1.Prevention and treatment of acute graft-versus-host disease in mice by bortezomib

Objective To observe the effect of bortezomib on acute graft-versus-host disease (aGVHD) in an aGVHD model of mice and investigate the related mechanism. Methods Male C57BL/6( H-2Kb)mice were used as donors and female Balb/c (H-2Kd) mice used as recipients. Balb/c mice received total body irradiation (TBI) by 7.0 Gy X-radiation, and randomly divided into five groups. normal (group A), TBI (group B), TBI + bortezomib (group C), TBI + bone marrow cells (BMC) + spleen cells (SC) (group D) and TBI + bortezomib + BMC + SC (group E). The physical signs and the pathological damage of aGVHD, mean survival time, and chimerism were observed in recipients. The NF-κB p65 levels in nuclei of the liver and small intestine tissues of groups A,B and C were analyzed by Western blot. Results ( 1 ) The clinical aGVHD score in group D was (7.37±0. 32), significantly higher than in group E (5.85 ± 0.40) (P<0. 05). Histopathology of the gut, liver and skin illuminated that the Ⅲ-Ⅳ degree GVHD occurred in group D. The occurrence of aGVHD in group E was later than in group D. The symptoms and the pathological damage of aGVHD in group E were milder than in group D. The average survival time in group E was significantly longer than that in group D (P<0.05). The percentage of donor-derived cells in recipient mice was above 90% at day 12 after transplantation; (2) NF-κB p65 levels in nuclei of the liver and small intestine tissues in group B was significantly higher than in group C on the day 1,3 and 5 (P<0. 05). Conclusion Bortezomib can inhibit the activation and expression of NF-κB,which may be the underlying mechanism for it to relieve aGVHD.
2.Effects ofYushen Zhuyun Prescription on Ovarian Morphology and Sex Hormone of Rats with Decreased Ovarian Reservation Induced by Tripterygium Glycosides

ObjectiveTo observe the effects ofYushen Zhuyun Prescription on ovarian morphology and sex hormone of rats with decreased ovarian reservation (DOR) induced by tripterygium glycosides; To explore its mechanism of action.Methods Tripterygium glycosides solution was used to establish DOR model rats by gastric gavage. 48 female SD rats were randomly divided into blank group, model group, positive medicine group andYushen Zhuyun Prescription low-, medium- and high-dose groups.Yushen ZhuyunPrescription groups were given the low, medium, high dosagesYushen Zhuyun Prescription liquid; positive medicine group were given estradiol valerate liquid; model group and blank control group were given normal saline with the same amount, by gavage 2 times per day for 14 consecutive days. The general condition of the rats was observed; ovarian was weighed and the viscera index was calculated; ovarian morphology was observed by HE staining; the levels of serum estradiol (E2), follicle stimulating hormone (FSH) and testosterone (T) were detected by radioimmunoassay; the levels of inhibin B (INHB) and anti Mullerian hormone (AMH) were detected by ELISA.Results Compared with the blank group, ovarian tissue atrophied; the number of follicles was reduced; ovarian index and the level of E2, INHB and AMH decreased; the levels of FSH and LH increased in the model group (P<0.01). Compared with the model group, ovarian tissue morphology improved significantly; the number of follicle and corpus luteum increased; follicular atresia was reduced; ovarian index and the levels of E2, INHB, and AMH increased; the levels of FSH and LH decreased in Yushen Zhuyun Prescription high-dose group (P<0.01). INHB and AMH had significant correlation (P<0.01). ConclusionYushen Zhuyun Prescription can regulate hormone levels, promote the growth and secretion of follicle, and inhibit follicular atresia, thereby improve ovarian reserve function.
3.The relationship between the personality and the mindfulness level of the nursing college students

