Chinese Journal of Radiation Oncology 2008;17(5):344-348
Objective Renal papillary cell carcinoma(RPCC) has been historically regarded as a radio-resistant malignancy. However, the molecular mechanism underlying its radio-resistance is not well understood. Recently,STAT1, a transcription factor downstream of the IFN-signalling pathway, has been implicated in radioresistance. This study is to investigate the role of STAT1 in "radio-resistant RPCC". Methods The expression of STAT, in 137 human RPCC samples compared with 15 normal kidney tissues was examined by micrearray expression profiling using the Affymetrix HGU133 Plus 2.0 GeneChip oligonucleotide arrays. For in-vltro experiments, human RPCC cell line(SKRC-39), human fibroblast(CCL-116) and human Wiim's tumor cell line(CRL-1441) were used. Western blotting was performed to evaluate total and phosporylated STAT1 expression. RPCC cells were irradiated and compared to controls in clonogenic assays. STAT1 inhibition either with fludarabine or siRNA was done and their effects on radiation-induced cell survival were investigated. Results The STAT1 expression data shows that there was a significant increase in human RPCC when compared to normal kidney tissues (t=44.38,P=0.000). Similarly, the expression of STAT1 was higher in the RPCC cell line when compared to firbroblast and Wilm's tumor cell lines. STAT1 expression was inhibited by beth fludarabine and siRNA. Radiosensitivity in RPCC cell lines was enhanced by both fludarabine and siRNA induced STAT1 inhibition. Conclusions This is the first study reporting the over-expression of STAT1 in human RPCC tissues and human RPCC cell line. Radiesensitization of RPCC is observed via inhibition of STAT1 signaling by fludarabine and siRNA techniques. Our data suggests that STAT1, through IFN-signalling pathway , may play a key role in RPCC radioresistance and manipulation of this pathway may enhance the efficacy of radiotherapy.

Objective: To investigate the relationship between serum leptin concentration and diet nutrition, age and gender. Method: One hundred fifty-eight men and women aged 18-72 y were initially drawn from a community in Jinan City. The parameters measured included height, weight and serum leptin concentrations. In addition, a 3-d food record was used to represent usual food intake by the participants. Results: The mean serum leptin concentration in males was 8.38?6.31ng/ml, while in females 24.98?13.29 ng/ml. Serum leptin level in males was significantly correlated with age, but not in females. Respectively, serum leptin level in males and females were not correlated to dietary factors. After adjusted for BMI, serun leptin level in females was positively correlated with dietary intake of fat, MUFA and PUFA. Conclusion: Serum leptin level in female is higher than that in male. Age is correlated to serum leptin level in males. The relation of dietary intake of fat, MUFA and PUFA to serum leptin concentration only existed in women.

Abstract: Objective To investigate the impacts of vaccination with inactivated SARS-COV-2 vaccine on the clinical manifestations and serological responses of COVID-19 patients infected by Delta and Alpha variants. Methods Clinical and experimental data of 341 confirmed SARS-COV-2 patients were collected from The Eighth Affiliated Hospital of Guangzhou Medical University May 1- September 30, 2021. The subjects were divided into Delta and Alpha variant group according to virus variants, and were divided into vaccinated group and unvaccinated group according to whether they had received inactivated COVID-19 vaccine or not. The clinical manifestations and serological responses of patients with Delta and Alpha variant, and vaccinated and unvaccinated patients with Delta and Alpha variants were compared. Results Totally 253 patients were infected with Delta variant (103 vaccinated and 150 unvaccinated patients), and 88 patients were infected with Alpha variant (21 vaccinated and 67 unvaccinated patients). The proportion of asymptomatic infection in Delta variants group was significantly lower than that in Alpha variants group (P<0.01). Delta variant group of vaccination rates and vaccine breakthrough infection rate was 40.7% (103/253) and 22.9% (58/253), were higher than Alpha variant group was 23.9% (21/88) and 8.0% (7/88), difference was statistically significant (χ2= 8.009, 9.484, P<0.01). The proportion of cough and fever in Delta variant group was higher than that in Alpha variant group (both P<0.01), the peak viral load was higher than that of Alpha variant group (P<0.01), the virus duration was longer than that of Alpha variant group (P<0.01), the levels of SAA, CRP and IFN were higher than those of Alpha variant group (all P<0.05), CD4+T cell count was lower than that of Alpha variant group (P<0.05), IgG and IgM levels were lower than those of Alpha variant group (both P<0.01). The proportion of moderate COVID-19 in the vaccinated group was lower than that in the unvaccinated group (P<0.01). In these two variants, the peak viral load of vaccinated group was lower than that of the unvaccinated group (both P<0.01), the duration of virus was shorter than that of unvaccinated group (both P<0.01). The levels of SAA, CRP and IL-6 in the vaccinated group were lower than those in the unvaccinated group (all P<0.05), CD4+T cell level was higher than that of unvaccinated group (both P<0.05), IgG and IgM level were higher than those in unvaccinated group (both P<0.05). Conclusions Delta variant can lead to higher viral load and more severe disease course, which is associated with vaccine breakthrough infection. Inactivated vaccines for COVID-19 can reduce severe illness and death by reducing viral load, disease duration and inflammatory response through humoral and cellular immune mechanisms.
