1.Determination of Adenosine in Chongcaoqizhi Granule by HPLC

Objective To develop a RP-HPLC method to determine adenosine in Chongcaoqizhi granule. Method The separation was performed in a Kromasil ODS-1 column (250 mm?4.6 mm, 5 ?m) with a mobile phase of phosphate buffer solution (pH 6.5) - methanol (85∶15). The flow was 0.7 mL/min and the detective wavelength was set at 260 nm. Results The standard adenosine showed a good linear correlation at range of 1.016~12.192 ?g/mL. The linear regression equation and correlation coefficient (r) were Y =169339X -82318, r =0.9990, respectively. The average recovery of the loading was 98.02% and the RSD was 1.72%. The average content of adenosine was 12.45 ?g/g. Conclusion This method is accurate, reproducible and highly selective, and can be used for quality control of Chongcaoqizhi granule.
2.The analysis of the quality of life of elders in the community of Shangqiu and the affecting factors

Objective To understand the quality of life of elders in the community of Shangqiu and analyze the affecting factors to provide a scientific evidence which could improve their quality of life.Methods The general questionnaire and World Health Organization Quality of Life Brief Evaluation From were used among 116 elders in two communities of Shangqiu.Results The quality of life of elders was in low level.The mainly factors that influencing the quality of life of elders in the community of Zhengzhou were age,education status,economic income,hobbies and interest,family relation,physical exercise.Conclusions The factors that influence the elders in community are various.To improve quality of life of elders,community workers should take relevant measures and encourage family members to participate in caring elders.

6.Intervention effect of Dachengqi Granule on apoptosis of small intestine smooth muscle cells in rats with multiple organ dysfunction syndrome.

OBJECTIVETo observe the intervention of Dachengqi Granule (DG) on the apoptosis of small intestine smooth muscle cells (SMCs) in rats with multiple organ dysfunction syndrome (MODS) and its mechanisms.
METHODSHealthy 100 adult Wistar rats were randomly divided into the control group (n =20), the MODS model group (n =40), and the DG group (n =40).E. coli suspension was peritoneally injected to rats in the model group and the DG group to establish bacterial peritonitis induced MODS model. DG at 1 mL/100 g was administered by gastrogavage to rats of the DG group, twice daily for 3 successive days. Twenty-four hours after modeling, the proximal segment of intestine was taken and stained by using terminal-deoxynucleotidyl transferase mediated nick end labeling (TUNEL) and immunohistochemistry. Changes of apoptosis quantity of SMCs and the expression of Bcl-2 associated X protein (Bax), B cell lymphoma/leukemia-2 (Bcl-2) and cytochrome c protein (Cyt c) in mitochondrial apoptotic signaling pathway were observed.
RESULTSCompared with the control group, the apoptosis quantity of SMCs and the expression of Bax and Cyt c protein significantly increased, and the expression of Bcl-2 protein significantly decreased in the MODS model group (P <0.01). Compared with the MODS model group, the apoptosis quantity of SMCs and the expression of Bax and Cyt c proteins significantly decreased, and the expression of Bcl-2 protein significantly increased in the DG group (allP <0.01).
CONCLUSIONDG could inhibit apoptosis of SMCs through suppressing activation of mitochondrial apoptotic signaling pathway in intestinal SMCs, thus promoting the recovery of the gastrointestinal motility function in rats with MODS.

7.Study on Inpatient Oxygen Transfer Monitoring System Based on Wireless Network

Objective To design a wireless monitoring system for locating oxygen supply terminals and measuring oxygen intake value of patient.Methods The locating and measuring system was built by using Mesh network of Zigbee technology and AODV algorithms based on hardware featuring MC9S08 microprocessor and MC13192.Results The oxygen supply terminals sent the location and oxygen flux information to monitoring nodes,while the monitoring server read data from all nodes in turn and save to the database.Conclusion The wireless network based on Zigbee technology can effectively monitor the utilization information about oxygen supply terminals and the value of patient oxygen intake.
8.Neuroprotective effect of Gangliosides on cerebral ischemia reperfusion injury in rats

Objective To investigate the effect of Gangliosides on brain protection after cerebral ischemia reperfusion injury in rats. Methods The middle cerebral artery (MCA) focal cerebral ischemia /reperfusion rat models were used by thread occlusion method. The cerebral infarct volume, neurologic function deficits and neurons apoptosis in cerebral ischemic region were observed following focal cerebral ischemia 90 min (IR 0 h) and ischemia 90 min / reperfusion 24 h (IR 24 h) by injecting Gangliosides and normal saline intraperitoneally, respectively. Results A significant reduction of infarct volume was found in the Gangliosides-treated group compared with the control group. 9. Panning and identification of humanized antinuclear antibody Fab fragment

Objective: To prepare humanized antinuclear antibody Fab fragment. Methods: The reconstructed humanized antinuclear antibody (ANA) Fab phage display library was enriched by 4 rounds of panning and was identified by indirect ELISA method. Phasmid DNA isolated from positive clones was deprived of g III gene. After self-ligation the recombinant plasmid was used to transform E. coli. XL1-Blue, then XL1-Blue was induced by IPTG to product soluble human antinuclear antibody Fab fragment. Finally, soluble human antinuclear antibody Fab in the supernatant was identified by indirect ELISA method and immunofluorescence. Results: The eluted phages were enriched by more than 200 folds after 4 rounds of panning. Two positive clones were isolated from the ANA Fab library. Electrophoresis after Xho I digestion proved that the self-ligation was successful after deletion of g III gene. The results of indirect ELISA indicated that the 2 positive clones of Fab had specific anti-dsDNA activity. Indirect immunofluorescence showed homogeneous fluorescence within nuclei of Hep2 and monkey hepatic cells and in the Crithidia kinetoplast. Conclusion: We have successfully prepared soluble, specific human antinuclear antibody Fab fragment, which pares a way for preparation of high affinity antinuclear antibody Fab fragment.
10.The review of newborn hearing screening program in neonatal intensive care unit.

The incidence of hearing impairment in neonatal intensive care unit (NICU) was much higher than that of well-baby nursery. The incidence of the former was 2%-4%, whereas that of the latter was 0.1%-0.3%. Furthermore, the incidence of auditory neuropathy spectrum disorder, progressive and delayed hearing loss was also higher than those of other infants. Therefore, the newborn hearing screening program in NICU has become an important part of pediatric audiology. In this paper, we reviewed the previous studies and suggested the special procedure of hearing screening and following-up which based on the physiological and pathological characteristics of NICU in order to detect hearing impaired as early as possible.
