Objective:To discuss the effect and mechanism of autophagy inhibitor 3-MA on arsenic trioxide inducing apoptosis of acute T-cell leukemia cell line Jurkat cells. Methods:Proliferation inhibition of Jurkat cells treated with arsenic trioxide was detected by XTT.Morphological characteristics of Jurkat cells treated with different concentrations arsenic trioxide were observed by electron mi-croscope.Microtubule-associated protein 1 light chain 3B (LC-3B) protein expression was detected by Western blot and flow cytome-try.Apoptosis rates of Jurkat cells treated with 3-MA combining arsenic trioxide were detected by flow cytometry using AnnexinV-FITC/PI double staining. Results:Arsenic trioxide inhibited the growth of Jurkat cells in a dose and time dependence. We observed different morphological characteristics of autophagy , apoptosis and necrosis accompanying more autophagosomes in Jurkat cells which were treated with arsenic trioxide 2.5,5,10 μmol/L after 24 h.LC3B mean fluorescence intensity (MFI)relative multiples were(3.1±0.2) fold,(4.6±0.31)fold,(34.2±4.5)fold with 5 μmol/L arsenic trioxide treated Jurkat cells 0,24,48 h,and the P values between each of the two groups were less than 0.05,which increased depending time consistently with the growth inhibition rates.LC-3B protein expression gradually increased treated Jurkat cells with arsenic trioxide after 24 h,48 h.The growth inhibition rate (60.6±8.3)%was significantly different treated with arsenic trioxide combining 3-methyl adenine ( 3-MA ) while it was ( 33.4 ±9.1 )% treated with arsenic trioxide alone, however, LC-3B protein expression gradually decreased.Jurkat cell apoptosis rate ( 44.96 ±3.60 )% was significantly increased treated with arsenic trioxide combining autophagy inhibitor(3-MA) while it was (2.94±0.26)% treated with arsenic trioxide alone, and this difference was statistically significant. Conclusion: 3-MA increased apoptosis rates of Jurkat cells inducing by Arsenic trioxide and it may be related with inhibition of autophagy and induction of apoptosis.

Objective To investigate the effect of interferon-α (IFN-α) on expression of major histocompatibility complex (MHC) class I chain-related protein A (MICA) in the malignant hematopoietic cell lines. Methods The myeloid leukemia cell lines K562 and B-cell lymphoma cell line Raji were treated with IFN-α.The expression of MICA was measured by RT-PCR and immunohistochemical staining at mRNA and protein level. The cytotoxicity of human NK cells to the IFN-α treated malignant hematopoietic cells was detected by MTT method. Results After being treated with 1000 U/ml IFN-α,up-regulation of MICA mRNA in Raji and K562 cells were respectively found in 24 h and 48 hours (t =17.016,P <0.05; t =5.616,P <0.05).After being treated with IFN-α 72 hours,up-regulation of MICA mRNA in K562 and Raji were found at the dose of 500 U/ml and 1000 U/ml dose (t =6.622,P <0.05; t =9.071,P <0.05).The mRNA and protein expression of MICA were up-regulated by IFN-o in the two malignant hematopoietic cells in a time and dosedependent manner. After being treated with 1000 U/ml IFN-o for 72 hours, the susceptibility of the two malignant hematopoietic cells to NK cytolysis was significantly increased (t =20.016,P <0.05; t =7.969,P <0.05).Moreover,the up-regulated susceptibility can be blocked by anti-MICA antibody. Conclusion IFN-α can up-regulate the MICA expression in the malignant hematopoietic cell lines and thereby enhance the susceptibility to cytolysis of NK cells.

Objective To provide microanatomic basis for direct microsurgical operation on the region of Dorello's canal. Methods The structures of the region of Dorellos canal were observed and measured on 30 sides of adult cadaverical heads under microscope. Results ①Dorello's canal was an osteofibrous canal composed of Gruber ligament, petrous apex and the lateral border of the upper part of clivus. The roof of Dorello's canal was mainly composed of Gruber ligament. The maximal distance from roof to bottom of Dorello's canal was 2.14 mm±0.70 mm. ②Abducens nerves passed through the lateral 1/3(56.7%) or intermedial 1/3(43.3%) of Dorello's canal to the cavernous sinus. The nutrient arteries of abducens nerves were originated from the dorsal meningeal arteries, and 83.3% of them traversed Dorello's canal by one stem, medially to abducens nerves. ③The point of abducens nerves entering cavernous sinus was 3.97 mm±1.83 mm posterior to the ascending portion of intracavernous carotid artery, 3.11 mm±0.95 mm medial to the Meckel's cave. Conclusion In the region of Dorello's canal, the positions of abducens nerves and their nutrient arteries are steady, and they should be protected during operation.

Action of electromagnetic radiation exerting on human body has been a concerned issue for people. Because electromagnetic waves could generate an electric stress in a discontinuous medium, we used the finite difference time domain (FDTD) as calculation methods to calculate the electric stress and its distribution in human head caused by high-frequency low-power electromagnetic environment, which was generated by dual-band (900 MHz and 1 800 MHz) PIFA antennas with radiated power 1 W, and we then performed the safety evaluation of cell phone radiation from the angle whether the electric stress further reached the human hearing threshold. The result showed that there existed the electric stress at the interface of different permittivity organization caused by the two kinds of high-frequency low-power electromagnetic environment and the maximum electric stress was located at the interface between skin and air of the phone side, and the electric stress peak at skull did not reach the threshold of auditory caused by bone tissue conduction so that it can not produce auditory effects.

Acta Anatomica Sinica 1953;0(01):-
40 human fetuses were used in this study. The range of fetal age varied from 3 months to full terms. All fetuses used were normal and their parents were healthy. The CFU-C from bone marrow, liver, spleen and peripheral blood were studied in vitro with agar culture technique modified by Metcalf.Experiments proved that the CFU-C of bone marrow may be observed in the fetuses in 3rd month, however, it increased rapidly in 4th month and maintained higher level until the end of fetal life. The progenitor cell population was detected at very high level in liver from 4th month to 6th month fetuses. The number of CFU-C of fetal liver were low in 7—10th month. The circulating progenitor cells in peripheral blood of 4, 5 and 6-month fetuses Were on high level but it was lower than that of liver and bone marrow.
