Objective:To study the dynamic frictional resistance in the buccal segment between preadjusted brackets coupled with different archwires,at different times under artificial saliva condition. Methods: The dynamic friction of different combinations of 4 kinds of preadjusted brackets and 4 archwires were tested simulating archwires sliding in the buccal segments at 4 different times(0,15,30,45 d)under artificial saliva condition by using Instron universal testing machine. Results: The dynamic frictional resistance of combinations of 3M MBT bracket and all archwires were the smallest. The combinations of 0.46 mm inch stainless steel round wire and all brackets produced significantly lower dynamic frictional resistance than that of other archwires. The 0.48 mm?0.64 mm inch stainless steel rectangular wire produced the highest dynamic frictional resistance combined with all preadjusted brackets. The dynamic frictional resistance of combinations of archwires and brackets were highest at the 15th day and smallest at the 30th day under artificial saliva condition. Conclusion: 0.46 mm stainless steel round wire produces the smallest dynamic frictional resistance combined with all preadjusted brackets.3M MBT brackets are more favorable to sliding mechanics compared with other brackets. As for time, the lowest dynamic frictional resistance exhibits at the 30th day under artificial saliva condition.
2. Inhibition of lung cancer by transfer of p53 gene using Tie2-mediated gene delivery system
Tumor 2007;27(4):256-259
Objective: To explore the therapeutic efficacy of transduction of p53 gene with Tie2-mediated gene delivery system in the treatment of lung cancer and provide experimental basis for future clinical application. Methods: GA3, a small peptide against Tie 2, was conjugated with PEI by bifunctional crosslinking agent SMCC to construct Tie2-targeted gene vector. Then pCMV-luciferase cDNA was combined with GA3-PEI to form an electrostatic complex. GA3-PEI/luciferase cDNA complex was transduced into Tie2-positive SPC-A1 lung cancer cells and Tie2-negative SMMC7721 hepatic cancer cells in vitro. The activity of luciferase was measured 24 h later. The GA3-PEI/β-gal complex was injected into peri-tumorous tissues of the transplanted SPC-A1 tumors in nude mice in vivo. Meanwhile, PEI/β-gal gene was injected as control. The nude mice were decapitated 24 h later. The heart, liver, spleen, lung, kidney and tumor tissues were excised and stained with X-gal. Human lung cancer SPC-A1 cells were inoculated subcutaneously into the 4-week-old female athymic mice (BALB/c). The mice were randomly divided into 5 groups (NS, GA3, p53, PEI/p53, and GA3-PEI/p53) with 6 mice in each group. GA3-PEI/ p53 1 μg was subcutaneously injected into peri-tumorous tissues once every two days. The tumor volume was calculated according to formula (V = πab2/6). The tumor-inhibiting ratio was calculated regarding NS group as control. Results: The activity of luciferase of GA3-PEI/luciferase was significantly higher in SPC-A1 cells than that in SMMC7721 cells (P<0.05). A lot of blue-stained cells were observed in transplanted tumor tissues but not in heart, liver, spleen, and kidney tissues except lung bronchial mucosa. Compared with NS group, GA3-PEI/p53 significantly inhibited the growth of tumor by 61.29%. Conclusion: Transfer of p.53 gene by GA3 gene delivery system remarkably inhibits tumor growth.

With inlet temperature, specific gravity, feeding speed as independent variables, the comprehensive evaluating indexes of content of schisandrin and arctiin as dependent variable, the experimental data were fitted to a second order polynomial equation. Based on establishing the mathematical relationship between the comprehensive evaluating indexes and respective variables, Box-Benhnken central composite test and response surface analysis method was employed to optimize the spray drying technology of Biqiu granules ethanol extract. The optimal drying parameter was as follows: the inlet temperature was 175 degrees C, the specific gravity was 1.10, feeding speed was 32 r x min(-1). Under these conditions, the comprehensive evaluating indexes of spraying dry processes was 92.68, which was close to the model prediction. The spraying dry technology of Biqiu granules ethanol extract optimized by response surface methodology was accurate and feasible, which provided theoretical experiment basis for the industrialization production.

Journal of Southern Medical University 2008;28(6):953-955
OBJECTIVETo investigate the expression of FSCN1 in human epithelial tumors and their clinical significance.
METHODSFSCN1 expression was examined immunohistochemically in specimens of human epithelial tumors, including 26 cases of lung cancer, 33 cervical cancer, 22 ovarian cancer, 38 esophageal cancer, 24 pancreatic cancer, 23 gastric cancer, 29 laryngocarcinoma, 17 primary hepatocellular carcinoma, 34 colorectal cancer, 33 breast cancer, 24 nasopharyngeal carcinoma and their corresponding normal tissues.
RESULTSThe positivity rates of FSCN1 expression in epithelial tissues and epithelial tumors were 6.3% (5/80) and 58.7% (178/303), respectively. FSCN1 showed higher expressions in cervical cancer, ovarian cancer, esophageal cancer, pancreatic cancer, gastric cancer, laryngocarcinoma, colorectal cancer, breast cancer and nasopharyngeal carcinoma, but lower or no expression in the corresponding normal tissues (P<0.05). In gastric cancer and nasopharyngeal carcinoma, the edges of the tumors were more strongly stained for FSCN1 than the interior of the tumor.
CONCLUSIONFSCN1 expression is significantly upregulated in human epithelial tumors in close correlation with tumor occurrence and progression.

First with the qualified rate of granules as the evaluation index, significant influencing factors were firstly screened by Plackett-Burman design. Then, with the qualified rate and moisture content as the evaluation indexes, significant factors that affect one-step pelletization technology were further optimized by Box-Behnken design; experimental data were imitated by multiple regression and second-order polynomial equation; and response surface method was used for predictive analysis of optimal technology. The best conditions were as follows: inlet air temperature of 85 degrees C, sample introduction speed of 33 r x min(-1), density of concrete 1. 10. One-step pelletization technology of Biqiu granules by Plackett-Burman design and Box-Behnken response surface methodology was stable and feasible with good predictability, which provided reliable basis for the industrialized production of Biqiu granules.

Lycii Cortex, a popular herb medicine in traditional Chinese medicine, is used to treat different inflammation-related diseases. The aim of our work is to find the key constituents inhibiting NF-kappaB, a key regulator of inflammation. In the investigations of cell-based in vitro assays of extracts, we found that both ethyl acetate extract and methanol extract of Lycii Cortex inhibited the TNF-alpha-induced activation of NF-kappaB. Through bioassay-guided fractionation, we identified 4 phenolic amides including trans-N-(p-coumaroyl) tyramine (1), trans-N-feruloyltyramine (2), trans-N-caffeoyltyramine (3), and dihydro-N-caffeoyltyramine (4). Four phenolic amides showed differently inhibitory activities on TNF-alpha-induced NF-kappaB activation. Trans-N-caffeoyltyramine (3) was identified as the key component with an IC50 of 18.41 micromol x L(-1). It was suggested that the hydroxyl group at C-3 in trans-N-caffeoyltyramine might be a key binding site and its C-7,8-double bond might play an important role on NF-kappaB inhibitory activities as the link of the conjugation of pi electrons leading to a partial planar conformation. It might be inferred that the biological activity of compound 3 is attributed to the structure of Michael reaction acceptor containing alpha, beta-unsaturated ketones and benzene along with hydroxyl group in o-diphenol.
