3.Study on the corretation between serum HBV.M,alanine transaminase and hepatic tissue pathology in patients with chronic hepatitis B

Objective To investigate the relationship between serum HBV.M.alanine transaminase(ALT) and hepatic tissue pathology in patients with chronic hepatitis B.Methods The serum HBV.M.liver function in 133 patients with chronic hepatitis B were measured,and these patients underwent percutaneous liver biopsy by colour Echo.All these patients were divided into eight groups according to HBeAg and HBV DNA positive or negative,ALT normal or not.Hepatic necrosis inflammation grade and hepatic fibrosis stage in the groups were compared.Results Hepatic histology of all these patients showed inflammation and necrosis and different degree fibrosis.In serum ALT normal patients,liver biopsy still showed different degrees inflammation and hepatic fibrosis degrees partly,even showed hepatocirrhosis.In serum ALT abnormal and HBeAg negative patients with chronic hepatitis B patients,hepatic tissue inflammation and fibrosis degrees were more serious,hepatic tissue pathology was out of proportion to the degree of serum HBV reproduction.Conclusion Judgment of disease activity doesn't only depend on the serum transaminases and the degree of serum HBV DNA replication.When clinical choosing indication of antiviral therapy,value of liver biopsy should not be substituted.Liver biopsy in chronic hepatitis B should be regarded as main basis of judgment the hepatitis activity.It also should be regarded as main basis if antiviral therapy is conducted.
4.Blood coagulation disorder in acute pancreatitis of the elderly

Objective To explore the significance of blood coagulation disorder in acute pancreatitis(AP) of the elderly and its treatment with prostaglandin E_1.Methods Prothrombin time(PT),partially activated thromboplastin time(APTT) and fibrinogen(FIB) in cases of AP with ages over 60 were checked and compared with those in groups of AP with ages under 60 and normal control subjects.10 cases of AP of the elderly with elevated FIB were treated with prostaglandin E_1.Results Compared with groups of AP with ages under 60 and the normal subjects,FIB was significantly higher in cases of AP of the elderly with the highest in patients of SAP.8 out of 10 aged AP patients with elevated FIB was successfully treated with prostaglandin E_1.Conclusion Patients of AP may present with blood coagulation disorders especially in aged patients.Aged patients with elevated fibrinogen may predispose to SAP.Prostaglandin E_1 may be effective in this occasion.
5.Effect of alendronate on the healing of replanted dog teeth after extended dry time

Objective:To investigate the effect of alendronate(ALN) solution on the healing of replanted dog teeth after extended dry time. Methods:The first, second incisor and first premolar in maxilla and the second, third incisor and first premolar in mandible in two mature mongrel dogs were used for the test. The right tested teeth were included in experimental group, and the left in control. Firstly, all tested teeth were endodontically treated. Seven days later, they were extracted, then naturally dried for 60 min. The teeth in experimental group were soaked for 5 min in a 1 mmol/L solution of ALN before replantation. The teeth in control group were not treated with any other methods before replatation. Bone masses including teeth were taken out 3 monthes after replantation. Objective To explore the clinical curative effect of the bipolar artificial femoral head replace-ment in the treatment of advanced aged patients with femoral neck fracture .Methods 32 advanced aged patients with femoral neck fracture were treated with the bipolar artificial femoral head replacement ,the effect was observed . Results 32 patients were stage Ⅰincision healing ,and after two weeks all could ambulate .Followed up for 6 to 36 months,two cases of dislocation ,two cases of postoperative pain ,3 cases of deep venous thrombosis ,one case of hip limited functionality.Harris score function assessment:excellent 24 cases(74%),good in 5 cases(16%),3 cases (10%).Conclusion Bipolar artificial femoral head replacement is the effective treatment of advanced aged patients with femoral neck fracture,with shorter operation time,small trauma,early and activities,and reduce the complications and reduce mortality rate etc .

BACKGROUND: Previous reseamh has proved that RNA interference can inhibit vascular endothelial growth factor (VEGF) gene expression of colon carcinoma, carcinoma of prostate, and retinoblastoma. However, RNA interference inhibiting VEGF of carcinoma of gallbladder was not reported. OBJECTIVE: To construct a plasmid expression vector coding for the short hairpin RNA (shRNA) targeting hVEGF165 mRNA. DESIGN, TIME AND SETTING: A gene engineering study was performed at National Hepatobiliary & Enteric Surgery Research Center, Xiangya Hospital, Central South University from 2008 to 2009.MATERIALS: Human GBC-SD was provided by Tumor Research Institute of Tongji University. METHODS: Four pairs of shRNAs that targeted at VEGF gene were designed. The eukaryotic expression plasmids (named shRNA1-4) were constructed and identified using restriction enzyme analysis. The plasmids were then transfected into GBC-SD cells via liposome2000. The transfection rate of recombinant plasmids was measured at 48 hours after transfection. MAIN OUTCOME MEASURES: Enzyme analysis of recombinant plasmid; transfection rate; VEGF mRNA expression determined using fluorescent polymerase chain reaction. RESULTS: shRNA plasmid vector targeting at VEGF gene was successfully constructed, in particular, pDC316-EGFP-U6-shRNA2 was the most effective. The expression plasmids were confirmed by restriction enzyme analysis. The transfection rate of recombinant plasmids in GBC-SD cells was approximately 58.6%. shRNA could inhibit VEGF mRNA expression, in particular, the inhibitory rate of RNA2 was the highest by 86%.CONCLUSION: The shRNA eukaryotic expression plasmid targeting at VEGF gene is constructed and selected successfully, and it can remarkably inhibit VEGF expression of GBC-SD cells. Additionally, the inhibitory effect of RNA2 is the greatest.
9.Expression of cathepsin B and cystatin C and its potential role in diabetic rats

AIM:To observe the expressions of cathepsin B (CB) and cystatin C (CC) in different stage of diabetic rats and to investigates their potential roles.METHODS:Sixty rats were divided into diabetes mellitus group induced by intravenous injection of streptozotocin (55 mg/kg) and normal group injected with citrate buffer. Ten rats were sacrificed respectively at the end of fourth week,eighth week and sixteenth week in both groups. 24 h urine excretion was collected in rats before sacrifice. The blood and the kidney were also collected. 10.Effects of low-glucose on long-term potentiation in the hippocampal slices of immature and adult rats

AIM: The effects of low-glucose on long-term potentiation (LTP) in the CA1 region of hippocapal slices of immature (15-16 days old) and adult (56-63 days old) rats were examined. METHODS: The technique of electrophysiology was used, and the slopes of the field excitatory postsynaptic potentials (S-EPSP) were measured. RESULTS: When slices were exposed to glucose medium at concentrations of 3 or 1.5 mmol/L, S-EPSP decreased significantly. In the slices from adult rats, only short-term potentiation was elicited by high frequency stimulation in the medium of 3 or 1.5 mmol/L glucose. However, in the slices from immature rats, LTP was still induced in the medium of 3 mmol/L glucose. CONCLUSION: Low-glucose medium depressed the synaptic transmission. In terms of the synaptic plasticity, the low-glucose endurance in immature rats was stronger than that in adult rats.
