1.Change of pancreatic blood flow and blood miR indexes in pancreatic cancer

Objective To study and observe the changes of pancreatic blood flow and blood miR indexes of patients with pancreatic cancer.Methods 70 patients with pancreatic cancer were selected as the observation group,70 health persons of the same age at the same period were selected as the control group, then the blood flow indexes and blood miR of observation group’s lesion and tumor adjacent tissue and control group’s healthy pancreas were detected,and the detection results of control group and observation group were compared,then the above indexes of observation group with different Hemrick stages were compared.Results The lesion blood flow indexes of observation group were all obviously worse than those of control group(P<0.05),blood miR indexes were all higher than those of control group(P <0.05),the lesion and tumor adjacent tissue blood flow indexes all had obvious differences(P<0.05),and the above indexes of observation group with different Hemrick stages all had significant differences(P<0.05).Conclusion The pancreatic blood flow and blood miR of patients with pancreatic cancer all show obvious abnormal state,and the differences of indexes of patients with different Hemrick stages are all prominent, so the pancreatic blood flow and blood miR indexes all have higher value for the patients with pancreatic cancer.

PPAR? is a kind of nuclear receptors responsible for the regulation of glucose and lipid metabolisim and adipocyte difference,which is related with the pathogenesis of obesity,insulin resistance and type 2 diabetes mellitus.PPAR? agonists as insulin sensitizer have been used in the clinical treatment for type 2 diabetes for more than decades.Recently,with the further research plunging into the PPAR signaling pathway and insulin sensitizer,the recognition of this crucial nuclear receptor has been renewed and developed.The aim of this review is to report the update research outcomes.
3.Clinical observation on prevention of chemotherapy infection in gastric cancer by moxa-stick moxibustion plus rhG-CSF and its effect on immune function

Objective: To observe the effect of moxa-stick moxibustion plus recombinant human granulocyte-colony stimulating factor (rhG-CSF) in preventing chemotherapy infection in gastric cancer and its effect on immune function. Methods: A total of 70 patients with gastric cancer treated by chemotherapy were randomly divided into an observation group and a control group, with 35 cases in each group. The control group was given rhG-CSF, and the observation group was given additional moxa-stick moxibustion on the basis of rhG-CSF. Both groups were treated for 2 chemotherapy cycles, totally 6 weeks. The number of patients with infection, the duration of infection and the duration of continuous use of antibiotics were observed. The leukocytes and granulocytes counts, the levels of tumor necrosis factor-α (TNF-α) and interferon-γ (IFN-γ) were measured before and after treatment, and the levels of CD4, CD8 and natural killer (NK) cells were analyzed. Results: The infection rate of the observation group was significantly lower than that of the control group (P<0.05), and the duration of infection and the duration of continuous use of antibiotics were also shorter (P<0.05). After treatment, the leukocytes and granulocytes counts in the two groups were higher than those before treatment (all P<0.05). After treatment, the levels of TNF-α and IFN-γ of the patients in the two groups were improved (all P<0.05), and there were significant differences between the observation group and the control group (P<0.05). After treatment, the numbers of CD4, CD8, and NK cells in the observation group increased significantly (all P<0.05), but the changes in the control group were not significant (P>0.05). Conclusion: Moxa-stick moxibustion plus rhG-CSF can significantly reduce the incidence and severity of chemotherapy infection in gastric cancer, increase the leukocytes and granulocytes counts, and regulate the levels of inflammatory factors, which may be related to the improvement of the immune function of the patients.
4.Clinical research of single glucocorticoid treatment and glucocorticoid joint cyclophosphamide treatment on thyroid-associated ophthalmopathy

