1. Protection of puerarin on impaired vascular endothelial function induced by glycosylated protein

OBJECTIVE: To investigate the effects of puerarin on impaired endothelium-dependent relaxation induced by glycosylated bovine serum albumin (GBSA) in rabbit thoracic aorta and its mechanisms. METHODS: The rings were incubated with GBSA for 30 min to induce endothelial dysfunction, and with puerarin(0.25, 0.5 and 1 g · L-1), A-nitro-L-arginine methyl ester (L-NAME, 30 νmol · L-1), and indomethacin(10 μmol · L-1) to investigate the protective effect of puerarin on impaired vascular endothelial function elicited by GBSA, and the effect of L-NAME and indomethacin on the protective effect of puerarin. Moreover, the content of nitric oxide (NO) and malonaldehyde (MDA) and the activity of superoxide dismutase(SOD) in the rings were measured. RESULTS: Exposure of aortic rings to GBSA (200 mg · L-1) for 30 min resulted in a significant inhibition of endothelium-dependent relaxation, but had no affect on endothelium-independent relaxation. GBSA significantly decreased the level of NO and the activity of SOD but hugely increased the content of MDA in vascular tissues. Pre-incubation of aortic rings with puerarin markedly attenuated the inhibition of endothelium-dependent relaxation induced by GBSA. This protective effect of puerarin (1 g · L-1) was partially inhibited by L-NAME but not by indomethacin. Puerarin obviously increased NO level and SOD activity but evidently decreased MDA content in rings. CONCLUSION: Puerarin can protect against vascular endothelial dysfunction caused by GBSA and its mechanisms may be related to enhancing NO synthesis and its anti-oxidation.
2.Analysis of IgH gene rearrangement and its significance in non-Hodgkin lymphoma

Objective To explore the significance of detecting IgH gene rearrangement in diagnosis of non-Hodgkin lymphoma. Methods By using family-specific primers mixtures (VH1,VH2,VH3) for IgH FR3A,FR2A,FR1 genes,clonal IgH gene rearrangement was detected by heminested and multiplex PCR in 44 cases of B-NHL,1 undifferentiated lymphoma,15 T-NHL and 5 lymphonode reactive hyperplasia. Results Clonal IgH gene rearrangement was detected in 36 of 44 B-NHL (positive rate 82%) by using FR3A primers,27 of 44 B-NHL (positive rate 61%) by using FR2A primers. By using FR1 family-specific primers mixtures (VH1,VH2,VH3) and JH (JHb,JHc),clonal IgH gene rearrangement was demonstated respectively in 34/33 of 44 B-NHL (positive rate 77%,75%). The total positive rate was 95% (42/44) by combined family-specific primers mixtures (VH1,VH2,VH3) of FR3A,FR2A,FR1 gene. Four of 15 T-NHL revealed clonal IgH gene rearrangement,while 5 LRH had no clonal IgH gene rearrangement. One NHL unclassified by immunohistochemical methods was classified definitely as B cell-derived lymphoma by PCR. Conclusion The detection rate of IgH gene rearrangement can be improved by combined usage of several pairs of primers,especially family-specific primers. Detecting clonal IgH gene rearrangement is significant in the procedure of differential diagnosis for B/T-NHL and reactive hyperplasia.
3.Establish Assessment Model of 18 Years of Age in Chinese Han Population by Mandibular Third Molar.

OBJECTIVE:
To explore the value of estimating chronologic age based on the grades of mandibular third molar development. To evaluate whether mandibular third molar could be used as an indicator for estimating the age under or over 18 years.
METHODS:
The mineralization status of mandibular third molar of 1 845 individuals aged 10 - 30 was graded and marked based on Demirjian's classification of grades reformed by Orhan. Gender difference was examined by t-test. A cubic regression model was established to analyze the correlation between third molar and chronologic age. Each grade of age cumulative distribution diagram and ROC curve was respectively performed to evaluate the relationship between third molar and the age of 18. Using Bayes discriminant analysis, an equation was established for estimating the age of 18.
RESULTS:
The inner-rater reliability was 0.903. Statistical analysis showed a moderate correlation between age and grade. Significant differences of both genders were found only in grade D and H (P < 0.05). Males at the grades from 1 to D and females at the grades from 1 to C were under 18 years old, and both males and females at grade H were over 18 years old. The area under the ROC curve was 0.797 (P < 0.05).
CONCLUSION
Third molar development shows a high correlation with age, and combined with other indicators, it can be used to estimate the age of 18.

