Aim To compare the inhibition of lipid peroxidation of ethyl acetate extract( EAE) and n-buta-nol( BE) extract from Coreopsis tinctoria Nutt. in vitro. To investigate the parameters such as body weight, bio-chemical indexes in plasma, and viscera indexes on type 2 diabetes mice by intraperitoneal injection of streptozotocin ( STZ ) . Methods The extracts were prepared by response surface methodology. The ex-tracts were suspended in distilled water and defatted with petroleum ether. The aqueous layer was succes-sively extracted with ethyl acetate and n-butanol. The inhibition of lipid peroxidation activity was determined by thiobarbituric acid method. The effects of extract BE on diabetic mice were observed at the dosage of 0. 2,0. 4,0. 8 g·kg-1 ( ig) for 4 weeks. The parame-ters were observed such as weight of body changes, or-gan coefficients of liver, pancreas and kidney, bio-chemical indexes in plasma and viscera pathological sections. Results In the linoleic acid reaction system, the SC50 value of the EAE and BE was ( 443. 96 ± 11. 24) mg·L-1, (840. 29 ± 16. 38) mg·L-1, re-spectively, and that in rat liver homogenate was (23. 59 ± 3. 67 ) mg · L-1 , ( 60. 37 ± 4. 27 ) mg · L-1 , respectively. Compared with diabetic model group, BE could significantly improve the trend of weight loss, and increase viscera indexes. The patho-logical sections showed that BE had the recovery and improvement effects on the damage of liver, pancreas and kidney. Conclusions The extracts of C. tinctoria have a certain anti-lipid peroxidation activity in vitro. And BE has a certain capacity to improve and repair damaged organs for DM mice.

Objective:To investigate the effect of elevated low-density lipoprotein(LDL)cholesterol on the release of endothelial microvesicles into the circulatory system in elderly patients with obstructive sleep apnea(OSA).Methods:This was a case-control study.A total of 69 elderly inpatients aged ≥ 60 years were divided into the OSA group(n=48)and the normal control group(n=21)based on the apnea/hypopnea index(AHI). Sleep monitoring was carried out in the two groups, and venous blood samples were taken after the monitoring.Density gradient centrifugation was used to extract microparticles and flow cytometry was used to analyze patients' blood levels of endothelial microvesicles.Blood lipid levels of patients were measured by biochemical analysis methods. The correlation between LDL cholesterol levels and endothelial microvesicles in OSA patients was analyzed. Results:LDL cholesterol levels were higher in OSA patients than in normal controls(2.6±0.6 mmol/L vs.2.2±0.7 mmol/L, t=2.089, P=0.041). Endothelial microvesicle levels were elevated in OSA patients compared with the normal controls[(28.2±12.2)×10 -6events/L vs.(19.6±7.8)×10 -6 events/L, t=-3.164, P=0.003]. Endothelial microvesicle levels increased in OSA patients with elevated LDL cholesterol compared with OSA patients with normal LDL cholesterol[(39.3±12.6)×10 -6 events/L vs.(28.2±12.2)×10 -6 events/L, t=2.899, P=0.006]. There was a significant correlation between LDL cholesterol levels and endothelial microvesicle levels in OSA patients( r=0.497, P=0.032), while levels of HDL, total cholesterol, triglycerides, blood pressure and blood glucose had no significant correlation with endothelial microvesicle levels. Conclusions:The level of endothelial microvesicles released into the circulation is higher in elderly OSA patients than in normal elderly people, and the elevated concentration of LDL can further promote the release of endothelial microvesicles in OSA patients.

Objective To investigate the clinical and prognostic significance of ABO promotor methylation level in adult patients with leukemia and myelodydysplastic syndrome(MDS). Methods ABO promoter methylation level of 182 malignant hematological disease patients and 68 normal controls were detected by bisulfite sequencing PCR. Then clinical features and outcome were compared between hypermethylation group and hypomethylation group. Results The median methylation rate of ABO promoter in newly diagnosed acute myeloid leukemia (AML) and acute lymphocytic leukemia (ALL) were 46.98% and 11.01% respectively, which were both higher than that in controls (2.30%, P<0.05). The methylation rates in remission AML and ALL were 1.58%and 2.30%respectively, which were comparable with that in normal group (P>0.05). As to relapse AML and ALL, methylation rates were 41.26% and 17.50%respectively, also significantly higher than that in controls (P<0.05).In patients with chronic myeloid leukemia (CML) chronic phase, the median methylation rate was 1.00%, which was similar to normal group. But a CML patient who transformed to ALL hadextremely high methylation rate 92.56%. The median methylation rate in patients with MDS significantly elevated as 5.81% compared with that in controls (P<0.05). The median overall survival (OS) of ALL and AML (non-M3) patients with hypermethylation were 12.5 months and 15.3 months, which were significantly shorter than those with hypomethylation (24.0 months and 20.0 months) (P<0.05). The median disease-free survival (DFS) of ALL and AML (non-M3) patients with hypermethylation were 9.9 months and 12.0 months, which were significantly shorter than those with hypomethylation (22.3 months and 18.5 months), (P<0.05). Multivariable analysis suggested that ABO promoter methylation level was an independent predictive factor of OS and DFS in ALL and AML (non-M3) patients. Conclusion ABO promoter hypermethylation is closely related to genesis, development and prognosis of leukemia and MDS. Hypermethylationis related to a clinical poor prognosis compare with hypomethylation.
