Aim To Study the inhibition mechanism of edelfosine on cytokinesis in Schizosaccharomyces pombe. Methods To observe the inhibition of cytokinesis and growth, the experiments of inhibition of cyto-kinesis and parallel growth were carried out. To explore the inhibition mechanism of edelfosine, the experiment of retransfection of yeast genes was carried out. Results Edelfosine inhibited the cytokinesis of S.pombe in low dosage and cell growth in high dosage. The mid2△、spm1△ and pmp1△ strains were resistant to edelfosine in high dosage. However, these strains retrieved the sensitive to edelfosine in high dosage by retransfected the relevant genes, respectively. A reciprocal action, which was the Mid2p induced the phosphorylated Spm1, while the Pmp1 dephosphorylated Spm1, existed among the Spm1、Mid2 and Pmp1 in S.pombe treated with edelfosine. Conclusion The inhibition of edelfosine on cytokinesis and cell growth due to the effect of Spm1、Mid2 and Pmp1.

AIM:To analyze the recovery of visual cortex activation range of color vision in anisometropia amblyopia children after treatment by using blood oxygen level dependence-functional magnetic resonance imaging ( BOLD- fMRI ) and SPM8 software.
METHODS:Self-control study. This fMRI study directly compared activity in visual cortex produced by color monocular stimulation in 13 monocular anisometropia amblyopia children. The project of study was blocked design. The data of functions and anatomical MRI was stimulated by three-primary colors and was preprocessed and analyzed by SPM8 that based on MATLAB software. According to the data, we compared the change of the central area of color vision when first visit and 1, 2 ,4wk after treatment.
RESULTS: The BA17, BA18, BA19 and BA37 of amblyopia children were different activated after they accepted the three-primary colors stimulation (P<0. 01). After 4wk treatment, amblyopia children accepted red stimulation, the visual cortex activation range without expanding; after accepting green stimulus, on the left side of the visual cortex BA37, BA19, BA20, the average t value were 3. 5210, 3. 2716, 3. 1534 ( P< 0. 01 ); after accepting blue stimulation, the scope of the right side of visual cortex at BA19, BA18 were expanded to different extent, the average t value were 3. 7345, 3. 2701 (P<0. 01).
CONCLUSION: After 4wk treatment, visual cortex activation of color vision of anisometropia amblyopic children is expanded, but with low extent, after the short-term treatment of amblyopia, visual cortex activation of color vision has a certain degree of recovery but not obvious.

Objective To investigate the effect of Danzhijiangtang capsule on serum, fat in type 2 diabetic rats. Methods The model of type 2 diabetes mellitus rats were established by injecting small does streptozotocin(STZ)into abdomen and feeding high sucrose-fat diet. All the rats were divided randomly into five groups. The level of blood glucose, serum insulin, insulin sensitivity index, blood fat(triglyceride, cholesterol,high-density lipoprotein,low-density lipoprotein),free fatty acid after treated were examined. Results Danzhijiangtang capsule could decrease the level of blood fat. Conclusion Danzhijiangtang capsule could improve the blood fat in type 2 diabetic rats.

Aim To investigate the effect of the inhibitors of 3-hydroxy-3-methylglutaryl coenzyme A(HMG-CoA)reductase enzyme, simvastatin on E-Selectin ex-pression and adhesive function assay induced by CD40L.Methods Human umbilical vein endothelial cell(HUVEC)was cultured from ECV304 cell strain. Then E-Selectin expression and adhesion of lymphocytes to endothelial cells induced by CD40L and interfered with different concentrations of simvastatin or simvastatin(5 ?mol?L-1)+mevalonic acid were determined by RT-PCR and FCM analysis. Results Preincubation of HUVECs with simvastatin(0~10 ?mol?L-1)decreased the expression of E-Selectin induced by CD40L in a concentration-dependent manner.Mevalonic acid(400 ?mol?L-1)inhibited the restrain of simvastatin on E-Selectin expression. Moreover, preincubation with simvastatin, decreased significantly adhesion of lymphocytes to endothelial cells induced by CD40L.Conclusions Simvastatin inhibits activation of endothelial cells induced by the CD40L/CD40 pathway through HMG-CoA dependent effect.

Objective To investigate the clinical effect of low frequency electrical pulse therapy combined with α-lipoic acid on diabetic gastroparesis in elderly patients with type 2 diabetes. Methods A total of 65 patients diagnosed as diabetic gastroparesis were selected from our hospital and divided into three groups according to random number table:the control group(n =23,the α-lipoic acid treatment),the conventional treatment group (n =16),and the experimental group (n =26,treating with α-lipoic acid combined with low-frequency electrical pulse therapy). All patients received the conventional diabetic therapy.Clinical effects, gastric emptying rate and serum gastrin(GAS) and fasting blood glucose levels were compared before versus after treatment among the three groups. Results The cure and total effective rates were higher in the experimental group than in the control group [46.15% (12 cases) vs.30.43% (7 cases),80.76% (20 cases) vs.65.21% (15 cases),x2 =0.867,P<0.05].There were significant differences in gastric emptying rate, serum gastrin and fasting plasma glucose levels among the 3 treatment groups before versus after treatment. And the gastric emptying rate and serum gastrin level were better improved in the experimental groups compared with the conventional treatment and control groups. Conclusions The low-frequency electrical pulse therapy combined with α-lipoic acid has a significant clinical efficacy, which can improve clinical effects, promote gastric emptying, decrease fasting plasma glucose levels in elderly patients with type 2 diabetes.

Objective To observe the antioxidative capability and the mRNA expression of sodium pump αl-subunit in kidney of hypothyroid rats by iodine deficiency and illuminate the pathogenesis of kidney damage. Methods Wistar rats were randomly divided into two groups as control group (NT) and hypothyroid group (HT).The rats were all fed with low-iodine diet derived from an endemic goiter area and drank deionized water containing different potassium iodide to duplicate hypothyriod animal models. We determined the morphometric parameters of kidney by routine histology method. The contents of malondialdehyde and free radical scavengers (GSH-PX and SOD) in kid-ney,as well as the activity of Na~+-K~+ ATPase were measured in two groups. The mRNA expression of sodium pump α1 subunit was determined by reverse transcription-polymerase chain reaction. Results Compared with that in the control group,in hypothyroid group (1) serum free T3,free T4,total T3 and total T4 were markedly lower.(2) mean glomerular area and volum diminished markedly.(3)the content of MDA and activity of PGx increased markedly, but the activity of SOD decreased significantly,as well as the one of Na~+-K~+-ATPase.(4)the mRNA expression level of sodium pump α1 subunit was lower. Conclusion In a hypothyroid state, the decrease of antiox-idative capability of kidney resulted in lipid peroxidative damage, atrophy of kidney, decreased activity of Na~+-K~+ -ATPase and degression of sodium pump α1 subunit mRNA expression.

China Biotechnology 2006;0(01):-
The methods for aptamer selection have been modified and developed in recent years. Selection efficiency has been increased due to the improvement in separation of bound and unbound nucleic acids; applicability of aptamers has been improved due to the development of primer-free SELEX and the utilization of short oligonucleotide library; more research requirements have been satisfied due to the functional modifications of aptamer selection methods. All the methodological advances enhance the potential value of aptamers in basic researches and clinical applications.
