Chinese Journal of Biochemical Pharmaceutics 2017;37(9):53-54
Objective To observe the clinical effect of psychological intervention combined with somatostatin in the operation of pancreas. Methods From January 2016 to August 2017, 68 cases of pancreatic surgery were treated in our hospital. The patients were randomly divided into the observation group (n=34) and the control group (n=34). All patients were treated with somatostatin after the operation of pancreas. The patients in the observation group were treated with positive psychological intervention, and the effect of the application was observed. Results After the intervention, the two groups of patients with SAS and SDS were improved, the improvement is more obvious in the observation group (P<0.05);the observation group anal exhaust time, patients with gastrointestinal peristalsis time, bed time, hospitalization time were significantly shorter than the control group (P<0.05). Conclusion The combination of psychological intervention and somatostatin can improve the psychological status and promote postoperative recovery of patients after pancreatic surgery.

Objective To investigate the expression of phospho-histone H3 (PHH3) protein in pancreatic neuroendocrine neoplasms (PNENs) and explore its potential value for pathological grading of PNENs.Methods Clinical and pathological data of 283 patients with PNENs treated in Changhai Hospital from December 2000 to May 2016 were retrospectively analyzed.PNENs tissue chip was prepared, and immunohistochemistry was used to examine the expression of PHH3 and Ki67.The receiver operating characteristic curve for PHH3 was drawn and the area under the curve(AUC) was calculated to determine the cutoff value for PNENs grading.Ki67 was used for PNENs grading according to domestic and intemational criteria for PNENs classification. The relationship of PHH3 and Ki67 classification with clinical pathological features and prognosis of PNENs as well as the correlation between the two classification methods were analyzed. Results Of 283 patients,132 were male and 151 were female, aged from 16 to 78 years old.The average age was (50 ± 1) years old. There were 44 cases with functional PNENs and 239 with non-functional PNENs.The diameter of tumors ranged from 1.1 cm to 17 cm and the average diameter was (4.1 ± 1.2)cm.According to the Ki67 standard, there were 127,116,33 and 7 cases of Grade G1,G2,pancreatic neuroendocrine tumor(PNET) G3 and pancreatic neuroendocrine cancer(PNEC) G3.According to the PHH3 standard, there were 118,119,3 8 and 8 cases of Grade G1,G2,PNET G3 and PNEC G3.There was a positive correlation between the two grading criteria (r =0.941,P <0.001).PHH3 expression and PNENs grading were not correlated with age, gender, tumor functional or not and tumor locations (all P > 0.05),but were both positively correlated with tumor size, histological grade, lymph node metastasis,TNM stage and prognosis (all P <0.05).The average time of observing PHH3 and Ki67 staining per case in the immunohistochemistry of tissue chip was 9 min and 45 min, respectively. Conclusions Detection of PHH3 expression in PNENs can be used for the pathological diagnosis, grading and prognostic evaluation, which was more accurate and convenient than Ki67 criteria.

[Objective]To study NGF,BDNF gene modified bone marrowe stem cells transplantation with vein on spinal cord injury.[Method]Spinal cord injured rats were divided in to 3 groups.Group C consisted of the rats treated with the transplantation of modified NGF,BDNF genes BMSCs;group B of the rats with transplantation of BMSCs without genetic modification;group A of the rats without treatment and group D was the normal contral. At 2,6,10 weeks the rats were examined with GBS score and neurological function were tested.[Result]The vein with tintracord transplantation of modified BMSCs group was improved by the GBS score and neurological outcome of injured spinal cord was achieved well.[Conclusion]The vein with transplantation of modified NGF,BDNF genes BMSCs can recover of spinal cord injury.
7. Interference of curcumin in cycle of HepG2 cells by regulating microtubule system
Chinese Traditional and Herbal Drugs 2013;44(9):1144-1148
Objective: To investigate the effect of curcumin (Cur) on cycle in HepG2 cells and the action of microtubule system for it. Methods: The proliferation inhibition of Cur on HepG2 cells was evaluated by MTT assay; The effect of Cur on the cycle of HepG2 cells was analyzed by flow cytometry; Confocal laser scanning microscopy (CLSM) was used to observe the changes of microtubule in HepG2 cells after treated with Cur; Western blotting was performed to determine the expression of α-tubulin; The in vitro effects of Cur on tubulin polymerization and depolymerization activities were studied. Results: Cur inhibited the proliferation of HepG2 cells in a dose- and time-dependent manner. With Cur concentration increasing, the proportion of HepG2 cells arrested in G2/M phase was also increased. CLSM showed that Cur could seriously destroy the cell microtubule structure and change the polymerization state of tubulin. Western blotting showed that the decrease of the expression level of α-tubulin was related to Cur concentration. Cur could also interfere the microtubule polymerization and depolymerization activities. Conclusion: Cur could inhibit the proliferation of HepG2 cells by destroying the microtubule structure and downregulating the expression of α-tubulin, and make HepG2 cells arrested in G2/M phase.

The challenge of the emergence of drug-resistant influenza strains, which is caused by wide spread utilization of direct-acting antivirals (DAAs), accelerates the research and exploration towards host targeted agents. In contrast to DAAs targeting viral replication components, host targeted agents, which regulate host factors and pathways linked to viral replication, can interfere the replication of influenza. Additionally, the innate immune system is activated by influenza during the early stage of infection, so manipulating the innate immune response may prevent the viral infection. However, the excessive inflammatory response induced at the late phase of influenza infection would lead to severe tissue injures. Thus, it is very important to explore drugs with anti-inflammatory actions to suppress these immune imbalances and tissue injures. Here we overview the current progresses about host targets related to anti-influenza drugs.

In recent years, antibiotics have been playing more and more important roles in maintaining human health with the increasing variety and quantity. However, under the influence of different reasons caused by subjective or objective factors, antibiotics irrational use has induced bacterial resistance and adverse reactions, which are becoming more and more serious. The paper put forward the countermeasures of antibiotics irrational use and provided reference for rational use of antibiotics based on the present situation analysis in China and foreign experience in order to control the irrational use.
