1.Animal experimental study on incorporation of silk fibroin reducing the risk of cardiovascular complication during vertebroplasty

[Objective]To develop an animal pulmonary cement embolism model,by which the effect of polymethylmethacrylate(PMMA),calcium phosphate cement(CPC) and silk fibroin/calcium phosphate cement composite(SF/CPC) on the hemodynamic change,respiratory function and the antithrombin activity were measured after the pulmonary embolism caused by cement injection.The efficiency of SF reducing the risk of cardiovascular complication was evaluated.[Method]Twenty-four pigs were assigned to 3 groups randomly,with 8 pigs in each group.After general anesthesia,animals were endotracheal intubated and mechanically ventilated by a respirator.The pulmonary trunk was exposed via sternotomy and each kind of cement was injected into the main pulmonary artery.Hemodynamic parameters were continuously monitored.Blood samples were drawn at certain time points for blood gases analyzing and measurement of AT Ⅲ.Postmortem lungs were subject to computer tomography(CT) examination along with three-dimensional reconstructions of cement casts.[Result]The injection of CPC caused severe pulmonary hypertension and arterial hypotension,with a maximum elevation of 20.93?5.04 mmHg in PAP(20 min post-injection) and a maximum decrease of-26.59?6.8 mmHg in MABP(50 min post-injection).The changes in SF/CPC group were relatively slighter,which is similar with PMMA group,having a statistically significant difference from CPC groups(P
2.Photodynamic therapy with 5% 5-aminolevulinic acid in the treatment of acne: a randomized, controlled trial

Objective To investigate the clinical efficacy of photodynamic therapy using low concentration of photosensitizer, short incubation time and red light source in the treatment of ache. Methods Thirty patients with facial ache were equally randomized into two groups, one group applying 5% 5-aminolevulinic acid (ALA) to the facial lesions on the right side and placebo agent on the left side as control, the other applying the same concentration of ALA to the facial lesions on the left side and placebo agent on the right side as control. The whole face of every subject was irradiated with red light once a week for 4 sessions. The lesions were counted by a dermatologist, sebum production was measured by a sebum meter (SHP88) and pigmentation index by Mexameter MX 18, at the baseline and on week 2, 4, 8 after the last treatment. Adverse effects were also recorded at each visit. Results Two weeks after the last treatment, a sixty percent or more clinical improvement was achieved in the ALA-treated side of 25.9% patients, but no patients experienced sixty or more percent clinical improvement in control side, and the efficacy of ALA was superior to that of the placebo (P<0.05). A significant reduction was noticed in the count of every kind of lesions, including comedo, papules, pustules, cysts/nodules, as well as the total count of these lesions compared with the baseline (all P<0.05); meanwhile, only the pustule count and total count of lesions were decreased in the control side (both P<0.05). Conclusions Both ALA (5%)-PDT and red light-irradiation alone can reduce the number of acne lesions, but the former is more effective than the latter with few side reactions.
3.Up-regulation of TLR4 in the gut of rats after traumatic brain injury

Objective:The aim of the current study was to explore the alterations of TLR4 after traumatic brain injury. Methods: 20 Male ICR rats were randomly divided into four groups(5 rats each group)including control group without brain injury and traumatic brain injury groups(4,24 hours and 7 days).All rats were decapitated at corresponding time point and mid-jejunum samples were taken.The intestinal mucosa structure was detected by histopathological examination.Immunohistochemistry and realtime RNA were used for detection of TLR4 expression in ileum samples.Results: After traumatic brain injury,the histopatholocal alterations of gut mucosa occurred at 4 hours and progressed to a serious state.Compared to control group,TLR4 was significantly up-regulated on the endothelia of microvessels in villous interstitium and lamina propria by 4 h following TBI and maximally expressed at 24 h post-injury.The endothelial TLR4 immunoreactivity in ileum mucosa still remained strong on 7 d post-injury.There was a very low mRNA expression of TLR4 in the gut of control rats and significantly increased at 4 h following TBI(P
4.The experimental study of selective arterial embolization in the lumbar spine of dogs

