2.Highuric acid induced insulin resistance in 3T3-L1 adipocyte and its mechanism

Objective To investigate the effect of high uric acid(HUA) on insulin sensitivity (IS) in 3T3-L1 adipocytes and its mechanism. Methods 3T3-L1 adipocytes were pretreated with HUA with or without NAC,and then stimulated by insulin. The cell viability of 3T3-L1 adipocytes was detected by CCK-8 assay. The glucose consumption was measured by glucose oxidase method. The levels of phospho-IRS-1,phospho-Akt and GluT4 protein were tested by western blot. Results HUA could inhibit insulin-induced glucose consumption,phosphorylation of Akt (Thr308),dephosphorylation of IRS-1 (Ser307) and the GluT4 protein expression (P<0.05). All the above could be blocked by NAC(P<0.05). Conclusion HUA can inhibit inulin stimulated IRS-1/Akt signaling and GluT4 expression,and thus induced insulin resistance in 3T3-L1 adipocytes.
3.Establishment of Determination Method for Drug Release of Phencynonate Hydrochloride Extended Release Tablets

Objective:To establish the drug release determination conditions and method for phencynonate hydrochloride extended release tablets. Methods:The drug release of the tablets was determined by HPLC using a Diamonsil C18 (250 mm × 4. 6 mm, 5 μm) column with the mobile phase of acetonitrile-water-phosphoric acid-triethylamine (270∶400∶1. 3∶2), the detection wavelength was 220 nm, the flow rate was 1. 0 ml·min-1 , the column temperature was 30 ℃ and the injection volume was 20 μl. The effects of different release apparatus, release media and rotation speeds on the release of phencynonate hydrochloride extended-release tablets were studied as well. Results:The established drug release determination method had a good linear relationship within the range of 0. 3-5. 0 μg· ml-1(r=0. 999 8), and the average recovery was 100. 6%(RSD=1. 16%, n=15). Under the conditions of 900ml pH 3. 0 phos-phate buffer solution as the release medium, rotation speed of 50 r·min-1 and the settlement basket as the apparatus, the release be-havior of the product was complied with a zero-level model in vitro, and the release equation was as follows:Q=6. 141 2t-9. 328 7(r=0. 996). Conclusion:The method is simple, accurate and reliable, and suitable for the quality control of phencynonate hydrochlo-ride extended-release tablets.
4.Spiral CT Scanning Method and Technique Applied to Chest Examine:1000 Cases Analysis

Objective To evaluate the diagosis value of different spiral computed tomography(CT) chest scanning methods in the differentiation of various pulmonary diseases,in order to avoid x-ray irradiation damage and reduce patients economic burden,but the diagnosis criterion should be guaranteed still.Methods 1000 cases of chest scan by spiral CT were randomly analyzed.In 927 conventional scan cases,including 155 cases of simple plainscan,474 cases of direct contrast scan,25 cases of plain plus contrast scans,73 cases of CT angiography.In addition,high resolution CT(HRCT) for 105 cases,thin layer scan for 86 cases,dynamic scan for 6 cases,and 40 cases bronchgraphy diagnosis were also carried out in the conventional scanning cases.Results The different CT scanning methods were adapted for diffenent pulmonary diseases.An adaptive scanning method and technique can decrease X-ray irradiation dosage and rise accuracy of diagnosis.Conclusions Aptimal scanning method and technique should be alternative according the clinical need and the patients status.
5.Determination of 16 Metallic Elements in Water for Injection by ICP-MS

Objective:To establish the methods for the determination of Mg,Al,K,Ca,Cr,Cu,Zn,As,Se,Ag,Cd,Sn,Ba,Pb,Na and Hg in water for injection. Methods:The sixteen metallic elements in water for injection were determined by ICP-MS. The collision cell technology ( CCT) was used to minimize the multiple atomic interference, and the matrix effect and signal drift were compensated by using Li, Sc, Ge, In, Ir and Bi as the internal standards and the samples were directly acidified to detect the metallic elements. Results:The detection limit range of the 16 metallic elements were from 0. 009 to 0. 165 ng·ml-1. The calibration curves showed good linearity (r≥0. 999 0). The recoveries were within the range of 80%-120%(n=6). Conclusion:The method is simple, rapid and accurate. It can be used to determine the contents of metallic elements to reduce the potential exceeding limit risk of toxic metal el-ements in water for injection. The methods also can provide reference for the more strict quality evaluation of water for injection.
6.Analyzing Financial Security Situation of Disease Prevention and Control System Construction in Jiangxi

