1.THE EFFECT OF CHIP ON THE BIOSYNTHESIS OF DNA, RNA, AND PROTEIN OF EAC CELLS IN VITRO

The effect of CHIP [cis-dichloro-trans-dihydroxy-bis-isopropy-lamine platinum (Ⅳ)] on the biosynthesis of DNA, RNA, and pro-tein of Ehrilch ascites carcinoma ( EAC ) cells in vitro was studied. 3H-TdR, 3H-UR, 3H-Leu were used as precursors of cellular macro-molecules to study the effect of CHIP on the biosynthesis,with CDDP [cis-dichlorodiammineplatitinum (Ⅱ)] as a positive control. The experimental results showed that CHIP, similar to CDDP, inhibited the biosynthesis of DNA, RNA and protein, and has a strong antitumor activity. The inhibitory effect on DNA biosynthesis lasted 40 hours indicating that CHIP might induce permanent damage in DNA template and thus reduced the mitotic division of tumor cells. Furthermore, in contrast to the general view, the in vitro experimental results showed that CHIP, a tetravalent platinum coordination compound, exerted its antitumor activity directly in vitro.
2.Role and mechanism of gastric cancer stem cells in gastric cancer invasion, metastasis and angiogenesis

BACKGROUND:Although tumor stem cel s and their differentiated vascular cel s, which are not sensitive to chemotherapy and molecular targeted therapy, reduce overal blood supply of the tumor, these cel s facilitate the invasion and metastasis of the tumor, eventual y resulting in treatment failure. OBJECTIVE:To investigate the effect of gastric cancer stem cel s in stomach cancer invasion and metastasis as wel as its mechanism in angiogenesis. METHODS:A total of 20 gastric cancer stem cel s and 20 normal gastric cancer cel s SGC7901 were cultured in the medium with DMEM/F12 (1:1), 2%B27 and 20μg/L epidermal growth factors or cultured in 10%PBS, respectively. Invasion of gastric cancer stem cel s was detected by sphere formation assay, colony formation assay and Transwel invasion assay. Levels of mRNA and protein expression of CD44 and E-cadherin were measured to observe tumor metastasis. Besides, angiogenesis was observed by cel scratch test, Transwel migration assay and ring test. RESULTS AND CONCLUSION:Gastric cancer cel s adhered to the medium wal , and gastric cancer stem cel s suspended in the medium, which appeared to be spindle interstitial form. Number of gastric cancer tumor spheres was significantly lower than that of gastric cancer stem cel spheres in the sphere formation assay and colony formation assay (P<0.05). Number of gastric cancer stem cel s through the basement membrane was significantly higher than that of gastric cancer cel s (P<0.05). And the cel migration distance and ring number were significantly increased in the gastric cancer stem cel s as compared with the gastric cancer cel s (P<0.05). In conclusion, the invasion, migration and angiogenesis abilities of gastric cancer stem cel s are stronger than those of gastric cancer cel s, which can further promote the occurrence and progression of gastric cancer.
3.Monitoring of Color Doppler Ultrasound in Arteria-Venous Fistula in Patients with Uremia.

Objective To evaluate the effect of color Doppler ultrasound monitoring in artificial arteria-venous fistula in patients with uraemia.Methods 25 cases were examined with upper limbs arteriovenous fistulas with uremia and analysed by CDFI and 2D.The fistula and blood vessel was observe and hemodynamics changes were examined.Results Among 25 patients,18 cases showed good filling state of the vessel,3 cases with anastomoses stenosis,2cases with thrombus and 2cases with venous dilatations.Conclusion CDFI is an effective supplementary detective methods in treating hemodialysis patients with uremia.
4.Expression of ICAM-1,LFA-1 and VEGFR-3 in Brest Cancer and its Relationship with Lymphatical Metastasis

