1.The postoperative secondary xerostomia′s uncomfortable experience of patients with fating in gastrointestinal surgery:a qualitative study

Objective To explore the postoperative secondary xerostomia′s uncomfortable experience of patients with long fating in gastrointestinal surgery, providing basis for medical staff to understand comfort needs of patients and to take targeted nursing intervention. Methods The phenomenological methodology was used in the study. Totally 12 patients with postoperative secondary xerostomia′s uncomfortable experience were interviewed, and tape recording was used. The data was analyzed by Colaizzi analysis method. Results Four themes were extracted: emotional response to patients with postoperative xerostomia, uncomfortable experience of mouth, the insufficient cognition of fating in perioperative period, desire for attention to solve the problem of dry mouth. Conclusions Medical staff should strengthen the awareness of the patients′experience with secondary and temporary dry mouth, and improve the nursing intervention measures according to need, in order to increase patients′comfort and to promote them recover soon.
2.Effect evaluation of family sickbed comprehensive nursing intervention on stroke patients

[Objective] To study the effect of family sickbed comprehensive nursing mode on patients with stroke.[Methods] 92 cases of discharged patients with cerebral apoplexy were selected between October 2010 and October 2011,and were divided into the observation group(52 cases)and the control group(40 cases).Among them,the observation group received the family sickbed comprehensive nursing intervention,which was conducted by community nurses.The control group received general discharge mode and did not accept community nursing intervention.As designed,patients were followed weekly or monthly after discharge.After three months the treatment efficacy and quality of life were assessed.[Results] After follow-up for 3 months,HAMD,NIHSS scores in the intervention group were lower than those of the control group,MMSE,FMA and BI scores were higher than those of the control group.The physical,psychological,social and environmental scorcs in the intervention group were higher than those in the control group.[Conclusions] Family sickbed comprehensive nursing of cerebral apoplexy patients during rehabilitation stage can not only save medical resources,but also facilitate to recovery of patients.
3.Effects of Aristolochic Acid on Renal Function and Pathological Changes of Renal Tissue in Rats

Objective To investigate the effects of aristolochic acid on renal function and tissues morphology in rats.Methods Forty-six male Wistar rats were divided into the normal control group(n=20) and the tested group (n=26) at random.The tested group was treated with the extract of Caulis Aristolochiae Manshuriensis(CAM) (aristolochic acid 20 mg?kg-1?d-1) by gastric gavage,the control group with the same volume of potable water.The body weight of rats was meausured weekly.At the end of 4th,8th,12th weeks,the 24-hour-urine were collected,and blood samples were taken from abdominal artery for detection of indexes of renal function.The pathological changes of renal tissues were also observed.Results The body weight increased slowly and urine volume increased in tested group.The urine albumin level and N-acetyl-?-glucosaminidase (NAG) excretion increased significantly,and the ratio of serum creatinin and body weight increased progressively,indicating that renal function has been impaired.The renal pathological changes under light microscope were as follows:degeneration and necrosis of renal tubule in the early stage and serious tubular structure disorders of focal atrophy and interstitial inchoate fibrosis in the late stage.The changes of ultrastructure of renal tissues under the electronic microscope were as follows:obvious organelle impairment and nucleus variation,interstitial inchoate fibrosis occurring at the 8th week and fibrosis rate arriving 31.36 %at the 12th week.Conclusion Aristolochic acid in CAM has renal toxicity,which is shown as remarkably renal pathological changes,tubular-interstitial injury,interstitial inchoate fibrosis,and cell nuclear variation.
4.Genetic polymorphisms of the pentanucleotide repeat loci Penta D and Penta E in Wuhan Han population by re-designed primers

Objective To study the primer design for Penta D and Penta E genotyping and investigate the genetic polymorphisms of these two loci in Chinese Han population in Wuhan.Methods 281 unrelated Chinese individuals living in Wuhan were typed by hot-start PCR with re-designed Penta D and Penta E genotyping primers and PAGE technique,and the results were compared with those by the PowerPlex TM 16 system of Promega.Results The amplified fragment size of Penta D and Penta E loci with re-designed primers ranged between 153~198bp and 107~212bp respectively,which were consistent with the results obtained by the re-designed primers and PowerPlex TM 16 system,and the re-designed primers had a higher sensitivity than PowerPlex TM 16 system (0.2ng vs 0.5ng) by silver staining.10 and 21 alleles were observed for Penta D and Penta E in Chinese Han population in Wuhan,and the genotype distributions of the two loci were in accordance with Hardy-Weinberg equilibrium.Family studies confirmed Mendelian inheritance of alleles.The power of discrimination (DP) for Penta D and Penta E were 0.926 2 and 0.986 0,and the power of exclusion (PE) were 0.665 1 and 0.832 5 respectively.Conclusion The re-designed primers for Penta D and Penta E genotyping are reliable.These two loci are highly polymorphic in Chinese Han population and can be used in forensic identification and paternity test.
5.The Study of Cell-cycle Specificity of Tamoxifen-induced Apoptosis in the ER(+) Breast Cancer Cells

