Objective To investigate the effect of probucol on Sertoli cell injury induced by oxygen-glucose deprivation/reoxygenation(OGD/R)and mechanism. Methods Primary Sertoli cells treated with oxygen glucose deprivation for 2 h and reoxygenation for 3 h were modeled. We incubated Sertoli cells with probucol in a concentration of 5,10,and 20 μmol/L from reoxygenation to the end. The cell viability was evaluated using Cell Proliferation Assay kit. ROS production was observed by flow cytometry. The levels of MDA ,iron and GSH or the activity of glutathione-dependent peroxidase were measured by Assay Kit. Results The ferroptosis inhibitors pre-vented cell death(P < 0.01). The cell death rate was decreased by probucol in time- and dose-dependent manner (P < 0.01). Meanwhile, probucol caused increase of ROS,MDA,GSH content and GPXs activity(P < 0.05). Conclusion Probucol protects Sertoli cells from OGD/R damage via inhibiting lipid peroxidation-mediated ferroptosis.
4. Development of researches in intracranial atherosclerosis based on the high-resolution magnetic resonance imaging
Chinese Journal of Cerebrovascular Diseases 2019;16(11):607-612
Intracranial atherosclerosis (ICAS) is a common cause of ischemic stroke. The symptomatic ICAS is closely associated with frequent stroke recurrence, while the asymptomatic ICAS has evidently higher risk of future stroke onset. High-resolution magnetic resonance imaging is regarded as a significant emerging method. In addition to non-invasive identification of intracranial artery stenosis causes, accurate stroke mechanism judgment and plaque stability evaluation, it has great advantages and clinical value in observing the dynamic evolution of lesions and characteristics of perforating arteries, evaluation of drug efficacy and guidance of intravascular treatment of intracranial artery stenosis. The article aims to review the recent development of researches in the mentioned aspects.

Objective To study the effects of bioartificial kidney(BAK)treatment on cytokines interleukin-10(IL-10)and tumor necrosis factor-α(TNF-α),biochemistry indexes, and survival time in acute uremic pigs with multiple organ dysfunction syndrome(MODS).Method Pigs with MODS and acute renal failure(ARF)were treated with BAK(group A,n = 6)or sham BAK containing no cells(group B,n = 6)or received no treatment(group C,n = 5).Data on blood pressure, hepatic and renal functions, serum IL-10 and TNF-α levels, arterial blood gas and survival time of all the pigs were recorded.Comparisons of values were done using Student's t-test or repeated-measures analysis of variance(ANOVA).ResultsMean arterial pressure(MAP,mmHg)responsed more rapidly and reached higher values in group A (91.82±5.73)compared with group B and C at 24 hours(P<0.01).The peak level of serum EL-10(pg/mL)ingroupA(249.57±43.51)was significantly higher than that in group B and C(132.06± 17.53,104.25 ±13.42,P<0.01).Serum TNF-a level(pg/mL)in group A dropped gradually to(402.91 ± 32.47)at 24 hours, and had significant discrepancy compared with that before the treatment(537.16 ± 38.45)and that in groupB(P<0.05).The average survival time(hours)in group A(113.01 ± 14.32)was significantly longer than that in the group B and C(P<0.01),which was prolonged by 35.93%and 63.90%,respectively.ConclusionsTreatment with BAK can prolong the survival time of uremic pigs with MODS,possibly through ameliorating the MAP,increasing the level of IL-10 and reducing the concentration of TNF-α.

Objective To study the relationship between polymorphism of NAT2 gene and the susceptibility of osteosarcoma. Methods Using serum samples of patients with osteosarcoma , NAT2 gene polymorphism was determined by PCR-RFLP to observe M1, M2 and M3 mutant genes, NAT2 genotype and allele distribution. Slow acetylation and rapid acetylation genotype between groups in the distribution were also detected. Moreover , the relationship between the clinical characteristics of osteosarcoma and NAT2 genetic polymorphism were analyzed. Results From January 2010 to September 2015, 126 patients with osteosarcoma were enrolled as a case group, and 119 healthy persons in the same period were as the control group. In the control group, the frequency of NAT2 genotype (homozygous wild type (wt/wt and miscellaneous synthetic mutant WT/ MX, homozygous mutant MX/MX) were 30.95%, 50.79% and 18.25% respectively; In the experimental group were 47.06%, 46.22% and 6.72% respectively. The frequency of the two groups was statistically significant (P <0.05). Rapid NAT2 acetylator genotype of suffering from the risk of osteosarcoma 1.782 times slow acetylation genotype (P < 0.05). Compared to rapid acetylation genotype patients with osteosarcoma, slow acetylation genotype patients has smaller tumor volume (P = 0.008),lower differentiation degree of tumor (P = 0.011) and less occurrence of distant metastasis (P = 0.001). Conclusion The rapid acetylation genotype of NAT2 gene may be a risk factor for osteosarcoma.

Neonatal intrahepatic cholestasis caused by citrin deficiency (NICCD) is an autosomal recessive disease caused by SLC25A13 gene mutations, and is characterized by delayed jaundice clearance, liver dysfunction, and elevated aminoacidemia. The confirmed diagnosis depends on gene analysis. Citrin deficiency is one of the important causes of neonatal intrahepatic cholestasis in China. Recently more and more researches about NICCD were reported. The paper summarized the epidemiology, pathogenesis, clinical characteristics, and progresses in diagnosis and treatment of NICCD.

Objective To explore the association between the signal transducer and activator of transcription 4 (STAT4) gene polymorphism and Chinese Han systemic lupus erythematosus patients. Methods Pyrosequencing technique was used to genotype the 3 single nucleotide polymorphisms (SNP) in the samples of patients and normal individuals. Results Not only these 3 SNPs, but also the haplotypes composed by them showed significant difference between the SLE patients and normal individuals[rs11889341:P=0.012 02, OR (95%CI)=1.22( 1.044~1.424); rs7574865:P=0.003 454, 0R(95%CI)=1.25(1.076~1.451 ); rs8179673: P=0.004 275, OR (95%CI)=1.274 (1.079~1.505)]. Conclusion rs11889341, rs7574865 and rs8179673 of STAT4 are associated with the pathogenesis of Chinese Han SLE, and the STAT4 gene is a susceptibile gene for SLE in a few racial cohorts.

Objective To study the effects of rehabilitation training on the expression of synaptophysin in the cortex after cerebral ischemia. Methods A model of focal cerebral ischemia was created in rats. The rats were divided into 4 groups at random 24 h after the cerebral ischemia was induced. The rehabilitation group was given bal-ancing, grasping, rotation, walking and other training every day. An immobilization group was fixed in their cages. The model control group and cerebral ischemia group were kept in general cages, taking food and moving freely. Im-munohistochemisty method was used to detect synaptophysin expression at the 1st, 7th, 14th, 21st and 28th day after the cerebral ischemia was induced. Results In the model control group, synaptophysin immunoreactive positive products were observed. In the cerebral ischemia group, the expression of synaptophysin decreased gradually. After rehabilitation training, synaptophysin immunoreactive positive products decreased slowly, but remained more abun-dant than among the immobilized rats. Conclusion After cerebral ischemia, the expression of synaptophysin de-creases. This demonstrates that the synaptic terminals were less injured. Rehabilitation training can enhance synapto-genesis in the cortex after cerebral ischemia.
