1.Detection of circulating tumor cells and its clinical application

Circulating tumor cell (CTC) is a kind of malignant tumor cells shed from the primary tumor into the circulation caused by spontaneity or surgical operation and puncture. With the development of CTC analysis recently, the detection methods have been gradually diversified. As a new biomarker, CTC plays a significant role in the diagnosis, therapeutic effect monitoring and prognosis of tumors. This paper mainly introduces the enrichment and analysis technology of CTC, and explore the relationship between CTC and tumors.

4.Expression and significance of PTEN in esophageal squamous cell carcinomas

Objective To study on the relationship of PTEN with clinicopathological parameters and prognosis of esophageal squamous cell carcinoma patients.Methods The expression of PTEN was detected in 62 cases of esophageal squamous cell carcinomas by immunohistochemistry.Results(1)PTEN expres- sion is negatively correlated with the depth of invasion,lymph node metastasis,distant metastasis,pTNM stage and degree of differentiation.(2)The difference of survival is significant between high and low expres- sion groups.Conclusion PTEN is correlated negatively with the clinicopathological parameters reflecting the malignant biological behavior,and is one of the significant prognostic predictors by univariate analysis.
5.Self-identity and security of patients with obsessive-compulsive disorder

Objective To study the self-identity and security of patients with obsessive-compulsive disorder (OCD) Methods According to the Chinese Classification of Mental Disorder,Third Edition,63 inpatients and outpatients with OCD and 61 healthy controls were recruited to investigate with self-identity scale(SIS),security questionnaire (SQ),Yale-brown obsessive compulsive scale (Y-BOCS),and self-made questionnaire.Results Compared with controls,patients had lower self-identity (50.81 ± 7.52 vs 55.10 ± 6.86,P < 0.01)and security(50.09 ± 11.65 vs 58.87 ± 10.22,P < 0.01)than healthy controls.Female patients had lower self-identity (48.31 ±7.68 vs 52.57 ±6.98,P<0.05) than male patients.There were correlation between self-identity and Y-BOCS (r =-0.404,P < 0.01),security and Y-BOCS (r =-0.314,P < 0.05),self-identity and course of disease (r =-0.284,P < 0.05),security and course of disease (r =-0.259,P < 0.05) self-identity and security (r =0.698,P< 0.01) Conclusion OCD patients have lower self-identity and security.The self-identity and security reduce with the symptoms getting worse.Low self-identity and security maybe characteristic of OCD patients.
6.Dynamic change of peripheral blood cell in patients with differentiated thyroid cancer before and after 131I treatment

Objective To evaluate the kinetics of peripheral blood cells in DTC patients before and after 131I treatment.Methods A total of 64 patients were divided into 2 groups with different therapeutic doses:high-dose group (3.70-5.55 GBq,n =24) and low-dose group (1.11 GBq,n =40).The WBC,neutrophils (NEUT),lymphocytes (LY),RBC and PLT were counted before operation,before 131I treatment,and on 3 d and 7 d after 131I treatment.One-way analysis of variance and two-sample t test were used to analyze the data.Results The counts of WBC and NEUT in both groups along with the LY in high-dose group varied significantly before,and on 3 d and 7 d after 131I treatment(WBC:high-dose group,(6.30±1.04),(8.86±2.07),(6.59±1.64) × 109/L;low-dose group,(6.65±1.48),(10.17±3.04),(7.17± 1.57) ×109/L; NEUT:high-dose group,(3.75±0.88),(6.42± 1.91),(4.53± 1.54) × 109/L; low-dose group,(3.88±0.90),(7.12±2.77),(4.40±1.17) × 109/L;LY:(2.11±0.67),(2.06±0.74),(1.59±0.49) × 109/L;F values:3.88 to 30.20,all P<0.05).The counts of WBC and NEUT in both groups were significantly higher on 3 d after 131I treatment than that before treatment (all P<0.05).The counts of WBC and NEUT in both groups along with the LY in high-dose group decreased significantly on 7 d compared to that on 3 d after 131I treatment (all P<0.05).The counts of LY in high-dose group also significantly decreased on 7 d after 131I treatment than before treatment(P<0.05).The counts of RBC before 131I treatment and LY on 7 d after 131I treatment were significantly different between the 2 groups(t=2.36,-4.30,both P<0.05).Compared with the counts before operation,LY,RBC and PLT were significantly higher (t values:from-4.92 to-2.45,all P<0.05) during hypothyroid state induced by thyroxine withdrawal before 131I treatment.Conclusions Short-term kinetics of WBC and NEUT present as an increase first followed by a decrease after 131I treatment; while LY of high-dose group presents as a gradually decrease.Hypothyroid state induced by levo-thyroxine withdrawal leads to increased counts of LY,RBC and PLT before 131I treatment.
7.Determination of residual aluminium Ion in Huoxiang Zhengqi pellets by GFAAS with EDTA complexation extraction.

