2.Construction of lentiviral vector of DAB2IP gene and its expression in prostate carcinoma PC3 cells

Objective To construct recombinant lentiviral vector pSin-EF2-Puro-DAB2IP transfect prostate carcinoma PC3 cells,and to observe its transfection rate and expression level in PC3 cells.Methods The cDNA sequences specifically targeting the DAB2IP gene were designed and cloned into lentiviral vector PC3-pSin using DNA recombinant technique.Using PC3-pSin-EF2 as control,the 293T cells were transfected by Lipofectamine reagent for lentiviral particles packaged, and viral titer was determined. The DAB2IP mRNA and protein expression levels were examined by RT-PCR and Western blotting methods. Results The PCR and sequencing analysis results confirmed that the DAB2IP gene sequence was consistent with the sequence in GeneBank. The number of DAB2IP gene copy in PC3-pSin-EF2-DAB2IP cells and its control PC3-pSin-EF2 cells were 0.001±0.000 and 0.158 ± 0.013,respectively.Compared with control cells,the overexpression of mRNA in PC3-pSin-EF2-DAB2IP cells upregulated (179.370 ± 15.891)times,the difference was statistically significant (P<0.001). Compared with internal reference and control cells, the expression levels of DAB2IP protein in PC3-pSin-EF2-DAB2IP cells upregulated (2.431 ± 0.892)times and (2.415 ± 0.961)times respectively,the differences were statistically significant (P<0.001).Conclusion The lentiviral vector of the DAB2IP gene pSin-EF2-Puro-DAB2IP is successfully constructed,and its targeted gene is stably expressed in the targeted cells,which provides a basis for the further functional study of DAB2IP.
5.Periodization of international spread of acupuncture-moxibustion and their characteristics at each period.

The history of international spread of Chinese acupuncture and moxibustion is divided into three sta ges in this paper, and the spreading characteristics are analyzed. The first stage is approximately from the 6th century to the end of the 15th century, during which acupuncture and moxibustion were spread to neighboring countries by personnel exchanges; the spread towards Korean peninsula, Japan and Vietnam was considered the most successful communication. The second stage lasts from the beginning of 16th century to 1970. At the early time of this stage, the employees of the Dutch East Indian Company introduced acupuncture and moxibustion to European countries through Indonesia and Japan, leading to a short and small fashion; also the United States and Australia were involved. At the late time of this stage, by medical aid teams dispatched by China government, acupuncture and moxibustion were introduced to African countries. The third stage starts from 1971. With the establishment of Sino-US diplomatic relations as an opportunity, acupuncture and moxibustion were being spread rapidly to the world through radio, TV and internet. So far it has been introduced to more than 140 countries and areas. Performing serious studies on the spreading characteristics of three stages will promote the international communication of acupuncture and moxibustion, by which the world will have a better understanding onthe broad and profound traditional cultures of China.

6.Efficacy of Iron Dextran Combined with EPO in the Treatment of Renal Anemia

Objective:To compare the intravenous (Ⅳ) administration with an oral iron agent administration in their efficacy and side effects in hemodialysis patients with renal anemia. Method:50 renal anemic patients were randomly grouped as the treatment group (TG) and controlled group (25 cases for each). 100 mg iron dextran injection was intravenously dropped twice weekly for the TG patients in the dialysis course, totally 10 times. 150 mg iron polysaccharide complex capsules was orally used qd in the controlled group. All the patients were treated in combination with 8000u of erythropoietin (EPO) weekly and observed for 8 weeks. The changes in Hb, serum ferritin level and agent side effect were monitored. 7.Relationships between the polymorphism of methylenetetrahydrofolate reductase gene C677T and hypertension,cardiac structure and function

Objective Elevated plasma homocysteine(Hcy) level has been associated with the increased risk for cardiovascular disease.The level of plasma Hcy is related to methylene tetrahydrofolate reductase(MTHFR) gene C677T polymorphism.The aim of this study is to evaluate the relationships between MTHFR gene C677T polymorphism and hypertension,cardiac structure and function.Methods Genotype of MTHFR gene C677T polymorphism was detected by polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP) analysis and decided by electrophoresis and sequencing in 695 patients with essential hypertension and 509 age-matched normal controls.Echocardiography was performed in patients with essential hypertension.Results In hypertension group,the frequencies of TT genotype and T allele were significantly higher than those in control group(26.5% vs 20.6%,48.7% vs 42.4%;P=0.015,0.002).The essential hypertension odds ratio(OR) associated with MTHFR 677T allele was 1.300(95%CI:1.166 to 1.450).Logistic regression analysis indicated that MTHFR gene C677T polymorphism was an independent variable of hypertension(P=0.027).End diastolic dimension(EDD),interventricular septal thickness(IVST),left ventricular posterior wall thickness(LVPWT),left ventricular mass(LVM) and left ventricular mass index(LVMI) were higher in hypertensive patients with T allele than that in those with CC genotype,but all had no statistical significance(P=0.428,0.118,0.064,0.077,0.054).Conclusion The 677T allele in MTHFR gene may be a risk factor for hypertension.Hypertensive patients with MTHFR 677T had a trend of hypertrophy,but the relationship has no statistical significance.The relationship between MTHFR gene C677T polymorphism and left ventricular hypertrophy need to be further studied.
9.Progress in the study of choroidal thickness on diabetic retinopathy by OCT

? The choroidal vasculature provides oxygen and nutrients to the outer retina, and is responsible for maintaining the highly metabolically active photoreceptor cells. The normal structure and function of its vascular system is very important for the retina. So it is more meaningful to observe the choroid morphology for tracking pathological changes in diabetic retinopathy. With the application of the high resolution optical coherence tomography ( OCT ) technology, spectral -domain optical coherence tomography ( SD - OCT ) enhanced depth imaging technology can be used to measure the choroidal thickness quantitatively, which provides a new idea for the diagnosis and treatment of diabetic retinopathy. Therefore, the research and progress of choroidal thickness on diabetic retinopathy by OCT are summarized as follows.

The metabolism study of radiolabeled drugs plays an important role in the development of new drugs. It provides information on drug absorption, metabolism, tissue distribution and excretion, and plays an irreplaceable role in the metabolite safety evaluation and mass balance of new drugs. The new guidance draft on clinical trials of radiolabeled drugs recently released by the US FDA puts forward higher standards and has been widely concerned by the industry. In recent years, in the research and development of new drugs in China, 14C labeled drugs have been used to carry out clinical metabolism studies, which has overcome key technical bottlenecks and accumulated experience. This paper summarizes the above research progress, analyzes the existing problems, and preliminarily looks forward to the future technological development and application.
