2. Identification of metabolites of Pulsatilla saponin D in intestinal microflora of rats in vitro by UPLC-Q-trap-MS

Objective: To identify the metabolites of Pulsatilla saponin D (PSD) in the intestinal microflora of rats in vitro. Methods: The sample of PSD incubated in intestinal microflora of rats in vitro was analyzed by a ultraperformance liquid chromatography/hybrid triple quadrupole linear ion trap mass spectrometry (UPLC-Q-trap-MS). The data were processed with Analyst 1.5 software, and the metabolites of PSD were confirmed by comparing the retention time and MS/MS spectra. The possible structures of metabolites were proposed by analyzing the proposed fragmentation pathway. Results: PSD (M0, parent) and seven metabolites were identified, including hederagenin 3-O-α-L-rhamnopyranosyl-(1→2)-α-L-arabinosyl (M1), hederagenin 3-O-α-L-glucopyranosyl-(1→4)-α-L-arabinosyl (M2), hederagenin (M3), hydroxylation of PSD (M4), methylation of PSD (M5 and M6), and dehydrogenation of PSD (M7). Conclusion: PSD has a wide range of metabolic in the intestinal microflora of rats in vitro.
3. Identification of the metabolites and effective components of flos scabiosae extract in rats using UHPLC-Q-Tof-MS/MS method

OBJECTIVE: To identify the metabolites of Flos scabiosae extract in rats after oral administration for exploring its metabolism mode in vivo. METHODS: Welch Ultimate UHPLC C18 column (2.1 mm×100 mm, 1.7 μm) was used for the analysis, and the column temperature was maintained at 25℃. Gradient elution was conducted with mobile phase consisting of 0.1% acetic acid water (A) and acetonitrile (B) at a flowing rate of 0.35 mL·min-1. The injection volume was 6 μX. Urine, plasma, and feces samples were collected during 0-24 h or 0-12 h after oral administration of Flos scabiosae extract to rats. The samples and control samples were analyzed by UHPLC-Q-T MS/MS in the scanning mode to acquire the full-scan chromatograms. The data acquisition and processing analysis were conducted using compound retention time, precise molecular mass, elemental composition, isotopic abundance, and mass spectra with Peak View v1.2 data processing software. Metabolites were identified by comparison of chromatograms of the urine, feces and plasma samples after administraion of Flos scabiosae extract with those of the blank biological samples and the herb extract. RESULTS: The developed method is applicable to the analysis and identification of metabolites of Flos scabiosae extract in biological matrices after oral administration. Based on the investigation of Flos scabiosae extract in rats, 28 parent compounds with 31 metabolites were detected in rat urine, feces, and plasma. CONCLUSION: In this study, the developed method is simple and efficient. The main metabolism pathways of Flos scabiosae extract in rats may include oxidation, demethylation, glucuronidation, and sulfation. This study provides a novel pattern and illumination for the research of material basis of Flos scabiosae.

5.The Quality Control of Leukodepleted Red Cell

Objective To explore the quality control method of leucodepleted red cell concentrate (LDRC) preparation ,to choice an optimal processing , in order to raise the quality of LDRC,so as to ensure the efficiency and safty for the preparation application.Methods 62 bag red cells concentrates suspensin(each 150ml,preparation from 200ml whole blood) were investigated.By linear regression multianalyzed the effects of some factors on efficiency for removal of leukocytes from red cell concentrates.Free hemoglobin value(n=8) was detected before and after leukodepleted filtration.1% samples were tested and bacterial cultured,and observed the clinical adverse effects after LDRC tranfusion.Results ⑴The plasma and platelet in red cell contentrate before filtration,the time of filtration had influence significantly on the result of leukocyte depletion.⑵The percentage of leukodepleted red cell in which residual leukocyte less than 5?10 6/unit and 1?10 6/unit were 100% and 46.9%. The percentage of leukodepleted red cell in which the leukocyte removal efficiency more than 2log(99%)and 3log(99.9%) were 100% and 56.1% .After controlling the time of filtration,the percentage of leukodepleted red cell in which residual leukocyte less than 1?10 6/unit was up from 46.9% to 90.91% and the percentage of leukodepleted red cell in which the leukocyte removal efficiency more than 3log(99.9%)/bag was up from 56.1% to 78.1%.⑶Red blood cell recovery rate was (90.48?4.73)%. There was no significant change of free hemoglublin before and after filtration.⑷No bacteria were detected by germiculture.⑸Evident side effects was no seen after transfusion.Conclusions During the course of removal of leukocytes from red cell concentrates by filtration, the sustained quality control is necessary for improving the quality of leukodepleted red cell.So enforcing aseptic manipulation rigorously and controlling the components of blood,the time of blood storage and filtration appropriately are quite important.
6.A bibliometrics study of literature on medical image processing for the past ten years.

