Objective To explore the optimal dose and ways of anesthesia for creating a rat model of spastic paralysis with intermittent bilateral common carotid artery ligation. Methods 60 Wistar rats were randomly divided into groups A, B, C, D, E and F. Group A was anaesthetized with 10% chloral hydrate 5 ml/kg injected subcutaneously, while group B with 4 ml/kg subcutaneously, group C with 4 ml/kg intraperitoneally, group D with 3 ml/kg subcutaneously, group E with 3 ml/kg intraperitoneally, group F with 2 ml/kg subcutaneously. The onset and duration of anesthesia, and intraoperative and postoperative mortality were compared. Results All the rats in the group A died during anesthesia, while the group F did not achieve the depth of anesthesia, and abandoned. The onset time was (6.5±0.7) min, maintained (121.4± 3.9) min, mortality was 0 in the group B, and it was (5.5±1.1) min, (122.0±3.6) min, 30% in the group C; (9.6±0.8) min, (106.7±3.7) min, 0 in the group D, (7.4±1.2) min, (105.3±3.5) min, 20% in the group E. The overall mortality rate was 0 in the groups accepted subcutaneous injected and 25% of intraperitoneal injection. Conclusion Anesthesia with 10% chloral hydrate 4 ml/kg subcutaneous injection is optimal of lower mortality, faster onset and longer maintaining in rats for spastic paralysis model with intermittent bilateral common carotid artery ligation.

Objective To investigate the DNA methylation of skeletal muscle in spastic paralysis rats and correlation with the muscle fiber configuration. Methods 100 5-day old Wistar rats were randomly divided into model group and control group. The former was established the spastic paralysis modle and reared for 30 days. Then, tissues from the gastrocnemius of all the rats were observed with triplicate DNA methylation, myosin heavy chain-I (MHC-I) mRNA with RT-PCR and transmission electron microscopy. Results The DNA methylation was (4.95±0.83)×10% in the model group, significantly less than (6.59±0.75)×10% in the control group (P<0.001); while the MHC-I mRNA was (1.23±0.31), significantly more than (0.44±0.29) in the control group (P<0.001). The Z-line was disordered, and the mitochondria near the Z-line increased, with edema and partially broken in cristae. The balance between the thick and thin filaments was broken, and myofibrils envelope fused. Conclusion Hypomethylation and hyperexpression of MHC-I mRNA have been found in skeletal muscle of spastic paralysis rats, which may result in type I fibers increase. However, there was no sufficient evidence to support the correlation between the DNA methylation and the secondary pathological changes.

Objective To explore the effects of hand-arm bimanual intensive training (HABIT) in group on functional independence and quality of life of children with hemiplegic cerebral palsy. Methods From November, 2015 to September, 2016, 20 children with hemiplegic cerebral palsy were randomly divided into control group (n=10) and experimental group (n=10). Both groups received routine occupational therapy, and the experimental group received HABIT in group one hour per day additionally, for eight weeks. They were assessed with the Carroll Upper Extremities Functional Test (UEFT), Functional Independence Measure Children's Edition (WeeFIM) and Chinese Version Pe-diatric Quality of Life Inventory (PedsQLTM) 3.0 Cerebral Palsy Module respectively before and after treatment. Results After treatment, the scores of UEFT, WeeFIM and the PedsQLTM3.0 improved in both groups (t>3.970, P<0.05), and were higher in the experimental group than in the control group (t>2.306, P<0.05). Conclusion HABIT in group could improve the fine function of upper extremity and hands coordina-tion of children with hemiplegic cerebral palsy, and enhance the functional independence and quality of life.

Liver sinusoidal endothelial cells (LSECs) are the highest proportion of liver non-parenchymal cells with fenestrae structure and high endocytic ability maintaining liver homeostasis and playing an indispensable role in the physiology and patholo-gy of the liver.LSECs are involved in the regulation of patholog-ical process in nonalcoholic fatty liver disease(NAFLD),alco-holic fatty liver(AFL),hepatocellular carcinoma(HCC),liverregeneration and liver fibrosis mainly via antiinflammation,endocytosis,secretion of angiocrine signals and maintaining thequiescence phenotype of HSCs.This review highlights the physiological function of LSECs and the different roles in different pathological conditions,which aims to provide a new perspectivefor the treatment of liver diseases through targeting LSECs.

Objective To investigate whether there is correlation exists in the effection of chronic unpredictable stress on active avoidance behavior and electroencephalogram ( EEG) of rat. Methods Twenty male SD rats (weight～200 g) were randomly divided into control group( n= 10) and model group( n- 10). Rats in model group were subjected to chronic unpredictable stress . Active avoidance response were observed with GeminiTM avoid system and EEG power spectrum was evaluated with Powerlab system. Results The failure rate in active avoidance test was correlation with delta wave power in EEG. Compared with control group, rats in model group had reduced failure rate in active avoidance task(42 ±36)% vs (82 ±30)% , P＜0.05). The EEG power spectrum of model group rats increased in delta band((47.09 ±22.86)μV2 vs (22. 55 ± 11. 57) μV2, P＜0. 05), which had the significant difference between the two group rats. The failure rate of active avoidance task in model group rats was positive correlation with EEG power spectrum of delta wave (r = 0. 717, P ＜ 0. 05) . Conclusion Chronic unpredictable stress facilitated active avoid behavior and changed spontaneous EEG, which suggested chronic unpredictable stress actived the neurotransmitter network system involved with active avoidance task in central nervous system. Brain cognitive function correlated positively with brain electrophysiology activity measured in chronic unpredictable stress induced rat model.

