1.Simultaneous Determination of Five Major Lignans from Schisandrae Sphenantherae Fructus and Schisandrae Chinensis Fructus by Microemulsion Liquid Chromatography

Objective To develop a stable and sensitive microemulsion liquid chromatography method by using oil-in-water microemulsion mobile phase;To simultaneously determine five major active lignans, namely, schizandrin, schisandrol B, schisantherin A, deoxyschizandrin, and schisandrin B, in Schisandrae Sphenantherae Fructus and Schisandrae Chinensis Fructus. Methods The quantitative determination was conducted by using Kromasil C18 column (250 mm × 4.6 mm, 5μm). The influence of the composition of the microemulsion system on separation effect and retention time was studied. The method was optimized to sodium dodecyl sulfonate 3.3%(SDS, W/V), n-octanol 1.2% (V/V) and n-butanol 6.6% (V/V) in 20 mmol/L phosphate buffer (adjusted to pH 7.0 with orthophosphoric acid and sodium hydroxide). The flow rate was set at 0.7 mL/min. The compound peaks were detected by UV at 254 nm. The column temperature was 25 ℃. Results Under the optimized conditions, baseline separation of 5 lignans was achieved in 20 min, and the separation was satisfying. Schizandrin, schisandrol B, schisantherin A, deoxyschizandrin, and schisandrin B were in linear relation within the ranges of 2.0-155.0, 8.3-166.7, 8.8-175.8, 8.3-166.7, 8.9-177.1μg/mL (r>0.999). The limit of detection ranged from 0.06μg/mL to 0.18μg/mL, and the limit of quantitation ranged from 0.17μg/mL to 0.55μg/mL. The average recovery values were between 97.22%and 100.8%. Conclusion This method is simple, accurate and reliable, and can be successfully applied to simultaneous determination of five major active lignans in 13 batches of Schisandrae Sphenantherae Fructus and Schisandrae Chinensis Fructus from different producing areas in China.
2.Lobectomy by video-assisted thoracoscopic surgery versus thoracotomy for clinical stage Ⅰ / Ⅱ non-small cell lung cancer: a multi-center comparative study

Objective To compare safety,completeness and long-term results between lobectomy by video-assisted thoracoscopic surgery(VATS) and thoracotomy for clinical stage Ⅰ / Ⅱ non-small cell lung cancer(NSCLC).Methods Between July 2010 and December 2011,673 patients with clinical stage Ⅰ / Ⅱ NSCLC from 11 hospitals underwent pulmonary lobectomy(425 VATS,248 thoracotomy).Patients were matched by propensity score matoh method to create two comparable groups,402 patients remained eligible for analysis,with 201 patients in each group.Perioperative variables and follow-up results were compared between the two groups.Results Patients characteristics were comparable between the two groups,except for a slight lower preoperative FEV1,FEV1% and FEV1/FVC in the VATS group (P < 0.05).There was one postoperative death in each group.There was no significant difference in operative time,mediastinal lymph nodes stations/numbers dissected and postoperative drainage time between the two groups.The operative blood loss was lower in the VATS group(P < 0.001),the mean hospital stay was shorter in the VATS group(P < 0.001).Postoperative pneumonia/pulmonary atelectasis rate was lower in the VATS group than in the thoracotomy group (5.1% vs.10.4%,P < 0.05).Median follow-up time was 20 months in both groups.One-year and two-year recurrence-free survival was 90% vs.88% and 80% vs.70%,respectively(P =0.163).One-year and two-year overall survival were 96% vs.95% and 88% vs.85%,respectively(P =0.226).Conclusion For clinical stage Ⅰ / Ⅱ NSCLC,VATS lobectomy is a feasible procedure with shorter hospital stay and less complications,while its surgical and oncologic outcome is comparable to that of thoracotomy lobectomy.
3.Changes of lens thickness, anterior chamber depth and influence factors in normal subjects