Objective To study the cell-cycle specificity of tamoxifen-induced apoptosis in the ER(+) breast cancer cells in vitro and in vivo. Methods ER(+) MCF-7 cell line (in vitro) and primary cultured ER(+) breast cancer cells (in vivo) were treated with tamoxifen, and the cell cycle specificity of cell apoptosis and the apoptotic rate were dertermined by flow cytometry. Results Both MCF-7 and primary cultured breast cancer cells were induced to apoptosis with G 0/G 1 phase specificity by tamoxifen. And the apoptotic rate in MCF-7 was higher than that in primary cultured breast cancer cells. Conclusion Tamoxifen could induced G 0/G 1 phase specific apoptosis in the ER(+) breast cnacer cells, and tamoxifen-induced apoptotic rate was higher in vitro than in vivo.
6.The Neuroprotection of Mild hypothermia in the Acute Phase of Subarachnoid Hemorrhage

There are a lot of reports about the neuroprotection of mild hypothermia in the acute phase brain injury of aneurysmal subarachnoid hemorrhage. There are many mechanisms of brain damage involving in the development of brain damage in the acute phase of aneurysmal subarachnoid hemorrhage. Mild hypothermia can protect against various brain damages in the early stage of cerebral infarction. It may play a role in brain protection when it is used in the acute phase of aneurysmal subarachnoid hemorrhage.
7.Changes of cerebral mitochondrial respiratory function and ultrastructure after traumatic brain injury in response to hypothermia

Objective To study the effect of hypothermia on cerebral mitochondrial respiratory function and ultrastructure after traumatic brain injury(TBI). Methods Sprague-Dawley rats were subjected to moderate brain injury by using lateral fluid-percussion(LFP)and randomly divided into sham operation group,normothermic TBI group(rectal temperature for 36-37℃)and hypothermic TBI group(rectal temperature for 31-32℃ lasting for two hours).The ipsilateral brains were dissected and homogenized brain tissues were extracted to obtain mitochondfia by density-centrifugation and speed-centrifugation at 2,24 hours and at days 3 and 7 after TBI.The mitochondrial uhrastructure was studied by electron microscope.The indices of respiratory control rate(RCR)and P/O ratio of mitochondrial respiratory function were measured after oxygen consumption was determined with a Clark-type electrode.Results The mitochondrial uhrastructure of normothermic TBI group was damaged severely while that of hypothermic TBI group kept relatively integrated.The RCR and P/O ratio were markedly decreased two hours after TBI and reached the lowest level at the 24th hour(P<0.01).At day 7,RCR kept at a lower level compared with sham operation group but P/O ratio recovered to normal.Change of RCR was similar in hypothermie TBI group and normothermic TBI group.However,RCR of the hypothermic TBI group was significantly higher than that of the normothermic TBI group within three days after TBI.In the meantime,P/O ratio recovered to normal three days after TBI. Conclusion Hypothermia can improve cerebral mitochondrial respiratory function and protect the mitochondrial structure after TBI.
8.Impact of mild hypothermia on changes of somatosensory evoked potential and synaptophysin mRNA level following traumatic brain injury

Objective To investigate effect of mild hypothermia on changes of somatosensory evoked potential (SEP) and synaptophysin mRNA level after traumatic brain injury (TBI) and determine hypothermia-induced neuroprotection.Methods Forty-five SD rats were allocated into mild hypothermia group,TBI group and sham operation group with 15 rats per group according to the random number table.Left-side fluid percussion impact was performed to induce models of TBI.Rats were exposed to hypothermia environment (32-35℃) for 6 hours in mild hypothermia group after TBI.Rats in sham operation group were treated by only drilling on left side of the head,rather than hitting.To evaluate function outcome,modified neurological severity score (mNSS),SEP and synaptophysin mRNA level were measured at 6 hours,24 hours and 7 days postinjury.Results The mNSS in mild hypothermia group lowered compared with TBI group,especially at 24 hours and 7 days (P < 0.05).SEP in mild hypothermia group was significantly shortened at 6 and 24 hours compared with TBI group (P < 0.05),but SEP revealed no significant difference among the 3 groups at 7 days (P > 0.05).Level of synaptophysin mRNA in mild hypothermia group increased at 6 hours postinjury compared with TBI group [(0.08 ± 0.02) vs (0.12 ±0.04)],with further increase at 7 days postinjury[(0.06 ± 0.01) vs (0.33 ± 0.10)] (P <0.05).Conclusion The shortage of nerve conduction time of the injured side and promotion of nerve regeneration suggest the neuroprotective role of mild hypothermia following TBI.
