1.STUDIES ON THE ULTRASTRUCTURE OF LAMELLAR BODIES, THE ORIGIN AND STUCTURE OF THE PULMONARY SURFACTANT SYSTEM OF RATS

Lamellar bodies, tubular and lattice myelin figures (TMF and LMF)of the Type Ⅱ epithelial cells lavaging from the lungs of adult male rats following injection of a single dose of 10mg of active carbon powder, silica dust, uranium ore dust and U_3O_8 powder were observed by scanning and transmisson electron microscope as well as freeze-etching technique. According to the membrane split theory and our observations, we suggested that the lamellar bodies were composed of a lot of subunits, the square crystalls. These crystals linked up each other to form a cuboidal structure and arranged regularly into ring-like lamellae. A central core was surrounded by the lamellae to constitute a lamellar body, which can be distingushed into three types, namely the wollen ball-shaped, the circular concentric shaped and the lattice-shaped. When the circular concentric lamellar bodies were dissociated from the type Ⅱ epithelial cells, they then extended to form the pulmonary surfactant system (LMF). The amount of the pulmonary surfactant system substances in the rats administrated the above-mentioned foreign substances was obviously higher than that of controls.
2.Observation on efficacy of endoscopic submucosal dissection and endoscopic mucosal resection in the therapy of esophageal mucosal disease

Objective To evaluate the outcome and safety of endoscopic submucosal dissection (ESD) versus endoscopic mucosal resection (EMR) in the management of esophageal mucosal lesion.Methods The clinical data of 70 patients with esophageal mucosal lesion were analyzed.Twenty-five patients were treated by ESD (ESD group) and 45 patients were treated by EMR (EMR group).The enbloc resection rate,tissue healing resection rate,operation time,complication and recurrence rate were compared between two groups.Results The enbloc resection rate,tissue healing resection rate in ESD group were significantly higher than those in EMR group [60.0%(15/25) vs.48.9%(22/45),48.0%(12/25) vs.40.0% (18/45)] (P <0.05).The local recurrence rate in ESD group was lower than that in EMR group [4.0% (1/25) vs.20.0% (9/45)] (P < 0.05).The severe complication such as delayed hemorrhage,perforation,stenosis between two groups had no significant difference (P > 0.05).The operation time in ESD group was (64.3 ±27.1) min,and significantly longer than that in EMR group [(27.6 ± 14.1) min] (P <0.05).Conclusions Compare with EMR,ESD has better enbloc resection rate,tissue healing resection rate,and lower recurrence rate.It is more safe and effective in the therapy of esophageal mucosal disease.
3.Effect of Shexiang Baoxin Pill on endothelial in stable angina pectoris patients and its underlying mechanisms

AIM: To investigate the effect of Shexiang Baoxin Pill(SXBXP) on endothelial in stable angina pectoris patients and its underlying mechanisms. METHODS: Sixty-eight stable angina pectoris patients were randomly divided into two groups,the conventional group(34cases),SXBXP treatment group(34cases).The conventional group was treated with standard treatment,and SXBXP group was treated with standard treatment and SXBXP.FMD,NO,ET,TXB_2,6-Keto-PGF-1a were determined before and after three months treatment. RESULTS: FMD,NO,6-Keto-PGF-1a of SXBXP group were (9.35%?0.78%)、(77.25?6.36)?mmol/L、(93.87?(10.28))?/(ng/L) after treatment,ET、TXB_2 were(81.15?5.43) pg/mL、(43.02?4.19)?/(ng/L).There were significant improvement as compared with in SXBXP group before treatment and in conventional group after treatment(P
4.Implementation of sector imaging mode with high signal-to-noise ratio in the high frame rate ultrasonic imaging system.

The high frame rate (HFR) ultrasonic imaging system, which is developed with limited diffraction beams, constructs images at a high frame rate. However the rectangular imaging area, to some extent, restricts the far field imaging information. At the same time, because of one transmission for constructing image, the system suffers from low SNR. In this paper we present a computationally efficient method to construct sector mode image and to increase the SNR in HFR system. The method uses Golay complementary sequence as excitation to realize two transmission events. Each event simultaneously transmits two plane waves with different transmission angle. Then the received echo signals related to different angle are separated according to orthogonality of Golay complementary sequence and used to construct two images of different area by HFR method. Finally the two images are synthesized to one frame of sector mode image.

5.Targeting Kindlin-2 in adipocytes increases bone mass through inhibiting FAS/PPARγ/FABP4 signaling in mice.

Osteoporosis (OP) is a systemic skeletal disease that primarily affects the elderly population, which greatly increases the risk of fractures. Here we report that Kindlin-2 expression in adipose tissue increases during aging and high-fat diet fed and is accompanied by decreased bone mass. Kindlin-2 specific deletion (K2KO) controlled by Adipoq-Cre mice or adipose tissue-targeting AAV (AAV-Rec2-CasRx-sgK2) significantly increases bone mass. Mechanistically, Kindlin-2 promotes peroxisome proliferator-activated receptor gamma (PPARγ) activation and downstream fatty acid binding protein 4 (FABP4) expression through stabilizing fatty acid synthase (FAS), and increased FABP4 inhibits insulin expression and decreases bone mass. Kindlin-2 inhibition results in accelerated FAS degradation, decreased PPARγ activation and FABP4 expression, and therefore increased insulin expression and bone mass. Interestingly, we find that FABP4 is increased while insulin is decreased in serum of OP patients. Increased FABP4 expression through PPARγ activation by rosiglitazone reverses the high bone mass phenotype of K2KO mice. Inhibition of FAS by C75 phenocopies the high bone mass phenotype of K2KO mice. Collectively, our study establishes a novel Kindlin-2/FAS/PPARγ/FABP4/insulin axis in adipose tissue modulating bone mass and strongly indicates that FAS and Kindlin-2 are new potential targets and C75 or AAV-Rec2-CasRx-sgK2 treatment are potential strategies for OP treatment.