Objective To explore the prevalence of the personality and mindfulness level of the nursing college students and investigate their correlation.Methods 180 nursing college students were investigated by the scale of NEO and FFMQ.Results The scores of personality trait were in the moderate level.The correlation between the score of the adaptability items and the mindfulness was negatively correlated,the moral sense items,sociability items and agreeableness items and the mindfulness was positively correlated.Conclusions The personality of nursing college students is closely related with their mindfulness level.To perfect the personality of the nursing college students can improve their mindfulness level.
4.Therapeutic effect of dihydroarteminsinin/piperaquine phosphate on Pneumocystis pneumonia in rat model

Objective To research the therapeutic effect of dihydroarteminsinin/piperaquine phosphate(duo-cotecxin)on rats with Pneumocystis pneumonia(PCP),and explain its potential mechanism.Methods The PCP rat model was established through the subcutaneous injection with cortisone acetate twice a week for 6 weeks.The model rats were orally treated with duo-cotecxin at a dose of 40 mg/kg once a day for 3 days,and with cotecxin at a dose of 60 mg/kg once a day for 6 days,respectively.The therapy control group was treated orally with SMZco(sulfamethoxazol 250 mg/kg +Trimethoprim 50 mg/kg),positive control group and normal group were established,respectively.The drug efficacy was based on rat survival rate,increase rate of body weight,lung weight/body weight ratio,mean numbers of the cysts per field in the lung print smear,and the changes of CD4+ T cells,CD8+ T cells,NO,IFN-? and TNF-? in the blood.Results The survival rates and the body weight of rats treated with duo-cotecxin and cotecxin were higher and heavier than those of the positive control group respectively,which approached to those of the group treated with SMZco.The lung weight/body weight ratio,and mean cyst number per field of lung print smear of the rats treated with duo-cotecxin and cotecxin were lower than those of the positive control group,respectively.NO and TNF-? in the blood of the rats treated with duo-cotecxin and cotecxin were lower than those of the positive control group,respectively(P
5.Analysis of epidemic situation of malaria in Jiangsu Province from 2005 to 2014

Objective To understand the epidemic situation and epidemiological characteristics of malaria in Jiangsu Prov?ince from 2005 to 2014,so as to provide the evidence for formulating and adjusting the strategies and measures on malaria elimi?nation in this province. Method The epidemiological data of malaria in Jiangsu province from 2005 to 2014 were collected, and the prevalence situation,infection sources as well as the temporal,regional and population distribution of the cases were an?alyzed. Result A total of 5 069 malaria cases were reported in Jiangsu Province form 2005 to 2014,there were 3 422 cases (67.51%)of vivax malaria,1 497 cases(29.53%)of falciparum malaria,and 150 cases(2.96%)that were unclassified. Among all the reported cases,2 139(42.20%)were local cases,1 131 were(22.31%)imported cases from other places in Chi?na ,and 1 799(35.49%)were imported cases from abroad. There were 3 809 male cases and 1 260 female cases,with a sex ratio of 3.02∶1. The patients were mainly aged from 25 to<60 years(accounting for 67.15%),and were mainly young adult farmers (40.26%)and migrant workers(23.63%),who mainly distributed(accounting for 60.58%)in Xuzhou,Suqian,Huai’an,Su?zhou and Nantong cities. The predominant malaria type from 2005 to 2009 were vivax malaria,while after 2010,it changed to falciparum malaria. Conclusions The malaria epidemic situation has been effectively controlled in Jiangsu Province,which re?flects the initiative achievements of malaria elimination. However,there are still many imported malaria cases from abroad,and the species of the Plasmodium are diverse. Therefore,the imported malaria from abroad still remains the key of malaria control in Jiangsu Province.
6.Analysis of outcome and prognosis-related factors of MA regimen for acute myeloid leukemia

