The methods for aptamer selection have been modified and developed in recent years. Selection efficiency has been increased due to the improvement in separation of bound and unbound nucleic acids; applicability of aptamers has been improved due to the development of primer-free SELEX and the utilization of short oligonucleotide library; more research requirements have been satisfied due to the functional modifications of aptamer selection methods. All the methodological advances enhance the potential value of aptamers in basic researches and clinical applications.
4.Accuracy of femoral components sizing predicted by standardized digital templating in total knee arthroplasty

Objective To determine the accuracy of femoral components sizing predicted by standardized digital templating in total knee arthroplasty (TKA).Methods Fifty consecutive patients (50 knees),who underwent primary TKAs for endstage osteoarthritis,were prospectively studied.The intra-operative and radiographic data were collected.All operations were performed by the same surgical techniques with PS type,open box Vanguard Complete Knee System.All patients underwent lateral and AP radiography of the involved knee under fluoroscopy before and after surgery.The distal femoral anteroposterior dimension (DFAP) were measured and the femoral components size were predicted on preoperative radiographs by two different methods:measurement of DFAP did not include (group A) the cartilage thickness of the medial posterior condyle or included that (group B).Cutting errors were corrected gradually,and DFAP was measured consequently.The most appropriate size was chose after each step respectively based on postoperative radiographs.The accuracy of femoral size predicted under different conditions was compared within two groups.Results During correction of cutting errors,the correct rate ranged from 18% to 44% in group A and from 26% to 34% in group B,the accuracy within one size ranged from 54% to 84% in group A and from 58% to 84% in group B.The cartilage thickness of medial posterior condylar,external rotation of femoral component,under-resected of anterior condylar,flexion of femoral component,and over-resected of posterior condylar can change the DFAP by 1.97±0.85 mm,1.56±2.06 mm,1.15±1.31 mm,-2.86±1.52 mm,and-0.87±0.77 mm,respectively.Conclusion Variation of intraoperative cutting errors and the cartilage thickness of medial posterior condyles can influence the accuracy of templating to some extent.Standardized digital radiography templating cannot predict femoral sizes accurately.
5.Determination of content and encapsulation efficiency of leuprolideacetate microspheres

Objective To establish a determination method for leuprolide acetate microspheres. Methods HPLC was performed on Inertsil ODS-SP (150×4.6 mm×5μm) with mobile phase consist of 0.1 mol/L of Ammonium Dihydrogen Phosphate(adjust its pH to 7.00±0.05 with Ammonium Hydroxide) and Acetonitrile in ratio of 3:1(V/V),and the flow rate was 1.0 mL/min.The wavelength was 220 nm and column temperature was 30℃. The injection volume was 20 μL. Results The linear range of leuprolide acetate was 20.0-160.0μg/mL (r=0.9999) with an average recovery of 99.80%,RSD=0.63%(n=9). Conclusion The method of HPLC was accurate,reliable and specific, which could be used to determinate the assay of leuprolide acetate microspheres and for quality control of microspheres.
6.Design of a controlling switch set based on RS-232.

As a common computer keyboard and a mouse can't meet the requirements of some special occasions, this paper introduces the design of a controlling switch set based on RS-232 for capturing the image at the medical image workstation. Its working principles, circuit, assembly together with its associated software controller are given in detail. The control switch set features easy manufacturing, low cost and a good reliability.

7.Design of print out function for a multi-parameter patient monitor.

This paper describes the design of the printing output function for a multi-parameter patient monitor by using the graphic plot function of a Laser Printer. The ECG data are preprocessed with a notch filter and interpolation algorithm. A logical page is constructed in the extended memory for virtual page printing. This monitor is able to output a satisfied printing with ECG waveforms of high quality.

