1.The application of different linear evaluation method for clinical chemistry measurement

Objective To explore the application of different linear evaluation methods for clinical chemistry measurement .Meth‐ods The analytical measurement ranges of clinical common items ,including alanine aminotransferase(ALT) ,creatine kinase(CK) , glucose(Glu) ,urea(Urea) and uric acid(UA) were established by using visual method ,modified Doumas method ,and according to′Guide to linearity evaluation of clinical chemistry analyzers′(health profession standard method) and Clinical and Laboratory Stand‐ards Institute(CLSI) EP6‐A document(EP6‐A method) .Results The visual method shown that the expected values and the meas‐ured values of these inspection items were probably in a straight line .The modified Doumas method shown that nonlinearity be‐tween expected values and measured values of CK was found ,but linearity was evaluated when discarding the last two points ;and the rest of inspection items all had linearity .The health profession standard method indicated that nonlinearities between expected values and measured values of CK and Urea were found ,but could not determine whether the nonlinearities of the two items were clinically acceptable ;and the rest of inspection items all had linearity .The EP6‐A method shown that linearities were directly found between expected values and measured values of ALT ,Glu and UA ,while CK and Urea had clinical acceptable linearities and linear‐ities were evaluated when discarding the maximum values .Conclusion The linearity evaluation is integrated with clinical objectives through EP6‐A document .Through setting methodology allowable deviation ,the EP6‐A method could expand the scope of accredi‐tion of linearity within the clinical acceptable range ,which has higher clinical practicability .

Objective:To observe the anti inflammatory action of Yangfeihuoxue Decoction and its effects on inflammatory mediums. Methods: The mouse and rat inflammatory models were established by dimethylbenzene, carrageenin and paper strip investing method, and the rat model of pulmonary fibrosis was established by bleomycin. The inhibition of Yangfeihuoxue Decoction on various inflammatory reactions (with swelling rate, swelling degree and height of granuloma as markers) and its effects on levels of histamine (H) and tumor necrosis factor (TNF) in blood were studied. 5.Pathological Analysis of liver Biopsies in 115 Patients with Chronic Asymptomatic Carrier

Objective To analyze the pathological features of Liver in chronic asymptomatic carrier (AsC) and to diagnose and treat AsC correctly.Methods 115 patients with AsC were examined with serological markers of B-hepatitis,liver function serum,HBV-DNA level,liver biopsy and immunohistochemistry.Results Of 115 patients with AsC,there were 8.7% patients with invisibly pathological lesions of hepatic tissue (G_0S_0),29.6% patients with chronic hepatitis,1.7% patients with cirrhosis.Through immunohistochemistry HBsAg and HBcAg test,there were 66.96% patients with HBsAg(+) and HBcAg(+),33.04% patients with single HBsAg(+).Conclusions Liver biopsy and immunohistochemistry can help to correct diagnose.It has important directional significance to treat AsC and establish treatment project of AsC.
6.Detection and separation of side population cells in haematological tumor cell lines

BACKGROUND:Side population(SP)cells,with varied contents,are widely distributed in adult tissues,embryos,and certain tumor cells.Haematological tumor is one of the main pathological conditions,which endangers human life.Thus,it is important to recognize SP cells in haematological tumor cell lines.OBJECTIVE:To identify whether hematologic tumor cell lines contain SP cells,and to explore a stable detection and separation methods.METHODS:Cells with the characteristics of stem cells being capable of fluorescent dye Hoechst33342 were isolated by flow cytometry;whether there were SP cells in logarithmic growth period of NB4,Raji,K562/ADM and K562 or not were detected.After sorting K562 subpopulations,the purity of sorted cells was detected.RESULTS AND CONCLUSION:After Hoechst33342 staining,flow cytometry results showed that the SP cells appeared in the haematological tumor NB4,Raji,K562/ADM and K562 cell lines,which accounted for 0.8%,2.7%,1.3% and 2.7%,respectively.These cells could be blocked by Verapamil.The purity was greater after a second detection of SP and Non SP cells in K562 cells.
7.MRI study of acute high voltage electric injury in forearm muscle

