1.Inhibition of dedifferentiation of human articular chondrocytes by transfection with recombinant TGF-?_2 in vitro

Objective In cartilage tissue engineering,it was a difficult problem to keep chongdro-cytes phenotype for long time.The aim was made to investigate the dedifferentiation of human articular chon-drocytes,and to verify the inhibition to dedifferentiation by transfection with recombinant pcDNA3.1(+)/TGF-? 2 in vitro.Methods Free chondrocytes were isolated from healthy articular cartilage harvested from6adult patients underwent operation because of other condition.The recombinant pcDNA3.1(+)/TGF-? 2 con-structed in advance was transferred by liposome mediated transfection to monolayer cultured articular chon-drocytes.The first,sixth and ninth generation chondrocytes being transfected and non-transfected were as-sayed by histological examination of HE and Safranine-O staining to observe their morphological changes.RT-PCR,ELISA,immunohistochemistry analysis and prime hybridization were performed to these cells to i-dentify the expression and biosynthesis of TGF-? 2 and cartilage-associated genes and proteins.Results The non-transfected articular chondrocytes in monolayer culture demonstrated the tendency of dedifferentia-tion,and lost the characteristics of their phenotype progressively in subculture.The expression of TGF-? 2 ,collagen typeⅡand proteoglycan in non-transfected cells were decreased,but expression of collagen typeⅠincreased gradually with time going on.On the contrary,as for the transfected chondrocytes,the trans-ferred gene was expressed in cells of all examined generations,and the expression levels were higher than that of non-transfected cell of same generations.The transfected chondrocytes maintained their phenotype and characteristics of articular cartilage.They expressed and produced collagen typeⅡand proteoglycan in a higher level,and collagen typeⅠin a lower level than that of non-transfected cells.Conclusion Human articular chondrocytes in monolayer culture have the tendency of dedifferentiation.The results suggested that chondrocytes tranfected by pcDNA3.1(+)/TGF-? 2 could produce endogenous TGF-? 2 .Transfection of pcD-NA3.1(+)/TGF-? 2 is able to provide transient and long lasting expression of TGF-? 2 in chondrocytes,and may be a feasible method to inhibit dedifferentiation of articular chondrocytes in vitro.
2. Experimental study on the therapeutic effects of nimustine combined with cisplatin on C6 brain glioma

Objective: To observe the synergistic effects of nimustine(ACNU) and cisplatin(CDDP) in the treatment of glioma, and investigate the theoretical basis for the combined chemotherapy in clinic. Methods: The cell cycle and apoptosis of C6 glioma cells were examined by flow cytometry after treatment with ACNU plus CDDP for 24 h in vitro. C6 glioma cells were transplanted into the brain of Wistar rats to establish the C6 brain glioma model. The Wistar rats were treated with ACNU plus CDDP for 3 days, and then some rats were sacrificed. Morphological changes of tumor tissues were examined by HE staining. The protein expression of proliferating cell nuclear antigen (PCNA) and apoptosis-associated gene bcl-2 were tested by immunohistochemical method. The survival time of the Wistar rats were observed. Results: The FCM test found that there was no significant difference in cell cycle distribution between combined therapy group and single ACNU therapy group. The apoptotic rate increased after combined therapy [(2.10 ± 0.14)%] compared with single CDDP therapy group [(1.72±0.21)%] and ACNU single therapy group [(0.57±0.01)%]. The difference was significant (P < 0.01). Histological examination revealed the decreased number of the invaded focuses in the margin of the tumor body. Immunohistochemistry analysis indicated that the protein expression levels of PCNA and bcl-2 decreased compared with single therapy group (P < 0.01). The rats in the combined therapy group had longer survival time compared with those received single therapy. The difference was significant (P < 0.01). Conclusion: The combination of ACNU plus CDDP has apparent synergistic effects in the treatment of malignant brain glioma compared with single drug therapy.
4.Survey of infection situation and knowledge of high-risk HPV among women working in entertainment places in Gaoqiao area

Objective To investigate infection situation and knowledge of high‐risk human papilloma virus(HPV)among women working in entertainment places in Gaoqiao area ,and provide theory evidences for making health education measures related to high‐risk HPV .Methods According to the systematic random sampling method ,entertainment female in Gaoqiao area were extracted , and normal women who participated in community gynecology census were extracted as control group .Then questionnaire investiga‐tion and laboratory testing were carried out .Results The infection rate of high‐risk HPV in entertainment female was 35 .18% , while in normal woman was 15 .84% ,there was significant differences of infection rate between the two groups (P<0 .05) .There were were only 16 women(14 .81% )and 17 women(16 .83% )had heard of information about HPV in two groups respectively ,and there was no statistically significant difference(P>0 .05) .The cognitive situation of whether a person with family history of cervi‐cal cancer should be regularly attend screening among the two groups of women was different ,and had significant difference(P<0 .05) .Conclusion Women who worked in entertainment places had higher infection rate of high‐risk HPV ,both of the two groups had poor knowledge of HPV .So the propaganda and screening of HPV should be strengthen in order to raise consciousness in pre‐venting cervical cancer .
5.CT features of liver abscesses caused by the fasciola hepatica infection

Objective To study CT features of liver abscesses caused by the fasciola hepatica infection , and discuss its pathologic basis.Methods CT images of 15 Patients were collected. All patients underwent both unenhanced and biphasic enhanced CT scanning, then its CT performances were analyzed. Results round and nodular lesions were observed in 15 cases, branching and stripping lesions like dilated bile duct in 9 cases. The density of lesions was inhomogeneous, and the lesions were multifocal and multiform. The liver abscesses caused by the fasciola hepatica infection had no “rim sign” or “target” sign, Liver abscesses were less than 3.0 cm in diameter, and the dilation of the bile duct were not observed. Conclusion Liver absessed caused by the fasciola hepatica infection have characteristic CT features. Combined with clinical examination and laboratory test, the reliability of diagnosis will be considerably increased.