Objective To explore the influence of ICAM-1 and LFA-1 on the breast cancer metastasis and provide scientific base for studying the molecular mechanism of breast cancer metastasis by observing the expression of ICAM-1(intercellular adhension molecule1) and LFA-1(Lymphocyte function related antigen 1) in different time different metastasis stage of breast cancer. Methods We observed the expression of ICAM-1 and LFA-1 in breast cancer tissue as well as the expression of ICAM-1、LFA-1 and VEGFR-3 in Lymphatic vessel endothelial cell of nearby the breast cancer tissues from the patients of the operation in the defferent stages with the methods of HE staining and immunohistochemistry staining. Results The positive rate of ICAM-1 and LFA-1 expression in cancer cell and cancer nest of breast cancer was increased gradually along with the increase of clinical stages and the occurrence of lymph node metastasis. And the positive rate of ICAM-1、LFA-1 and VEGFR-3 expression in lymphatic vessel endothelial cell of nearby the breast cancer tissues was increased gradually too along with the increase of clinical stages and the occurrence of lymph node metastasis,which has the positive correlation,and it has the positive correlation that the ICAM-1 and LFA-1 expressed in Lymphatic vessel endothelial cell of nearby the breast cancer tissues(P0.05).Conclusion The ICAM-1 and LFA-1 were related with breast cancer lymph metastasis,and the ICAM-1 and LFA-1 promoted breast cancer lymph metastasis through synergism each other.The increase or decrease of ICAM-1 and LFA-1 expression in the breast cancer tissue and lymphatic vessel endothelial cell of nearby the breast cancer tissues could as the data target to display certain instruction function for prevention and treatment of breast cancer lymph metastasis.The Lymphatic vessel quantity of nearby the breast cancer tissues was increased along with tumor progress, which may be related with breast cancer lymph metastasis.VEGFR-3 has the high specificity in the lymphatic vessel endothelial cell expression,and may act as the lymphatic vessel as the lymphatic vessel endothelial cell specific marker.
5.Effect of Xiao Chaihu Decoction on the Expression of Cyclooxygenase-2 in Endometriosis Rats

Objective To explore the therapeutic mechanism of Xiao Chaihu Decoction for endometriosis in rats. Methods The rat models of endometriosis were treated with Xiao Chaihu Decoction, Danazol, and Xiao Chaihu Decoction plus Danazol for 4 weeks,respectively. And then cyclooxygenase-2(COX-2) expression in endometrium and endometriotic tissue was observed by immunohistochemistry method. Results The expression of COX-2 in the endometriotic tissue was lower than that in endometrium in Xiao Chaihu Decoction group and Xiao Chaihu Decoction plus Danazol group, while the results in Danazol group and control group were opposed to those in the above two groups. Conclusion The mechanism of Xiao Chaihu Decoction for rats endometriosis may be related to the decrease of COX-2 expression in ectopic Endometrium, thus reducing the formation of prostaglandins.
6.Chemical constituents in immature fruits of Momordica charantia

Objective To study the chemical constituents in the immature fruits of Momordica charantia. Methods Isolation and purification were carried out by macroporous absorption resin and silica gel, and compounds were identified and elucidated by spectral and chemical methods. Results Ten compounds were obtained from alcohol extract and five of them were determined as germanicyl acetate (Ⅰ), aglycone of momordicoside Ⅰ (Ⅱ), aglycone of momordicoside L (Ⅲ), charantin (Ⅳ), and ?-sitosterol (Ⅴ). Conclusion For all the first time, compound Ⅰ is found in the plants of Momordica L., compound Ⅱ is found as novel natural product in this plant, and compound Ⅲ is found in China.
7.Expression of plasma bone morphogenetic protein-4 in patients with coronary heart disease and intervention effect of rosuvastatin

