Chinese Pharmacological Bulletin 2003;0(08):-
Livin is a novel member of IAPs (inhibitors of apoptosis protein) family, which is expressed highly in a variety of tumors but not in majority of normal tissues. This protein contains a BIR domain and a RING finger domain just like other members of IAPs. It will be of great sigificance to investigate the relationship between Livin and tumors, and it might be a potential target for anti-cancer drugs.

AIM: Antisense oligonucleotides against livin were designed with computer software. METHODS: Antisense oligonucleotides were designed according to the secondary structure of livin mRNA which was simulated with RNAdraw and Sfold. And then these oligonucleotides were transfected into Hela cells for inducing apoptosis. RESULTS: Five antisense oligonucleotides were designed using RNAdraw and Sfold, and effectively induced Hela cell apoptosis. CONCLUSION: The approach is effective and feasible to design antisense oligonucleotides by means of calculation with two kinds of software.

Objective The preservation of all subvalvular apparatus for chronic mitral regurgitation has been reported in many studies. In this paper we analyzed the effects of complete chordal preservation for mitral stenosis versus mitral regurgitation on postoperative recovery. Methods Thirty-nine patients undergoing mitral valve replacement(MVR) with preservation of all subvalvular apparatus were divided into two groups, mitral stenosis (MS) group(n=23) and mitral incompetence (MI) group(n=16), according to the preoperative diagnosis. Echocardiography was performed preoperatively and at the time of discharge to determined left ventricular end diastole dimension (LVEDD),ejection fraction(EF) and the hemodynamics of the prosthesis. Results The LVEDD and EF did not change in MS group, declined in MI group postoperatively. No hemodynamic disorder of the prosthesis was detected in either group. Conclusions The valve replacement with preservation of all subvalvular apparatus could be used in both the mitral stenosis and mitral regurgitation. The early-term effects of two groups are similar, and the long-term effects are still to be observed.

?-MSH is an endogenous neuropetide that is effective of all categories of expreimental inflammation. The mechanisms underlying the anti-inflammatory influence of ?-MSH are reviewed in the article. ?-MSH suppresses inflammation in the CNS and periphery by downregulating the activation of NF-?B, then inhibiting production of proinflammatory cytokines, nitric oxide, chemokines and adhension molecules, and increasing synthesis of anti-inflammatory cytokines. ?-MSH is useful in the treatment of many pathological situations in humans.

α-MSH is an endogenous neuropetide that is effective of all categories of expreimental inflammation. The mechanisms underlying the anti-inflammatory influence of α-MSH are reviewed in the article. α-MSH suppresses inflammation in the CNS and periphery by downregulating the activation of NF-κB, then inhibiting production of proinflammatory cytokines, nitric oxide, chemokines and adhension molecules, and increasing synthesis of anti-inflammatory cytokines. α-MSH is useful in the treatment of many pathological situations in humans.

AIM:To study the expression of glycine receptorα1 subunit in neonatal rat myocardial cells and to investigate the effect of lipopolysaccharide (LPS), hypoxia/reoxygenation, isoproterenol (ISO) and high concentration of glucose (HG) on the expression of glycine receptorα1 subunit in the neonatal rat myocardial cells.METHODS:Neonatal rat myocardial cells were cultured in vitro. The expression of glycine receptorα1 subunit was detected by Western blotting. The neonatal rat myocardial cells were treated with LPS (20 mg/L), ISO (100 μmol/L) or high concentration of glucose (25 mmol/L) for 24 h, or were exposed to hypoxia for 3 h followed by reoxygenation for 3 h.Subsequently, the cell viabil-ity was measured by CCK-8 assay, and the expression of glycine receptorα1 subunit was determined by Western blotting. RESULTS:The expression of glycine receptor α1 subunit in the neonatal rat myocardial cells was positively detectable by Western blotting.Compared with control group, no significant difference of the cell viability ( P>0.05) in LPS group, ISO group, hypoxia/reoxygenation group and HG group was observed. The expression of glycine receptor α1 subunit was in-creased (P<0.01) in LPS group, ISO group and hypoxia/reoxygenatio group, but decreased (P<0.01) in HG group. CONCLUSION:Glycine receptorα1 subunit exists in the neonatal rat myocardial cells.A certain concentration of LPS or ISO, or hypoxia/reoxygenation for a certain period upregulate the expression of glycine receptorα1 subunit, but HG down-regulates the expression of glycine receptor α1 subunit in cultured neonatal rat myocardial cells.

Objective To explore the application of cancer pain standardized treatment in patients with lung cancer pain and clinical results. Methods 120 cases of lung cancer pain patients as the research object in Our hospital from April 2015 to June 2016,The patients were randomly divided into study group and control group,60 cases in each group, the patients in the control group were treated with routine therapy and nursing, and the patients in the study group were treated with normalized treatment and care of cancer pain, and then compared the two groups after treatment Pain status changes and adverse events and patient satisfaction statistics. Results There was no significant difference in pain status and VAS score between the two groups before treatment, but the pain status was relieved after treatment. The pain status of patients in the study group after standardized treatment of cancer pain changed significantly, the VAS score was lower than the control, the difference between the two groups was statistically significant(P<0.05).After treatment, the adverse events of the study group including non-on-time medication, syncope pressure sores and the incidence of self-mutilation or suicide were statistically significant And the difference was statistically significant(P<0.05).The satisfaction of the patients in the two groups showed that the satisfaction of the patients in the study group was significantly higher than that of the control group,The difference between groups was statistically significant(P<0.05).Conclusion The standardized treatment of cancer pain is effective in the treatment of patients with lung cancer pain.It can effectively relieve the pain and improve the quality of life and satisfaction of patients with lung cancer pain.It has broad clinical application and popularization value.

AIM: To assess the effect of recombinant human bone morphogenetic protein-2 (rhBMP-2) combined with type I collagen on pulp capping in cats. METHODS: Freshly exposed pulps of thirty cats were capped with four different pulp capping agents: rhBMP-2/collagen I, calcium hydroxide, type I collagen and bovine serum albumin as control. The reparative dentinogenesis and osteocalcin content were assessed after 1 week, 2 weeks and 4 weeks by means of histology and radioimmunoassay. RESULTS: the pulpal tissues of the teeth capped with rhBMP-2/collagen showed mild inflammation and odontoblast differentiation with tublar dentin bridge on the exposed site. The level of bone ?-carboxyglutamic containing protein(BGP) in the group of rhBMP-2 with collagen I was significantly higher than that of the other three groups (P
