1.Cryopreservation of test-tube shoot tips of Siraitia grosvenorii by vitrification and plant regeneration

Objective To get a new approach for conserving the germplasm of Siraitia grosvenorii Methods Shoots, 0.8—1 cm in length, excised from test-tube plantlets which were subcultured 15 d, were precultured. Then its shoot-tips about 2—3 mm in length were dissected and loaded with 60% PVS2 at 25 ℃, and dehydrated with 100% PVS2 at 0 ℃, changed for fresh 100% PVS2 prior to directly plunging into liquid nitrogen. After cryopreservation for 24 h, the shoot tips were thawed, rinsed in MS+1.2 mol/L sucrose medium, blotted with filter papers, then plated on the MS + 1.0 mg/L 6-BA+0.05 mg/L NAA + 0.1 mg/LGA3 + 0.8% agar + 45 g/L sucrose medium at 25 ℃ for 7 d in dark prior to exposure to the light. The root medium for regeneration plantlets was 1/2 MS + 0.2 mg/L NAA + 30 g/L sucrose. Results Shoots were precultured for 3 d on MS + 0.7 mol/L sucrose medium, then its shoot-tips loaded for 40 min, dehydrated for 50 min. After cryopreservation, the shoot tips were rapidly thawed in water at 40 ℃, then rinsed for 40 min. The survival rate of shoot-tips plated on the recovering medium in one week was 100%, and regeneration rate after 30 d was 78.33%, which was the highest. The regeneration plantlets inoculated on root medium were reconstructed integrating plantlets. Conclusion The method of vitrification to cryopreserve the germplasm of S. grosvenorii is a simple way with high survival rate and normal regeneration plants.
2.Some Visible Properties of T Lymphocyte Activation in Human Umbilical Cord Blood

Objective To explore visible properties of T lymphocyte activation in human umbilical cord blood.Methods T lymphocytes of human umbilical cord blood were cultured in RPMI 1640 complete medium and divided into two groups: the experimental group activated with CD3 McAb(10mg/L)plus PHA and the control group cultured without these reagents. Afcer the cell viability being measured, T cell activation was observed with inverted microscope. And then ultrathin section of the cells sample was prepared tobe observed with transmission electron microscope(TEM).Results Morphologically, activated T cells were obviously bigger, their nuclei clear, and most of divided cells had irregular outline;there were cell colonies to be seen in growing regions. Under TEM, there were more vacuoles (visible property of structure) in typically activated T lymphocytes , chromatic agglutination and nucleoli to be observed.Conclusion Main properties of T cell activation of human umbilical cord blood were chromatic agglutination, more vacuoles in cytoplasm and cell colonies formation.
3.Bond strength between titanium alloy with different surface roughness and dentin

OBJECTIVE: To compare the difference in bond strength between titanium alloy with different surface roughness and dentin. METHODS: Thirty titanium alloy cylindrical specimens were fabricated and randomized to 6 groups. All groups of specimens were rubbed separately with 60, 80,100,150, 240, and 320 grit silicon carbide (Si-C) sand paper. The surface roughness of each group was measured. The occlusal surfaces of 30 human molars freshly extracted within 1 month were flattened to expose an adequate area of lower dentin. The dentin bonding surface was produced using 320, 400, and 600 grit Si-C sand paper, respectively. Titanium alloy and dentin specimens were bonded with Ketac~(TM) Cem Easymix ionomers. All bonded specimens were stored for 24 hours at 37 ℃. The microtensile bond strength was measured through the use of universal testing machine. The fractured surface of titanium alloy was observed under scanning electron microscopy.RESULTS: The titanium alloy group rubbed with 80 grit Si-C sand paper yielded the strongest bond strength, 6.65 MPa, compared with other groups (P < 0.05). The surface roughness of titanium alloy in this group was (2.69±0.32) μm. Scanning electron microscopy results revealed that all fractured surfaces appeared at the interface of dentin and ionomer cement. CONCLUSION: The surface roughness could influence the bond strength between titanium alloy and dentin. Titanium alloy with (2.69±0.32) μm surface roughness bonding to dentin showed the maximum bond strength.
4.Effect of different pressure time on bond strength of glass ionomer cement

