1.THE DIFFERENTIATION OF MESENCHYMAL STEM CELLS INDUCED BY THE SUPERNATANT OF CULTURED HAIR FOLLICLE

Objective To observe the possibility of inducing mesenchymal stem cells(MSCs)to differetiate into hair follicle stem cells by the supernatant of cultured hair follicle in vitro and to investigate the transdifferentiation potentiality of MSCs. Methods MSCs were isolated and cultured from rat bone marrow by complete adherence.MSCs of passage 3 were characterized with markers CD44 and CD29 by immuohistochemical staining technique.The stem cells were induced by the supernatant of cultured hair follicle.The morphological character was observed by inverse phase-contrast microscopy.The expression of keratin 15 was detected by immunohistochemical staining technique and immunofluorescence staining technique.RT-PCR was further used to detect the expression of keratin 15. Results The isolated and separated MSCs were immunostaining positive in CD44 and CD29.After induced by the hair follicle conditioned medium in vitro,MSCS could be partially identified by the positive staining for keratin 15,a specific antibody of hair follicle stem cells.After 3 weeks′ induction,keratin 15 was detected by RT-PCR in MSCs induced by supernatant of cultured hair follicle.Conclusion MSCs have the potential to differentiate into hair follicle stem cells induced by the supernatant of cultured hair follicle in vitro.
2.Analysis of influencing factors and strategies on archiving and sharing of S & T resources in a medical organization in Beijing

In Scientific Research Institutions,it has great significance of the S&T resources archiving and sharing to promote the deepening reform of S&T system and realize the great leap-forward development of S&T innovation.In this paper,we conduct empirical analysis on the conditions of S&T resources archiving and sharing in a Medical Organization in Beijing.The results show that the S&T resources archiving and sharing in Medical Organization have achieved initial success,however,the achievements are not ideal and the waste and shortage of S&T resources still coexisted.In view of the deficiencies of the idea,system,laws and regulations of S&T resources archiving and sharing,this paper puts forward corresponding countermeasures and suggestions.
3.Multidisciplinary team confirmed the simultaneous diagnosis of CLL and AML-M5

A 54-year-old man diagnosed with chronic lymphocytic leukemia (CLL) was admitted in our department in June 2014. After one cycle of FC chemotherapy, a bone marrow examination revealed normalized lymphocyte count and another acute myeloid leuke-mia (AML)-M5 clone. The patient refused sequential treatment and only received follow-up examination. He had continuous hemato-logically stable disease and died of pulmonary infection on July 2015. After a multidisciplinary team discussion, we confirmed the si-multaneous diagnosis of CLL and AML-M5. Through this discussion,tumor second hit model,tumor evolution model,andtumor heterogeneitywere further defined.
4.EFFECT OF KONJAC FOOD ON THE BLOOD GLUECOSE LEVEL IN DIADETICS

Seventy two diabetic subjects type Ⅱ were given konjac food for 65 days.The data analysed by multiple F-test indicated that the fasting blood glucosc(FBG), 2-h postprandial blood glucose (PBG) at the 30th and 65th day after consuming the food were significantly reduced (P = 0.001, P200mg%) decreased on the average by 51.8 and 84.6 mg% respectively, those with FBG-O 150-200 mg% by 24.1 and 68.7mg%, and those with FBG-0200 mg%).It could be concluded that konjac food was very useful in both prevention and treatment of hyperglycemia.

6.Association between TOX gene expression level and radiosensitivity in lower-grade gliomas