4.Correlation between B beta-Fibrinogen Gene Polymorphism,Plasma Fibrinogen Level and Henoch-Schoenlein Purpura in Children

Objective To investigate the association of B?-fibrinogen gene-455G/A polymorphism and plasma fibrinogen levels with Henoch-Schoenlein purpura(HSP) in children.Methods Sixty-seven children(including 40 boys and 27 girls) with HSP were served as HSP group,age ranging from 5-14 years,with the average age of 9 years.Seventy healthy controls(including 37 boys and 33 girls) were served as healthy controls.Age ranging from 5-13 years,with the average age of 9 years.The B?-fibrinogen gene-455G/A polymorphism was detected in all subjects by polymerase chain reaction-restrictive fragment length polymorphism technique with restrictive enzyme HaeⅢ.Results There were a greater proportion of individuals with the AA,GA,GG genotype in the HSP group comparied with those of the healthy controls(?2=29.5 P0.05].Conclusions The B?-fibrinogen gene-455G/A mutation is correlated with HSP in children,A allele is susceptibility gene of HSP in children.The plasma fibrinogen level is related to HSP in children.
5.Relationship between Body Height and Craniofacial Lines Measured by CT in Southwest Han Males.

OBJECTIVE:
To establish regression model between craniofacial lines and body height by measuring craniofacial lines in Southwest Han males using CT and to accumulate data for the study of forensic anthropology.
METHODS:
Head CT data of 273 Han males in Southwest were collected and 7 craniofacial lines were determined. Multiplanar reconstruction and volume rendering were performed by image post-processing software and the selected lines were measured. The relationship between each measuring indicator and body height was analyzed using SPSS 21.0 software. The regression equation of body height estimation was established and 50 samples were selected again and put into the mathematics models to verify its accuracy.
RESULTS:
The linear regression equations of 7 lines were established (P < 0.05). The correlation coefficients of the unary linear regression equations were 0.190-0.439 and the standard errors of the estimate (SEE) were 4.597-5.023 cm. The correlation coefficients of the multiple linear regression equation were 0.494-0.524 and the SEE were 4.418-4.458 cm. The return tests showed that the highest ± 1SEE accuracy of the multiple regression equation: y = 83.959+3.589 x6+2.573 x2, were 30%; and the highest ± 2SEE accuracy of the multiple regression equation: y = 72.646+3.316 x6+1.586 x2+1.553 x4+2.211 x3, were 92%.
CONCLUSION
There is significant linear correlation between 7 selected lines and the stature in this study, and the plural linear regression equation established could be applied for estimating the stature of Southwest Han males.

6.Initial study of optimal single-voxel ~1H-MR spectroscopy parameters on femoral bone marrow

Objective To choose proper proton magnetic resonance spectroscopy(~1XH-MRS) parameters to fit for practical femoral marrow cavity and to produce short-timed,well-repeated and excellent ~1H-MRS images.Methods The tentative study of ~1H-MRS on the normal femoral bone marrow in 26 volunteers was performed with a 1.5 T MR after the informed consent.The single-voxel spectroscopy and stimulated echo acquisition mode were used for ~1H-MRS collection.~1H-MRS parameters for 12 volunteers were 128 acquisitions,1 cm?1 cm?1 cm volume of interest(VOI)size and repeatedly 2—3 times within the same location.~1H-MRS parameters for another:14 volunteers were different numbers of acquisition (128 and 256 times,respectively)and different VOI sizes(2 cm?2 cm?2 cm and 1 cm?1 cm?1 cm, respectively).Results For ~1H-MRS with 1 cm?1 cm?1 cm size of VOI and 128 times of acquisition with the full width haft max of water≤8—12 Hz,the base-line was steady and the signal-noise ratio was high up to 11.31.~1H-MRS was different in the different femoral locations showing the maximum peak sites at near 0.90 ppm(?10~(-6))or 1.65 ppm,but~1H-MRS within the same location was always same or similar with different VOI sizes(1 cm?1 cm?1 cm or 2 cm?2 cm?2 cm)or different numbers of acquisition(128 or 256 times).~1H-MRS acquisition time was not related with the size of VOI but with the numbers of acquisition.128 and 256 times of acquisition cost 199 s and 391 s,respectively.Conclusion With the technique of small size of VOI(1 cm?1 cm?1 cm)and decreased numbers of acquisition(128 times),it is propable to get well-repeated and excellent ~1H-MRS within less time.It is also more practical for clinics to achieve ~1H-MRS of the femoral marrow with the proper technique.
7.The effect of web-based histologic laboratories for general practical students