Objective: To understand current situation and existing problems in financial security of the center for disease control and prevention system in Jiangxi, to provide scientific references for advancing continuous, fast and healthy development of the system. Methods: combining three methods of questionnaire, group discussions and field surveys to investigate the situation of financial income from 112 centers of disease control and prevention in Jiangxi. Results: The government finance investment increased by year, but the funds of daily work and personnel is insufficient, public and regular business funds of some institution still depend on paid service income. Conclusion: It needs to further enhance financial structure adjustment and disease control investment, increase disease control input, change government appropriation method for disease prevention and control institutions, change the government ’ s funding mode to “purchase system” , reasonably pricing public product, the government on behalf of the people buying services according to quality and quantity from the disease prevention and control institutions.
7.The expression changes in IL-6,GP130,JAK1 and STAT1 genes in PC12 cell after combined soman and hypoxia injury

Objective To investigate the complex effect of soman and hypoxia on PC12 cell line as shown by the expression changes in IL-6/GP130, JAK1 and STAT1. Methods A cell model of intoxication by combining effect of hypoxia and soman was reproduced, and it was divided into four groups: control, soman intoxication, hypoxia combined with soman intoxication, and Genistein inhibition groups. PC12 cells were cultured in RPMI 1640 and were treated with NGF (50ng/ml) for seven days. The differentiated PC12 cells were then exposed to hypoxia in an incubator containing 5% CO2, 95% N2 and (or) incubated with soman (20?mol/L) for 2, 6, 12 or 24 hours. The expression levels of IL-6/GP130, JAK1, STAT1 mRNA and protein were assessed by RT-PCR and Western blot in PC12 cells. The products were sequenced by Sanger's double strand DNA sequence determination. Results In soman intoxicated group, the expression levels of IL-6/GP130, JAK1, STAT1 mRNA and protein were elevated in PC12 cell, reaching the peak level at 12 hours, and then lowered at 24 hours, but remaining higher than that of control group. In combined soman intoxicated and hypoxia group, the expression levels of IL-6/GP130, JAK1, STAT1 mRNA and protein reached the peak value at 6 hours, being higher than that of control group, soman intoxicated group and Genistein inhibition group. It was shown that the sequences of the products as amplified by RT-PCR were the same as that found in the GenBank. Conclusion Soman intoxication or (and) hypoxia up-regulate the expression of IL-6/GP130. Both hypoxic condition and soman treatment can up-regulate the expression of JAK1/STAT1 mRNA and protein in PC12 cells. JAK-STAT pathway may play a role in the mechanisms of brain injury resulted from hypoxia and soman poisoning.
8.Simultaneous Determination of Ibuprofen and Indomethacin in Human Urine by HPLC

OBJECTIVE:To establish a method for simultaneous determination of ibuprofen and indomethacin concentration in human urine. METHODS:The urine samples were precipitated by acetonitrile. HPLC method was adopted. The determination was performed on Discovery C18 column with mobile phase consisted of acetonitrile-20 mmol/L ammonium acetate solution(85:15,V/V, pH value adjusted to 3.5 with glacial acetic acid)at the flow rate of 1.0 mL/min. UV detection wavelength was set at 220 nm. The column temperature was room temperature,and sample size was 80 μL. RESULTS:The linear range of ibuprofen and indometha-cin were both 0.1-50.0μg/mL(r=0.9996,0.9995,n=3). The limits of quantitation were both 0.1μg/mL,and the limits of detec-tion were both 0.03 μg/mL. RSDs of inter-day and intra-day were all lower than 10%(n=5),and accuracy ranged 94.7%-97.2%. The extraction recoveries of ibuprofen and indomethacin were 89.5%-91.8% and 90.2%-92.4%(all RSDs<10%,n=15),respec-tively. CONCLUSIONS:The method is simple and rapid with high selectivity,sensitivity and accuracy. It is suitable for simultane-ous determination of ibuprofen and indomethacin concentration in human urine.

Elsholtiza Bodinieri Vaniot has the food and medicinal value. The whole plant contains aromatic oils, flavonoids, triterpenoids, phenolic constituents, β-carotene, vitamin C and inorganic elements, and is characterized by antibacterial, antioxidant, hypolipidemic and other kinds of activity. However, few researches are on the extraction method and activity of the effective ingredient of Elsholtiza Bodinieri Vaniot. This article reviewed the chemical constituents of Elsholtiza Bodinieri Vaniot, with a purpose to provide references for further study and development.
