Objective To study the molecular mechanism of acute myocardial infarction(AMI),searching for protective genes and injury-related ones,aimed to fight myocardial infarction disease.Methods Rat model of AMI was established with left anterior descending (LAD) coronary artery ligation.Gene chip was used to verify the mechanism of AMI at molecular level.Results The author screened out 190 differential expression genes between AMI rat myocardium and sham operated rat myocardium in the gene expression profile study;In the Model/Sham operation gene expression profile,there were 89 genes depressed expression and 101 genes elevated expression.Most of them involve in the 12 known categories,and relate to the metoblolism,signal and trandport pathways.Conclusion The major possible mechanism of AMI is to regulate the local expression of related gene through a multi-pathway.

Objective Aim to explore how severe chyle blood serum effects on the End-point method of colorimetry,velocity method,immunological transmission turbidimetry(TIA)three types of common biochemical tests and their different influ-ences.Methods Collected 20 normal appearance serums,divided each pooled serum into A,B,C,D,E,F and G 7 experimen-tal groups and one control group.Each group contented for 1ml serum.Added intralipid 10,20,40,80,160μl and 320μl to the experimental groups from A to G in turn to prepare into different concentrations of simulate chylous samples.Tested each group by the End-point method of colorimetry,velocity method and TIA which represented by Glucose (Glu),uric acid (UA),Total bilirubin (TBil),aspartate aminotransferase (AST),gamma-glutamyl transpeptidase (GGT),lactate dehydro-genase (LDH),hypersensitive C reactive protein (hs-CRP),Apolipoprotein A1 (apoA1 )andβ2-microglobulin (β2-MG). Used pair T test for both experimental group and their control group.Results In terms of TIA,there were statistical differ-ences between all the experimental groups and their control groups in the three projects (t=-2.842~29.465,P<0.05).In terms of the End-point method of colorimetry,there were statistical differences between all the experimental groups and their control groups (t=-34.942~13.884,P<0.05)except for A group in Glu experiment (t=1.875,P=0.068>0.05).In terms of velocity method,there were no statistical differences between all the experimental groups and their control groups (t=-1.532~1.619,P>0.05)except for the B and C groups in GGT experiment (t=2.234&5.006,P<0.05).Conclusion Severe chyle blood serum had significant influence on the End-point method of colorimetry and TIA,as well as less influ-ence on velocity method.

[Objective] To investigate the feasibility of posterior interbody parvule-impacted bone graft fusion.[Method] A retrospective study was performed in 31 patients admited to our hospital in 2003~2006(M=14,F=17).The ages were 55 to 80,with an average of 66.2 years.There were 9 cases of spondylolisthesis,6 of degenerative unstability and 16 of developmental stenosis.All patients underwent posterior interbody impact bone graft fusion and pedicle screw fixation.The pre-and post-operative JOA score were used to calculate the improvement rate.The postoperative bending-extending plain films were taken to observe whether the fusion was successful.[Result]All patients were followed up for 12~18 months.Three patient had leg symptoms at postoperation and were released 3 months later.Thirteen patients had fusion at 6 month,16 had union at 12 month,and 2 had union at 18 month.The lumbar lordosis angle and intervertebral height got significant restoration.The spinal fusion rate was 100%.No dura tear leakage of cerebrospinal fluid or wound infection was found.No graft absorption,displacement or collapse occurred.There were 16 cases excellent,12 cases good,and 3 cases fair.JOA score was 24~29(average 27.4?1.9)at postoperation.The improvement rate was 89.4%.[Conclusion] Posterior interbody parvule-impacted bone graft is a feasible fusion method.The method is simple and cheap,with advantages of less injury and high fusion rate.

Objective To evaluate the life quality in the elderly who underwent internal fixation with cannulated screws and bipolar hemiarthroplasty for the displaced femoral neck fractures. MethodsA retrospective study was performed on 141 cases older than 60 years treated from 1993 to 2004. All cases were followed up for mean 40.2 months to evaluate the differences in regard of pain and daily living at one and three years. Results The daily living in hemiarthroplasty group possessed better outcome than that in internal fixation group at one year, with statistical difference but without statistical difference at three years. There was no statistical differences in postoperative pain relief between both groups. Conclusions Both internal fixation and hemiarthroplasty can relieve pain and revive the life quality of the elderly.