To establish an EDTA complexation extraction pretreatment combining with GFAAS method for the determination of residual aluminium ion in Huoxiang zhengqi pellets without digestive treatment, systematical investigation was made on sample preparation, and EDTA was used for the complexation extraction of residual aluminium ion in samples. The pH, concentration and volume of extraction solution, the temperature and time of microwave extraction, and graphite furnace temperature program were investigated. The results were compared with the microwave digestion. It was showed that, 0.1 g of sample weight was added in 20 mL 0.05 mol x L(-1) EDTA solution (pH 3.5), followed by heating at 150 degrees C for 10 min in the microwave extraction device. The determination of GFAAS was performed at optimized detection wavelength (257.4 nm) as well as graphite furnace temperature program, the detection limits and quantification limits were 2.37 μg x L(-1) and 7.89 μg x L(-1), respectively. The precision (RSD) was less than 2.3%. The average recovery was 96.9% -101%. The present method is easy, rapid and accurate for the determination of residual aluminium ion in Huoxiang zhengqi pellets.

9.Effect of APRIL siRNA in Nude Mice with SW480

Objective To investigate the role of APRIL in SW480 cell line.Methods The CRC model was established in the nude mice,all the mice were divided into 3 groups,the mice were separately treated with APRIL siRNA,pGC-vector and PBS solution.The APRIL mRNA was detected by RT-PCR and the APRIL protein was surveyed by the way of immunohis-tochemistry (IHC).The proteins of TIMP-3,Syndecan-1and MMP-9 also were assessed by IHC.Results ①Tumor mass in the group of nude mice injected with PBS (2.15±0.30 g)was significantly higher than the injection APRIL siRNA group (0.95±0.15 g,P<0.05),and was not statistically significant (P>0.05)compared with the injection of empty vector group (2.20±0.25 g).②APRIL mRNA/18S rRNA ratio (2.48±0.25)in the group of mice injected with PBS was signifi-cantly higher than the injection APRIL siRNA group (0.39±0.15,P<0.05),and was not statistically significant (P>0.05)compared with the injection of empty vector group (2.51±0.30).③SW480 cells injected with APRIL siRNA signifi-cantly inhibited invasion and metastasis.TIMP-3 Allred scores in three groups were 7.70±0.35,1.10±0.16 and 1.15± 0.12,Syndecan-1 protein was 7.80±0.30,1.05±0.20 and 1.10±0.22 MMP-9 protein was 1.20 ±0.10,8.00±0.25 and 8.20±0.20,respectively.Conlusion APRIL was closely connected with the growth and metabasis of CRC.
10.Curative effect of L-carnitine on neonatal abnormal cardiac troponin I caused by asphyxia

Objective To explore the protective effect of L-carnitine on neonates with myocardial injury caused by as?phyxia. Methods Forty-four neonates with myocardial injury caused by asphyxia were randomly divided into L-carnitine treatment group (21 cases) and control group (23 cases). Patients in control group were received routine treatment and pa?tients in treatment group were given L-carnitine 0. 1 g/(kg · d) on the basis of routine treatment for 7 days. Symptoms and physical signs were observed before therapy and during the treatment in two groups. Before and after the treatment, plasma levels of free L-carnitine and cardiac troponin I (cTnI) were detected with the method of colorimetric assay and chemilumi?nescent, respectively. Results The clinical effective rate was significantly higher in treatment group than that of control group (90.48%vs 60.87%, P<0. 05). Compared with the control group, there was a significantly higher plasma concentra?tion of free L-carnitine in treatment group after treatment [(27.00±5.69)μmol/L vs (13.20±3.04)μmol/L, P<0.05]. In treat?ment group, plasma concentration of free L-carnitine was significantly higher after treatment than that of pre-therapy [(14.87 ± 3.95)μmol/L,P<0.05]. Compared with the control group, there was a significantly lower plasma concentration of cTnI after treatment in treatment group [(0.025±0.006)μg/L vs (0.046±0.010)μg/L, P<0.05]. In the treatment group, there was a significant correlation between decreased plasma concentration of cTnI and increased plasma concentration of free L-carnitine (r=0.899, P<0.05). Conclusion Administration of L-carnitine can effectively decrease the abnormal plasma lev?el of cTnI in neonates with myocardial injury caused by asphyxia, and thereby protect the myocardium.
