1.Progress on community management models of hyperuricemia

Hyperuricemia is the second largest metabolic disease next to the diabetes and is an independent risk factor for other chronic diseases. With the increase of incidence rate, it has become one of the common chronic diseases in general practice. For the implementation of hierarchical medical system, it is necessary to establish a sound and effective community model for the management of hyperuricemia in China. This article reviews the literature at home and abroad to provide information for building a community management model of hyperuricemic patients.
2.Effect of environmental enrichment on depressive-like behaviors induced bv chronic stress in rats

ObjectiveTo observe the effects of enriched environment on depressive-like behaviors induced by chronic stress in rats.MethodsAdult rats were randomly divided into stress group and the control group,then after 21 days of stress,rats were raised in an enrich environment (EE) or standard environment (SE)for 28 days.Open field test,forced swimming test and sucrose preference test were used to detect depressive-like behavior in rats.Results ( 1 ) Open field test showed that grid crossing,standing and rearing in stress ( no gap)rats were significantly reduced ( 13.88 ±4.38 vs 45.00 ± 10.19,9.13 ±2.54 vs 16.38 ±4.11 and 4.78 ± 1.39vs 10.51 ± 2.52 ; n =8 ; P< 0.01 respectively) ; but grid crossing and standing in stress + EE rats were significantly increased than stress + SE rats (41.61 ± 10.53 vs 26.25 ± 6.18 and 16.79 ± 3.49 vs 11.25 ± 3.12 ; n =8 ; P<0.01 and P<0.05 respectively).(2) Sucrose preference tests showed that sucrose consumption and sucrose preference% in stress ( no gap) rats were significantly decreased than those in the control group ( 5.22 ± 0.94 vs 10.61 ±2.59 and (49.11 ±6.77)% vs(63.38 ±8.36)% ; n=8; P<0.01 respectively).(3) Forced swimming test showed that immobility time was increased in stress ( no gap) rats ( ( 19.5 ± 5.43 ) s vs ( 12.75 ± 3.9 ) s; n =8 ; P < 0.05 ) and was restored by EE compared to SE rats ( ( 10.25 ± 3.57) s vs ( 17.75 ± 5.45 ) s; n =8 ; P < 0.05).ConclusionChronic stress can reduce depressive-like behaviors,and EE can restore depressive-like behaviors induced by chronic stress.
3.Association between γ-Aminobutyric Acid B2 Receptor Gene Polymorphism rs3750344 and Bone Mass Content or Bone Mass Density in Chinese Hans Population

ObjectiveTo investigate the association of γ-aminobutyric acid B2 receptor (GABABR2) gene polymorphism rs3750344 and bone mass (bone mass content, BMC, and bone mass density, BMD) in population of Chinese Hans lives in Jinan. Methods425 volunteers were recruited. BMC and BMD of all subjects were measured with dual energy X-ray absorptiometry (DXA). Genotyping of polymorphism rs3750344 was conducted using Taqman assays. ResultsBMD and BMC increased with the copy of the C allele of rs3750344 in whole body. ConclusionThe C allele of GABABR2 rs3750344 increases the levers of BMD and BMC in Chinese Han population.
4.Study on the changes and clinical significance of the DNT cells and T lymphocyte subtype in children with infectious mononucleosis

Objective To study on the changes of the DNT cells and T lymphocyte subtype in children with infectious mononucleosis (IM) and its clinical significance.Methods The flow cytometry (FCM) was used to detected the DNT cells and other T lymphocyte subtype in 48 cases of IM.Results The study showed that DNT cells( 9.39 ± 4.89 )% were greatly increased in comparison with normal controls (NC) (4.26 ± 1.68)% ( P <0.01 ).CD4 cells(21.45 ±9.87)% were decreased ( P <0.01 ) and CD8 cells increased in comparison with NC (32.43 ± 5.07) % ( P < 0.01 ).Conclusion DNT cells and T lymphocyte subtype can be used to evaluate the immune function of children with infectious mononucleosis (IM) and provide guidance for adoptive immunotherapy.
5.The role of HPV type 16 E6 variant in persistent infection and recurrence of cervical HSILs after conization

Objective To explore the role of humall papillomavims (HPV) type 16 E6 variants in the persistent infection and recurrence of the cervical high-grade squamous intraepithelial lesion (HSIL) after conization. Motheds During May to June 2012, 100 HSIL patients with HPV16 positive but negative at the margin after Loop electrosurgical excision procedure (LEEP) were selected from Shanghai People′s Hospital of Pudong District. The gene sequencing was done to the E6. The patients were followed up for 2 years. The E6 gene sequencing was done again to detect the HPV16 variants among those with persistent infections and recurrence. Results Among the 100 cases, E6 variants were detected, 81% of them HPV E6 variants Asian prototype, 14% European variant; 2% European prototype (EP) and 3% African variant 1. LEEP for Asian prototype was susceptible to the clearance of HPV16 (P < 0.001). The European variants of T350G, and A442C were correlated with persistent infection and recurrence (P < 0.001). Conclusion The HPV16 variant is closely related to the HSIL incidence. HSIL caused by HPV16 in Pudong New Area of Shanghai belong to the Asian type variant. The European variants of E6 , T350G/A442C were likely related to the recurrence of HPV16.
6.Analysis of clinic pathological characteristics and prognosis between Uyghur and Han people with rectal cancer

