1.The expression and clinical significance of miR-150-5P in patients with hepatocellular carcinoma

Objective To explore the expression and clinical significance of miR-150-5P in patients with hepatocellular carcinoma (HCC).Methods Expressions of miR-150-5P were analyzed by quantitative real-time polymerase chain reaction (qRT-PCR) and in situ hybridization (ISH) in cancerous and adjacent tissues of 91 HCC patients.Statistical analysis was conducted by SPSS 22.0 software.Results qRT-PCR revealed that the mRNA expression level of miR-150-5P was significantly lower in HCC tissues than adjacent tissues.ISH revealed that the positive rate of miR-150-5P was downregulated in HCC tissues compared to adjacent tissues(x2 =4.958,P =0.046).Chi-square analysis revealed that high miR-150-5P expression was inversely associated with lymph node status of HCC patients,which was statistically significant (x2 =8.617,P =0.006).Kaplan-Meier curves revealed that high miR-150-5P expression was positively correlated with 5-year overall survival rates of HCC patients after surgery,which was also statistically significant(x2 =6.184,P=0.013).Conclusions miR-150-5P expression is downregnlated in HCC,and is negatively correlated with lymph node metastasis and positively correlated with cumulative survival of HCC patients.

4.Study on Microbial Oil Production with Chlorella pyrenoidosa

Chlorella pyrenoidosa No.2 was screened from five species of microalga Chlorella sp. for its higher lipid yield. Effects of medium components and culture conditions on cell growth as well as lipid ac-cumulation of C. pyrenoidosa No.2 were investigated and the results showed that the optimum medium rec-ipe was 20.0 g/L glucose,0.08 g/L glycine,1.0 g/L K2HPO4?3H2O,0.4 g/L MgSO4?7H2O and 0.004 g/L FeSO4?7H2O. The optimum culture temperature,initial pH,shaking rate and light intensity were 28℃,6.0,130 r/min and 650 Lux,respectively. Biomass and lipid content increased from 3.73 g/L and 40.15% to 6.56 g/L and 59.90% when Chlorella pyrenoidosa No.2 was cultivated under the above optimal conditions for 7 days,with lipid yield raised by 162%. Chlorella pyrenoidosa No.2 could produce lipid with xylose as carbon source,and so is potential for lipid production from renewable materials such as lignocellulose. GC analysis demonstrated that the fatty acid composition of the lipid was similar to that of vegetable oil and its unsaturated fatty acid content reached around 71%,thus it is a promising material for biodiesel production.
5.Effects of tetramethyl pyrazine (TMP) on noise induced hearing loss in hypobaric environment

Objective To evaluate the effect of tetramethyl pyrazine(TMP)on noise induced hearing loss (NIHL) in hypobaric environment. Methods Normal guinea pigs were randomly divided into two groups, in which the animals were exposed to noise for 1 day or 7 days, respectively. Each group was randomly divided into 3 subgroups: control, TMP-treatment and TMP-pretreatment groups. All animals were exposed to 110dB white noise in hypobaric chamber to repreduce a low pressure environment of 5500m altitude. The mechanism underlying TMP protection was studied by detecting the auditory brainstem response (ABR). Results ABR threshold shift in different degrees occurred in every group, especially in animals exposed to 4kHz. The threshold shifts increased in extent along with prolongation of exposure time. The longer they were exposed, the more ABR threshold shifts. ABR threshold shifts occurred more severely in control group compared with the two TMP-utilized groups. Conclusion TMP has certain protective effect on NIHL in a hypobaric environment.
6.Mutation analysis of STK11 gene in patients with Peutz-Jeghers syndrome

Objective To study the mutation of STK11 gene in a Chinese family and a sporadic patient with Peutz-Jeghers syndrome (PJS),and to provide a basis for genetic diagnosis and counseling.Methods One sporadic patient and two patients from a family with PJS were collected,all of whom had typical mucosal pigmentation and gastrointestinal polyposis.Blood samples were obtained from the two patients and six unaffected relatives in this family,the sporadic patient,and 100 healthy controls.DNA was extracted,and PCR was performed to amplify nine exons and their adjacent introns in the STK11 gene followed by direct sequencing.The sequencing results were aligned to the published sequence of STK11 gene from Genbank.Results No mutation was found in the STK11 gene of any of the patients,unaffected relatives,or healthy controls.Conclusions Genetic heterogeneity exists in Peutz-Jeghers syndrome,hinting that there may be other causative genes or sites for this entity.

9.Determination of Paraquat Adsorbability to Four field Soils of Guangxi Province by High Performance Liquid Chromatography Method

Objective To study paraquat adsorbability of different field soils in Guangxi province of China. Methods HPLC method was adopted to measure the peak area of paraquat in three different media of four types of soils. Chromatographic column was Kromasil C18 column (4. 6 mm×200 mm, 5 μm); mobile phase was acetonitrile-water (including 0. 03 mol·L-1 sodium heptanesulfonate and 0. 24 mol·L-1 phosphoric acid) at a ratio of 397 (pH adjusted to 2. 0 by triethylamine). Detection wave length was 258 nm; column temperature was 25 ℃; the injection volume was 20 μL; flow rate was 0. 8 mL·min-1 . The peak areas of paraquat before and after being adsorbed were compared to calculate the adsorption rate of paraquat in different soils. Results All tested soil samples possessed the adsorption rate of paraquat over 99. 0%. Conclusion Four common field soils in Guangxi province can be used as temporary effective absorbents for the first-aid of paraquat poisoning.
10.Application value analysis of the method cut-open performing umbilicus abdominal access during laparoscopy

Objective To introduce a new method performing umbilicus abdominal access during laparoscopy. Methods In a prospec-tive,362 patients received laparoscopic appendectomy in October 2012 to October 2014 were oberserved in this study. The clinical data,the time of the progress of making umbilicus abdominal access,complications and the state of the wound were recorded. Results The performing time of the trocar in umbilicus was (42. 3 ± 27. 1)s. Conclusion The method Cut-open,is simple,fast and save,which is worth to promoting in laparoscopic surgery.
