1.Personal Exposure to PM2.5 of Children Living near Traffic Road and the Influencing Factors in Beijing

Objective To evaluate personal exposure level to PM2.5 of children living in one of districts of Beijing and to explore the possible influencing factors. Methods Thirty-six pupils were chosen from a primary school in the district. The PM2.5 level of classrooms,campuses and children's living rooms were monitored by Aerosol Monitor and 24 h time-active pattern and environmental information of living room of these pupils were collected at the same time. Each child's exposure level of PM2.5 was calculated by combining the environment PM2.5 monitor data and the children's 24 h time-active pattern. The possible influencing factors were analyzed by single factor analysis(Kruskal-Wall rank sum test and Spearman rank correlation analysis) and multiple linear regression models. Results The 24 h average exposure level of PM2.5 of 36 pupils was 98.13 ?g/m(341.22~121.13 ?g/m3) . 2.A case of giant cavernous hemangioma of the nasal septum.

To report a case of an adult male with a huge cavernous hemangioma arising from the nasal septum, and the lesion was widely spread to hard palate, sphenoid sinus, ethmoid sinus and right maxillary sinus. After giving embolization, it was completely excised via transnasal endoscopic approach and lateral rhinotomy, histologic examination reported cavernous hemangioma. There was no recurrence at 1 year's follow-up.

3.MUC-1 EXPRESSION IN GASTRIC MUCOSA OF HUMAN FETUSES

Objective To explore the expression of MUC-1 (one mucin) in the gastric mucosa of human fetus at different developmental stages and elucidate the relationship between MUC-1 expression and the cytodifferentiation of gastric epithelia. Methods HE staining and immunohistochemistry methods were used. Results The differentiation of the fetal gastric epithelial cells could be divided into three stages, the undifferentiated (S1), the differentiating (S2), and the growing stages (S3). MUC-1 appeared in cell apex of all undifferentiated cells in S1. In S2, MUC-1 appeared in cell apex and cytoplasm in the differentiating cuboidal epithelial cells.In the S3, MUC-1 focused on the glands, especially in the basal cells of glands.Conclusion There was different expression of MUC-1 at various developmental stages of gastric epithelia.
4.Hand-assisted laparoscopic hepatectomy for hepatic hemangioma

Objective To evaluate the feasibility, safety, and outcome of hepatectomy by hand-assisted laparoscopic surgery (Lapdisc system) in patients with hepatic hemangioma. Methods Eight patients with hepatic hemangioma underwent hand-assisted laparoscopic hepatectomy. Procedures included introduction of Lapdisc system, isolation of the liver from the ligments, occlusion of the hepatic porta, dissection of the hepatic parenchyma by harmonic scaple and removal of the samples. ResultsIn all patients, the hand-assisted laparoscopic hepatectomy were successfully performed. The operation time was(196.3?81.2)min. Blood loss was (307.5?224.7)ml, and postoperative hospital stay was (7.9?2.9)d. There was no significant postoperative complication such as bile leakage, bleeding or infection. ConclusionsLapdisc system could be safely used for hepatectomy in cases of hepatic hemangioma.
5.Analysis of 1540 neonates with birth defects related to different assisted reproductive technique

Objective To investigate the incidence of and clinical factors influencing neonatal birth defects from different assisted reproductive technology. Methods Between October 1998 and December 2006,1271 newborns from mothers treated by in vitro fertilization techniques [ including in vitro fertilization (IVF), intracytoplasmic sperm injection (1CSI) and thaw embryo transfer (Thaw-ET) ] matched with 269 newborns from mothers treated by artificial insemination were enrolled in Reproductive Medicine Center in First Hospital Affiliated to Zhengzhou University. Their medical information was analyzed retrospectively to compared neonatal characteristics, the incidence of birth defect and anomalous organs involved between in vitro fertilization group and artificial insemination group. Results In group of in vitro fertilization, those newborns with low birth weight from IVF, ICSI and Thaw-ET were 20. 0% ( 134/671 ), 22. 4% (92/410), 18.9% (36/190)respectively, which were more than 11.5% (31/269) cases in group of artifical semination with statistical significance (P < 0. 05 ). The rates of multiple pregnancy of 23.8% ( 160/671 ), 25.4% (104/410) ,21.1% (40/190) in subgroup of 1VF, ICSI and Thaw-ET were significantly higher than 10. 0% ( 27/269 ) in group of artifical insemination( P < 0. 05 ). The rate of macrosomia in group of in vitro fertilization was significantly lower than that of artificial insemination group (3.9% vs 8. 2%, P <0.05). However, the incidence of birth defect involved in various organs did not show significant difference between two groups ( P>0.05 ). Conclusions The incidence of multiple pregnancy demonstrated obviously increasing trends born with various In Vitro Fertilization techniques, which pave the way to high risk pregnancy. However, the incidence of newborn birth defect was not increased significantly. Thus, to lower occurrence of multiple pregnancy was the key approach to obtain neonates health.
6.Association of serum adiponectin and resistin with type 2 diabetes mellitus and its macrovascular complications

The concentration of serum adiponectin [(2.51±1.42)mg/L] was lower in the group of patients with type 2 diabetes mellitus as compared with that in normal controlgroup [(5.26±0.78)mg/L ,P<0.01]. The concentration of serum adiponectin was lower in the diabetics with macrovascular complications (MVC) [(1.38±0.77)mg/L] as compared with those without macrovascular complication [(3.66±0.91)mg/L]. The concentration of serum resistin was higher in the diabetic group as compared with that in control group[(7.07±1.11 vs 6.09±0.47)μg/L, P<0.01]. It was also higher in patients with MVC [(7.96±0.65)μg/L] compared with those without MVC [(6.10±0.43)μgL, P<0.01].

