1.Influence of Serum Containing Hechan Tablets on Proliferation and Apoptosis of Human Lung Adenocarcinoma Cell Line

【Objective】To investigate the influence of serum containing Hechan Tablets(HT)on the proliferation and apoptosis of human lung adenocarcinoma cell(LAC)line.【Methods】Serum containing HT was prepared.Human LAC line was cultured by the routine method firstly,and then was planted respectively in the culture medium including serum containing HT at the concentration of 30%(group A),in the culture medium including blank serum at the same concentration(group B)and in RPMI-1640 culture medium(group C).The growth of LAC was monitored under inverted phase contrast microscope,histological features of apoptotic LAC nucleus examined under fluorescence microscope and the apoptosis of LAC observed by flow cytometry with the methods of DNA content analysis and terminal-deoxynucleotidyl transferase mediated nick end labeling(TUNEL).【Results】After treatment with serum containing HT,the apoptotic changes of LAC were as follows: apoptotic rate was 5.6% in group A,lower than 0.9% in group B and 0.1% in group C;DNA content analysis showed that the number of LAC which was in phase S and phase G2-M was less in group A than that in groups B and C 72 hours after the culturing,so did the cell proliferation index;the results of TUNEL examination showed that the incidence of cell apoptosis was 12.3% in group A,5.7% in group B and 1.2% in group C 72 hours after the culturing.【Conclusion】Serum containing HT has an inhibitory effect on the proliferation of LAC,and its mechanism may be related to inhibiting DNA synthesis and division of LAC,restraining LAC at the phase G0 and G1 and promoting LAC apoptosis.

Objective To establish an improved model of exercise-induced glycometabolism in type II diabetic rats,and to provide a theoretical reference for the establishment of exercise prescription for type II diabetes.Methods Forty-five 8-week old SPF male Wistar rats were used in this study.Of which 32 were fed with high-fat diet for 7 weeks,and intraperitoneal injection of 30 mg/kg STZ was given to establish the rat model of type II diabetes.The normal rats and successful model rats were divided into four groups:The normal control group (C group),normal exercise group (CE group),diabetic group (DM group) and diabetic exercise group (DME group).The exercise group was assigned by the Ploug training protocol,6 days/week,60 min/day,for a total of 8 weeks.After the high fat diet fed for 7 weeks,blood sample was taken from the tail vein,FBG and serum insulin were detected after baseline and 8 weeks exercise,and blood sample was collected from the tail vein to determine the FBG.Serum insulin (FINS) was detected by orbital blood sampling at the end of 8 weeks of exercise,and HOMA-IR was calculated.Results 1.After 7 weeks of high fat diet,compared with the groups C and CE,the levels of FBG,FINS and HOMA-IR were significantly higher in the DM and DME groups.2.After 8 weeks of exercise intervention,compared with the groups C and CE,FINS was significantly lower in the groups DM and DME,but the FBG and HOMA-IR were higher.Compared with the DM group,the level of FINS was significantly higher in the DME group,and the levels of FBG and HOMA-IR were significantly lower.The body weights of DM and DME groups were significantly lower than those of the groups Cand CE,the body weight had no significant difference between the DME and DM groups,and similar result was between the groups CE and C.Conclusions 1.The rat model of type II diabetes is successfully established with high fat diet for 7 weeks plus STZ injection(30 mg/mL).2.Aerobic exercise 60 min/day for a total of 8 weeks can improve the glycometabolism in type 2 diabetic rats,to be an ideal animal model for study of the mechanism of prevention and amelioration of type II diabetes.
5.The reliability of hand-held electronic dynamometer tests for lower limb muscle strength measurement in children with spastic cerebral palsy

