Objective To establish a method for rapid determination of chlorinated benzene compounds in drinking water.Methods Chlorinated benzene compounds were extracted by purge-and-trap and separated by GC then determined by MS.Results The correlation coefficients of 6 chlorinated benzene compounds were 0.999 1-0.999 8 and the average rates of recovery were 93.0%-97.5%(n=6).The relative standard deviation was 2.45%-4.36%(n=6) and limit of detection 0.007-0.05 ?g/L.Conclusion The method established in the present paper is simple,precious,accuracy and sensitive and applicable to the determination of chlorinated benzene compounds in water.

Objective To take nursing measure through analysis on the risk factors of recurrent stroke in the aged.Methods Total 96 subjects were analyzed retrospectively.The risk factors were compared on the urban and the rural through analyzing their clinical characteristics.Results There were more diabetes mellitus and blood lipid disorder and obesity in urban than those in rural.There was also a difference in hypertension between the urban and the rural.Conclusion The recurrence of stroke in the aged is owing to many factors.The different measure should be taken by different reasons.

6.Preliminary assay of beta-amyloid binding elements in heart-beneficial recipe

OBJECTIVE: To explore whether there are beta-amyloid protein (Abeta) binding elements in heart-beneficial recipe (HBR, a compound traditional Chinese herbal medicine), which can ameliorate the cytotoxicity of Abeta. METHODS: The extract of HBR and Abeta(1-40) were co-precipitated, and the Abeta(1-40) in pellets was detected by immunoblotting. Affi-gel-Abeta(1-40) was constructed, and Affi-gel-Abeta(1-40) affinity elements from the extract of HBR were analyzed by high-performance liquid chromatography (HPLC). The assay of lactic dehydrogenase (LDH) release from the primary cultured rat cortex neurons was used to evaluate the cytotoxicity of Abeta(1-42), and the protection effects of the HBR serum and the Affi-gel-Abeta(1-40) treated HBR serum. RESULTS: Immunoblotting examination showed Abeta(1-40) could be co-precipitated with components of HBR following co-incubation, and the amount of Abeta(1-40) within pellets decreased when the HBR extract was diluted. Abeta(1-40) affinity elements from the extract of HBR, eluted from Affi-gel-Abeta(1-40) by glycine solution (pH=2.5), could be detected by HPLC-fluorescent detector system. The analysis of LDH release showed that exposure of neurons to 5 micromol/L Abeta(1-42) for 48 h caused a significant increase of LDH release in either a serum free or 10% serum contained culture condition (P<0.01). The rat HBR serum was able to suppress Abeta(1-42) induced LDH release (P<0.05), whereas Affi-gel-Abeta(1-40) treated HBR serum still maintained the ability to attenuate Abeta(1-42) induced LDH release although the effect was somewhat decreased compared with Affi-gel treated HBR serum. CONCLUSION: There are Abeta affinity components in HBR, which could not increase the Abeta cytotoxicity, but might be able to inhibit the cytotoxicity of Abeta. The results implied that the exploration of Abeta affinity elements from Chinese medicinal recipe which is effective for Alzheimer disease, might be an important direction in Alzheimer disease therapeutic research area.

Objective To study the function of the X\|zone cells in the 5 days old male mouse adrenal gland and the relationship between the function and the sexual differentiation. Methods The technique of in situ RP\|PCR was used to detect the expression of type Ⅰ and Ⅲ 17? HSD mRNA. Results In the cells of X\|zone which located in the interface between adrenal crotex and medulla, there were expressions of type Ⅰ and Ⅲ 17? HSD.Conclusion\ The X\|zone cells of adrenal gland in immature mouse could synthesize androgen, enhanced the male sexual differentiation together with the androgen synthesized from the testis.\;[
8.THE REGULATORY EFFECT OF hCG ON STEROIDOGENIC ENZYMES IN ADULT MOUSE LEYDIG CELLS

Objective To study the regulation role on molecular level of hCG on P450scc,P450c17 and 3?HSD in adult mouse Leydig cells. Methods Leydig cells of adult mice were cultured in vitro for 1 and 8 hours with or without hCG,then P450scc,P450c17 and I,VI total 3?HSD mRNA levels were measured respectively by the semi quantitative RT PCR as well as Scal restriction endonuclease incised methods.Meanwhile,the testosterone contents were measured in two groups. Results 1.In stimulated group Leydig cells cultured for 1 and 8 hours with hCG,the testosterone contents were higher significantly than those in control groups(1 hour P 0.05),but the ratio of type VI/I 3?HSD mRNA gradually increased. Conclusion hCG can stimulate the transcription of P450scc,P450c17 and type VI 3?HSD in adult mouse Leydig cells.
9. Effects of Taxus polysaccharide on myocardial NADPH oxidase mRNA, and SOD, and MDA in myocardial ischemia-reperfusion injury model of Beagle's dogs

Objective: To investigate the influence of Taxus polysaccharide on myocardial NADPH oxidase mRNA, SOD, and MDA in myocardial ischemia-reperfusion injury (MIRI) model of Beagle's dogs. Methods: Beagle's dogs (30) were randomly divided into Sham operation, model, low-dose Taxol polysaccharide, high-dose Taxol polysaccharide, and Carvedilol groups, six in each group, and MIRI model was established after 7 d administration. Stained with HE, pathological changes in myocardial tissue were observed with light microscopy; NADPH oxidase p47phox (NCF-47K) mRNA expression was detected with in situ hybridization; Myocardial SOD and MDA contents were detected with colorimetry. Results: Under light microscope, serious cardiac cells disorders, fracture, karyopyknosis, and significant eosinophilic cytoplasm existed, mild to moderate hyperemia and edema and inflammatory cell infiltration appeared in the model group. However, in the high-dose group, myocardial cells disorders and destruction existed in a lesser extent, karyopyknosis and eosinophilic cytoplasm decreased significantly, and some organizations were still mild hyperemia and edem. Myocardial NCF-47K mRNA expression of Beagle's dogs in the model group and low-dose group were significantly higher than that in the Sham-operated group (P<0.01), NCF-47K mRNA expression of Beagle's dogs in the high-dose group was significantly lower than that in the model group (P<0.05). And SOD content in myocardial tissue of the model group and the treatment group was significantly lower than that in the Sham-operated group (F<0.01). SOD content in the high dose group was significantly higher than that in the model group (P<0.01). MDA contents in myocardial tissue in the model and low-dose groups were significantly higher than those in the Sham-operated group (P<0.05). MDA content in high-dose group was significantly lower than that in the model group (P<0.05). Conclusion The Taxus polysaccharide may reduce ischemia-reperfusion myocardial injury by reducing the expression of NADPH oxidase, increasing SOD activity, and reducing injury of O2.- and other oxygen free radicals in myocardial cell.
10.Experimental study on preventive effect of Radix Paeoniae Rubra to restenosis after carotid balloon injury in high fat-diet rabbits.

OBJECTIVETo observe the preventive effect of Radix Paeoniae Rubra (RPR) to restenosis after carotid balloon injury in rabbits.
METHODSThe rabbit model of carotid balloon injury was established adopting Clowes method, and treated with extract of RPR. Component of new genesic intima and expression of proliferating cell nuclear antigen (PCNA) and macrophage was determined by immunochemical stain. The collagen of type I was detected by special staining for blood vessels and the area of new genesic intima was measured by image assay system.
RESULTSRPR could remarkably decreased the PCNA positive expression and inhibit the proliferation of collagen type I and reduce the generating of new intima.
CONCLUSIONRPR has significant preventive effect on the restenosis after carotid ballon injury in high fat-diet induced atherosclerotic rabbits.