8.Agreement between cardiac index measured with arterial pressure-based cardiac output and pulmonary artery catheter during cesarean section in parturients with congenital heart disease and severe pulmonary hypertension

Objective To determine if the cardiac index (CI) measured with arterial pressure-based cardiac output (APCO) agrees with that measured with pulmonary artery catheter (PAC) during cesarean section in parturients with congenital heart disease and severe pulmonary hypertension.Methods Thirty parturients with congenital heart disease and severe pulmonary hypertension,scheduled for elective cesarean section,were enrolled in the study.APCO and PAC were used for cardiac output and other parameters of hemodynamics monitoring during operation in all patients.Continuous epidural anesthesia was performed.CI was recorded immediately before epidural administration (T1),at 5 and 10 min after epidural administration (T2-3),immediately before delivery (T4),and at 2 and 5 min after delivery (T5-6).The correlation was tested by Pearsone correlation analysis.CI derived from APCO and PAC was compared by Bland-Altman analysis for agreement.Results CI measured with PAC was significantly higher than that measured with APCO (P < 0.05).Compared with the baseline value at T1,no significant difference in CI measured with PAC and APCO was found at T11-T4 and T6 (P > 0.05),and CI measured with PAC and APCO was significantly increased at T5 (P < 0.05).Bland-Ahman analysis showed poor agreement between CI measured with the two methods.The correlation analysis showed that there was positive correlation between CI derived from the two methods at T1-6 (the correlation coefficient was 0.93,0.95,0.94,0.89,0.96 and 0.91 at T1-6,respectively,P < 0.05).Conclusion CI value obtained with APCO agrees poorly with that obtained with PAC during cesarean section in parturients with congenital heart disease and severe pulmonary hypertension,but agrees well in monitoring the changing trend of CI.
9.Effect of different degrees of intermittent hypoxia on inflammatory cytokines and adipokines in rats

To study the effect of different levels intermittent hypoxia on inflammatory cytokines and adipokines in a rat model. 160 male Wistar rats were divided into five groups, three groups with intermittent hypoxia (5% O2:IH-1 group;7. 5% O2: IH-2 group;10% O2: IH-3 group), continuous hypoxia group (10% O2, CH group);and the intermittent normal oxygen control group(IN). Before the exposure and at the second, fourth, sixth, and eighth week after the exposure, eight rats in each group were selected randomly with blood samples collected. Blood glucose was measured by glucose oxidase-peroxidase ( GOD-POD ) reagents, double sandwich enzyme-linked immunosorbent assay ( ELISA) was used to detect serum concentration of insulin, tumor necrosis factor alpha( TNF)-α, interleukin ( IL)-6, leptin, and adiponectin and adipocyte nuclear factor kappa B ( NF-κB) levels. In IH-1, IH-2, and IH-3 group, blood glucose, insulin, TNF-α, IL-6, leptin, and nuclear NF-κB in rats showed consecutive increment after exposure to intermittent hypoxia, and those findings in IH-1 group were higher than those in IH-2 and IH-3 group (P<0. 05), but the level of TNF-α dropped slightly on the 8th week. The level of adiponectin showed consecutive decrease from the outset and its level in IH-1 group was significantly lower than that in IH-2 and IH-3 group(P<0. 05). Cytoplasmic NF-κB levels did not have obvious difference in all these groups(P>0. 05). These results demonstrate that intermittent hypoxia could activate NF-κB and result in extensive changes of proinflammatory cytokines and adipokines in the serum of rats. The release of pro-inflammatory cytokines and adipokines keeps pace with the degree of intermittent hypoxia.
10.Clinical study on treatment of pulmonary interstitial fibrosis with ginkgo extract.

OBJECTIVETo observe the effect of ginkgo extract on pulmonary interstitial fibrosis.
METHODSForty-five patients with pulmonary interstitial fibrosis were randomly divided into two groups, the treated group (n = 30) received ginkgo biloba extract 1 g, three times a day; the control group received prednisone 30 mg, once a day, the therapeutic course for both groups was 3 months. Changes of clinical symptoms, pulmonary function, arterial partial pressure of oxygen, computerized tomography (CT), interleukin-6 (IL-6), interleukin-8 (IL-8) and tumor necrosis factor-alpha(TNF-alpha) in the two groups were observed before and after treatment.
RESULTSThe efficacy of treatment in the two groups showed insignificant difference, clinical symptoms, pulmonary function, arterial partial pressure of oxygen were improved after treatment (P < 0.05), and the levels of IL-6, IL-8 and TNF-alpha significantly decreased after treatment as compared with those before treatment in the two groups. The occurrence of pulmonary infection was less in the treated group than that in the control group (P <0.05).
CONCLUSIONGinkgo is effective in treating pulmonary interstitial fibrosis.