Background The study on influences of gender,age and axial length (AL) on lens thickness (LT) and anterior chamber depth (ACD) in normal population is still lack in China.Objective This study was to measure LT and ACD in normal subjects and discuss their changes with age,gender and AL.Methods AL,ACD and LT were measured on 866 eyes of 433 normal population with Lenstar LS900 in Tianjin Eye Hospital from March through December in 2013,including 390 eyes in 195 males and 476 eyes of 238 females.ACD/AL,LT/AL and (ACD+LT)/AL were calculated after measurement.The subjects were grouped to the 10-20 years group,21-30 years group,31-40 years group,41-50 years group,51-60 years group and ≥61 years group based on age,or the 22 mm≤AL<24 mm group,24 mm≤AL<26 mm group and AL≥26 mm based on AL,and the measured parameters were compared and analyzed among the groups.Oral informed consent was obtained from each subject or child contutor prior to any medical examination.Results AL,ACD and LT values were (24.41±1.37),(2.89±0.42) and (4.05±0.52) mm in the males,and those in the females were (23.83±1.31),(2.72±0.40) and (4.20±0.44) mm,showing significant differences between them (t=6.278,6.102,-4.595,all at P=0.000).No significant differences were found in ACD,LT,ACD/AL,LT/AL and (ACD+LT)/AL values between the right eyes and the left eyes (t=-0.713,0.186,-0.754,0.296,-0.168,all at P>0.05).As the increase of the AL,ACD was remarkable deepened but LT was declined,with the considerable difference among the 22 mm ≤ AL<24 mm group,24 mm ≤ AL< 26 mm group and AL ≥ 26 mm group (F =176.640,94.569,both at P =0.000).ACD value was higher,but LT value was lower in the 24 mm≤AL<26 mm group or AL≥26.00 mm group than that in the 22 mm≤AL<24 mm group (all at P<0.05).ACD value was gradually reduced,while LT value was elevated with aging,with significant differences among the different age groups (F =104.531,373.225,all at P =0.000).Significant correlations were found between age and ACD,LT or (ACD + LT)/AL,with the regression equations of ACD (mm) =-0.015 × age + 3.434 (F =-22.979,P =0.000);LT (mm) =0.024 × age + 3.122 (F =44.856,P =0.000),LT/AL =0.001 × age + 0.122 (F=37.982,P=0.000),(ACD+LT)/AL=0.001×age+0.259 (F=22.451,P=0.000).Conclusions Compared with normal females,males have thinner LT and deeper ACD.In the normal eyes,longer AL is,thinner LT is and the deeper ACD is.LT thickens and ACD deepens with aging.

Purpose:To explore the expression of metastasis suppressor gene Kai1 in ovarian epithelial tumor and its significance.Methods:The expression of Kai1 protein in 108 samples of epithelial ovarian tumors and 12 of normal ovarian tissues was examined by S-P immunohistochemical staining.Results:The positive rates of Kai1 protein expression among malignant,borderline,benign and normal tumors were 26.6%,71.4%,73.3% and 83.3% respectively. The Kai1 protein expression in malignant tumors was obviously less than that in benign ones ( P 0.05). 5.EFFECTS OF CYCLOOXYGENASE-2 ON ALCOHOL-INDUCED LIVER INJURY IN RATS

The effects of cyclooxygenase 2 (COX 2) on alcohol induced liver injury in rats were investigated. Rats were randomly divided into three groups: Control group was given with corn oil plus dextrose; Model group with corn oil plus ethanol; Treatment group with corn oil plus ethanol and a selective COX 2 inhibitor celecoxib. All materials were injected into stomach through intragastric tubes. Liver samples were analyzed with histopathology, fatty staining and immunohistochemistry. Glutathione s transferase (GST) activity of liver tissue and plasma were detected. Levels of aspartate aminotransferase (AST) and alanine aminotransferase (ALT) in serum were measured. Compared with the control group, positive fatty staining cells and expression of COX 2 in liver, level of ALT and AST in serum and activity of GST in plasma were all significantly increased in the model group, but the GST activity in liver was decreased. However, these changes were all reversed in the treatment group. The results show that the model of alcohol induced liver injury in rats has been set up successfully, the selective COX 2 inhibitor celecoxib can protect against alcohol induced liver injury in rats and COX 2 may play a role in alcohol induced liver injury
6.Intervention of Tert-butyl Hydroperoxide-Induced Injury in Human Umbilical Cord Derived Mesenchymal Stem Cells with Ginkgo Biloba Extract

