1.Expression of adhesion-related cytokines in the uterine fluid after transcervical resection of adhesion

Objective To study the concentration of adhesion-related cytokines,transforming growth factor (TGF) β1,platelet-derived growth factor (PDGF) BB,b-fibroblast growth factor (bFGF) in intrauterine fluid and their roles in the intrauterine-adhesion reformation.Methods From August 2009 to February 2010,18 patients with moderate or severe intrauterine adhesions underwent transcervical resection of intrauterine adhesion as study group,in the mean time,20 patients with incomplete septate uterus underwent transcervical resection of septum as control group.The charge of each patient's intrauterine fluid was collected at the time of 3,6,9,12,24,48 and 72 hours,respectively,then the concentration of the three cytokines was measured by enzyme-linked immunosorbent assay(ELISA).Results The concentration of the expression of TGF-β1 in the intrauterine fluid at 3,6,9,12 hours after operation in study group was (3.6±0.9),(10.4±1.1),(7.6±1.2),(7.2±1.3) ng/ml,respectively; PDGF-BB was(2.6±0.6),(3.5 ±0.5),(5.4±1.0),(5.7 ±0.8) ng/ml,respectively; bFGF was(16.9±1.3),(95.8±17.8),(330.9 ± 70.5),(1303.3 ± 117.4) ng/ml,respectively.The expression of TGF-β1 in control group was (3.0 ± 0.6),(7.5 ± 0.6),(5.4 ± 0.6),(4.6 ± 0.8) ng/ml,respectively ; PDGF-BB was (2.5 ± 0.4),(2.6±0.5),(4.7±0.6),(4.4±0.4) ng/ml,respectively; bFGF was (19.1 ±2.4),(82.9±21.8),(249.0 ±54.2),(775.6 ± 178.8) ng/ml,respectively.The concentration of TGF-β1,PDGF-BB and bFGF reached the peak during the time of 3-24 hours,6-48 hours,6-72 hours after surgery,respectively.The concentration of TGF-β1,PDGF-BB and bFGF in study group were significantly higher than that in control group (P <0.05).Conclusions Concentration of TGF-β1,PDGF-BB and bFGF was positively associated with area of transcervical resection of intrauterine adhesion,those cytokines might be involved in reformatin of intrauterine-adhesion.
2.Study on effect of phacoemulsification combined with goniosynechialysis on corneal endothelial cells for the treatment of angle closure glaucoma

AIM:To observe the effect of phacoemulsification and intraocular lens ( IOL ) implantation combined with goniosynechialysis on corneal endothelial cells for the treatment of primary angle closure glaucoma ( PACG ) combined cataract, and to analyze the relative factors.METHODS: Ninety-five eyes of 95 patients with PACG combined cataract were documented in this study. Twenty-two patients were male, and 73 were female. The age ranged from 46 to 85y old with a mean of(66±7) y. All patients were examined for endothelial cell count ( ECC ) , intraocular pressure ( IOP ) and best corrected visual acuity ( BCVA ) 1wk, 1, 2, 3 and 6mo after operation. Meanwhile, the range of anterior chamber closure and anterior chamber depth ( ACD ) were recorded before operation and postoperative 6mo.
RESULTS: The mean IOP was 36. 1±4. 3mmHg ( 28-42mmHg) preoperatively and 15. 8±3. 5mmHg ( 8-28 mmHg)(1mmHg=0. 133kPa) 6mo after operation. There was a decreasing trend in IOP after operation( t=17. 173, P<0. 01). There was an increased trend in BCVA after operation(Z=-8. 351, P<0. 01). The mean ACD was 1. 95±0. 34mm preoperatively and 3. 11±0. 33mm 6mo after operation. There was an increased trend in ACD after operation(t=11. 483, P<0. 01). There were 74 eyes in the range from 90o to 180o of anterior chamber and 21 eyes less than 90o before operation. There were 12 eyes in the range from 90o to 180o of anterior chamber and 83 eyes less than 90o postoperatively. The most of anterior chamber were open(Z=-9. 013, P<0. 01). ECC was 2304±135 cells/mm2 before operation and 2 243±152 cells/mm2 , 2 135±177 cells/mm2 , 2 028±172 cells/mm2 , 2 017±181 cells/mm2 , 2 006±143 cells/mm2 1wk, 1, 2, 3 and 6mo after operation respectively. ECC loss rates were 2. 6%, 7. 3%, 12. 0%, 12. 5% and 13. 0%. ECC had a decreased trend 2mo after operation(F=5. 568, P<0. 01) and then kept stable(P3mo=0. 067, P6mo=0. 073).
CONCLUSION: Phacoemulsification and IOL -implantation combined with goniosynechialysis is an effective method to treat PACG combined cataract. It can increase BCVA and decrease IOP. ECC decreases after operation, but it is in the normal range. It is a safe and effective operation mode.
3.The expression and clinical significance of miR-150-5P in patients with hepatocellular carcinoma

Objective To explore the expression and clinical significance of miR-150-5P in patients with hepatocellular carcinoma (HCC).Methods Expressions of miR-150-5P were analyzed by quantitative real-time polymerase chain reaction (qRT-PCR) and in situ hybridization (ISH) in cancerous and adjacent tissues of 91 HCC patients.Statistical analysis was conducted by SPSS 22.0 software.Results qRT-PCR revealed that the mRNA expression level of miR-150-5P was significantly lower in HCC tissues than adjacent tissues.ISH revealed that the positive rate of miR-150-5P was downregulated in HCC tissues compared to adjacent tissues(x2 =4.958,P =0.046).Chi-square analysis revealed that high miR-150-5P expression was inversely associated with lymph node status of HCC patients,which was statistically significant (x2 =8.617,P =0.006).Kaplan-Meier curves revealed that high miR-150-5P expression was positively correlated with 5-year overall survival rates of HCC patients after surgery,which was also statistically significant(x2 =6.184,P=0.013).Conclusions miR-150-5P expression is downregnlated in HCC,and is negatively correlated with lymph node metastasis and positively correlated with cumulative survival of HCC patients.
4.Overview of Chinese medicine in prevention of non-small cell lung cancer recurrence and metastasis after surgery