Objective To determine the reliability of hand-held electronic dynamometer (HHD) tests for lower limb muscle strength measurement in children with spastic cerebral palsy (CP). Methods Twenty-eight children ( 15 boys and 13 girls; mean age 5 years 8 months) with different types of CP (2 with spastic quadriplegia,8 spastic diplegia, 6 hemiplegia, 1 triplegia and 1 monoplegia) , and at different functional levels ( 19 graded at gross motor function classification system level Ⅰ, 6 level Ⅱ, 2 level Ⅲ and 1 level Ⅳ ) were recruited from the Rehabilitation Center of the Children's Hospital of Fudan University. Standardized HHD protocols were used to measure the strength of their hip, knee and ankle muscles. The HHD test was performed by the same examiner twice with an interval of 10 min in between. The HDD test was also performed with 15 randomly selected CP children by 2 examiners with an interval of 10 min in between. The test-retest and inter-rater reliabilities of the HDD readings were determined by calculating the intra- and inter-class correlation coefficients. Results The HHD measurements showed fine testretest reliability ( ICC = 0.74-0.97 ) and inter-rater reliability ( ICC = 0.63-0.97 ) in measuring lower-limb muscle strength of children with spastic CP, with the highest test-retest reliability for the hip flexion, foot plantar flexion and knee extension muscle groups. The highest inter-rater reliability was achieved with the hip flexion and foot dorsiflexion muscle groups. Conclusions Standardized HHD testing of lower-limb muscle strength in children with spastic CP shows fine test-retest and inter-rater reliability. The HHD can reliably assess the lower-limb muscle strength of children with spastic CP.
6.Development of the general chapters of the Chinese Pharmacopoeia 2020 edition:A review

The Chinese Pharmacopoeia 2020 edition was reviewed and approved by the National Medical Products Administration and the National Health Commission of the People's Republic of China in July 2020.The current edition was officially implemented on December 30,2020.The general chapters of the Chinese Pharmacopoeia discuss the general testing methods and guidelines,which are the common re-quirements and basis for the implementation of drug standards in the Chinese Pharmacopoeia.Owing to adherence to the principles of scientificity,versatility,operability,and sustainable development,there is an improvement in the general chapters of the 2020 edition over those of the previous editions.Further,the application of advanced and mature analytical techniques has expanded,the development of testing methods for exogenous pollutants in traditional Chinese medicines has been strengthened,and technical requirements are now better harmonized with international standards.The updated edition provides technical and methodological support to ensure safety,effectiveness,and control of pharmaceuticals in China and will play an important and active role in encouraging the application of advanced technolo-gies,improving the quality control of medicines,and strengthening the means of drug regulation in China.This review provides a comprehensive introduction of the main features of and changes to the general chapters in the Chinese Pharmacopoeia 2020 edition and aims to provide reference for its correct understanding and accurate implementation.
7.Quality of life and its related factors in caregivers of patients with breast cancer

Objective To exam the quality of life of the caregivers of patients with breast cancer, to explore their related factors, including medical characteristics of the patients, demographic characteristics of the caregivers, relationship with the patients, social support and psychological resilience, and to test the mediating and moderating roles of psychological resilience between social support and quality of life. Methods A total of 179 hospitalized patients diagnosed with breast cancer were recruited in Qilu Hospital of Shandong University. The self-designed demographic questionnaire, Medical Outcome Study 12-Item Short Form Health Survey,Version 2(SF-12V2), Perceived Social Support Scale(PSSS), and 10-item Connor-Davidson Resilience Scale(CD-RISC-10) were used to collect information about demographic characteristics and measure the quality of life, perceived social support and psychological resilience of the caregivers. The factors affecting the quality of life of the caregivers were analyzed by multiple linear regression analysis, and the mediating and regulating effects of mental resilience on social support and mental health were analyzed. Results The age of the caregivers was negatively correlated with the physical health (P=0.010). The score of the body health of the family members of the middle and late stage cancer patients was lower than that of the family members in the early stage (P=0.003). There was a positive correlation between friend support and resilience and mental health (P<0.01). Resilience played a moderating role and mediating role in the relationship between friend support and mental health. Conclusions Attention should be paid to the quality of life of caregivers in patients with breast cancer. The medical staff should provide more social support for the caregivers, and should pay more attention to improve the psychological resilience of the caregivers, guide them to better cope with the burden and pressure of care, and improve their mental health with half the effort.
8.Relationships between perioperative depression, anxiety symptoms and fear of cancer recurrence in breast cancer patients

