1.Findings of styloid Process and clinical value of 16-slice spiral CT postprocessing in investigating styloid process in vivo

[Objective]To investigate the value of 16-slice spiral CT(MSCT)postprocessing in styloid process in vivo and provide the vivisection data for diagnosing and curing styloid process syndrome.[Method]Totally 100 volunteers with normal styloid processes underwent 16-slice spiral CT(MSCT). Based on the initial datum,4 kinds of postprocessing including multiple planar reconstruction(MPR), curve planar reconstruction(CPR),surface shaded display(SSD) and volume rendering (VR) were performed.The length and inclination angle of stytoid processes were measured. The distance of between styloid processes and carotid canal external aperture,jugular foramen, stylomastoid foramen ,mastoid process, internal carotid artery and internal jugular vein were surveyed. [Result]The average length of styloid processes was 24.51 mm?3.05 mm, left was 24.33 mm?3.15 mm and right was 24.69 mm?2.95 mm. The anterior inclination angle was 19.35??2.92? and medial was 24.00??4.07?.The distance of between styloid processes and carotid canal external aperture, jugular foramen,stylomastoid foramen ,mastoid process , internal carotid artery and internal jugular vein were 8.53 mm?2.45 mm, 2.87 mm?0.88 mm, 2.34 mm?0.74 mm, 17.23 mm?1.85 mm,6.49 mm?1.58 mm and 3.70 mm?1.46 mm respectively. [Conclusion]Sixteen-slice Spiral CT postprocessing demonstrate the stytoid processes in vivo accurately and fully, among which multiple planar reconstruction and surface shaded display are more valuable, because they can evaluate the length and inclination of the styloid processes and the distance between the processes and their surrounding structures in vivo more accurately.
2.Human umbilical cord mesenchymal stem cells under atomic force microscope: Correlation between biological characteristics and ultrastructure

BACKGROUND: The relationship between cellular morphosis and function is undetachable, but there is little investigation about ultrastructure of human umbilical cord mesenchymal stem cells (hUCMSCs).OBJECTIVE: To study the relationship between the functions of hUCMSCs and their ultrastructure obtained by atomic force microscope (AFM). METHODS: hUCMSCs were isolated, cultured, expanded after enzyme digestion. P3 cells were observed under AFM. Immunophenotype and cell cycle were analyzed by flow cytometry, as well as induction of the adipogenic, osteogenic differentiation of hUCMSCs were identified using Oil red O staining and alkaline phosphatase staining. RESULTS AND CONCLUSION: hUCMSCs at passage 3 were strongly positive for CD44 and CD29, weakly positive for CD106, but negative for hematopoietic marker CD34. Cells in G_0/G_1 phase accounted for 80%. Proliferation index was 19.9%. Following adipogenic induction, alkaline phosphatase staining demonstrated brown cytoplasm in cube and polygonal cells. AFM showed hUCMSCs were spindle shape, obvious cytoskeletal filament that connected into nets, which fit for the strong capacities for proliferation, migration and differentiation.
3.Primary culture and identification of rat skeletal muscle-derived stem cells

