5.Recombinant human erythropoietin inhibiting apoptosis of human mesenchymal stem cells induced by H2O2 in vitro
Chinese Journal of Organ Transplantation 2012;33(8):470-473
ObjectiveTo investigate the effects of recombinant human erythropoietin (rhEPO) on the apoptosis of human mesenchymal stem cells induced by H2O2 and related cell signaling pathways. Methods After being cultured in vitro, human MSCs were treated with rhEPO.Phosphorylation of ERK1/2,p38 MAPK and PI3K/Akt was detected by using Western blotting. One h after pretreatment with1U/ml rhEPO,cells were cultured in the presence of1mmol/L H2O2 for1h.Cell morphology was observed under the inverted microscopy.Cell apoptosis was determined by using flow cytometry, migration assay was performed in Transwell chambers, and adhesion assay was performed by plastic dishes.ResultsRhEPO could increase phosphorylation of PI3K/Akt pathway in human MSCs,but reduce phosphorylation of p38MAPK.RhEPO had no obvious effect on ERK1/2 pathway and total proteins of Akt and p38MAPK.RhEPO could decrease apoptosis of human MSCs induced by H2O2 (P<0.01) and the inhibitory effect was abrogated by Ly294002 but not anisomysin. Conclusion RhEPO could protect MSCs from apoptosis. Activation of PI3K/Akt pathway was involved in the effect of rhEPO on apoptosis.
6.Social Connectedness in Physical, Mental and Social Health
The Singapore Family Physician 2016;42(1):31-35
We are constantly reminded to take care of ourselves. We need a healthy and balanced diet, with regular exercises, and good quality rest. However, research has shown that social connectedness also plays an essential and critical function to our physical and mental well-being. Many studies have shown that lack of social connection is a predictor of social isolation, resulting in greater detriment to health. Strong social connectedness is associated with increased longevity, strengthening of ones’ immune system, faster recovery from disease and, hence, a lengthening of one’s life. Studies have also shown that people who feel more connected to others in relationships tend to generate a positive feedback loop in communicating and maintaining healthy social and emotional well-being with others in the family, workplace and community. This decreases the propensity to antisocial behaviour, isolation behaviour or poor mental health. Individuals with strong connectedness were observed to have lower rates of anxiety and depression; and higher self-esteem and were more empathic towards self and others, resulting in better emotional and psychological well-being. Social connectedness is a broad topic and can be reviewed from various perspectives. This article will review the concept from a psychological perspective of self, exploring the importance of practicing compassion intervention in social connectedness and self-care.
7.Effects of panax notoginseng saponins pretreatment on the expression of wnt4 protein in renal ischemia reperfusion injury in rats
Tianjin Medical Journal 2017;45(5):前插2,472-476
Objective To observe the effect of panax notoginseng saponins (PNS) pretreatment on the expression of wnt4 protein in renal ischemia reperfusion injury in rats. Methods Seventy-two male SD rats were randomly averaged into control group (CON group), renal ischemia-reperfusion group (IR group) and PNS group. Rats in CON group were removed the right kidneys, and the left kidneys were exposed for 30 minutes, and then the incisions were closed. Rats of IR group were removed the right kidneys, and the left renal artery were clamped for 30 minutes, and then the incisions were closed. Rats in PNS group were injected PNS (150 mg/kg) via tail vein for 3 days before experiment, and the rest steps were the same with IR group. After 24 h, 48 h and 72 h reperfusion, 8 rats in each group were killed, and blood and kidney samples were collected to examine the serum creatinine (Scr) and blood urea nitrogen (BUN). The left renal tissues were stained with HE. Pathological changes were observed under light microscopy. The score of renal tubular injury was evaluated. Immunohistochemistry was performed to detecte the expression of wnt4 in kidney tissue. The expression of wnt4 was detected by Western blot assay. Results There was significant renal injury in IR group. The renal tubular injury score, Scr and BUN reached the peak levels of injury at 24 h, which were then decreased gradually, but at 72 h still significantly higher than those of CON group (P<0.05). The renal tubular injury score, Scr and BUN values at different time points were significantly lower in PNS group than those of IR group (P<0.05). The results of wnt4 immunohistochemistry and Western blot assay were consistent, which showed that the expression of wnt4 peaked at 24 h in IR group and PNS group, but the expression levels were significantly higher in PNS group than those of IR group at each time point (P<0.05). Conclusion PNS could protect kidney from ischemia-reperfusion injury, which may be related to the enhanced expression of wnt4 protein.
8.The linear position tracking servo system using a linear voice-coil motor
Journal of Pharmaceutical Analysis 2008;20(3):164-167
More and more linear servo systems have been used in servo applications. Direct drive technology can greatly increase the bandwidth and the tracking accuracy. A position servo-system based on linear voice-coil motor was designed for one linear oscillation movement application. Besides the conventional position, speed and current control loops, the speed and acceleration feed-forward control of command position signal were also used. The experimental test proved the correctness of the design, and the system can track the given periodic sinnsoid position command signal of 15Hz with high accuracy. The linear voice-coil motor is very suitable for short stroke position tracking application with high dynamic response.