Objective:To investigate the predictive effect of perioperative depression and anxiety symptoms in breast cancer patients on the fear of cancer recurrence.Methods:A total of 194 patients with breast cancer during perioperative period from May 2019 to May 2020 in Qilu Hospital of Shandong University were selected. The depression, anxiety and fear of cancer recurrence were investigated by the general information questionnaire, the Generalized Anxiety Disorder-7 (GAD-7), the Patient Health Questionnaire-9 (PHQ-9) and the Fear of Progression Questionnaire-Short Form (FOR-Q-SF) at admission and two weeks after surgery, respectively.Results:Perioperative depression and anxiety symptoms of breast cancer patients were significant predictors of fear of cancer recurrence ( B value was 2.325, OR value was 10.22, P<0.05; B value was 2.570, OR value was 13.07, P<0.05), and patients with depression and anxiety symptoms after surgery were at higher risk of fear of cancer recurrence ( OR values were 7.653-25.403, P<0.01). Conclusions:For breast cancer patients with negative emotions and fear of disease progression, it is necessary to help them improve their psychological coping ability, encouraging patients to actively cooperate with follow-up treatment, improving the prognosis, and improving their overall quality of life.
9.Application of in situ amniocyte culture for prenatal diagnosis.

OBJECTIVE:
To explore the value of in situ amniocyte culture for prenatal diagnosis.
METHODS:
2716 amniotic fluid samples were cultured in situ on slides. After the culture, the slides were stained, photographed and analyzed.
RESULTS:
All samples were successfully analyzed, with the success rates for primary culture and subculture being 98.42% and 1.58%, respectively. 224 samples (8.25%) were detected with chromosomal aberrations, which included 125 cases with trisomy 21, 31 with trisomy 18, 3 with trisomy 13, 4 with 45,X, 17 with 47,XXY, 5 with 47,XYY, 1 with 48,XXY,+18, 1 with 48,XXYY, 26 with structural chromosomal aberrations, and 11 with mosaicisms for aneuploidies.
CONCLUSION
In situ amniocyte culture is stable and has a high success rate, and is capable of identifying true and false mosaicisms, which can improve the accuracy of prenatal diagnosis.
10.Correlation Research of Plasma Glucose, Blood Pressure Level and TCM Syndromes in Shanghai Community Residents

Objectives: The purpose of this study is to understand the relationships among plasma glucose, blood pressure level and Traditional Chinese Medicine (TCM) syndromes in Shanghai community residents, and provide a theoretical basis for the prevention of community chronic disease based on TCM syndrome differentiation.Methods: Residents above35 years old will attend the Type 2 Diabetes Mellitus (T2 DM) risk assessment at Community Health Center. By distributing questionnaires and performing glucose testing, we screened the residents at high risk of T2 DM, and conducted a physical examination of them. Further, a body constitution questionnaire was required to be completed by the residents. Results: In total, 933 residents were screened. The plasma glucose and blood pressure levels related to age, waist circumference, hip circumference, body mass index (BMI) and waist-to-height ratio (WHtR) . Residents with increasing blood pressure have an increased risk of T2 DM (P < 0.01) . Total 529 questionnaires were completed, and 129 subjects (24.4%) have single TCM syndromes, 75 subjects (14.2%) have at least two TCM syndromes and 325 subjects (61.4%) have no TCM syndromes. Conclusion: Plasma glucose and blood pressure are associated and interacted with several physical indexes. TCM syndromes distribution was found no significant change among subjects with different plasma glucose and blood pressure.