Objective To analyze the outcome and prognosis-related factors of MA (mitoxantrone+cytarabine) regimen for acute myeloid leukemia(AML). Methods 102 untreated AML patients were treated with MA. All patients were divided into two groups according to age, blood white cell count(WBC), FrenchAmerican-British (FAB) morphology, level of lactate dehydrogenase (LDH) and immunophynotype respectively.Analyze the prognosis-related factors. Results The complete remission (CR), partial remission (PR), nonresponse (NR) rate, and remission rate (CR+PR) of all the 102 cases were 63.73 % (65/102), 17.65 % (18/102), 18.62 % (419/102)and 81.38 % (83/102) respectively. The patients younger than 60 years old, WBC<100×109/L, LDH≤600 U/L, FAB-M2 morphology group had higher CR and remission rate. The CR rate of patients with CD7 positive had statistical difference from that of patients with CD7 negative (P <0.05), but the remission rate not. However, the CR and remission rate of patients with CD19 positive had no statistical difference from that of patients with CD19 negative (P >0.05). Conclusion These results suggest that use of MA regimen was effective and safe for AML. Age, WBC, FAB morphology, level of LDH and CD7 expression are prognosis-related factors for clinical outcome.
7.Diagnostic knowledge and skills of parasitic diseases based on competition of professional personnel in Jiangsu Province,China

Objective To understand the integrated ability of parasitic disease prevention and control of professional person-nel of Jiangsu Province through the contest. Methods Totally 56 players from the whole province were selected,and all the players participated in the contest. The theory knowledge and skill scores were collected and the statistical analyses were con-ducted. Results The average theoretical score of the participants was 88.86±15.56 and the passing rate was 91.1%. The aver-age skill operating score was 69.16 ± 16.01 and the passing rate was 67.9%. The average Plasmodium microscopy score was 16.54±8.09 and the passing rate was 50%. The average helminth egg microscopy score was 34.27±10.66 and the passing rate was 67.9%. There were statistical differences among the age groups and different levels of schistosomiasis endemic situation (F =5.10,6.39,both P<0.01). The theoretical knowledge including schistosomiasis,malaria,hydatid disease and others and the score rates were 91.07%,90.94%,85.83%and 90.93%,respectively. The hydatid disease score rate was lower(χ2=19.17, P<0.01). The radar chart displayed that the score rates of tabletting and microscopy test in Kato-Katz film production ,malaria blood film production and microscopy test were all low. Conclusion In Jiangsu Province,the participants have higher score in the theory test. However,they have lower skill test score,especially in the parasite species identification. The operational skills still need to be strengthened for center for disease control(CDC)participants.
8.Epidemiological analysis of malaria prevalence in Jiangsu Province in 2015

Objective To understand the malaria epidemic situation and characteristics in Jiangsu Province in 2015,so as to provide the evidence for malaria elimination. Methods The data of malaria cases in Jiangsu Province in 2015 were collected from China's routine diseases surveillance information system. Results Totally 405 imported cases were reported in Jiangsu Province in 2015,and the cases increased by 14.08%compared with those in 2014. All the malaria cases were imported,and 5 cases(1.23%)were from Southeast Asia and 400(98.77%)were from 25 African countries or regions. The imported malaria cases were reported in 13 cities across Jiangsu Province,where Taizhou,Lianyungang,Nantong,Huaian and Yangzhou cities accounted for 68.64%of all the cases in the province. Jiangsu Institute of Parasitic Diseases(JIPD)reference lab checked all the cases and classified 299 falciparum malaria cases,13 vivax malaria cases,18 quartan malaria cases,71 ovale malaria cas-es,and 4 mixed Plasmodium infection cases. Conclusions In Jiangsu Province,there are no local malaria cases for 4 consecu-tive years,but the imported cases are on the rise. Compared to 2014,the imported vivax cases increased significantly in 2015. It should be the key points to strengthen the surveillance of imported malaria cases,improve malaria diagnosis and treatment abili-ties for all levels of medical institutions,and promote the abilities of Plasmodium check,and focus survey and disposal of all Center for Disease Control(CDC)staffs across Jiangsu Province.