BACKGROUND: Skeletal muscle-derived stem cells (MDSCs), another adult pluripotent stem cells, have become a hot topic in the field of gene therapy and cell-based tissue engineering. It has wide prospect in treating stress incontinence by periurethral injection of patients' MDSCs.OBJECTIVE: To investigate the method of culturing MDSCs in vitro so as to provide experimental basis for treating stress incotinence by injecting MDSCs.DESIGN: Repeated observation.SETTING: Department of Reproductive Medical Center and Department of Urology, Renmin Hospital of Wuhan University.MATERIALS: This experiment was conducted at the Laboratory of Department of Urology , Renmin Hospital , wuhan University from December 2003 to May 2004. Totally 20 female SD rats , aged 4 to 6 weeks , were involved . Type- Ⅺ pancreatin, collgenase , pancreatin ,polylysine (Sigma), dispase enzyme (Gibco) , chick embryo extract (self made).METHODS: ① Rats were anesthetized intraperitoneally with 10 g/Lpentobarbital sodium (30 g/kg). Under aspetic condition, gastroeneminus were isolated and immediately put into pre-cooled DMEM media (Gibco)containing antibiotics and then removed into the hood. After washed with D-hank's solution three times, muscle biopsies were removed of fascia,tendon, nerve and blood vessels and minced into small pieces about 1-3 mm3,and then transferred into a centrifugation tube. 0.2% collgenase-type Ⅺ(Sigma) and 0.1% pancreatin were added to the left tissue . Skeletal muscle cells of the rats were isolated with collagenase. ② Differential attachment was used to purify the skeletal muscle cells of the rats. After screened with cell screen cloth , cell suspension was transferred into a polylysine-coated flask and cultured at 37 ℃ in a humid atmosphere with 0.05 CO2 in air for 1 hour. All the cells that did not adhere to the flask were then transferred to another flask with new culture medium and cultured for approximately 1 hour at 37 ℃ .Again,the non-adhering cells were transferred to another flask and were incubated at 37 ℃ overnight.The technique was carried out for an additional 4-5 days. This operation was repeated every 24 hours until the fifth day. Those cells attached on days 5-6 were MDSCs.③ After they grew with 70% confluence , the cells were digested with trypsin and passaged at the ratio of 1:2. After digestion, the generative cells were inoculated to the culture plate with 6-well cover glass. Growing culture medium was added and cell slide was prepaared 24 hours later. The expression of specific protein antigen and stem cell antigen-1 of the cultured cells were identified with immunohistochemical staining.MAIN OUTCOME MEASURES: Morphology and identification of MDSCs.RESULTS: ① Morphology of MDSCs: Cells isolated from skeletal muscle tissue took the form of spheres and had high refraction. When subcultured,they began to adhere at the 12th hour, at that time they were still round,and complete their attachment at the 48th hour. Then they began to expand and ultimately became fusiform-shaped or spindle-shaped, having two poles and small size. As time going, they fused with each other to form mature poly-nuclear myotubes. ② Identification results of muscle-derived stem cells: MDSCs were desmin and stem cell antigen-1 (Sca-1) positive specific for some stem cells. Red fluorescence effluenced from cytoplasm was found under the fluorescence microscope and the positive rate reached 90%.CONCLUSION: MDSC belongs to adult stem cells and has the advantages of rich source, low immunogenicity and long survival after transplantation. High-purity muscle-derived stem cells can be obtained through primary culture , and immunohistochemical technique can identify muscle-derived and specific characteristics of stem cells, so it has a broad application perspective in tissue engineering and gene therapy.
4.The role of HBXIP overexpression in the prognostic evaluation of breast cancer

Objective To investigate the role of HBXIP overexpression in the prognostic evaluation of breast cancer. Methods HBXIP expression was detected in the tissues of 60 breast cancer cases and 15 cases of ductal cancer in situ (DCIS) as well as in the adjacent non-tumorous tissues of 27 cases of breast cancer using EnVision immunohistochemical staining method. The correlations between the HBXIP overexpressions and the clinical pathological characters of the patients with breast cancer were also analyzed. Results The HBXIP proteins showed a major cytoplasmic staining pattern in breast cancer. The strongly positive rate of HBXIP was75.0%(45/60) in breast cancer, significantly higher than in DCIS (20.0%, 3/15) and adjacent non-tumorous tissues (14.8%, 4/27). High-level expression of HBXIP was correlated with late clinical stage, lymph node metastasis and HER-2 positive expression in breast cancer. Conclusions The expression level of HBXIP is closely related to the progression and prognosis of breast cancer. It might be a potential biomarker and therapeutic target of breast cancer.
5.Present situation investigation and literature analysis of unique copy TCM ancient books

Objective To investigate the present condition of unique copy of TCM ancient books, and analyze the literature value. Methods Bibliographic research was underwent first, then investigation table was made, investigation team for spot investigation was organized, the bibliography was sorted, and on this basis analysis of the literatures was performed. Results 59 collection units at home and abroad and 1370 unique copy TCM ancient books were investigated, which including 57 books published by photocopy and 37 books published by proof reading. Conclusion The unique copy TCM ancient books have important academic value and can be effectively saved and sorted through in-depth investigation and analysis.
6.Discussion about the application of topic maps technology on knowledge organization of traditional Chinese medicine with the example ofShanghanlun