Objective To evaluate the effects of delayed transplantation of bone marrow mononuclear cells (BM-MNCs) after myocardial infarction (MI) on the expressions of bcl-2/bax mRNA and proteins in myocardial cells, and to explore the possible mechanism of cardiomyocyte apoptosis. Methods The MI rat model was reproduced by ligating the left anterior descending coronary artery. Two weeks later 5?106 of BM-MNCs were injected into the infarct zone and the peri-infarct zone (BMT group). TUNEL was used to determine the cardiomyocyte apoptosis, immunohistochemical method was employed to detect the expressions of bcl-2 and bax protein, and the technique of hybridization in situ was applied to assess the changes in of bcl-2/bax mRNA expression. Objective: To evaluate the testing level of the relevant items in food inspection agency laboratory objectively, analyze the existing problems and help them enhance the ability of detection.Methods: The blind sample assessment of sodium saccharin in drinks was organized and performed.The test samples at high and low concentrations were prepared and studied by the tests of homogeneity and stability.Those met the requirements of blind sample assessment were randomly distributed to 193 laboratories and the returned data were analyzed statistically.Results: Totally 182 valid data were collected.Among the reported results, those from 85 laboratories were satisfied in the low concentration group with the satisfaction rate of 87.6% , those from 12 laboratories were not satisfied, and no results were suspicious;as for the high concentration group, the above data was 67(78.8%), 12 and 6, respectively.The overall satisfaction rate was 83.5%.The results, especially the dissatisfied and suspicious data, were analyzed by the original records of each participating laboratory, the analysis focused on 8 aspects including detection method, instrument brand, control product variety, pretreatment method, recovery rate, column selection and the other influencing factors, and suggestions were given to every laboratory to improve its own situation.Conclusion: The statistical data reflect that our food testing laboratory has strong detection ability in the determination of saccharin sodium in beverages, and can provide powerful technical support for the regulatory authorities.

ObjectiveTo evaluate whether β-tricalcium phosphate (β-TCP) combined with bone marrow mesenchymal stem cells (BMSCs) can be used for lumbar posterolateral spine fusion (PLF) instead of autogenous bone graft. Methods6 crab-eating macaques underwent bilateral PLF at L4-5, and divided into 3 groups that implanted β-TCP/BMSCs composite, autogenous bone, and β-TCP. Monkeys were sacrificed 12 weeks after implantation. Manual palpation, micro computed tomography, peripheral quantitative computed tomography (pQCT), and histology were used to assess bone formation. ResultsManual palpation showed that 75% of β-TCP/BMSCs composite group and autogenous group achieved solid spine fusion, whereas none of β-TCP group fused. Histological analysis showed that all of the β-TCP/BMSCs group achieved massive bone formation. Bone mineral density (BMD) evaluated with pQCT in the β-TCP/BMSCs group increased by additional new bone. Conclusionβ-TCP/BMSCs composite can be used for PLF instead of autogenous bone graft.

Objective To evaluate the diagnosis and surgical treatment of primary retroperitoneal extraadrenal pheochromocytoma(PREAP). Methods Clinical data of 17 cases with PREAP from 1976 to 2006 were analyzed retrospectively. Results It was not difficult for the diagnosis of PREAP based on the primary symptoms, including paroxysmal hypertension or persistent hypertension. PREAP is usually deep, large and invading on adjacent organs. B-uhrasound was used for preliminary screening. CT and MRI with high soft tissue resolution and multi directional imaging are of great value for correct diagnosis and determination of exact extent of the tumor in diagnosis of PREAP. 24 h urinary VMA output imcreased in 12 cases (92. 3%) and urinary catecholamine level was up to 526. 1 μg- Precise location of PREAP by 123I-MIBG is superior to that by 131I-MIBG. The diagnosis was tentative in 4 cases and definite in 13 cases. Complete tumor resection was achieved in 15 cases, among them 2 cases died on table due to inadequate preparation before surgery in 1970's. In the remaining 2 cases with huge tumors partial resection, silver clips were put in during operation to locate the extent of the tumors for further management after operation. Conclusions Locating and qualitative diagnosis before operation are valuable and once the patients are diagnosed as PREAP, they should receive exploratory laparotomy after an adequate preparation.

Objective To investigate the expression of VEZT and its clinical pathological significance in gastric cancer tissues,and to construct the VEZT over-expression vector.Methods The expression of VEZT was examined in 119 cases of gastric carcinoma and their corresponding normal mucus tissues by SP immunohistochemical staining.The relationships between the VEZT expression levels and its clinicopathological characteristics,prognosis were also investigated.VEZT cDNA was extracted and amplificated from 293 cells by polymerase chain reaction (PCR).Using DNA recombinant technique,the target gene was connected to the pEGFP-N1 vector to construct recombinant plasmid vector after the target gene and pEGFP-N1 were purified.The recombinant plasmid was identificated by 1％ enzyme electrophoresis and DNA sequencing.Results The VEZT-positive expression in gastric carcinoma was 30.3％ and 60.5％ in normal gastric tissues.VEZT expression in high differentiated cancer tissues was higher than that in lower differentiated tissues(x2 =5.002,P ＜ 0.05),expression of VEZT in early gastric cancer was significantly higher than that in patients with advanced gastric cancer (x2 =5.551,P ＜ 0.05),expression of VEZT in patients without lymph node metastasis was significantly higher than that of lymph node metastasis (x2 =5.878,P ＜ 0.05).Patients with positive VEZT expression have better prognosis than the negative patients(x2 =6.908,P ＜ 0.01).The target fragment,consistent with the theoretical value,was verified by gel electrophoresis.DNA sequencing confirmed that the gene sequence had no mutation and the eukaryotic expression plasmid vector pEGFP-N1-hVEZT was successfully constructed.Conclusions VEZT protein was correlated with TNM staging,tumor stage,invasion depth and lymph node metastasis,positive VEZT expression predicting better five year survival.The VEZT over-expression plasmid vector was successfully constructed.