Objective To establish the model of acute spinal infarction, to evaluate the relative factors affecting results in spinal embolization, and to provide the theorial basis with the preoperative embolization of spinal tumors. Methods Through the SAE of the lumbar arteries, the neuro function of the posterior legs of dogs, MRI findings, and pathologic changes of the spinal specimen were observed in 12 dogs. The embolizing agents was gelfoam(GF). Results The significant ischemia changes of spinal column and the corresponding muscles at the occluding spinal after embolizing more than one segmental arteries occurred in 9 dogs, but there were no paraplegia or obvious changes in 3 dogs having been embolized single lumbar arteries no matter they sent out the radiculomedullary artery(RA) or not. Paraplegia occurred in one dog after embolizing the multisegmental arteries. Conclusion (1) The method of SAE in dog can be used to set up the experimental model of the acute ischemia of spine. (2)The occlusion in single segmental arteries can not result in the infarction of the whole spine. (3)The serious complication may result from embolizing multisegmental spinal arteries (especially sending out RA) (4) The protecting embolization should be carried out in order to decrease the reaction during SAE in spine.
5.Establishment of a human keratinocyte cell line (HaCaT) stably expressing human papillomavirus type 16 E7 (HPV16E7) protein

Objective To establish a human keratinocyte cell line (HaCaT) stably expressing HPV16E7 protein. Methods HPV16E7 gene was amplified from CaSki cells using PCR and inserted into the eukaryotic expression plasmid pcDNA3.1. Then, the recombinant expression plasmid pcDNA3.1-HPV16E7 was transfected into HaCaT cells followed by G418 selection and identification by RT-PCR and Western blot. Results The recombinant eukaryotic expression plasmid pcDNA3.1-HPV16E7 was successfully identified by restriction enzyme digestion pattern and sequence analysis. Agarose gel electrophoresis of RT-PCR products detected the 297-bp fragment of HPV16E7 cDNA, and Western blot confirmed the stable expression of HPV16E7 protein. Conclusion A human keratinocyte cell line (HaCaT) stably expressing HPV16E7 protein is successfully established.
6.Mutation analysis of GJB2 gene in a patient with keratitis-ichthyosis-deafness syndrome

Objective To investigate the clinical features of and GJB2 gene mutations in a Chinese Han patient with keratitis-ichthyosis-deafness syndrome (KID syndrome),in hope to offer evidence for the clinical and genetic diagnosis of KID syndrome.Methods Clinical data were collected from a patient with KID syndrome.DNA was extracted from peripheral blood of the patient and his two family members (mother and brother).PCR was performed to amplify the exon 2 and its flanking splicing sites of GJB2 gene followed by bidirectional direct DNA sequencing. Results The patient presented with the typical triad of vascularizing keratitis,ichthyosis and congenital deafness.A G148A mutation in the exon 2 of GJB2 gene,resulting in the substitution of aspartic acid by asparagine at position 50 of the junction protein connexin 26 (Cx26),was identified in the patient,but not in either of his family members.Conclusion The G148A mutation in GJB2 gene may be responsible for the clinical phenotype of KID syndrome in this Chinese patient.
7.Clinical study on in-stent restenosis after middle cerebral artery wingspan stenting

Objective To observe long-term follow-up results of in-stent restenosis by digital subtraction angiography(DSA) method after angioplasty and stenting with the Gateway-Wingspan stenting system in middle cerebral artery(MCA).Methods Consecutive patients with ischemic stroke and Wingspan stent placement were enrolled into our study.The proportion of in-stent restenosis and ischemic stroke associated with restenosis were evaluated by DSA after 6 and 12 months of stent placement.Results 30 patients with stenosis/occlusion of MCA underwent Wingspan Stent successfully.All of the patients finished follow-up except two patients(6.7% ) who died in the first three months after stenting.At the sixth months follow-up,in-stent restenosis was observed in 7 patients( 23.3% ) with average (69.0 ± 9.8 )% in-stent restenosis degree.However,no additional in-stent restenosis was found at the twelfth month,two patients among the 7 with in-tent restenosis were suffered with artery occlusion in stent.Conclusion In-stent restenosis after Wingspan stenting in middle cerebral artery was more common during the first six months,and 85.7% with ischemic stroke.It was worthy of paying attention to prevent in-stent restenosis at the first six months after stenting.
8.Efficacy of light-emitting diode irradiation at 530 nm, 630 nm and 850 nm in the treatment of photoaged skin