Objective To investigate the changes of plasma bone morphogenetic protein-4 (BMP-4) levels in patients with coronary heart disease (CHD) and rosuvastatin intervention effect on BMP-4 level.Methods Fifty-two patients with CHD and 35 health people were enrolled in this study as CHD group and control group.ELISA method was used to detect the concentration of plasma BMP-4.Analyzed the relationship between plasma BMP-4 and blood lipids,flow-mediated dilation (FMD),nitric oxide (NO),cyclooxygenase-2 (COX-2),malondialdehyde (MDA) and superoxide dismutase (SOD).And observed the changing of plasma BMP-4 before and after rosuvastatin intervention.Results Plasma BMP-4 level in CHD patients was (7.53 ± 1.20) μg/L,higher than that of control group ((3.81 ± 0.79) μ g/L,t =3.541,P =0.006).After rosuvastatin treatment,plasma BMP-4 level in CHD patient was decreased from (7.53 ± 1.20) μg/L to (5.40± 0.98) μg/L (t =1.436,P =0.001).Plasma BMP-4 level was positively correlated with COX-2,MDA,low-density lipoprotein cholesterol,total cholesterol (r =0.395,0.350,0.274,0.288 respectively,P < 0.01 or P <0.05).But,it was negatively correlated with NO,high-density lipoprotein cholesterol,SOD,FMD (r =-0.291,-0.253,-0.476,-0.320 respectively,P <0.01 or P <0.05).COX-2,SOD and FMD were independent risk factors of plasma BMP-4 in patients with CHD.Conclusion Oxidative stress and endothelial dysfunction are in patients with CHD.Rosuvastatin treatment can remarkably reduce plasma BMP-4 level,alleviate vascular endothelium injury induced by oxidative stress and improve endothelial function in patients with CHD.
8.Development of an HBV genotyping method by nucleic acid strip

Objective To develop an rapid, visuable method to detect the genotypes of HBV.Methods According to the published full-length sequence with known genotypes in GenBank, the specific primer and biotin-lablled probe were designed. We established a nucleic acid strip method for genotyping hepatitis B virus(HBV) isolates among 150 HBV infected patients and 20 healthy controls, and compared the results with those obtained by real-time PCR. Results There was 34.00% genotype B , 61.33% genotype C, 4.00% genotype B/C in 150 samples, respectively, while the remaining 0.67% unknown. The results for this assay were high comparable to the method of real-time PCR. Conclusion With the similar sensitivity and specifity when compared with real-time PCR, this rapid method is suitable to the clinical setting.
9.Effects of hydroxyethyl starch 130/0.4 used as priming fluid for cardiopulmonary bypass on plasma colloid osmotic pressure and lactic acid concentration in infants undergoing cardiac surgery

Objective To investigate the effects of hydroxyethyl starch 130/0.4 (HES) used as priming fluid for cardiopulmonary bypass (CPB) on the plasma colloid osmotic pressure (COP) and lactic acid (LAC) concentration in infants undergoing cardiac surgery.Methods Forty infants of either sex with age ≤6 yr undergoing cardiac surgery with CPB were randomly divided into 2 groups (n =20 each): HES group and control group. The left radial artery and the right internal jugular vein were cannulated for blood pressure (BP) and the central venous pressure (CVP) monitoring. Arterial blood gases, blood LAC concentration, hemoglobin (Hb), hematocrit (Hct), mean arterial pressure (MAP) and nasopharyngeal temperature were measured and recorded immediately before and 5 min after aortic cross-clamping, at the end of CPB and operation. Plasma COP was measured before induction of anesthesia (T1), at 5 and 30 min of CPB (T2 and T3, respectively), before routine ultra-filtration (T4), at the end of CPB (T5) and2 h in ICU (T6).Results The plasma LAC concentration was significantly lower and the COP significantly higher in HES group than in control group (P<0.05 or 0.01).The plasma LAC concentration increased after aortic cross-clamping, reached the peak at the end of CPB and then declined at the end of operation, but was still higher than that before aortic cross-clamping in both groups. Plasma COP was significantly decreased during CPB as compared with the baseline at T1, but increased at T6 in both groups.Conclusion Using HES 130/0.4 as pdming fluid for CPB can effectively improve plasma COP and reduce blood LAC level in infants undergoing cardiac surgery with CPB.