Objective To evaluate the selectively killing effect of adenovirus (Ad) mediated double suicide gene driven by KDR promoter on vein endothelial ECV304 cells.Methods KDR producing cells ECV304 and the KDR nonproducing cells LS174T were infected by Ad respectively, followed by treatment with 5-FC and GCV.Killing effects were evaluated and bystander effects were analyzed.Distribution of cell cycle was detected by flow cytometric assay and pathological character of cells was observed by electron microscopy.Results The infection rate of the resultant recombinant Ad to all the cells was not apparently different, and it increased gradually with the addition of multiple of infection (MOI) of Ads.ECV304 cells infected with Ad-KDR-CDglyTK were sensitive highly to the prodrugs, but the LS174T cells infected with Ad-KDR-CDglyTK appeared to be unsensitive to the two prodrugs ( P
8.Simultaneous determination of 4 major components in semen cassiae obtusifoline by HPLC.

OBJECTIVETo establish a HPLC method for the simultaneous determination of 4 major components in semen cassiae obtusifoline and provide valuable data for quality control of semen cassiae obtusifoline.
METHODKromasil 100-5 C18 column (4.6 mm x 250 mm, 5 microm). Eluted with water: acetonitrile: tetramethylene oxide: glacial acetic acid (100: 23: 5:1). Detecte at 278 nm. The flow rate was 1.0 mL x min(-1).
RESULTFour compositions were separated well. Good linearies were obtained within the range of 0.274-1.37 microg, 0.153-0.765 microg, 0.302-1.51 microg and 0.052-0.26 microg for rubrofusarin gentiobioside, casside, aurantio-obtusin-6-O-beta-D- glucopyranoside, casside B respectively. The average recoveries were 100.9% (RSD 1.8%),101.2% (RSD 1.2%), 99.40% (RSD 2.2%), 100.5% (RSD 1.6%).
CONCLUSIONThe method is accurate, reliable and convenient. It can be used to control the quality of semen cassiae obtusifoline and its products.

9.MEG3 LncRNA from Exosomes Released from Cancer-Associated Fibroblasts Enhances Cisplatin Chemoresistance in SCLC via a MiR-15a-5p/CCNE1 Axis

Purpose:
Long non-coding RNAs (lncRNAs) may act as oncogenes in small-cell lung cancer (SCLC). Exosomes containing lncRNAs released from cancer-associated fibroblasts (CAF) accelerate tumorigenesis and confer chemoresistance. This study aimed to explore the action mechanism of the CAF-derived lncRNA maternally expressed gene 3 (MEG3) on cisplatin (DDP) chemoresistance and cell processes in SCLC.
Materials and Methods:
Quantitative real-time PCR was conducted to determine the expression levels of MEG3, miR-15a-5p, and CCNE1. Cell viability and metastasis were measured by 3-(4, 5-dimethyl-2-thiazolyl)-2, 5-diphenyl-2-h-tetrazolium bromide and invasion assays, respectively. A xenograft tumor model was developed to confirm the effect of MEG3 overexpression on SCLC progression in vivo. Relationships between miR-15a-5p and MEG3/CCNE1 were predicted using StarBase software and validated by dual luciferase reporter assay. Western blotting was used to determine protein levels. A co-culture model was established to explore the effects of exosomes on MEG3 expression in SCLC cell lines.
Results:
MEG3 was overexpressed in SCLC tissues and cells. MEG3 silencing significantly repressed cell viability and metastasis in SCLC. High expression of MEG3 was observed in CAF-derived conditioned medium (CM) and exosomes, and promoted chemoresistance and cancer progression. Additionally, MEG3 was found to serve as a sponge of miR-15a-5p to mediate CCNE1 expression. Overexpression of miR-15a-5p and knockout of CCNE1 reversed the effects of MEG3 overexpression on cell viability and metastasis.
Conclusion
MEG3 lncRNA released from CAF-derived exosomes promotes DDP chemoresistance via regulation of a miR-15a-5p/CCNE1 axis. These findings may provide insight into SCLC therapy.
10.Genetic analysis of pseudohypoparathyroidism type Ⅰ a : report of a pedigree

The clinical and genetic data were retrospectively analyzed in a pedigree with pseudohypoparathyroidism type Ⅰ a.Clinically typical Albright hereditary osteodystrophy (AHO),hypocalcemia,hyperphosphatemia,and PTH- and TSH-resistance were manifested in the proband,but not in his brother and parents.The proband's symptom of epilepsy was alleviated by treatment with calcium and vitamin D,which was of no avail in regard to AHO.After GNAS1 genes were sequenced and compared with the GenBank data among the family members,a deletion of c.1107_1108 ( p.Glu370ArgfsX11 ) in exon l3 of GNAS1 gene leading to a frameshift mutation was found in the proband and his mother.It suggested that the GNAS1 gene mutation might be related to the pathogenesis of the disease.