Objective To observe the intervention effect of Ginkgo biloba extract EGb761 on tert-butyl hydroperoxide ( t-BHP)-induced injury in human umbilical cord-derived mesenchymal stem cells ( hUC-MSCs) . Methods Proliferation of hUC-MSCs after primary culture was detected by methyl thiazolyl tetrazolium (MTT) assay, and then the optimal concentrations of t-BHP and EGb761 for oxidative stress injured MSC model were screened. Cell apoptosis was assessed by flow cytometry after Annexin V-fluorescein isothiocyanatel propidium iodide ( Annexin V-FITC/PI) staining. Content of malondialdehyde ( MDA) and superoxide dismutase (SOD) activity in hUC-MSCs were evaluated, and the expression levels of p53 and p21Cip1/Waf1 were analyzed by real-time fluorescence PCR. Results Pretreatment with 10~200 mg/L of EGb761 for 3 hours reduced the sensitivity of hUC-MSCs to t-BHP ( 100 μmol/L) induced proliferation inhibition, while EGb761 over 100 mg/L had no significant effect on enhancing the protection of hUC-MSCs . EGb761 at 100 mg/L prohibited hUC-MSCs apoptosis and MDA accumulation in hUC-MSCs induced by 100μmol/L of t-BHP acting for 6 hours, maintained the enzymatic activity of SOD, and decreased the expression of p53 and p21Cip1/Waf1 in hUC-MSCs with t-BHP-induced injury. Conclusion EGb761 is capable of protecting hUC-MSCs against oxidative stress injury, and its mechanism is probably related with the modulation of p53/p21 signal pathway.
7.Analysis of the Impact of Rosuvastatin on Function Changes of Endothelial Cell

OBJECTIVE:To study and analyze the impact of rosuvastatin on function changes of endothelial cell. METHODS:30 patients with coronary heart disease were selected from our hospital during Jun. 2013 to Jun. 2014,and 15 healthy persons were included. They were divided into healthy control group,drug intervention of coronary heart disease group and coronary heart disease group with 15 cases in each group. The levels of asymmetric dimethylarginine (ADMA),NO and nitric oxide synthase (NOS) were determined by ELISA and compared among those groups. Its relationship with OD value was analyzed. After cultured for 4 h and 8 h,human umbilical vein endothelial cells (HUVEC) were divided into 5 groups,i.g. control group 0 μmol/L,0.1 μmol/L group,1 μmol/L group,10 μmol/L group,100 μmol/L group. The change of DDAH mRNA expression in endothelial cells treated with different concentrations of rosuvastatin was detected. RESULTS:The level of ADMA in drug intervention of coronary heart disease group were significantly lower than in coronary heart disease group,but still higher than that in healthy control group;NO and NOS levels were significantly higher than coronary heart disease group,but still lower than healthy control group,with statisti-cal significance (P<0.05). According to Pearson method correlation analysis,OD value was positively correlated to ADMA,NO and NOS. 4h after HUVEC culture,RT-PCR showed that mRNA expression of DDAH were increased in 0.1 μmol/L,1 μmol/L, 10μmol/L and 100μmol/L rosuvastatin groups,there was statistically significant difference between 10μmol/L and 100μmol/L ro-suvastatin groups and control group (P<0.05). 8 h after HUVEC culture,RT-PCR showed mRNA expression of DDAH were in-creased in 0.1 μmol/L,1 μmol/L,10 μmol/L and 100 μmol/L rosuvastatin groups,and there was statistically significant difference between 10 μmol/L rosuvastatin group and control group(P<0.05). CONCLUSIONS:Rosuvastatin is a new type of lipid-lowering drugs,which has a good protective effect on endothelial cells,it is worthy of attention in the clinic.
8.Determination of Ephedrine Hydrochloride in Zhike Pingchuan Tangjiang by HPLC