Objective:To investigate the relationship between the expression level of thymocyte selection-associated high mobility group box protein ( TOX) gene and the radiosensitivity of lower-grade glioma (LGG) patients. Methods:Using bioinformatics research methods, 474 LGG patients from The Cancer Genome Atlas (TCGA) database were selected as the test set (TCGA-474 set), and two different genetic data sets ( n=412 and n=171) from the Chinese Glioma Genome Atlas (CGGA) database were selected as the validation set (CGGA-412 set and CGGA-171 set). Patients were stratified based on whether received radiotherapy, and divided into the high and low TOX expression group according to the expression level of TOX gene in LGG. Survival curves of all patients were plotted. The overall survival (OS) and progression-free survival (PFS) of patients in the high and low TOX expression groups were compared and analyzed using log-rank test. Results:Multivariate analysis of OS in the TCGA-474 set showed that high expression of TOX was a protective factor for OS ( HR=0.061, 95% CI: 0.005-0.791, P=0.044). After stratification analysis based on radiotherapy and adjustment for confounding factors, the HR (95% CI) of patients with high TOX expression in the TCGA-474, CGGA-412, and CGGA-171 sets were 0.405 (0.261-0.629), 0.581 (0.418-0.806), and 0.464 (0.269-0.800), respectively, with P values of <0.001, 0.001, and 0.008, respectively. Among patients receiving radiotherapy in the TCGA-474 set, the OS and PFS of patients with high TOX expression were significantly longer than those in the low TOX expression group, and the differences were statistically significant (both P<0.001). The OS benefit of patients with high expression of TOX was significantly prolonged in both the CGGA-412 and CGGA-171 sets compared to those with low TOX expression, and the differences were statistically significant (both P<0.001). Conclusion:The high expression of TOX may be related to the radiosensitivity of LGG, which may be a gene marker of the radiosensitivity of LGG.
7.Effect of combined domestic clopidogrel and tongxinluo on major adverse cardiovacular events after PCI

Objective To study the platelet inhibition rate of foreign clopidogrel,domestic clopidogrel,combined domestic clopidogrel and tongxinluo and their effct on major adverse cardiovacular events (MACE) after PCI.Methods Two hundred and twenty patients after PCI were divided into foreign clopidogrel treatment group (n=77),domestic clopidogrel treatment group (n=80),combined domesticclopidogrel and tongxinluo treatment group (n =63).The high platelet reactivity (HPR) in 3 groups was detected by thrombelastography after PCI.The incidence of MACE in 3 groups was compared.Results The incidence of left anterior descending branch lesion was lower,the number of sacculi was smaller,and the incidence of HPR was higher in foreign clopidogrel treatment group than in domestic clopidogrel treatment group and combined domestic clopidogrel and tongxinluo treatment group after PCI (63.6% vs 87.5% vs 77.8%,P=0.002;2.3±1.1 vs 2.8±1.4 vs 2.7±1.5,P=0.026;24.7% vs 21.3% vs 11.1%,P=0.030).The incidence of HPR was significantly higher in foreign clopidogrel treatment group than in combined domestic clopidogrel and tongxinluo treatment group (24.7 % vs 11.1%,P =0.040).No significant difference was found in the incidence of MACE in 3 groups (P > 0.05).Conclusion The incidences of MACE of domestic clopidogrel and foreign clopidogrel are similar.Combined clopidogrel and tongxinluo can improve the platelet inhibition rate after PCI.
8.Application of gene editing technology in behavioral medicine and its regulatory policy

Gene editing technology has a broad development prospect in medical research and appli-cation,and it is a frontier hot spot in the field of biomedical research worldwide,especially in the field of be-havioral medicine,making it possible to treat and prevent major diseases such as brain diseases,neurodegen-erative diseases and behavioral psychology diseases. But we must recognize that gene-editing technology poses new threats to bio-security while benefiting humanity. This paper summarizes the development and application prospect of gene editing technology in the field of behavioral medicine,summarizes the ethical and relevant institu-tional problems and international regulatory policies faced by the rapid development of gene editing technology, and finally puts forward relevant regulatory suggestions for the development of gene editing technology.
9.Effect of baseline clinical characteristics of elderly female patients on their long-term ischemic cardiovascular event