Nicotinamide adenine dinucleotide phosphate oxidase ( NOXs) contributes to the production of reactive oxygen species ( ROS) in liver fibrosis, resulting in the activation of endoplas-mic reticulum stress ( ERS ) and IRE1α-XBP1 signaling path-way. ROS is a series of oxygen metabolites and its derivatives, produced by the single electron reduction of molecular oxygen ( O2 ) , including superoxide anion ( O2- ) , hydroxyl radical (-OH) , hydrogen peroxide ( H2 O2 ) , hypochlorite ion ( OCl-) and so on. They can interact with a large number of molecules, including small inorganic molecules, proteins, lipids, carbohy-drates and nucleic acids, resulting in lipid peroxidation of cell damaging molecules. And as a second messenger, ROS can also affect the proliferation and activation of HSC in liver fibrosis, and induce the hepatocyte apoptosis through a variety of cellular signal transduction. Here we review the current status of the study on the mechanism of NOXs in liver fibrosis.

Liver fibrosis is a major cause of morbidity and mor-tality worldwide which poses a great threat to public health. Con-siderable evidence suggests that the immune system is closely re-lated to the development of hepatic fibrosis especially the dendrit-ic cells ( DCs) . In recent years, many studies have showed that DCs play a key role in regulating the immune function of liver, which not only mediate the activation of the immune system and inflammation reaction in liver, but influence the occurrence and development of liver fibrosis. Further study has found that DCs exert different effects on liver fibrosis at different stages of the disease, and it exerts anti-fibrosis in early stages and recession period, while plays opposite effect in the middle of the disease. This article reviews the research progress of the role of DCs in liver fibrosis and discusses the underlying mechanisms of DCs in regulation of liver fibrosis, which may provide references for bas-ic and clinical studies of liver fibrosis.

Objective To investigate the effect of physician guidance on cigarette cessation in current smokers with chronic obstructive pulmonary disease(COPD).Methods Cigarette cessation education and routine telephone follow-up was provided for 70 COPD patients.Pulmonary function and exhaled carbon monoxide level were detected every 3 months.Results After 6 months' follow-up,28 participants(40.00%) successfully quitted cigarette smoking.Seventeen subjects (24.28%) experienced relapse,with mean cessation time(3.08±0.33) months.Once receiving physician guidance,5 re-attempted to quit cigarette smoking,and 12 reduced tobacco consumption. Sixteen subjects(22.86%) did not quit smoking,although the mean cigarette consumption per day was decreased by 60%.Nine participants (12.86%) showed no change in smoking habit. Conclusion Physician guidance could impreve the rate of cigarette cessation in COPD patients.Intensive intervention may be correlated with successful smoking cessation.

Objective To explore the correlation between psychosocial factors and adherence to anticoagulation therapy in patients after mechanical heart valve replacement.Methods Totally 176 patients who received mechanical heart valve replacement and admitted in Taizhou Hospital from January 2016 to December 2017 were selected.The Warfarin awareness and anticoagulation adherence questionnaire,Self-Anxiety Scale (SAS),Self-Depression Scale (SDS) and Social Support Rating Scale (SSRS) were used to evaluate their adherence to anticoagulation therapy and psychosocial factors.The correlation between their adherence to anticoagulation therapy and psychosocial factors was analyzed.Results The 176 patients showed good adherence to anticoagulation therapy.Their Warfarin awareness,SAS,SDS and SSRS scores averaged (13.27±3.59),(44.14±8.47),(45.36±10.11) and (44.38±8.81),respectively.According to Pearson correlation analysis,the patients' adherence to anticoagulation therapy was positively correlated (P＜0.01) with their Warfarin awareness (r=0.621) and social support (r=0.522) and negatively correlated (P＜0.01) with anxiety (r=-0.271) and depression (r=-0.319).Conclusions The adherence to anticoagulation therapy in patients after mechanical heart valve replacement is related to many psychosocial factors.It is positively correlated with their Warfarin awareness and social support and negatively correlated with anxiety and depression.

OBJECTIVETo explore the serological characteristics and molecular basis for an individual with para-Bombay phenotype.

METHODSBlood type of the proband was determined with routine serological methods. Exons 6 and 7 of the ABO gene and coding regions of the FUT1 and FUT2 genes were amplified by PCR and sequenced.

RESULTSThe para-Bombay phenotype was confirmed to be of Ah-secretion type. The genotype of the individual was determined as A102/O01. Position 328 of the FUT1 gene was mutated from A to G, resulting in replacement of Alanine (Ala) at position 110 by Threonine (Thr).

CONCLUSIONThe G to A mutation of nt328 of the FUT1 gene probably underlies the para-Bombay phenotype in this individual.