We searched and retrieved literature on the topic of medical image processing published on SCI journals in the past 10 years. We then imported the retrieved literature into TDA for data cleanup before data analysis and pro cessing by EXCLE and UCINET to generate tables and figures that could indicate disciplinary correlation and research hotspots from the perspective of bibliometrics. The results indicated that people in Europe and USA were leading researchers on medical image processing with close international cooperation. Many disciplines contributed to the fast development of medical image processing with intense interdisciplinary researches. The papers that we found show recent research hotspots of the algorithm, system, model, image and segmentation in the field of medical image processing. Cluster analysis on key words of high frequency demonstrated complicated clustering relationship.

7.Clinical significance of iron metabolism and variation of serum Prealbumin in patients with tumor associated anemia

Objective To explore the occurrence of anemia, anaemia type, iron metabolism situation,variation of serum Prealbumin (PA), and the relationship among them in cancer patients. Methods Three hundred and seventy cancer patients, admitted from July 2007 to April 2010 to our hospital, were enrolled into the study. The clinical data of all subjects were analyzed retrospectively. Results The incidence of anemia in 370 patients was 55.67% (206/370). Among all 206 anemia cases,129 cases had mild anaemia and 77 cases had middle to severe anaemia. In the last group ( n = 77 ), we found significant decrease in serum iron level ( [ 8. 37 ± 6. 09 ] μmol/L) and increase in serum ferritin level ( [ 474. 57 ± 327. 58 ] μg/L); and the correlation between serum iron and the anemia degree( Ps < 0. 05 ). However, we found no significant differences of serum ferritin level between the groups with different degree of anemia(P >0. 05). Among all 206 anemia cases ,187(90. 77% ) patients had a low level of serum PA, but no relationship between the degree of anemia and the drop of serum PA ( P > 0. 05 ). Conclusion The anemia was very popular in cancer patients, which had correlation with iron metabolism situation but not PA.

Technology readiness level (TRL) refers the general maturity level of a single technology or technical systems (including technology,component or subsystem) during the process of development.Base on the original TRL proposed by NASA,with reference to medical device TRL modified by US Department of Defense and NATO,this study modified TRL definition and description for advanced medical devices with consideration of R&D,review and approval characteristics of advanced medical devices in China.
9. Technology optimization for extracting total favonoids from Gynostemma pentaphyllum by cellulase

Objective: To optimize the extracting process of total flavonoids from Gynostemma pentaphyllum. Methods: The total flavonoids were extracted with ethanol 95% after the cellulose treatment to G. pentaphyllum, and the effects of enzyme dosage, enzymatic treatment temperature, enzymatic treatment time, and pH value on the total flavonoids yield was studied with response surface method. Results: The optimal enzymatic extracting process of total flavonoids was as follows: enzyme dosage 120 U/mL, enzymatic temperature 49 °C, enzymatic time 150 min, and pH value 3.9, and the total flavonoids yield reached 0.357%. Conclusion: Cellulase is suitable for the total flavonoid extraction from G. pentaphyllum.
10. Preparation and Preliminary Evaluation of Paclitaxel-loaded TPGS-CR Polymeric Micelles

OBJECTIVE: To prepare paclitaxel loaded D-α-tocopheryl polyethylene glycol 1000 succinate(TPGS)-modified carboxymethyl chitosan-rhein polymeric micelles (PTX/TPGS-CR PMs) and preliminarily evaluate their performance. METHODS: PTX/TPGS-CR PMs was prepared by dialysis method, and the preparation procedure of PTX/TPGS-CR PMs was optimized by single factor with the drug loading, encapsulation rate and particle size as the indicators, then the optimized preparation procedure was verified. The safety of PTX/TPGS-CR PMs was initially investigated by the hemolysis test and the vascular irritation test. The cytotoxicity of PTX/TPGS-CR PMs in Hela cells was studied by MTT assay. Cell uptake experiments were performed by laser confocal microscopy and flow cytometry to investigate the uptake of PTX/TPGS-CR PMs by Hela cells. RESULTS: The particle size and PDI of PTX/TPGS-CR PMs prepared by the optimized preparation were (197.3±4.4) nm and (0.131±0.021), respectively. The Zeta potential was (-31.8±0.5) mV. The drug loading and encapsulation efficiency were (48.20±3.03)% and (87.26±4.91)%, respectively. The hemolytic test results showed that the hemolysis rate was less than 1.71%. No obvious irritation was observed after intravenous injection. The cytotoxicity of PTX/TPGS-CR PMs in Hela cells was concentration-and time-dependent. Cell uptake experiments showed that PTX/TPGS-CR PMs could be efficiently uptake by Hela cells. CONCLUSION: The PTX/TPGS-CR PMs has high drug loading and encapsulation efficiency, good safety. And they exhibite slightly better antitumor activity in vitro than Taxol®.