Literature of TCM contains abundant information of Chinese medicine. Traditional Chinese medicine information is characterized by its high degree of knowledge-intensive and complex inter-knowledge, so it is difficult to integrate and share knowledge. TheShanghanlun is one of the Chinese medical classics, and it includes the rules and methods of syndrome differentiation and treatment and it showed good effects in the clinical application. However, it is difficult to master for beginners. As a kind of knowledge organization tool, topic maps can flexibly define the relationship between knowledge, which realizes the visualization of knowledge network, and integrates and shares the distributed knowledge. This paper focused on the current situation of topic maps researches. The topic maps was used to organize the original content ofShanghanlun, construct the concept model of it and explore the specific application of topic maps technology in the field of traditional Chinese medicine.
7.Efficacy of milrinone combined with dopamine in the treatment of elderly patients with refractory heart failure and the influence of levels of cardiac function and N-terminal B-type natriuretic peptide precursor

Objective To analyze and investigate dopamine combined with milrinone treatment effect on elderly patients with intractable heart failure and N-terminal B-type natriuretic peptide level and cardiac function.Methods 80 cases of elderly patients with heart failure according to the number table method randomly divided into two groups: control group and experimental group, and control group was given conventional drugs.The patients in experimental group were received dopamine +Milrinone on the basis of control group.Clinical efficacy, N-terminal pro-B-type natriuretic peptide levels ( NT proBNP ) and heart function condition between two groups are compared and analyzed.Results The total effective rate of experimental group (95.00%) was higher than that of control group (70.00%) (P<0.05).NT-proBNP(2013.31 ±295.84)ng/L、LVEDD(61.48 ± 10.11)mm、LVEF(59.69 ±8.44)% in the experimental group was significantly better than the control group(P <0.05).Conclusion Dopamine combined with milrinone in the treatment of elderly patients with intractable heart failure is remarkable, can relieve the level of NT proBNP, and promote the recovery of cardiac function.

9.Study on the compiling and arrangement of medical literature in local chronicles

10.Study on CD4~+CD25~+ regulatory T cells and CD4~+,CD8~+ T lymphocyte subgroup in peripheral blood of patients with chronic hepatitis B

Objective:To investigate the quantification of CD4~+CD25~+ regulatory T cells and distribution of CD4~+CD8~+ T lymphocyte subgroup in peripheral blood of patients in chronic hepatitis B (CHB),and to reveal relationship between CD4~+CD25~+ regulatory T cells,CD4~+CD8~+ T lymphocyte subgroup and HBV infetion as well.Methods:CD4~+CD25~(high),CD4~+CD25~+Foxp3~+Treg and CD3~+CD4~+CD8~+T lymphocyte subgroup in peripheral blood from 50 patients with CHB and 20 healthy controls was analyzed using flow cytometry.HBV DNA was detected by fluorescence quantitative PCR.Results:The number of CD4~+CD25~(high)Tregs in patients with CHB was obviously higher than that in healthy controls(P<0.01)and increased with copies of HBV DNA.The same with the change of CD4~+CD25~+Foxp3~+Tregs in patients with CHB and there was a positive correlation between CD4~+CD25~(high)Tregs and CD4~+CD25~+Foxp3~+Tregs(r=0.890,P<0.001).Compared with healthy controls,the frequency of CD4~+T cells and the ratio of CD4~+/CD8~+ in patients with CHB was declined,but there was no significant difference in the frequency of CD3~+T cells and CD8~+T cells between them(P>0.05).The variation in the number of CD4~+CD25~(high)Tregs was correlated positively with the copies of HBV DNA(r=0.782,P<0.001)and glutamic-pyruvic transaminase(ALT)(r=0.432,P<0.005)separately,but negatively with the frequency of CD3~+,CD4~+,CD8~+T cells and the ratio of CD4~+/CD8~+(P>0.05).The variation in the frequency of CD3~+,CD4~+,CD8~+T cells and the ratio of CD4~+/CD8~+ was also correlated negatively with the copies of HBV DNA(P>0.05).Conclusion:The number of CD4~+CD25~(high)Tregs increases in patients with CHB and is in accordance with the copies of HBV DNA and increased level of ALT.Further studies should be done to investigate weather CD4~+CD8~+ T lymphocyte subgroup could be used to monitor the state of community.