Objective To study the effect of baseline clinical characteristics of elderly female patients on their long-term ischemic cardiovascular events.Methods Of the 614elderly patients admitted to our center from April 2008to July 2010,144elderly female patients were divided into female cardiovascular events group(n=43)and female control group(n=101),470elderly male patients were divided into male cardiovascular events group(n=127)and male control group(n= 343).Their baseline clinical characteristics were compared.The patients were followed up for 4.5-6.7(5.6±0.9)years,during which the relationship between first ever acute ischemic cardiovascular events and their clinical factors were analyzed.Results The incidence of grade 2hypertension,grade 3hypertension and hypercholesterolemia,the ratios of antiplatelet drug therapy,CCB therapy and hypoglycemic drug therapy were significantly higher while the history of smoking was significantly shorter,the incidence of chronic renal insufficiency and the ratios of statins therapy,nitrates therapy and ACEI/ARB therapy were significantly lower in female patients than in male patients.The incidence of chronic renal insufficiency was significantly higher in female cardiovascular events group than in female control group(9.3%vs 1.0%,P=0.03).The age was significantly older,the incidence of CHD,ischemic cerebral stroke,chronic obstructive pulmonary disease and the ratio of nitrates therapy were significantly higher in male cardiovascular events group than in male control group.Cox multivariate regression analysis showed that the endpoint events were related with CHD,grade 2hypertension,grade 3hypertension,hypercholesterolemia,chronic renal insufficiency in female patients(HR=2.56,95%CI:1.19-5.54,P=0.017;HR=3.29,95% CI:1.14-9.52,P=0.028;HR=2.77,95%CI:1.08-7.12,P=0.034;HR=2.61,95%CI:1.27-5.36,P=0.009;HR=22.70,95%CI:5.22-98.78,P=0.000)and were related with age,CHD and ischemic cerebral stroke in male patients(P<0.05,P<0.01).Conclusion The effect of baseline clinical characteristics of elderly patients on their long-term ischemic cardiovascular events is different in males and females.The incidence of long-term cardiovascular events is closely related with the risk factors for cardiovascular disease in elderly females.
10.The research of pulmonary function changes after thoracoscopic lobectomy versus thoracoscopic segmentectomy based on propensity score matching method

Objective:To compare the effects of thoracoscopic anatomical segmentectomy and thoracoscopic lobectomy on patients' respiratory function.Methods:Retrospective analysis of 326 patients who underwent thoracoscopic surgery from July 2016 to July 2019(209 patients underwent anatomical segmentectomy, 117 patients underwent lobectomy). According to variables including gender, age, tumor location, smoking history and BMI, two propensity score-matched cohorts including 89 patients respectively were constructed. The patients’ baseline data and respiratory function date of the patients pre-operation and post-operation were analyzed. The measurement data that obey the normal distribution were described by mean±standard deviation, and the t-test was used for comparison between groups; the measurement data of non-normal distribution was described by the median value( P25, P75), and the Wilcoxon rank sum test was used for the comparison between groups; The data was described by frequency, and the chi-square test or Fisher's exact probability method was used for comparison between groups. Results:At the first-month follow-up after surgery, there was no significant difference in the variation of FVC[(0.48±0.40)L vs.(0.34±0.37)L, P=0.215)and FEV1[(0.52±0.46)L vs.(0.43±0.77)L, P=0.364), and in the change rate of FVC(%)[15.23(8.74, 21.25) vs. 14.58(7.75, 19.40), P=0.122], FEV1(%)[17.25(9.56, 22.78) vs. 16.42(9.15, 20.28), P=0.154]and DLCO(%)[18.54(10.88, 25.68)vs. 17.45(9.58, 23.75) P=0.245]. Between the segmentectomy group and lobectomy group, there was a significant difference in the alteration of FVC[(0.50±0.47)L vs. (0.29±0.31)L, P=0.031] and FEV1[(0.44±0.34)L vs.(0.24±0.23)L, P<0.001], the change rate of FVC(%)[14.27(7.87, 22.32) vs. 9.95(5.56, 17.24), P=0.008]、FEV1(%)[15.23(8.36, 22.17)vs. 10.05(5.15, 18.54), P<0.001]and DLCO(%)[13.74(6.24, 19.78) vs. 4.45(-2.32, 13.75), P=0.023]in the 6th month after surgery. The lobectomy group had a higher variation of FEV1[(0.34±0.49)L vs.(0.18±0.26)L, P=0.006] and change rate of FVC(%)[9.28(2.15, 18.94) vs. 5.24(0.52, 11.45), P=0.0032] and FEV1(%)[10.45(3.15, 21.32) vs. 6.50(1.55, 14.24), P<0.001] in the first year after surgery. However, the variation of FVC[(0.29±0.36)L vs.(0.21±0.24)L, P=0.176) and the change rate of DLCO(%)[8.35(2.15, 16.45) vs. 6.23(2.12, 14.54), P=0.143] didn't show a significant difference between the two groups. Conclusion:Whether in the short or the middle postoperative period, segmentectomy can preserve postoperative respiratory function than lobectomy.