Objective To investigate the features of MR imaging of acute high voltage electric injury in forearm muscle. Methods Nine patients (17 forearms, 8 males and 1 female, 15~36 years of age) with clinically and pathological proved acute high voltage electric injury were studied on MRI retrospectively. MRI studies were obtained within 72 hours on Siemens 1 0 T MR scanner. 2 forearms were examined with body coil, and 15 with head coil. The severe area was placed as near as possible to the isocenter in the magnet and was used as the center of the MR imaging acquisition. Spin echo T 1 weighted images, spin echo and fast spin echo T 2 weighted images were acquired in all patients. 14 out of 17 were performed with Ⅳ administration of Gd DTPA. Results All 17 forearms had fascistomy after MRI. 11 had only debridement. The lesions were mainly observed in the flexor digitorum supericialis or profunduds muscle appearing as isointense on T 1 weighted images, hyperintense on T 2 weighted images, and strongly enhanced after Ⅳ administration of Gd DTPA in 8. The proximal aspect of the lesion appeared as sharp knife in 11. There was a weaker twitch response to electrocauterization in the injury muscle than in healthy muscle. It was variably necrotic in histopathology. Two transitional zones accompanied with the suffered forearm in 2, and one transitional zone in 6. Both of them had well defined margin. 6 forearms had amputation after debriding. There was Ⅰ,Ⅱ,and Ⅲ mixture signal all over the forearms. The proximal lesions showed type Ⅰ changes. Distal to the zone of forearm showed type Ⅱ and Ⅲ pattern appearing as isointense on T 1 weighted images, hyperintense and hypointense on T 2 weighted images. It was hardly enhanced after Ⅳ administration of Gd DTPA. There was no twitch response to electrocauterization in the injury muscle. It was almost completely necrotic in histopathology. ALL amputated forearms had two transitional zones and ill defined margin. The second transitional zone was enhanced something like flower border. Conclusion MR imaging of acute high voltage electric injury in forearm appeared as three kinds of signal mode, which was closely related with histopathology. MRI was useful in dealing with clinic problem and in judging the prognosis.
8.Effects of glycyrrhizin-18? on expressions of CTGF and TGF-?1 in renal interstitium in rats with obstructive nephropathy

Objective To investigate the effects of glycyrrhizin-18? on the expressions of connective tissue growth factor (CTGF) and transforming growth factor-?1 (TGF-?1) in renal interstitium in rats with obstructive nephropathy. Methods A total of 100 Wistar rats were divided into experiment group (include prophylaxis group with glycyrrhizin-18?, glycyrrhizin-18? treatment group, and group with normal saline) and control group (sham operation group). Models of rats with obstructive nephropathy were established by unilateral ureteral ligation. The rats were sacrificed at 7, 14, 28, and 56 d after operation and specimens were taken from the renal cortex. The morphopathological changes in the renal interstitium were observed byr light and electron microscopy. CTGF, TGF-?1, and collagen type Ⅲ were detected by immunohistochemical staining. The measured data were evaluated by EIG image analysis system. Results In the renal interstitium, there was progressive fibrosis in the model. Semi-quantitative analysis indicated that the expression levels of CTGF, TGF-?1, and collagen type Ⅲ in the renal interstitium in glycyrrhizin-18? treatment and prophylaxis groups were markedly higher than those in the sham operation group (P0.05). 9.Epidemiological analysis of hand-foot-mouth disease in Shanghai Yangpu District during 2005-2008

Objective To know the epidemiological characteristics of hand-foot-mouth disease in Yangpu District and provide evidence for specific strategies and measures of hand-foot-mouth disease activity prevention and control. Methods Descriptive analysis of the data from hand-foot-mouth disease epidemic situation in Yangpu District from 2005 to 2008 was made. Results 1 348 cases were reported in the whole Yangpu District from 2005 to 2008, and no severe cases, no death. The average morbidity in Yangpu District was 27.48/100 000. The male to female ratio was 1.46∶1. The infection occurred to infants aged from 1 year to 5 years (85.39%). The incidence of the native population children aged 2 to 3 exceeded 10/100 000. The season peak appeared from May to July (70.18%), and outbreaks used to occur at nurseries and kindergartens. The typical clinical presentations mainly included fever and rash. The rash mainly occurred to hands, feet, mouth, buttocks and so on. Conclusions Incidence varied significantly between different sexes, seasons and ages. It can cause large-scale epidemic in a short period of time, the epidemic was very difficult to control, but the leaders attach importance to take the early warning and monitoring, accuratey deal with emergencies, health education promotion and training of comprehensive measures, the epidemic can be effectively controlled.
10.Expression and significance of transcription factor EBF3 in tissues of human primary hepatocellular cancer

Objective To investigate the expression differences of mRNA and protein of early B-cell factor 3(EBF3) in the tissues between hepatocellular carcinoma(HCC) and distant noncancerous tissues and the clinical significance.Methods The expression levels of EBF3 mRNA in 18 pairs surgical specimens of HCC and their distant noncancerous tissues were detected by real-time fluorescence quantitative polymerase chain reaction(FQ-RT-PCR).Western blotting was used to detect the expression levels of EBF3 protein in 5 pairs surgical specimens of HCC and distant noncancerous tissues.Results The ratios of EBF3 mRNA tobeta2 microglobulin mRNA in eighteen liver cancerous tissues(0.52?0.17) were significantly higher than those in distant liver noncancerous tissues(0.28?0.23,t=3.56,P=0.0011).The average gray scale level of EBF3 in five liver cancerous tissues(26.35?14.06)were significantly higher than those in distant noncancerous tissues(7.86?8.47,t=2.52,P=0.036).Conclusion Theexpression levels of EBF3 mRNA and protein up-regulated in primary hepatocellular carcinoma,suggestting that EBF3 may contribute to occurrence of primary hepatocellular carcinoma.