ObjectiveTo explore the effect of single glucocorticoid treatment and glucocorticoid joint cyclophosphamide treatment on thyroid-associated ophthalmopathy.MethodsRetrospective analysis of clinical data of 35patients used single glucocorticoid( single medicine group) and 16 patients used glucocorticoids joint cyclophosphamide(joint group) was carried out.ResultsThe effective rate of joint group(93.8% ) was significantly higher than single drug group (68.6%) ( x2 =3.87,P < 0.05).Simple using glucocorticoid had significant treatment effect at CAS score,prominent eyes degrees,soft tissue inflammation score,but the curative effect was significantly lower than the joint group ( P < 0.05 ).The adverse reaction rate had no significant difference between two groups.ConclusionCompared to single glucocorticoid treatment,the glucocorticoid joint cyclophosphamide treatment had better effect and lower adverse reaction rate,especially suitable for patients who had no effect or low effect on hormone therapy.
5.EXPRESSION OF ECTONUCLEOTIDE PYROPHOSPHATASE/PHOSPHODIESTERASE 1 IN HUMAN OVARY AND ITS RELATIONSHIP WITH POLYCYSTIC OVARY SYNDROME

Objective Insulin resistance is a possible cause of polycystic ovary syndrome(PCOS).It is presumed that ectonucleotide pyrophosphatase/phosphodiesterase 1(ENPP1) is associated with insulin resistance.The purpose of this study is to explore the expression of ENPP1 in granulosa cells of the ovary and its relationship with PCOS. Methods Twelve aliquots of follicular granulosa cells were isolated from 12 samples of the patients with PCOS and 22 aliquots from 22 samples of the patients without PCOS,respectively.ENPP1 expression was analyzed by reverse transcription polymerase chain reaction(RT-PCR) and in situ hybridization.The relative expression of ENPP1 mRNA was detected by real-time quantitative polymerase chain reaction(real-time PCR). Results ENPP1 was expressed in granulosa cells of the ovary.ENPP1 expression's 2~(-?Ct) in granulosa cells of PCOS was significantly higher than non-PCOS(1.67?0.89 vs.0.94?0.76,P=0.017).Conclusion ENPP1 expression plays a role in ovary function and may be closely associated with the development of PCOS.

Berberine, an isoquinoline alkaloid isolated from some Chinese medicinal herbs such as Coptidis rhizoma, has been used for the treatment of diarrhea and other gastrointestinal infections as an antibacterial drug in Chinese medicine. In recent years, it was reported to have beneficial effects on the metabolism disorders states of diabetes. The mechanisms involve many aspects of the diabetes, including regulating the blood cholesterol and triglyceride, lowering blood glucose, ameliorating the insulin resistant state and influencing the function of the pancreatic beta cell.
7.The expression of IFN-γ and FOXP3 in the microenvironment of renal cell cancer, and its role in the tumor infiltrating lymphocytes distribution

Objective To investigate the role of IFN-γ and FOXP3 expression in subpopulation distribution and functions of tumor-infiltrating lymphocytes (TILs) in the microenvironment of renal cell cancer.Methods 30 renal cell cancer tissue samples were freshly collected from the laparoscopic radical nephrectomy in the first hospital of Jiaxing.After frozen sectioning,immunofluorescent staining was conducted to detect the infiltrating CD4 positive and CDs positive cells,and the expression of FOXP3 and IFN-γ as well.In addition,TILs were isolated from the tumor tissues by density-gradient centrifugation.TILs from tumor center or tumor invasive edge were purified independently and measured for the mRNA levels of FOXP3 and IFN-γ by qRT (quantitative reverse transcription)-PCR.Results Tumor-infiltrating CD4+ and CD8+ T cells were concentrated in the invasive edge of renal cell cancer tissues.The expression of FOXP3 was found to be inversely related to that of IFN-γ from the immunofluorescent staining.The relative FOXP3 mRNA levels for the TILs from tumor center and invasive edge were 64.6 ± 9.4 and 36.2 ± 1.8,respectively,with significant difference(P <0.05).The relative IFN-γ mRNA levels were 631.8 ± 151.4 and 1 726.0 ± 344.1 (P < 0.05).The trend of relative expression of FOXP3 was reversed in terms of IFN-γ.Conclusions The study on the renal cell cancer tissue samples suggested that the tumor-specific cytotoxic immune cells relatively concentrated in the tumor invasive edge.
8.Development of UPLC-Q-TOF-MS/MS combined with reference herb approach to rapidly screen commercial sulfur-fumigated ginseng.