OBJECTIVE: To survey the immediate bond strength of the two glass ionomer cements with different pressure times.METHODS: Totally 70 teeth were prepared to expose the dentin, and then divided into the Ketac~(TM)Cem Easymix GIC glass ionomer cement group (3M group), and the TOKUSO glass ionomer cement (TOKUSO group). Each group was divided into 7 subgroups according to pressure times. The immediate tensile strengths of all specimens were measured after pressed for 5,10,15, 20, 25, 30 and 35 minutes.RESULTS: The immediate bond strengths of the two glass ionomer cements with different pressure time showed obviously significant differences (P < 0.01), which exhibited the greatest immediate bond strengths at 25 minutes after pressure. The bond strength of 25 minutes subgroup of 3M was higher than that of TOKUSO group, but the difference had no significance.CONCLUSION: Both Ketac~(TM) Cem Easymix and the TOKUSO glass ionomer cement can harvest the maximum bond strength after 25 minutes pressure.
5.Effect of atorvastatin on high-sensitivity C-reactive protein and interleukin-6 in unstable angina pectoris patients after intervention

Objective To explore the effect of atorvastatin on high-sensitivity C-reactive protein (hs-CRP) and interleukin-6 (IL-6) in unstable angina pectoris (UAP) patients after intervention.Methods Eighty cases with UAP who underwent percutaneous coronary intervention (PCI) were divided into group A and group B by random digits table method with 40 cases each after PCI.The patients in group A were given regular heart treatment and atorvastatin 20 mg treatment.The patients in group B were given regular heart treatment and atorvastatin 40 mg treatment.The serum hs-CRP and IL-6 was determined before treatment and 24 h,3 weeks after treatment and compared between two groups.Results There was no significant difference in the serum hs-CRP and IL-6 before treatment between group A and group B [(5.6 ± 1.2) mg/L vs.(5.6 ± 1.1) mg/L and (211.9 ± 21.2) ng/L vs.(209.6 ± 19.9) ng/L,P > 0.05].The serum hs-CRP and IL-6 24 h after treatment in group A and group B were increased compared with that before treatment,and there was significant difference(P < 0.05),but there was no significant difference between group A and group B[(8.1 ± 1.1) mg/L vs.(8.5 ± 1.2) mg/L and (311.1 ± 20.9) ng/L vs.(313.3 ± 18.5) ng/L,P> 0.05].The serum hs-CRP and IL-6 3 weeks after treatment in group A and group B were decreased compared with that before treatment,there were significant difference (P < 0.05),and there were significant difference between group A and group B [(3.1 ± 1.1) mg/L vs.(1.9 ±0.8) mg/L and (163.3± 18.5) ng/L vs.(123.3± 19.5)ng/L,P < 0.05].No obvious adverse reaction was observed in two groups after treatment and liver function was not seen obvious anomaly.Conclusions Atorvastatin treatment in UAP patients after PCI can significantly reduce the serum hs-CRP and IL-6,and the 40 mg treatment is better than the 20 mg treatment.It is worth of clinical application.
6.Study on Quality Standards for Lingyangqingfei Capsules

Objective: To establish the quality standards for Lingyangqingfei capsules. Methods: Microscopic identification of products, TLC identifications of Fructus Gardeniae and Cortex Moutan were carried out.Baicalin was determined by HPLC. Results: Baicalin in Lingyangqingfei capsules was about 1.5mg/capsule. The average recovery was 99.82% and RSD was 0.96%. Conclusion: The result is accurate and the reproducibility is good. This method can be used as the quality control of this preparation.
7.Application of LCSAS and SP to evaluate the doctor-patient communication ability of young doc-tors in our hospital

Objective To understand the status quo of our young doctors' doctor-patient communi-cation and related influencing factors. Methods Among the doctors under the age of 40 in various clinical departments, according to 30% proportion, we randomly selected 32 doctors, and combined with revised Liverpool communication skills assessment scale (LCSAS) and standardized patients (SP), the doctors' ability of doctor-patient communication was evaluated. Descriptive statistics, t test and variance analysis were per-formed by SPSS 21.0 lines. Results The average score of communication etiquette of young doctors was (2.00±0.15), and the average score of their communication skills was (1.74±0.29), and the average score of their communication technical ability was (3.74±0.39). The detection rate of communication etiquette, communication skills and communication skills evaluation (less than 2) was 33.3%, 81.5%, and 85.2%, respectively. Physicians and surgeons' scores in communication etiquette and communication skills dimen-sion had significant difference (P<0.05). Significant difference was found in different ages doctor communi-cation protocol (F=3.641, P=0.028), and 20 to 25 years old doctors' score in communication manners was significantly lower than other ages (P<0.05). There were significant differences in the scores of doctors of different title in communication etiquette (F=4.410, P=0.023), and the trainee doctors' communication etiquette score was significantly lower than the physician and surgeon (P<0.05). Conclusion The commu-nication skills of young doctors in our hospital are generally in the middle state, and their communication etiquette is good, communication skills are general. There are great individual differences. Hospital should begin from those whose communication ability are low, cultivate their biology-psychology-social medicine thinking mode of the doctor-patient relationship , help new employees complete the role transformation , emotion management, and professional commitment, and encourage patients to participate in the medical process, so as to improve the doctors' communication ability in all directions and at different levels.
8.Quantitative Tissue Velocity Imaging in Evaluation of Left Atrial Systolic Function in Patients with Acute Myocardial Infarction

