05), and the incidence of depression decreased significantly in the acupuncture with rehabilitation group compared with the control group(P<0.01). The score of Stroke Aphasic Depression Scale (SADQ) decreased significantly in the acupuncture with rehabilitation group compared with the control group (P<0.01).ConclusionTang's Approach may improve the depression of stroke patients with motor aphasia.
7.Stability Study of Tramadol,Fentanyl and Ondansetron Combined with 0.9% Sodium Chloride Injection in Analgesia Pump

OBJECTIVE:To study the stability of Tramadol hydrochloride,Fentanyl citrate and Ondansetron hydrochloride in-jection in 0.9% Sodium chloride injection. METHODS:At room temperature [(25±1)℃],three injections were poured into infu-sion bag of disposable analgesia pump,and diluted with 0.9% Sodium chloride injection. The changes in precipitation,turbidity, color and pH value of the mixture were observed. The relative percentage of three injections within 72 h were determined by HPLC method. RESULTS:There was no significant change in the pH value and appearance of the mixture. Relative percentage of tramad-ol hydrochloride,fentanyl citrate and ondansetron hydrochloride were all higher than 97% within 72 h. CONCLUSIONS:The mix-ture of Tramadol hydrochloride,Fentanyl citrate and Ondansetron hydrochloride injection in 0.9% Sodium chloride injection keep stable within 72 h under room temperature.

OBJECTIVE:To study the current situation and developing trend of the antidiabetics used in Guangdong re?gion.METHODS:The antidiabetics used in44hospitals in Guangdong region during the period2000~2003were analysed concerning the kinds of drug,sum of money for consumption and frequency of drug use.RESULTS:The sum of money for consumption of antidiabetics increased year by year so did its proportion in sum of money for consumption of total drug.Among them,consumption of insulin increased very quickly and oral antidiabetics assumed a tendency of increase as well.CONCLUSI_ ON:The variety of insulin is stable and new oral antidiabetics is going into market occasionally in this region,but frequency of use is primary to sulfonylurea,biguanides and glycosidase-inhibitors.

10.Expresson of the N-terminus truncated phosphotase D in Escherichia coli and characterization of its anti inflammatory activity

To investigate the immunological activities of the recombinant human phosphatase D2 (rhPLD2) in vitro and in vivo, especially its ability to reduce inflammatory reactions, the cDNA fragment encoding rhPLD2 was cloned into prokaryotic expression vector pET30a by RT-PCR and the recombinant protein rhPLD2 expressed in E.coli was purified from the inclusion bodies, while the anti inflammatory activity of rhPLD2 was determined by the amount of eosinophils in bronchoalveolar fluid(BALF) and blood and the expression of IL-5 and MMP-9 in lung tissues of guinea pig model of chronic asthma. It was found that the rhPLD2 recombinant protein was obtained from human Daudi cells by cloning to E.coli, which contained no membrane-binding site and signal peptide. The cDNA sequence encoded 631 amino acid residues (GenBank Accession Number: AY178289). The purity of the rhPLD2 approached up to 76% with a bioactivity of 50.9745 units/L (0.9212 g/L). In addition, the anti inflammatory effect of rhPLD2 protein could be demonstrated in the guinea pig model of chronic asthma after treatment with rhPLD2 protein, such as down regulation in the expression of the inflammatory cytokine IL-5. It is concluded that the anti-inflammator activity of the recombinant human truncated PLD2 protein produced from the E.coli plasmid can be demonstrated both in vitro and in vivo.