An ultra-performance liquid chromatography-quadrupole/time of flight mass spectrometry (UPLC-Q-TOF-MS/MS) combined with reference herb method was developed to rapidly screen commercial sulfur-fumigated ginseng. Sufur-fumigated ginseng reference herb was prepared using genuine ginseng by conventional procedure. Then the reference sulfur-fumigated ginseng sample was analyzed by UPLC-Q-TOF-MS/MS to identify characteristic marker components. 25-hydroxyl-Re sulfate with higher abundance was se- lected as marker compound from 8 characteristic components identified in sulfur-fumigated ginseng reference herb. The fragmentation of 25-hydroxyl-Re sulfate was extensively investigated, fragment ion m/z 879.44 with higher intensity was chosen as the characteristic ion of sulfur-fumigated ginseng. The response of ion m/z 879. 44 was improved by optimizing the MS conditions so that this ion could be used as the characteristic marker ion for screening purpose in ion extracting screening mode. The established approach was successfully applied to inspect 21 commercial ginseng samples collected from different cities in China It was found that the chemical profiles of 9 samples were similar to that of sulfur-fumigated ginseng reference herb, and the characteristic ion m/z 879. 44 of 25-hydroxyl-Re sulfate was also detected in these samples, suggesting that there were nearly 43% ginseng samples analyzed being sulfur-fumigated. This findng agreed well with the results of sulfur dioxide residues of these 21 commercial ginseng samples determined with the method documented in Chinese Pharmacopeia Compared with the method documented in Chinese Pharmacopeia, the proposed approach is more rapid and specific for screening sulfur-fumigated ginseng. SFDA of China should strengthen the enforcement to prohibit ginseng being sulfur-fumigated, so that ginseng and it preparations could be effectively and safely benefit to the health of human beings.

9.Effect of gonadotropin-releasing hormone agonist on fertility potential in patients with stage Ⅰ endometriosis after laparoscopy

Objective To investigate the impact on fertility potential of patients with stage Ⅰ endometriosis treated by gonadotropin-releasing hormone agonist (GnRH-a) treatment followed by laparoscopy surgery.Methods From Jan.2006 to Jun.2008, medical documents of 102 patients with minimal or mild endometriosis (stage Ⅰ) treated by laparoscopy and hysteroscopy surgery and excluded the other causes of infertility were reviewed retrospectively in Peking University People's Hospital.The pelvic endometriosis teisions were destructed by electric burning.Those patients were divided into GnRH-a treatment group(n = 60) and non-GnRH-a treatment group(n = 42) after surgery.The patients in GnRH-a treatment group were administered by GnRH-a injection once or twice after surgery, in the mean time, no adjuvant therapy were given for the patients in the other group.All the patients were followed-up for 24 months after surgery to evaluate the effect of postoperative GnRH-a treatment on pregnancy outcome and fertility potential(number of pregnancy per 100 women in one month, expressed as percentage).Results The total pregnancy rate of 78% (47/60) in GnRH-a treatment group is significantly higher than 55% (23/42) in non-GnRH-a treatment group(P =0.012).There is no significant difference in spontaneous abortion rate between the GnRH-a treatment group and non GnRH-a treatment group(19% vs.13%, P = 0.465).The fecundity rate of were 6.17% in GnRH-a treatment group and 3.26% in non-GnRH-a treatment group, which the relative risk is 1.9 (95% CI: 1.1-3.0).Conclusion Low-dose GnRH-a treatment after laparuscopy can improve the pregnancy rate and fertility potential of patients with stage Ⅰ endometriosis.
10.Expression of chlamydia pneumoniae DNA in peripheral blood in patients with atherosclerosis obliterans

Objective To investigate the association between chlamydia pneumoniae(Cpn)and atherosclerosis obliterans(ASO). Methods Sixty ASO patients and 40 healthy persons were selected,and their Cpn DNA expressions in venous blood determined by polymerase chain reaction(PCR). Results The ASO patients had significantly higher positive expression rate of Cpn DNA than that of healthy persons(78. 33% vs. 47.50% ,P <0.05). In all 60 cases the Cpn DNA expressions were positively correlated with the severity of ASO. Conclusions Cpn DNA expression closely relates with ASO severity,and it plays an important role in the ASO pathogenesis.