9.Interventional effect of irbesartan on vascular endothelial growth factor and its FIk-1 receptor expressions in the kidney of diabetic rats

BACKGROUND: Some studies have presented that vascular endothelial growth factor and its receptor system may take part in onset and development of diabetic nephropathy (DN).OBJECTIVE: To further verify the interventional effects of irbesartan on vascular endothelial growth factor (VEGF) and its Flk-1 receptor expressions in kidney of diabetes mellitus (DM) rat, and the possible mechanism of irbesartan.DESIGN: Randomized control animal study.SETTING: West China Hospital of Sichuan University.MATERIALS: Eighteen male closed colony SD rats weighing 150-200 g were cared in standardization.METHODS: This study was performed at Laboratory of West China Hospital of Sichuan University from August 2006 to April 2007. All rats were randomly divided into a DN group, an irbesartan group and a normal control group, with 6 rats in each group. 10 g/L streptozotocin (55 mg/kg) was intraperitoneally injected to establish models; rats in the control group were administrated with the same dosage of citric acid buffer solution; rats in the irbesartan group were administrated with 3 mg/(kg·d) irbesartan after model establishment. VEGF and FIk-1 expressions were detected using reverse transcription polymerase chain reaction (RT-PCR) and immunohistochemistry technique; while, urine protein level, area and volume of renal glomerulus were detected, and correlations of data were analyzed.MAIN OUTCOME MEASURES: ①Renal pathological indicators, urine protein level, area and volume of renal glomerulus; ②VEGF and Flk-1 expressions;③renal immunohistochemical examination;④ correlation analysis.RESULTS:① Renal pathological examination by HE staining indicated that renal glomerulus was remarkably enlarged in the DN group; mesangial matrix was increased; mesangial cells were also increased; renal tubule was expanded. The lesions in the irbesartan group were milder compared to the DN group. Urine protein level in the DN group was significantly higher compared to the control group (P < 0.01); renal weight/body mass, area and volume of renal glomerulus were significantly higher compared to the control group (P < 0.01); urine protein level, renal weight/body mass, area and volume of renal glomerulus in the irbesartan group were significantly lower compared to the DN group (P < 0.01). ② By the 16th week,VEGF and Flk-1 expressions in the DN group were significantly up-regulated compared to control group (P < 0.05); while,VEGF and Flk-1 expressions in the irbesartan group were also up-regulated compared to the control group by the 16th week (P < 0.05) but down-regulated compared to the DN group (P < 0.05). ③ VEGF staining in the DN group was darker compared to control group (P < 0.01), while the staining in the irbesartan group was also darker compared to the control group (P <0.01), but the staining in the irbesartan group was lighter compared to the DN group (P < 0.01). Flk-1 staining was similar to the VEGF. ④ VEGF and Flk-1 were positively correlated, with urine protein level, area and volume of renal glomerulus (P <0.05).CONCLUSION: VEGF and its Flk-1 receptor play important roles in DN pathogenesis. Over expressions may cause renal injury, but irbesartan (angiotensin Ⅱ receptor antagonist) has the protective effects on kidney through inhibiting abnormal expression of VEGF and Flk-1.
10.The impact of lupus flares on maternal and fetal outcomes in pregnant patients with systemic lupus erythematosus

ObjectiveTo determine the impact of lupus flares on maternal and fetal outcomes in pregnant patients with systemic lupus erythematosus(SLE).MethodsData was obtained from 46 pregnancies of 44 pregnant women with SLE.The relationship between lupus flares and pregnant outcomes,and the risk factors for adverse maternal and fetal prognosis were analyzed.T-test,X2 test or Fisher's exact test and Logistic regression were used for statistical analysis.Results① Lupus flares occurred in19(41%)pregnancies(group A) and stable lupus disease was observed in 27(59%) pregnancies(group B) during pregnancy.Compared to pregnancies in patients with stable lupus disease at the conception(n=32),pregnancies in patients with unstable lupus disease at the conception(n=8) had higher lupus flare during pregnancy( 100% vs 16%,P<0.05).(②) The common manifestations of lupus flares during pregnancy were lupus nephritis (LN) (11 cases),skin rashes (10 cases),arthritis (7 cases),and the common complication was infection ( 11 cases).(③) The incidence of premature labor,fetal growth retardation (FGR) and fetal loss in group A was 42%,47% and 26% respectively,which was significantly higher than that of the group B (7%,15% and 0 respectively)(P<0.05).There was no difference in the incidence of preeclampsia,fetal distress and neonatal asphyxia between the two groups ( 16% vs 7%,16% vs 19%,5% vs O,respectively,P>0.05).The incidence of premature labor and FGR in patients with active LN was higher than that of patients without active LN (55% vs 11%,64% vs 17%,respectively,P<0.05).(④)The binary Logistic regression analysis showed that renal impairment,hypocomplementemia,aPL and serum urea nitrogen level were independent risk factors for premature delivery,FGR,fetal loss and fetal distress.Conclusion(①) Lupus flares during pregnancy increase the incidence of premature labor,FGR and fetal loss.Active LN during pregnancy can increase the incidence of premature labor and FGR.② Renal impairment,hypocomplementemia,aPL and serum urea nitrogen level are associated with adverse fetal outcomes in pregnant patients with SLE.
