Objective To explore the effects oflosartan on brain natriuretic peptide(BNP)and C-reactive protein(CLRP)in diabetic cardiomyopathy patients. Methods Sixty diabetics were divided into diabetic control group(30 cases)and diabetic cardiomyopathy group(30 cases), and the levels of BNP and CRP in plasma were determined. Then, diabetic cardiomyopathy group were divided into conventional treatment group(15 cases)and losartan treatment group(15 cases)by random digits table to take corresponding treatment for 12 weeks. The changes of cardiac function, the levels of BNP and CRP in plasma were detected and compared. Results The levels of BNP and CRP in diabetic cardiomyopathy group [(331.27 ±65.64)ng/L,(26.10±10.13)mg/L]were significantly higher than those in diabetic control group[(76.09±39.14)ng/L,(7.03±2.71)mg/L](P<0.01). After treatment 12 weeks, the levels of BNP and CRP were significantly lower, left ventricular ejection fraction was significantly higher and left ventricular end-diastolic diameter was significantly lower in losartan treatment group than those in conventional treatment group(P<0.05).Conclusions The increasing levels of BNP and CRP can reflect clinical severity of diabetic cardiomyopathy.Losartan can decrease the levels of BNP and CRP, and improve heart function, and it has better effective on diabetic cardiomyopathy.

Objective To investigate the influence of VEGF expression on the apoptosis of pancreas subjected to ischemia/reperfu-sion injury. Methods Thirty male SD rats were randomly divided into three groups (rt = 10). Group A was served as sham-operation group. Groups B were subjected to 30 min of ischemia by clamping of celiac artery and superior mesenterie artery then releasing for 6 hours to produce ischemia/reperfusion injury model. Groups C were treated with VEGF antisense oligodeexynueleotide after isehemia. Rats were sacrificed, the pancreas was obtained to detect the VEGF expression by immunohistochemical method, and apoptosis was determined by TUNEL staining. Results Apoptosis appeared and VEGF expression up regulated in pancreas after Isehemia/reperfusion injury. Comparison between Groups C and Groups B showed a significant down regulation of VEGF expression (P <0. 05) and a notable increase of apoptotic index (P < 0. 05). Conclusion VEGF expression suppresses apeptosis of pancreas during the course of ischemia/reperfusion injury and may play an important role in protection of pancreas against the ischemia/repeffusion injury.

Objective To screen stage\|specific expression genes of plerocercoid of Spirometra erinacei\|europaei. Methods RNA was extracted by the acid guanidinium thiocyanate\|phenol\|chloroform from plerocercoid and adult worm of Spirometra erinacei\|europaei. Contaminated DNA in the RNA was digested by RNase\|free DNase. cDNA was synthesized by using T\-\{12\}MA, T\-\{12\}MC, T\-\{12\}MG and T\-\{12\}MT primers, and PCR was then done using the same T\-\{12\}MN and one random primers. The PCR products were fractionated on 8% denatured polyacrylamide gel, differential bands of plerocercoid found in the gel were cut out, amplified by PCR and sequenced after the gel was dried out and autoradiographed. Northern hybridization was conducted to identify the stage\|specific expression genes. Results Eleven differential bands were selected from the gel and classified into 3 kinds of gene fragments by hybridization after they were amplified by PCR. The fragments 1 and 2 were confirmed to express specifically in plerocercoid by Northern hybridization, but the fragment 3 was expressed in both plerocercoid and adult worm. When the 3 gene fragments were homologically analyzed in GenBank, the sequence which was homologous with the fragments 1 and 2 was not found, but the fragment 3 had high homology with many kinds of 28S rRNA. Conclusion The gene expression of plerocercoid was different from that of adult worm probably because they live in different hosts. Two kinds of different gene fragments in plerocercoid were identified by mRNA differential display technique.

AIM:ToinvestigatetheeffectofIkarosisoformsontheproliferationofhumanovariancancerSK-OV3 cells.METHODS:Three isoforms of Ikaros, IK1, IK2 and IK6, were transfected into ovarian cancer SKOV3 cells. CCK-8 assay and cell counting were used to detect the effects of Ikaros isoforms on the proliferation of SKOV 3 cells. The cell cycle was analyzed by flow cytometry .The cell cycle-related proteins were detected by Western blot .RESULTS:IK1 and IK2 expression inhibited SKOV 3 cells proliferation .Flow cytometry analysis indicated that IK 1 and IK2 induced SK-OV3 cell cycle arrest at the G 1 phase.IK6 isoform exerted no obvious effect on the proliferation or cell cycle of SKOV 3 cells.Compared with control EV group , IK1 group and IK2 group showed a dramatic elevation in the expression of the cell cycle inhibitor p21, along with a substantial decrease in the expression of the cell cycle inducers cyclin D 1 and cyclin D2, which did not change in IK 6 group.CONCLUSION:IK1 and IK2 significantly inhibit the proliferation of ovarian cancer SKOV3 cells and induce cell cycle arrest at G 1 phase by regulation of cell cycle-related proteins cyclin D1, cyclin D2 and p21, while IK6 isoform exerts no obvious effect on the proliferation and cell cycle of SKOV 3 cells.
