1.Preparation and identification of the monoclonal antibody against human PSMA7 antigen

Objective To prepare the PSMA7 hybridoma cell lines using classical hybridoma technique and to perform purification and elementary identification of the monoclonal antibodies(McAbs)against PSMA7 antigen for further study.Methods BALB/c mice were immunized with purified recombinant PSMA7 protein.Once the antibody titer of blood taken from mouse tail reached 1∶6 400,a cell fusion was performed between mouse splenic cells and myeloma cells(Sp2/0),and then the hybridoma cell lines secreting monoclonal antibodies against PSMA7 antigen were screened by indirect enzyme linked immunosorbent assay(ELISA).The immunoglobulin subtypes and titer of the monoclonal antibodies against PSMA7 antigen were identified and measured by Western blot analysis and enzyme linked immunosorbent assay,respectively.Results After cell fusion and subcloning,two hybridoma cell lines that stably secreted monoclonal antibodies against PSMA7 antigen were successfully obtained:B013 and B001,which belong to the subtypes of IgG1.The antibody titers in the hybridoma culture supernatant were 1∶128 and 1∶256,and those in the ascites fluid were 1∶25 600 and 1∶32 000,respectively.Western blot analysis and immunohistochemistry showed that the two antibodies can specifically bind with PSMA7 antigen derived from human eucaryotic cells or tissue.Conclusions Two monoclonal antibodies against PSMA7 antigen with high titers and specificity have been successfully prepared.These antibodies can be used for the identification of PSMA7 protein,and may be a useful tool for studying the biological properties of PSMA7 protein.
2.Clinical Observation of Rh-endostatin Combined with Cantharidin Sodium Vitamin B6 in the Treatment of Advanced Non-small Cell Lung Cancer

OBJECTIVE:To investigate the clinical efficacy and safety of rh-endostatin combined with cantharidin sodium vita-min B6 in the treatment of advanced non-small cell lung cancer (NSCLC). METHODS:180 patients diagnosed as advanced NSCLC were divided into group A,group B and group C ,with 60 cases in each group according to random number table meth-od. 3 groups were all given gemcitabine+cisplatin(GP)chemotherapy plan;Group B additionally received Rh-endostatin injection 7.5 mg/m2 intravenously for 3 h,d1-14;group C was additionally given Cantharidin sodium vitamin B6 injection 40 ml intrave-nously,qd,d1-14,on the basis of group B. every 21 days for a cycle,evaluation of therapeutic effect after 2 cycles. The clinical benefit rate,quality improvement rate of life,time to progression (TTP) and the occurrence of ADR were observed in 3 groups. RESULTS:The clinical benefit rate of groups A,B and C were 40.0%,58.3%,71.6% respectively,the quality improvement rate of life in 3 groups were 28.3%,41.7%,56.7% respectively,the differences were statistically significant among those groups(P<0.05). The median TTP of groups A,B and C were 126,190 and 195 days,TTP of groups B and C were significantly longer than that of group A,with statistical significance (P<0.05);there was no significant difference between group B and group C(P>0.05). The rates of leukopenia,thrombocytopenia,nausea and vomiting in group C were significantly lower than those of group A and B,with statistical significance(P<0.05);there was no statistical significance between group A and B(P>0.05). CONCLU-SIONS:Rh-endostatin combined with cantharidin sodium vitamin B6 can significantly improve the effectiveness of chemotherapy in patients with advanced NSCLC on the basis of the GP chemotherapy,while reduce the toxicity of chemotherapy drugs,improve the quality of life and prolong the survival time.
3.Short-term clinical effect of anterolateral minimally invasive approach of total hip replacement

Objective To investigate short-term clinical effect of anterolateral minimally invasive approach of total hip replacement.Methods A total of 35 patients treated by total hip replacement was divided into A and B groups.Group A used anterolateral minimally invasive approach,and group B used standard posterior Moore approach.The clinical data of operation time,amount of bleeding,blood transfusion volume,component position,Harris hip score,and complications of two groups were statistically compared.Results In terms of operation time,group A had certain disadvantages than group B.No significant difference was found between two groups in terms of operation time,annount of bleeding,blood transfusion volume,component position,Harris hip score,and complications.Conclusions In total hip replacement,anterolateral minimally invasive approach was safe and effective compared to standard posterior Moore approach.Doctors should choose their familiar surgical approach in operation.
4.Effects of Oligo-peptide I-C-F-6 in Carapax Trionycis on Rats with Liver Fibrosis Induced by CCl4