Objective:To evaluate the changes of the left atrial systolic function in patients with acute myocardial infarction(AMI)using quantitative tissue velocity imaging(QTVI).Methods:The systolic velocities of the middle of left atrial lateral wall,anterior wall,inferior wall and atrial septum were measured with QTV1 in 45 patients with AMI.The left atrial volume and active atrial emptying fraction(AA-EF)were measured using single-plane Simpson method.Results:(1)Compared with the control group(30 normal subjects),the diameter and volume of the left atrium,as well as AA-EF,increased obviously in patients with AMI(P < 0.01).(2)Compared with the control group,the systolic velocities of the middle of left atrial lateral wall,anterior wall,inferior wall and atrial septum,as well as the average value,increased significantly in patients with AMI (P < 0.05 or P < 0.01).In addition,the average value of left atrial systolic velocity was closely correlated with AA-EF(r=-0.906,tr=14.001,P < 0.01).Conclusion:QTVI could be used to evaluate the left atrial systolic function accurately in patients with AMI.
9.The relationship between serum complement C3 concentration and prediabetes in an adult population

Objective To evaluate whether serum complement C3 concentration was associated with the prevalence and incidence of prediabetes in an adult population. Methods A cross-sectional (n=10 539) and prospective cohort (n=3 064, followed up for-6 years, mean:2.8 y) study was performed on subjects recruited from the Health Management Center of Tianjin Medical University General Hospital in Tianjin. Measurements of serum C3 concentration, blood fasting glucose and other potential confounding factors were assessed at baseline and per year during the follow-up period. Prediabetes was defined according to the criteria of American Diabetes Association. Adjusted Logistic and Cox proportional hazards regression models were used to assess the associations between C3 quintiles and prediabetes. Results The prevalence and incidence of prediabetes were 19.9% and 99.5 per 1 000 person-year, respectively. In cross-sectional analysis, after adjusted for potential confounders, the odds ratios (95% confidence interval) of prediabetes for increasing quintiles of C3 were 1.00 (reference), 1.18 (0.98-1.42), 1.11 (0.92-1.34), 1.38 (1.15-1.65) and 1.63 (1.36-1.95) (P for trend<0.000 1). In cohort analysis, in the final multivariate models, the hazard ratios (95% confidence interval) for prediabetes across C3 quintiles were 1.00 (reference), 1.20 (0.94-1.54), 1.48 (1.16-1.88), 1.38 (1.09-1.76) and 1.53 (1.21-1.95) (P for trend <0.001), respectively. Conclusion The study suggests that the elevated C3 level is significantly associated with the prevalence and incidence of prediabetes, which means that C3 can be used as a biomarker in early prevention of prediabetes and diabetes.
10.Low frequency repetitive transcranial magnetic stimulation in the diagnosis and treatment of cranial dystonia

Objective To study the therapeutic effects of low frequency repetitive transcranial magnetic stimulation(rTMS)in cranial dystonia.Methods Twenty cranial dystoina patients were treated with low frequency rTMS.Their motor threshold,cortical silent period(CSP)were evaluated before and after the rTMS and after 1,2,6 months as well as the spares and Toronto Western Spasmodic Torticollis Rating Scale(TWSTRS)to evaluate the effects of rTMS in the treatment of cranial dvstonia.Results The patients scored(23.5±14.0)significantly lower after l and 2 months(17.6 ±14.3,18.5±14.2,t=2.632,2.149.both P<0.05).But there was an increasing tendeney of the score after 2 months.The 2-month efficient rate of low-frequency rTMS Was 60%(12/20),yet the long-term effect of rTMS was still to be studied.There was a very significant improvment of relaxed(46.5%±7.3%vs49.9%±9.2%,t=-3.235.P<0.05)and active threshold(40.2%±5.9%/)5 43.9%±8.8%,t=-2.339,P<0.05),prolongation of CSP((96.1±24.5)ms vs(121.6±27.7)ms,t=-7.223,P=0.000).Conclusion The low frequency rTMS is efficient to relieve the clinical symptoms of cranial dystonia.
