Journal of Applied Clinical Pediatrics 2004;0(09):-
Objectives To study health-related quality of life(HRQOL) of children with subclinical epileptiform electroencephalographic discharges. Methods Subjects were 30 children with subclinical epileptiform electroencephalographic discharges,50 children with clinical seizures,28 children without clinical seizures and 30 controls.Guardians completed a valid epilepsy-specific HRQOL questionnaire for children, the quality of life in children epilepsy questionnaire(QOLCE).Results Children with subclinical epileptiform electroencephalographic discharges have 8 subscales scores lower than control group,2 subscales scores higher than group with clinical seizures. Conclusions Quality of life of children with subclinical epileptiform electroencephalographic discharges is better than that of children with clinical seizures. However cognitive, behavior, daily life and social activity are impaired by subclinical epileptiform electroencephalographic discharges and lowered the HRQOL of children.

The maxillary first premolar is a transitional tooth between incisors and molars. Most maxillary first premolars have 2 root canals. However, the bilateral maxillary first premolar with 3 roots and 3 canals has not been described. This article reports a rare case:The bilateral maxillary first premolar with 3 roots and 3 canals.

China Oncology 2014;(4):279-283
Background and purpose:Studies have shown that ribosomal protein L8 (RPL8) is shared by melanomas, gliomas and ovarian carcinomas. A peptide of RPL8 signiifcantly stimulated proliferation and cytokine expression of the hepler T cell clone and lymphocytes in melanoma patients. RPL8 may stimulate anti-tumor immunity, making RPL8 an attractive candidate for therapeutic intervention. In this study, we prepared DC pulsed by RPL8 (RPL8-DC) and investigate the anti-tumor effect of RPL8-DC on melanoma in mice. Methods: The recombinant protein was achieved through IPTG induction in E. coli and identiifed with Western blot. Bone marrow-derived DC was loaded with RPL8 protein. RPL8 and CD11c, CD80, MHC-Ⅰ, MHC-Ⅱmolecules on dentritic cells were monitored by lfuorescence microscope and FACS analysis, respectively. The anti-tumor effect of T cells in vitro was detected by MTT assay. Subcutaneous tumors were induced in C57BL/6 mice using B16 cells. The tumor volumes were measured after injection with RPL8-DC. Results:The puriifed protein was combined with speciifc antibodies. DCs pulsed by RPL8 were visualized under lfuorescent microscopy. CD11c, CD80, MHC-Ⅰ, MHC-Ⅱmolecules on DCs were up-regulated after stimulation with RPL8 and LPS. B16 cells were inhibited by T cells stimulated with RPL8-DC. The inhibition rate of tumor cells was 70%in RPL8-DC group when effector-to-target ratio was 30∶1, which was higher than PBS and DC groups. Inhibition of growth could be observed more signiifcantly in mice after the treatment with RPL8-DC. The mice receiving the therapy of RPL8-DC were able to survive much longer than the mice receiving control therapy. Conclusion:The DC pulsed by RPL8 protein can inhibit the growth of melanoma.

Objective To study the effect and safety of dexmedetomidine on the intraoperative fentanyl requirements and cerebral hemodynamics in patients undergoing intracranial tumor surgery. Methods 62 patients scheduled for intracranial tumor surgery were selected and divided into observe group (31 cases) and control group(31 cases).Dexmedetomidine (0.5 μg/kg) was loaded in observe group, and followed by a continuous infusion of a rate of 1.0pμg · kg-1 · h-1 half an hour later.Saline was infused in the same time and the same way in control group.Dose of anesthetic and change of cerebral hemodynamics were recorded and compared. Results The mean heart rate and mean arterial blood pressure were lower in observe group than the underlying value when inducing anesthesia and during the operation (all P < 0.05),while that happened after operation starting 60 minutes later in control group(all P <0.05).The mean heart rate and mean arterial blood pressure were lower in observe group those that in control group (all P < 0.05).Dose of fentanyl and end-tidal sevoflurane concentration were lower in observe group(all P < 0.05).Conclusion Dexmedetomidine used for cerebral hemodynamics stabiliby in intracranial tumor surgery has obvious curative effect and good safety. And it could decrease the dose of anesthetics and anesthesia adjuvant.

Objective To determine the effects of Che A and CheY proteins of Campylobacter jejuni regulating the bacterial chemotaxis in vitro and colonization in vivo. Methods By using pET42a plasmid and E. coli BL21DE3 as expression vector and expression strain, respectively, prokaryotic expression systems of cheA and cheY genes of C. jejuni strain NCTC11168 was constructed. Rabbits were immunized with the target recombinant expression proteins, rCheA and rCheY, to prepare the antisera. rCheA-IgG and rCheY-IgG in the antisera were separated using DEAE-52 ion exchange column. pBluescript- II -SK was applied to construct suicide plasmid which used to generate cheA gene knock-out mutant (cheA-). A chemotaxis model in vitro of C. jejuni based on DOC-HAP, the chemotactic ability of cheA' mutant as well as the effect of rCheA-IgG and closantel inhibiting the bacterial chemotaxis were demonstrated. The phosphorylation levels of CheA and CheY after DOC treatment were examined by using either rCheA-IgG or CheY-IgG capture method. The difference of colonization ability between cheA- mutant and wild-type of C. jejuni in mice were checked and then compared. Results The constructed prokaryotic expression systems could efficiently express rCheA and rCheY. The data from PCR and sequencing confirmed the cheA gene knock out from cheA- mutant chromosome. cheA- mutant lost its chemotactic ability towards DOC. Both the rCheA-IgG and closantel could inhibit the chemotaxis of wild-type of C. jejuni (P < 0.05 ). When treatment of DOC, the phosphorylation levels of CheA and CheY in wild-type of C. jejuni rapidly decreased (P < 0. 05 ). The colonization ability in murine jejunum of cheA- mutant was also lower than that of the wild-type ( P<0.05 ) . Conclusion Chemotaxis-associated two-component signaling system (Che-TCSS) of C. jejuni are composed of CheA and CheY, and the two proteins are activated by dephosphorylation. CheA in the Che-TCSS play a critical role in chemotaxis in vitro and colonization in vivo of C. jejuni, and this protein can be used as a target for developing novel anti- C. jejuni drugs.