ObjectiveTo assess the satisfaction of general practical students' perception with the two modes of histologic laboratory instruction,and obtain the advantages and disadvantages of each mode.MethodsWeb-based database was established and in the questionnaires were delivered to the general practical students ( n=193 ).Results75% of students agreed that the case study component of the webbased laboratory instruction was arranged in order.82% of students identified with the detailed word explanation attached to every image presented.ConclusionStudents support the progression of web-based histologic laboratory instruction,but it is difficult to replace traditional microscope with web-based database in histologic laboratory instruction.
8.Necessity of setting up the course on clinical healthcare-associated infection according to case-based teaching method in medical higher education

Objective To explore the necessity of setting up the course on clinical healthcare-associated infection (HAI) and effectiveness of the case-based teaching method in medical higher education.Methods 198 students who underwent internship in a hospital between July 2015 and July 2016 were selected,according to whether they selected clinical HAI coures,they were divided into course selective group(n =107,adopted case-based teaching method) and unselective group(n =91),questionnaire survey on the knowledge and attitude of HAI was carried out.Results The average score on HAI in selective group was higher than that of unselective group([85.3 ± 1.6] vs [58.3 ± 1.8],t =111.72,P<0.01).Before and after conducted case-based teaching method,21.49% and 47.66% of students actively studied respectively,difference was significant (x2 =16.195,P<0.01).Conclusion It is necessary to carry out a systematic course on HAI,case-based teaching method can improve the learning enthusiasm of students.
9.Construction of the Bac-to-Bac System of Bombyx mori Nucleopolyhedroviru

To construct the Bac-to-Bac expression system of Bombyx mori nucleopolyhedrovirus (BmNPV), a transfer vector was constructed which contained an Escherichia coli (E. coli) mini-F replicon and a lacZ: attTN7: lacZ cassette within the upstream and downstream regions of the BmNPV polyhedrin gene. B. mori larvae were cotransfected with wild-type BmNPV genomic DNA and the transfer vector through subcutaneous injection to generate recombinant viruses by homologous recombination in vivo. The genomic DNA of budded viruses extracted from the hemolymph of the transfected larvae was used to transform E. coli DH10B. Recombinant bacmids were screened by kanamycin resistance, PCR and restriction enzyme (REN) digestion. One of the bacmid colonies, BmBacJS13, which had similar REN profiles to that of wild-type BmNPV, was selected for further research. To investigate the infectivity of BmBacJS13, the polyhedrin gene was introduced into the bacmid and the resultant recombinant (BmBacJS13-ph) was transfected to BmN cells. The budded viruses were collected from the supernatant of the transfected cells and used for infecting BmN cells. Growth curve analysis indicated that BmBacJS13-ph had a similar growth curve to that of wild-type BmNPV. Bio-assays indicated that BmBacJS13-ph was also infectious to B. mori larvae.
10.Changes of postmortem human blood biochemical indexes.

OBJECTIVE:
To investigate and analyze the changes of postmortem human biochemical indexes.
METHODS:
Subclavian venous blood samples were collected from 81 cases of traffic fatalities. Thirteen blood biochemical indexes including liver function (ALT, AST, TBIL and DBIL), renal function (UA and Cr), cardiac function (CK, CK-MB and LDH), electrolytes (K+, Na+ and Cl-), and glucose (GLU) were tested by Roche cobas c311 automatic biochemical analyzer. The descriptive analysis was made by SPSS 17.0 statistical software.
RESULTS:
The values of ALT, AST, CK, CK-MB, LDH and K+ were higher than normal reference values with more fluctuations. The values of TBIL, DBIL, UA, Cr, Na+, Cl- and GLU were relatively stable with less fluctuations.
CONCLUSION
The postmortem human blood biochemical indexes of liver function, renal function, cardiac function, electrolytes and glucose could be affected by the factors, especially hemolysis and autolysis. The biochemical indexes, particularly enzymes, increased significantly with higher standard deviation.