Objective To evaluate the efficacy of light-emitting diode (LED) irradiation at 530 nm,630 nm and 850 nm in the treatment of photoaged skin.Methods A total of 14 adults with photoaged extensor forearm were recruited in this study.Three areas were selected at the extensor forearm in each of these subjects and received LED irradiation at 530 nm,630 nm and 850 nm,respectively,thrice a week for 12 sessions.Dermoscopy was used to visualize these areas before and after the irradiation,the Corueometer CM 825 to measure stratum corneum (SC) hydration,Tewameter TM 300 to determine transepidermal water loss (TEWL),and CM-2600d spectrophotometer to measure L* value and melanin index (MI).Tissue specimens were obtained from these areas irradiated with LED at 530 nm (n =5),630 nm (n =5) and 850 nm (n =4) separately before and 6 weeks after the initial irradiation,and subjected to histopathological examination.Results Both skin texture and pigmentation were improved in 10 and 7 testees after 12 sessions of LED irradiation at 630 nm and 850 nm,respectively,but the skin texture became rougher and pigmentation was increased in 7 testees after LED irradiation at 530 nm.After LED irradiation at 630 nm and 850 nm,the testees experienced an obvious increase in SC hydration ((37.9 ± 7.7) au and (34.5 ± 7.1) au at 6 weeks after the initial irradiation vs.(33.1 ± 6.1) au and (32.0 ± 7.0) au before irradiation,both P < 0.05),but a significant decrease in TEWL value ((9.8-± 2.5) and (10.9 ± 2.5) g·m-2·h-1 at 6 weeks after the initial irradiation vs.(14.0 ± 1.8) and (14.2 ± 2.6) g·m-2·h-1 before irradiation),with no significant changes in L* value or MI (both P > 0.05).No evident alteration was observed in SC hydration or TEWL value (both P > 0.05),while the L* value significantly decreased (P < 0.05) and MI increased (P < 0.05) after LED irradiation at 530 nm.Histopathological examination showed photo-induced denaturation of collagen fibers and elastic fibers in the dermis of these subjects before irradiation,as well as nascent collagen fibers and elastic fibers in a more dense and regular arrangement after irradiation at the three wavelengths.Conclusions LED irradiation at both 630 nm and 850 nm can improve the appearance and barrier function of,and stimulate the proliferation and rearrangement of collagen fibers and elastic fibers in,photoaged skin,while LED irradiation at 530 nm increases skin pigmentation.
9.Application of neurophysiological monitoring and microsurgical technique in acoustic neurinoma resection

Objective To investigate the application of neurophysiological monitoring and microsurgi-eal technique in acoustic neurinoma resection, exploring the significance of neurophysiological monitoring in facial and auditory nerve reservation of acoustic neuronma microsurgery. Methods Accompanied with EMG and BAEP nerve monitoring, 113 patients harboring acoustic neuroma were treated surgically by the subocipi-tal retrosigmoid approach for reserving facial and auditory nerve. The facial nerve was stimulated to evaluate its function during late-operation. Postoperative facial and auditory nerve function were valuated in all the postop-erative following up. Results All of them were treated microsurgically via the suboccipitai retrosigmoid ap-proach. Total tumors resection was achieved in 102 cases (90.3%), subtotal resection in 6 cases(5.3%) and partial resection in 5 case (4.4%). The facial nerve was preserved anatomically in 98 cases (86.7%), the functional valuation of facial nerve according to the House-Brakman (H-B) postoperatively: 86 cases (76.1%) in class Ⅰ -Ⅱ , 12 cases(lO.6%) in class Ⅲ-Ⅳ and 15 cases (13.3%) in class Ⅴ-Ⅵ. The acoustic nerve was preserved anatomically in 40 cases (35.4%). It denoted the good function of facial nerve responsing sensi-tively to electrostimulation ≤ 4mA at the end of operation. Conclusion Assisted with the intraoperative electrophysiological monitoring techniques, it would be greatly facilitate the preservation of facial and auditory nerve in acoustic neuroma resection. Simultaneously, it would valuate the functional convalescence by late-op-erative electrostimulation in the postoperation.
10.Promoter methylation of Wilms' tumor gene on the X- chromosome in gastric cancer.

OBJECTIVETo investigate the changes in methylation levels of the promoters of the tumor suppressor gene Wilms' tumor gene on the X-chromosome (WTX) and its possible role in gastric cancer.
METHODSWTX promoter methylation levels were detected in 20 pairs of specimens of gastric cancer and matched normal tissues and in 3 gastric cancer cell lines (MGC803, SCG7901, and BGC823) using the Sequenom MassARRAY quantitative analysis system. The gastric cancer cell line BGC823 was treated with 5-aza-2'-deoxycytidine (5-aza-dC) for demethylation and the changes in the level of WTX promoter methylation were investigated.
RESULTSWTX promoter methylation levels were very low and showed no significant differences among normal gastric tissues, gastric cancer tissues and the 3 gastric cancer cell lines. In BGC823 cells, treatment with 5-aza-dC did not obviously affect the promoter methylation levels of WTX.
CONCLUSIONHigh methylation levels of WTX promoters are rare in gastric cancer.