Objective To study effects of oligo-peptide I-C-F-6 in carapax trionycis on rats with liver fibrosis induced by CCl4;To discuss its anti-liver fibrosis effects and possible mechanisms. Methods Forty-eight SD male rats were randomly divided into normal control group, model group, bifendate group, and oligo-peptide I-C-F-6 group, 12 in each group.CCl4 was injected intraperitoneally to build rat liver fibrosis model.Oligo-peptide I-C-F-6 group and bifendate group were given subcutaneous injection of oligo-peptide I-C-F-6 (0.12μg/g) or bifendate (0.12μg/g). At the same time, normal control group and model group were giventhe same volume of saline for seven weeks. The levels ofALT, AST,MDA, SOD, IL-4, IL-10 and TNF-α were tested.The histomorphology changes were observed under optical microscopeby HE, and the expressions of transforming growth TGF-β1 were determined by immunohistochemistry.Results Compared with model group, serum levels of ALT and AST were reduced evidently in oligo-peptide I-C-F-6 group. Hepatic content of MDA, IL-4 and TNF-α decreased, while SOD activity and IL-10 were found significantly increased. Liver fibrosis was ameliorated significantly. Hepatic expressions of TGF-β1 were weakly positive.Conclusion Oligo-peptide I-C-F-6 can ameliorate hepatocyte damage of model rats, thus it has anti-oxidative and anti-liver fibrosis effects on liver fibrosis in rats.
5.Analysis of the factors of acute ischemic stroke patients after endovascular treatment in the real world

Objective :
To investigate the 90 day prognosis of acute ischemic stroke patients treated with mechanical thrombectomy in the real world and analyze the related factors of prognosis.
Methods:
A total of 1 033 acute ischemic stroke patients treated with mechanical thrombectomy in tertiary hospitals of Anhui province were retrospectively investigated.The 90 day modified Rankin Scale (mRS) score was evaluated by follow-up telephone,0 ~2 points were good prognosis and 3 ~6 points were poor prognosis.
Results :
A total of 770 patients were followed up,with an average age of (67. 7 ± 11. 8) years.The good prognosis rate was 32. 1% and the mortality rate was 31. 8% . With the increase of age,the good prognosis rate showed a downward trend while the mortality rate showed an upward trend.The best cut-off value for age prediction of 90-day mortality was 69 years old.There was no gender difference in the good prognosis rate between women (70. 6 ± 10. 6) years and men (65. 6 ± 12. 2) years,but females experienced a higher mortality than males (37. 1% vs 27. 9% ) before adjusting for age. Comparison was made among provincial,municipal and county hospitals,only the good prognosis rate of provincial hospitals was significantly higher than that of county hospitals (37.2% vs 15. 6% ) .In the grouping of annual number of endovascular treatment cases,the good prognosis rate of only ≥40 cases group was significantly higher.There was no significant difference in mortality among the groups.
Conclusion
At present,the good prognosis is low and the mortality is high for the whole acute ischemic stroke patients treated with mechanical thrombectomy.Age was positively correlated with poor prognosis and mortality.The higher mortality rate in women than in men is due to the higher average age of onset in women.The good prognosis rate of provincial hospitals and hospitals with annual number of endovascular treatment ≥40 cases is higher than that of other hospitals.
6.Phosphorylated TDP-43 Staging of Primary Age-Related Tauopathy.

Primary age-related tauopathy (PART) is characterized by tau neurofibrillary tangles (NFTs) in the absence of amyloid plaque pathology. In the present study, we analyzed the distribution patterns of phosphorylated 43-kDa TAR DNA-binding protein (pTDP-43) in the brains of patients with PART. Immunohistochemistry and immunofluorescence double-labeling in multiple brain regions was performed on brain tissues from PART, Alzheimer's disease (AD), and aging control cases. We examined the regional distribution patterns of pTDP-43 intraneuronal inclusions in PART with Braak NFT stages > 0 and ≤ IV, and a Thal phase of 0 (no beta-amyloid present). We found four stages which indicated potentially sequential dissemination of pTDP-43 in PART. Stage I was characterized by the presence of pTDP-43 lesions in the amygdala, stage II by such lesions in the hippocampus, stage III by spread of pTDP-43 to the neocortex, and stage IV by pTDP-43 lesions in the putamen, pallidum, and insular cortex. In general, the distribution pattern of pTDP-43 pathology in PART cases was similar to the early TDP-43 stages reported in AD, but tended to be more restricted to the limbic system. However, there were some differences in the distribution patterns of pTDP-43 between PART and AD, especially in the dentate gyrus of the hippocampus. Positive correlations were found in PART between the Braak NFT stage and the pTDP-43 stage and density.