Objective To study the clinic pathologic characteristics and prognosis of rectal cancer between Uyghur patients and Han patients .Methods Data of 196 cases of rectal cancer (58 Uyghur and 138 Han) in the first affiliated hospital of Xinjiang medi‐cal university from January 2008 to February 2010 was collected and analyzed .Then analyzed the differences in their clinical patho‐logical features and prognosis .Results There were significant differences in sex ,age ,blood type ,occupation ,treatment time ,the first symptom ,histological type and DUKES between two nationals (P<0 .05);while there was no significant difference in educa‐tion ,concomitant diseases ,CEA ,tumor location ,tumor size ,gross type ,degree of differentiation and postoperative chemotherapy be‐tween two nationals(P>0 .05);the 5‐year overall survival rate of the patients from Uyghur was shorter than that of the patients from Han (P<0 .05);according to the multivariate analysis ,ethnic served as an independent prognostic factor in the patients who underwent radical resection of their rectal carcinoma (P<0 .05) .Conclusion The Uyghur rectal cancer patients are associated with significantly younger age ,later treatment ,later stages ,more aggressive histopathologic characteristics and significantly worse prog‐nosis than the Han patients .
7.Detection of FMS-like tyrosine kinase 3 internal tandem duplication in acute myeloid leukemia patients and its clinical significance

Objective To analyze the mutations of FMS-like tyrosine kinase 3 internal tandem duplication (FLT3/ITD) in bone marrow cells of patients with newly-diagnosed acute myeloid leukemia (AML).Methods The mutations of FLT3/ITD in 96 AML cases were detected by polymerase chain reaction (PCR)and the clinical features of FLT3/ITD positive patients were analyzed.Results FLT3/ITD mutations were identified in 18 patients (18.8 %),2 cases were M2,11 cases were M3,2 cases were M4,3 cases were Ms.Patients with FLT3/ITD mutations presented higher initial white blood cell count than that of patients without FLT3/ITD mutations [18.0×109/L (3.6×109-137.6×109/L) vs 6.3×109/L (4.5×109-113.0×109/L),t =3.04,P < 0.05].Out of FLT3/ITD positive patients,13/16 (81.3 %) obtained complete remission and 13 patients remained in first remission in a median follow-up of 10 months(6-15 months).Conclusion The mutations of FLT3/ITD are frequently identified in newly-diagnosed AML patients,patients with FLT3/ITD mutations present high white blood cell count.
8.Metabolic profiling analysis associated with acquired erlotinib resistance of lung adenocarcinoma cells based on liquid chromatography–high resolution mass spectrometry

Objective · To explore the change of metabolomic profiling after erlotinib (anepithelial growth factor receptor tyrosine kinase inhibitor)resistance of lung adenocarcinoma cells (PC9-ER), and find the differential metabolome associated witherlotinib resistance. Methods · Metabolic profiling of PC9-ER cells and homologous parent PC9 cells was acquired by the ultraperformance liquid chromatography tandem quadrupole time-of-flight mass spectrometry (UPLC-QTOF/MS). The data were analyzed by multi-dimensional statistical methods, such as partial least squares projection to latent structures-discriminant analysis (PLS-DA), to select and identify differential metabolites associated with erlotinib resistance. Results · A total of 14 differential metabolites were identified in PC9-ER cells. Seven up-regulated metabolites included N-acetylspermidine, phosphatidylethanolamine, AMP, pantothenic acid,proline, glutamate, and histidine, while seven down-regulated metabolites included citrulline, phosphorylcholine, glutathione, cysteinylglycine, glutathione oxidized, NAD, and S-adenosylmethionine, mainly participating in glutathione metabolism, glutamate metabolism, ammonia recycling, and protein biosynthesis. Conclusion · Metabolic profiling of erlotinib-resistant lung adenocarcinoma cells was changed. The information of differential metabolites associated with erlotinib resistance could provide clues for new resistance mechanisms and potential metabolism-related drug targets.
9.CysC specific peptides: bioinformatics analysis and mass spectrometry verification

Objective To analyze the specific peptide of cystatin C (CysC) and its characteristics by bioinformatics technology,and verify the predicted results by mass spectrometry.Methods Online software was applied to analyze the physicochemical properties and homology of CysC peptides hydrolyzed by trypsin and predict the associated parameters of ionized fragmentation of specific peptide by mass spectrometry.Precursor ion scan and product ion scan were conducted on the samples of synthetic specific peptide.The recombinant human CysC and serum samples were analyzed by mass spectrometry after trypsin digestion.The results of analysis were compared with the outcomes predicted by bioinformatics.Results T3 (ALDFAVGEYNK) was considered as the specific peptide of CysC by software analysis.When selecting[M + 2H] 2 + for product ion scan,almost all the y and b ions of fragmentation were observed using tandem mass spectrometry (MS/MS),showing consistency with Skyline predictions.Moreover,both the peptides from the human recombinant CysC and serum sample following the trypsin digestion were eluted at the same time with the isotope-labeled T3 * under the fixed conditions.Conclusion Bioinformatics technology could be available for picking out the specific peptides of target protein quickly and efficiently and predicting the ionized fragmentation precisely by mass spectrometry scanning.
10.Apolipoprotein C3-an Important Molecule in Hyperlipidemia and Endothelial Dysfunction

Apolipoprotein C3 (APOC3) is a multifunctional protein.Its concentration is positively correlated with triglyceride (TG) levels and is an independent risk factor for coronary heart disease (CAD) prediction and development.Recent studies showed that APOC3 could not only regulate triglyceride-rich lipoproteins (TRL) metabolism,but also regulated endothelial function,that is,it could induce endothelial dysfunction and disorders of lipid metabolism.Gradually,APOC3 induces atherosclerosis (AS),increases the risk of the occurrence of CAD and other related diseases.APOC3 polymorphisms are also closely related to related diseases.