Objective To establish the method for detemining the content of ephedrine hydrochloride in Zhike Pingchuan Tangjiang by HPLC. Methods Diamonsil ODS1 C_(18) Column was used with acetonitrile -0.1% phosphoric solution (with 0.1% triehylamine) (3 : 97) as the mobile phase, the detection wavelength as 205 nm, and flow rate was 1.0 mL/min. Results The calibration curve was linear at the range of 0.12~ 0.96 ?g for ephedrine hydrochloride and linear equation was Y= 109759X+3792.8, r=0.9998. The average recovery was 98.4% and RSD was 0.87% (n =5). Conclusion This method was simple, accurate and proper, with good reproducibility. It can be used for quantitative analysis of ephedrine hydrochloride in Zhike Pingchuan Tangjiang.
9.Effects of Tongxinluo capsule on sciatic nerve apoptosis in spontaneous type II diabetic KK/Upj-Ay mice and mechanism research.

To investigate the effects of Tongxinluo capsule on sciatic nerve apoptosis in spontaneous type II diabetic KK/Upj-Ay mice, in order to explore its mechanism for improving diabetic peripheral neuropathy (DPN). KK/Upj-Ay mice were selected as the DPN animal model and randomly divided into the model, Tongxinluo low, middle and high group (1, 2, 4 g x kg(-1)). C57BL/6 mice were selected as the control group. Mice were given intragastrically for 12 weeks. Paw withdrawal latency, motor nerve conduction velocity (MNCV) and sensory nerve conduction velocity (SNCV) were detected. Apoptotic rate were detected by FCM. Bcl-2, Bax, Caspase-3 mRNA and protein expression in sciatic nerve were examined by Real-time PCR and Western blot. p38MAPK, p-p38MAPK expression were examined by Western blot. In this study,the authors found that Tongxinluo capsule could increase paw withdrawal latency, MNCV and SNCV. Apoptotic rate of sciatic, the expression of Bax and caspase-3 were lower, while Bcl-2 expression was higher in Tongxinluo group than those in model mice. The expression of p-p38MAPK significantly decreased in Tongxinluo group. The results showed that Tongxinluo capsule has protective effects on diabetic peripheral neuropathy of mice via inhibiting cell apoptosis and suppressing the expression of p-p38MAPK.

10.Evaluation of echocardiagraphy in patients with aortic stenosis undergoing transapical aortic valve implantation

Objective To explore the value of echocardiography in patients with aortic stenosis who underwent transapical aortic valve implantation (TAAVI).Methods Fifteen high-risk patients with severe aortic stenosis were chosen in the present study,all of which received TAAVI in Fuwai Hospital Chinese Academy of Medical Sciences from June 2014 to March 2015.The left ventricular end-diastolic diameter (LVEDD),left atrial diameter (LAD),left ventricular ejection fraction (LVEF),effective orifice area (EOA),mean aortic pressure gradient (MPG),as well as artificial valve function of patients were measured at different time points (before operation,discharge,1 month and 3-6 months after operation) by echocardiography.The data were analyzed using one-way ANOVA analysis with SPSS software,and multiple comparisons were done using LSD student t test.Results The data from preoperative echocardiography indicated severe aortic stenosis in the 15 patients,with the average level of EOA as (0.55± 0.28) cm2 and MPG as (58.93± 14.96) mmHg (1 mmHg=0.133 kPa).Moderate paravalvular aortic regurgitation was observed in one patients,who was then received a second implantation.There was no significant difference between LVEDD,LAD,and LVEF when the patients were at discharge,1 month and 3-6 months after operation.The prosthetic valves were stable and conformed by echocardiography,while paravalvular leak regurgitation (1-2 mm) was observed in 7 patients.One patient died of other reasons.Compared with preoperative data,the EOA increased while MPG decreased when the patients were at discharge,1 month and 3-6 month after operation (t=6.619,7.357,6.401,all P < 0.001;t=9.523,9.687,5.932,all P < 0.001).Conclusion With careful patient screening and selection,TAAVI can be an effective treatment for high-risk severe aortic stenosis patients,in which echocardiography plays an important role during the surgery and follow up.