Recurrence and metastasis are main causes ofsurgical treatment failure of non-small cell lung cancer.In prevention of non-small cell lung cancer recurrence and metastasis,Chinese medicine has achieved a certain effect. It has been proved by clinical use of Chinese patent drugs, compound modification based on syndrome differentiation, integrated traditional Chinese and western medicine treatment, and molecular mechanism of anti-recurrence and metastasis, though there are still some problems. This article reviews the recent clinical and experimental research status.
5.Investigation and analysis of the cognitive level of occupational protection of staff in sterilization and supply center

ObjectiveTo investigate and understand the cognitive level of occupational protection of staff in sterilization and supply center. Methods180 staff in sterilization and supply center were taken for a retrospective questionnaire investigation.The survey included:general information of the object,cognition about occupational protection,wearing of protective equipment,influence of occupational hazards,sharp instrument injury and the handling of injury cases. ResultsThe cognitive level of occupational protection in the sterilization and supply center was low.Some of the staff did not fully implement the wearing of protective equipment rules in the decontamination areas.In the occupational hazards,sharp instrument injuries accounted the highest incidence.The situation of taking blood tests after injury,injection of high titer globulin,report to the higher-ups and follow-up investigation were rare,and even some staff gave no handling of the wound. ConclusionsThe cognitive level of occupational protection in the sterilization and supply center is low.It not only needs professional training to improve cognitive level of occupational protection but also effective protective measures.The management department should establish a complete protection regulations of occupational safety and strict supervision in order to prevent the occurrence of occupational exposure,ensure staff safety,and at the same time,health records should be established in order to understand health status of staff.
6.Application of case - based learning combined with problem -based learning teaching mode in the clinical teaching of ophthalmology

?AIM:To improve the efficiency and quality of teaching, and to cultivate students’ ability to analyze and solve clinical problems, the case - based learning ( CBL ) combined with problem-based learning ( PBL ) teaching mode is introduced into the clinical teaching of ophthalmology.?METHODS:Two classes ( total 131 students ) of 2013 grade major in clinical medicine, Kunming Medical University, were randomly selected as experimental group using CBL combined with PBL teaching mode, and control group using traditional teaching mode. The part of acute angle-closure glaucoma was taught. The scores of the experimental group were compared with that of control group at the end of term.?RESULTS:The mean scores of CBL combined with PBL teaching group were significantly higher than that of traditional teaching group.?CONCLUSION: The application of CBL combined with PBL in the clinical teaching of ophthalmology has obvious advantages in improving the learning interests and promoting students’ self-learning ability. It is helpful in improving the students’ ability of independent-thinking, analyzing and solving problem.

8.Pain of high-throughput screening--pan assay interference compounds.

High-throughput screening is a regular approach available for identitying new lead compounds for the growing validated drug targets in drug screening. However, it has also introduced a large number of peculiar molecules which interfere drug screening. Pan assay interference compounds (PAINS) interfere with the progress of drug screening in various ways, such as interfering with a biochemical assay, modifying the protein, aggregate-based inhibitors and so on. So it is of vital significance to remove them. This paper has consulted the concept, category of PAINS and reviewed the way of PAINS interfering and the countermeasures to cope with them to direct the approach of high through screening and improve the hits percent.

10. Signal transduction of atractylodin biosynthesis in Atractylodes lancea cell induced by endophytic fungal elicitor mediated with nitric oxide followed by salicylic acid

Objective: To investigate the signal molecules and signal transduction involved in endophytic fungal elicitor-induced atractylodin biosynthesis and the effect of an endophytic fungal elicitor on the key enzyme activity in Atractylodes lancea. Methods: Content changes of nitric oxide (NO), salicylic acid (SA), and atractylodin were detected under the endophytic fungal elicitor treatment by plant cell suspension culture technology. Results: The endophytic fungal elicitor remarkably promoted NO burst and the biosynthesis of SA and atractyodin by activating nitric oxide synthase (NOS), phenylalanine ammonia lyase (PAL), and acetyl coenzyme A carboxylase (ACC), respectively. NOS inhibitor PBITU could inhibit the NO and SA accumulation and the atractyodin biosynthesis induced by the elicitor. And atractyodin biosynthesis could also be triggered by exogenous NO or SA. The results indicated that NO and SA were the necessary signal molecules and NO burst was mediated by NOS induced by endophytic fungal elicitor. NO quencher cPITO could effectively remove NO burst in A. lancea cell induced by endophytic fungal elicitor and notably inhibit the biosynthesis promotion of SA and atractyodin in A. lancea cell induced by endophytic fungal elicitor. Exogenous SNP could reverse the cPITO inhibition on the activity of PAL and ACC and the synthesis of SA and atractylodin. This suggested that NO was an upstream signal molecule mediated endophytic fungal elicitor to accelerate the biosynthesis of SA and atractyodin. Conclusion: Endophytic fungal elicitor mediated through NO followed by SA could promote atractyodin biosynthesis by activating ACC in A. lancea.
